[TLC scanning determination of baicalin content in niuhuang qingxin pills].

Baicalin in the samples was extracted with ultrasonic wave, the extract was spotted on thin-layer plates of gel GF254 and developed with the mobile phase consisting of toluene-ethylacetate-formic acid (2:3:2.2). Determinations were carried out with Shimadzu CS-930 TLC scanner with lambda s = 280 nm and lambda R = 207 nm. The linear range was 0.46-4.6 micrograms/ml. The average recovery of baicalin from the samples was 98.35%, RSD = 2.82%.

A study of biological function of retinoblastoma gene (Rb gene).

The human wild-type Rb gene cDNA has been cis- or trans inserted into the retrovirus vector DOL, resulting in a sense-expression vector DOLRB and an antisense-expression vector DOLRBAS of Rb gene. By eletroporation transfection techniques, the vector DOLRB has been introduced into the human breast carcinoma cell line MDAMB468 and human hepatocellular carcinoma cell line SMMC7721 both of which have an inactivated Rb gene and the vector DOLBAS, into normal human embryonic lung fibroblasts HEL cells. With the expression of Rb protein, the growth rate of the MDAMB468 cells is decreased by about 50%, their colony formation ability in soft agar is repressed completely, and their tumorigenicity in nude mice is repressed partially. Meanwhile, the cell population of G1 phase of Rb+ MDAMB468 cells is increased markedly. About 75% of transfected SMMC7721 cells have been killed by Rb gene product. For HEL cells, with the transient expression of antisense Rb gene, the Rb protein synthesis is reduced and the growth rate of those cells increased, but no colonies of HEL cells are formed in soft agar.

[Treatment with autologous tumor-infiltrating lymphocytes and recombinant interleukin-2 in patients with lung carcinoma].

In 16 patients with lung cancer tumor infiltrating Lymphocytes (TIL) and peripheral blood lymphocytes (PBL) and their antitumor activities after IL-2 activation and were tested by 3H-TdR releasing assay. The results showed that the cytolytic activity of TIL against autologous tumor cells was lower than of IL-2 activated PBL (LAK cells) at the early incubation period but become markedly higher than that of LAK cells when the incubation lasted for 20 to 25 days. The subjectively most patients received systemic TIL treatment felt better and their immune functions were improved. Transiant Low-grade fever occurred in cases and drop of arterial blood pressure in 1. No potentially fatal toxic effect was observed.

[Determination of beta-phenyllactic acid in rat plasma by derivatization capillary gas chromatography (DCGC)].

The determination of beta-phenyllactic acid (PLA) in rat plasma by derivatization capillary gas chromatography is described. Quantitation was affected by 63Ni-electron capture detector after separation with a SE-54 quartz capillary chromatographic column. Calibration curves were linear within the range of 1.6-9.6 micrograms.ml-1. The limit of detection of PLA was considered to be 0.5 microgram.ml-1. PLA was extracted from plasma with ethyl acetate (rat plasma 0.1 ml). The recovery from plasma was high and the mean value was 98.11 +/- 3.81 (s)%, their within day and between day relative standard deviations were 5.13% and 5.99% respectively.

Morphology and morphogenesis of hepatitis E virus (strain 87A).

The morphology and morphogenesis of isolated hepatitis E virus (HEV, strain 87A) were observed by electron microscopy (EM) and immune electron microscopy (IEM). Progressively developing local vesicles, virions accumulation in crystalline arrays and viroplasmic focus were seen in cytoplasm of infected cells. Replication and assembly of the new generation viruses were closely associated with rough endoplasmic reticulum (RER), inclusion body (IB) and microfibrils. Condensation and margination of chromatin, dispersion of nucleolar material, nuclear membrane alteration and masses of threads, granular material, and fibrillar component of the nucleus were frequently found. These changes revealed that this strain virus was confirmed as a RNA virus. The shape of the virus particles appeared approximately spherical whether the specimens were from the tissue culture crude suspension or purified highly concentrated preparations. The size of the virion was about 30 nm in diameter. The viral particles appeared unsmooth and irregular in outline. The spike-like structures may be occasionally observed on the surface of some viral capsides. The diameter of the strain 87 A virus is larger than the picornavirus and smaller than the calicivirus. This strain virus is different from classical calicivirus in without the cup-shaped surface depressions. The new genus, heparnavirus genus of caliciviridae family should be proposed for HEV.

Cardiac MR imaging: principles and techniques.

Magnetic resonance (MR) imaging is becoming an important tool in diagnosing cardiovascular disease. Many technical advances have contributed to the improved capability of MR imaging in this arena. In this article, we review the technical concepts and recent advances for a variety of cardiovascular applications, including evaluation of congenital heart and great vessel disease, global cardiac function using cine acquisitions and automated ventricular volume segmentation, valvular heart disease, myocardial wall motion detection with magnetization tagging, two-dimensional and three-dimensional coronary artery imaging, and myocardial perfusion. Sequence diagrams are shown and important technical issues are discussed. Example images are shown to demonstrate the current status of research and future potential. Most results shown here are high-resolution scans acquired with new gradient coil systems, including coronary artery imaging using an in-plane resolution of 350 microns x 600 microns. This review serves as an introduction to the discussion of various clinical applications of cardiovascular MR imaging provided in this issue.

cDNA cloning and sequencing of 54kDa fragment of the 75kDa readthrough protein gene from beet necrotic yellow vein virus.

