Effect of cold exposure on peroxidase and iodinase activities of rat submandibulary gland.

Exposure of rats to 6 +/- 2 degrees C for 3 h caused an increase in peroxidase activity (55%) and tyrosine iodinase activity (40%) of the submandibulary glands. Thiocyanate, hydrogen peroxide and formation of triiodide from iodide were also elevated under the same conditions. Administration of alpha-receptor blockers and indomethacin prevented the rise of peroxidase activity during cold stress whereas beta-receptor blockers were less effective.

Activities of glucose-6-phosphate, 6-phosphogluconate, and isocitrate dehydrogenases from Leishmania donovani cultivated at 25 and 37 C.

The activities of glucose-6-phosphate dehydrogenase (G-6-PD) (EC No. 1.1.1.49), 6-phosphogluconate dehydrogenase (PGD) (EC No. 1.1.1.44), and isocitrate dehydrogenase (ICD) (EC No. 1.1.1.42) from promastigotes of Leishmania donovani strain 3S grown at 25 C in modified Tobie's (mT) medium and from promastigotes of the 37 C-adapted substrain of this strain cultivated in the mT at 37 C were assayed at 25 and 37 C. At 25 C ICD from both the strain and the substrain had the highest, and PGD, the lowest activity; the activity of G-6-PD was intermediate, but much closer to that of ICD. Irrespective of the temperature of the assay, the activities of G-6-PD and ICD from the 37 C substrain were significantly higher than those of these enzymes from the parental strain; however, the activity of PGD from the 25 C strain was slightly higher than that of this dehydrogenase from the 37 C-adapted stock. No significant activity losses of G-6-PD and ICD from either the strain or the substrain were noted after incubation of the extracts in the presence of 0.25 M sucrose at 37 C for 2 hr. PGD was unstable in such extracts, but it could be rendered stable by the addition of 4 mM 6-phosphogluconate, G-6-PD was the least and ICD the most dependent on Mg2+ ions. In the 15-25 C range, the Q10 values of the enzymes from the 25 C strain were 2.83, 2.5 and 2.63 for G-6-PD, PGD, and ICD, respectively. These values for the respective enzymes in the 25-35 C range were 2.06, 1.67, and 1.62. The Q10 values of the enzymes from the 37 C substrain in the 15-25 C range were 2.06 for G-6-PD, 3.25 for PGD, and 2.77 for ICD; in the 25-35 C range, the corresponding values were 1.67, 1.46, and 1.83. Cultivation of the 37 C substrain at 25 C was accompanied by a drop in G-6-PD and ICD activities.

Pharmacological actions of some simple analogues of reserpine. I. Analgesic effects.

In view of the fact that reserpine treatment lowers pain threshold, structural analogues of reserpine molecule were tested against thermal and visceral pain. Some compounds belonging to piperidino- and morpholino derivatives of acetanilide showed promising analgesic activity. These compounds in earlier studies had shown tranquillizing effect. Combination studies with meperidene, phenylbutazone, phenazone and meprobamate did not significantly change the analgesic activity in thermal methods but the test-compounds were potentiated by phenylbutazone and phenazone in writhing test.

Pharmacological actions of some simple analogues of reserpine. II. Anticonvulsant effects.

Reserpine analogues obtained from strip-down parts of the parent molecule were studied for their effects on experimental convulsive seizures and amphetamine antagonism. The unsubstituted and M-methoxy substituted piperidino- and morpholino acetanilides derived from A, B and C rings or reserpine showed prominent anticonvulsant activity as against the known lowering of threshold activity by reserpine. Methoxy morpholino derivative showed prominent activity with least sedative effect. gamma-Piperidino acetanilide showed a dual effect such that at higher doses it itself produced clocin covulsions. The effects were non-specific since they unpreferentially antagonized the maximal electrochock, Metrazol and strychnine convulsions Compounds derived from C, D and E rings of reserpine did not significantly increase or decrease the seizure thresholds.

Leishmania donovani: a chemically defined medium suitable for cultivation and cloning of promastigotes and transformation of amastigotes to to promastigotes.

A chemically defined medium using commercially available alpha-MEM supplemented with hemin, HEPES, L-glutamine, D-glucose, folic acid, D-biotin and adenine supports the luxuriant growth and propagation of Leishmania donovani promastigotes. A peak parasite population of about 7.0 x 10(7)/ml at stationary phase and a population doubling time of 11.4 h for high-subpassage promastigotes were obtained. The medium was suitable for transformation of isolated amastigotes from infected hamster spleen. Promastigotes could be detected by culturing kala-azar patients' bone-marrow aspirate or spleen puncture material in this medium. Four out of six freshly transformed isolates gradually adapted and grew well in this medium. Macroscopic colonies appeared on agar plates prepared with the medium within 16-20 days after inoculation. The cloning efficiency was increased about five-fold by glycerol supplementation.

Particle size distribution and respiratory deposition estimates of beryllium aerosols in an extraction and processing plant.

The mass size distribution of beryllium aerosols generated in the various operational areas of a typical extraction and processing plant was studied using an eight-stage impactor sampler. The total concentration of beryllium in the plant was found to be well below the threshold limit value. The mean value of mass median aerodynamic diameter of beryllium particles observed for various operations ranged from 5.0-9.5 microm. The alveolar deposition for various operational areas was estimated to be 3-5% for nasal breathing and 9-13% for oral breathing based on the International Commission on Radiological Protection (ICRP) human respiratory tract model. Deposition during oral breathing was higher than during nasal breathing by approximately a factor of two to three. This study on exposure characterization was useful for reducing the respirable fraction of beryllium aerosol by optimizing the capture velocity and improving the quality of other control measures.
