Preparation of benzyl 2-benzyloxycarbonylamino-4-dichloroacetamido-2,4-dideoxy-alpha- D-xylopyranoside, substrate for the synthesis of the heterocyclic analogue of chloromycetin.

The paper describes the eleven stage synthesis of the derivative (XI) of 2,4-diamino-2,4-dideoxy-D-xylose, containing amino groups selectively substituted by two different acyls. The initial substrate was L-arabinose. The compound obtained, after removing the blocking groups, constitutes a semiproduct for the synthesis of the pyrrole analogue of chloromycetin by the Knorr method.

Immunochemical studies on the O-specific side chains of the heterogeneous lipopolysaccharide from Proteus mirabilis O27.

Chemically and serologically identical O-specific fractions were isolated from two different lipopolysaccharide (LPS) preparations of Proteus mirabilis O27. The release of a labile constituent in mild hydrolysis deprived those fractions of their serological activity. The compound was identified as N-acetyl-D-glucosamine and its terminal position in the molecule was established by methylation analysis. The non-carbohydrate constituent of the specific fractions: lysine and alanine are linked via their amino group. Terminal residues of N-acetylglucosamine, have to be considered as immunodeterminants of Proteus mirabilis O27. The role of other carbohydrate and non-carbohydrate constituents for the serological specificity of P. mirabilis O27 is discussed.

Heterogeneity of the lipopolysaccharide from Proteus mirabilis O27.

Two lipopolysaccharides (LPS) were obtained by phenol-water procedure from Proteus mirabilis O27 strain -- LPS I as sediment and LPS II from the supernatant after ultracentrifugation. There was shown a distinct predominance of the O-specific material in LPSII, whereas in LPS I the core material prevailed. Heterogeneity of the P. mirabilis.O27 lipopolysaccharide was confirmed by the sodium dodecyl sulphate-polyacrylamide gel electrophoresis, in crossed immunoelectrophoresis and by the column chromatography.

[Contact allergy to epoxy resin hardeners].

447 patients suffering from eczema, occupationally exposed to epoxy resins, have been tested with Epidian 5 and five triethylenetetramine concentrations. By comparing the obtained results with literature data it has been found that in Poland those health damages are characterized by a considerable percentage of those sensitized to triethylenetetramine. The calculation of eczema incubation period and testing the allergen by several allergen concentrations demonstrated that the sensitivity to triethylenetetramine is sometimes very enhanced.

[Evaluation of occupational benzidine exposure of workers in automated benzidine plants].

The exposure to benzidine under old production conditions, during start-up and at normal work in the new benzidine production division in one of the plants in Poland was compared. In comparison with the old production conditions, in the automatized production process airborne benzidine concentrations were 10-fold lower so that the geometric mean of benzidine concentrations was 0.0008 mg/m3. The approximate annual dose of absorbed benzidine amounted to 0.065 mg, as estimated from urinary benzidine concentrations, and was three times lower than the one found in the old productive conditions, i.e. 0.2 mg. The benzidine absorption in the respiratory system was approx. 10%, the remaining part of the dose was probably absorbed by skin. This was proved by high concentrations of benzidine on the skin (epigastrium-up up to 300 micrograms/dcm2, palm-up to 680 micrograms/dcm2), and objects (up to 400 micrograms/dcm2). The results indicate a need to maintain the hygienic regime, introduced in the old productive conditions, including the establishment of the age up to which workers older than 50 may be employed. The cleanness in all the premises should be improved.

Contact sensitivity to phenylglycidyl ether.

From among 40 workers with dermatitis who did not have contact with phenylglycidyl ether but worked with epoxy resins, 5 were positive to Epidian 5 and phenylglycidyl ether. Cross reactions between these contactants were noted in 3 of the 17 guinea pigs with primary sensitization to epoxy resin or the tested diluent. For 58 persons coming into contact with phenylglycidyl ether the frequency of sensitization to the contactant was determined and the mean period between starting work and the occurrence of dermatitis was calculated.

New contact allergens and allergen sources.

In the report new contact allergens and allergen sources detected in Warsaw in the period 1975-1977 are described. They are divided into 3 groups: industrial allergens, remaining occupational allergens and cosmetics. There are given some data concerning the substances present in industrial oils, hardeners and epoxy resin solvents, drugs sensitizing nurses, several new sources of chromium allergens, essential oils and synthetic flavours. Results obtained with various star anise oil samples are described. Essential oils and synthetic flavours. Results obtained with various star anise oil samples are described. Essential oils and synthetic flavours are discussed as the main allergens in cosmetics.

Sensitivity to m-phenlenediamine.

Primary contact sensitivty to m-phenylenediamine was noted in 8 of 182 workers handling this substance as hardener. In these patients the period between the beginning of this work and the appearance of dermatitis was shorter if they became simultaneously sensitive to epoxy resin and/or triethylenetetramine. Moreover from among 150 consecutive patients one was positive to m-phenylenediamine and negative to p-phenylenediamine. From among the selected patients positive to p-phenylenediamine more than 40 per cent were also positive to m-phenylenediamine. A large number of the latter patients were as well positive to o-phenylenediamine. No patients were found who would be positive to o-phenylenediamine and negative to m-phenylenediamine. Nearly all those positive to m- and p-phenylenediamine were also positive to the meta isomers of other aromatic amines.