With viral RNA extracted from purified beet necrotic yellow vein virus (BNYVV) isolated from Inner Mongolia, the first strand of cDNA encoding a 54kDa fragment of the 75kDa readthrough protein on the RNA2 was synthesized by reverse transcription. A double-strand cDNA fragment of 1.5 kb was obtained after 30 cycles of PCR amplification. The fragment was ligated into and mapped on pGEM-7Zf(+). The result of sequence analysis shows that the 54kDa readthrough domain is 1509 nucleotides (nt). Compared with F13 isolate, the fragment shares 94.97% identity in terms of nucleotides with 3 nt deletion and 96.42% identity of deduced amino acids.

Changes of plasma endothelin and atrial natriuretic peptide during the onset and after termination of paroxysmal supraventricular tachycardia.

Radioimmunoassays were used to measure the concentration changes of plasma endothelin (ET) and atrial natriuretic peptide (ANP) during the onset and after termination of paroxysmal supraventricular tachycardia (SVT). 30 cases were reviewed and comparisons with 42 normal subjects were made. There are very significant differences (P < 0.0001) in the concentration changes of both plasma ET and ANP during the onset and 30 minutes after the termination of SVT. During the onset period of SVT, the plasma ET and ANP were markedly elevated and 30 minutes after its termination they were lowered significantly, but their concentrations were still 2-fold higher than those of the control group. As the biological effects of ANP and ET are antagonistic to each other, their parallel elevation and lowering of plasma concentrations during and after the termination of SVT reveal that these 2 hormones participate in the pathophysiological process of SVT. This phenomenon is possibly one of the homeostatic regulatory functions in the organism.

Establishment of a human T-lymphoma cell line (H-TL90) and analysis of its biological characteristics.

We established a human T-lymphoma cell line from the cancerous ascites of a male patient with prostate cancer which was named H-TL90. This cell line was characterized by its histological features, and by chromosomal and immunological analysis. Immunophenotypic analysis revealed that the cells expressed surface antigen CD-3CD-4CD+7CD-8. Biological analysis revealed that the cell can promote lymphocyte proliferation. This suggested that the cell line has an autosecretion function. Cytogenetic analysis revealed that H-TL90 was a hyperdiploid with 47 chromosomes and had characteristic translocation between chromosome 3 and 11, and the deletion of the long arm of chromosome 6. These results demonstrated the H-TL90 cell line can be a useful model for the study of human T-lymphoma.

Calcium channel blockers in cirrhotic patients with portal hypertension.

Four calcium channel blockers, i.e. nifedipine, verapamil, cinnarizine and tetrandrine are currently available and used widely in treating cardiovascular diseases. To confirm the effects, if any, of calcium channel blockers on cirrhotic patients with portal hypertension, a study was performed on esophageal variceal pressure and rebleeding rate of esophageal varices after 2 years by using calcium channel blocker in 321 patients from some 23 hospitals. The results demonstrated that the calcium channel blockers could significantly reduce the esophageal variceal pressure and the portal blood flow in cirrhotic patients with portal hypertension. The proportion of patients with no recurrent gastrointestinal bleeding after 2 years medication of tetrandrine was 87.9% in tetrandrine group, significantly higher than those in the other 4 groups (P < 0.05). It is suggested that tetrandrine should be more effective for cirrhotic patients with portal hypertension in preventing recurrent variceal bleeding.

[Preparation and characterization of liposomal adriamycin].

We established a more suitable methodology for preparing liposomal adriamycin (Lipo-ADM) by a remote loading technique. This procedure is convenient, simple with an entrapment efficiency of 95%. The mean diameter of liposome is about 100nm showing unilamellar spherical shape. The leakage ratio is about 10% within 24hr. Those characteristics fit for clinical demand.

[Use of nutrient density in etiologic studies on stomach cancer].

Role of nutrient density played in the genesis of stomach cancer was analyzed with a pair-matched case-control design to explore further the association between dietary nutrient and incidence of stomach cancer. Investigation of 88 pairs of cases with stomach cancer and controls showed high nutrient density of dietary vitamins C and A could protect from stomach cancer with OR of 0.34 and 0.46, respectively, and high of carbohydrate could increase risk of it with an OR of 2.20. Some problems in the use of nutrient density were discussed in the paper.

[Expression of proliferating cell nuclear antigen in the liver tissue of hepatitis B patients].

Liver biopsy and autopsy specimens of 153 cases, including 39 cases of acute severe hepatitis (ASH), 21 cases of subacute severe hepatitis (SSH), 11 cases of chronic severe hepatitis (CSH), 22 cases of active cirrhosis (AC), 20 cases of acute mild (AMH), chronic persistent (CPH) and active hepatitis (CAH), respectively, were immunohistochemically stained for proliferating cell nuclear antigen (PCNA) to compare the proliferative activity of hepatocytes by using labelling index (LI). LI is higher in CAH, SSH, and AMH than in AC and CSH (P < 0.01). In the latter, LI was completely negative in most of the specimens (25/33, 75.8%) and few scattered hepatocytes positive for PCNA were found in the remaining 8 cases (8/33, 24.2%) in the area next to the fibrotic septum and varied greatly in different pseudolobules. The results showed that the proliferative activity was very low in the AC and CSH. In some cases of ASH, the residual hepatocytes still kept prominent proliferating ability; it meant that the survived percentage would have been increased with regeneration of hepatocytes if those patients had not died early. In ASH and SSH, LI was significantly higher in patients who survived than in those who died (P < 0.01). The detection of PCNA in liver tissue with severe viral hepatitis is useful for prognostic evaluation.

[One-stage reconstruction of nipple-areola: the beta flap].

The main disadvantage of most methods for nipple reconstruction is shrinkage of the nipple. This paper presents a one-stage procedure using a local flap with a full-thickness skin grafting that has the advantages of high patient satisfaction. Since June 1993, we have been using a technique for nipple reconstruction by raising two opposing beta-shaped random skin flaps. This technique has been performed on 8 breasts in 4 patients.
