Functional and phenotypic changes associated with the in vitro development of human monocytes into activated macrophages.

Freshly isolated human peripheral blood monocytes had minimal cytotoxic effect in vitro on the schistosomula of Schistosoma mansoni. However, stimulation of the cells with either interferon gamma (IFN) or specific anti-parasite antiserum caused an increase in cytotoxicity. Additionally, the normal development of monocytes into macrophages over 7 days was associated with a sharp increase in cytotoxicity. The non-cytotoxic monocytes were compared with activated macrophages to assess whether cytotoxicity was associated with changes in immunophenotype. As monocytes developed into macrophages there were marked increases in transferrin receptors (HB21), macrophage cellular integrin (3.9), and Fc receptors (KB61). A further three markers showed increased expression in 7-day-old macrophages stimulated by IFN, namely a high affinity Fc gamma receptor (10.1), MHC Class II (1B5) and tumour necrosis factor (TNF).

The design and effectiveness of a graduate survey course in quality assurance and utilization review.

This paper reports on the design, presentation, and evaluation of a graduate university course in quality assurance and utilization review. The authors cite the need for expanded course offerings in this field and present descriptions of the course design and student participants, including program location, objectives, learning themes and content, instructor team, students, and course process. Student feedback is reported including comments on course strengths and needs, teaching methods, and learning evaluation. Additional work requirements are cited.

A method for the assessment of the output of irregularly shaped electron fields.

A simple approach to the calculation of dose in cone-collimated electron fields is presented. The method accounts for variations in lateral scatter with field size using a Clarkson Integration. The reduction in output by an irregularly shaped aperture is evaluated based on measurements of the output at various distances from the aperture in a field 2.5 cm in diameter. The first 14 months of clinical use were analysed and it is shown that in 42 out of 600 calculations, the cutout factor was less than 0.95 and that in 77% of these, the calculation method was accurate to +/- 3%.

Remnant model of renal failure in the dog: avoidance of second surgery by chemical nephrectomy.

Experimental chronic renal failure in the dog is usually studied by a two-step surgical procedure. However, it is becoming increasingly difficult, due to animal welfare concerns, to get permission for such procedures in Canada. We describe a method for obviating the need for second surgery by injecting absolute alcohol into a renal artery, which leads to immediate functional nephrectomy.

Effects of exocrine pancreatic insufficiency and replacement therapy on the bacterial flora of the duodenum in dogs.

The influence of pancreatic secretions on the bacterial flora of the small intestine in 6 dogs was investigated by determining effects of exocrine pancreatic insufficiency on numbers and types of bacteria in duodenal juice, and by examining the subsequent response to dietary supplementation with bovine pancreatic extract. Exocrine pancreatic insufficiency was induced by ligation of pancreatic ducts and was confirmed by indirect assessment of exocrine pancreatic function. Duct ligation was followed by large increases (P less than 0.01) in total numbers of bacteria, reflecting increased numbers particularly of Lactobacillus spp and Streptococcus spp, in 3 dogs accompanied by obligate anaerobes. Total numbers of aerobes and anaerobes decreased markedly (P less than 0.05) after supplementation with bovine pancreatic extract to values that were not significantly different from those determined before duct ligation. Exocrine pancreatic insufficiency therefore resulted in small intestinal bacterial overgrowth that was reversed by pancreatic replacement therapy, indicating that pancreatic secretions can have an important influence on the small intestinal bacterial flora of dogs.
