Visual habituation, sensorimotor development, and tempo of play in one-year-old infants.

The pattern of relationships between rate of visual habituation, sensorimotor development, play tempo, and motor activity was examined in 168 1-year-old infants. Subjects were exposed to 3 consecutive series of a repeated visual stimulus and the rate of habituation determined for each series. The measures of sensorimotor development included both standard psychometric and Piagetian scales. Measures of play tempo and motor activity were obtained from observations of free play. Longer fixation time to the initial stimulus presentation and faster habituation on the original series were associated with advanced sensorimotor development for girls. Longer initial fixation time in boys was associated with a slower tempo of play. The magnitude of these relationships, however, was found to be dependent on whether the subject completed the entire experimental session or was terminated early due to irritability or drowsiness. The results suggest that both the sex and state of the infant are important moderators of the relationship between measure of visual attention and other individual difference variables.

Therapeutic fasting in morbid obesity.

The weights of 207 morbidly obese patients were reduced via prolonged fasting. Half the patients fasted for close to two months, losing a mean of 28.2 kg; one fourth fasted for less than one month; and the other fourth fasted for more than two months, with a mean 41.4-kg loss. This latter group was heavier initially, and more than 50% attained near-normal weight. Patients with onset of obesity in childhood had the lowest tolerance for fasting and the lowest success rate in attaining normal weight. Over a 7.3-year follow-up period in 121 patients, the reduced weight was maintained for the first 12 to 18 months. Subsequently, regain proceeded equally in all groups irrespective of length of fast, extent of weight loss, or age at onset of obesity. Regain to original weight occurred in 50% within two to three years and only seven patients remained at their reduced weights. Regain to greater than original weight was more common in childhood-onset obesity.

Isolation of albumin from whole human plasma and fractionation of albumin-depleted plasma.

The dye Cibacron Blue F-3-GA was conjugated to Sepharose to provide an affinity column for serum albumin. Passage of whole human plasma through a column of Cibacron Blue-Sepharose results in the removal of approx. 98% of the albumin. The latter can be quantitatively recovered by desorption with NaSCN. Albumin-depleted plasma can be readily resolved into discrete fractions by a combination of conventional biochemical techniques. In particular, the resolution of plasma proteins with properties similar to those of native human plasma albumin can readily be accomplished by ion-exchange chromatography of the Sepharose-dye-treated plasma on DEAE-cellulose.

Epstein-Barr virus: experimental infection of Callithrix jacchus marmosets.

Eight common marmoset monkeys (Callithrix jacchus) were inoculated with about 10(4) transforming units of B95-8 virus; seven of the marmosets died 50-111 days post inoculation and all seven showed microscopic and/or macroscopic lesions compatible with a diagnosis of lymphoproliferative disease. Low levels of anti-VCA antibodies were detected in plasma from six marmosets. Attempts failed to establish continuous EBV-carrying lymphoblastoid cell cultures by cultivation in vitro of circulating lymphocytes or minced lymphoid tissues obtained at necropsy.

Histology of the endometrium in long-term use of a sequential oral contraceptive.

Non-atypical adenomatous hyperplasia was found in 13.5% of 111 biopsies obtained from long-term, predominantly black, users of Oracon. An association between length of time of use (95 cycles for those with adenomatous hyperplasia versus 73 for those without) and the development of adenomatous hyperplasia was noted, but there was no association with hypertension or obesity. Cystic glandular dilatation was seen in 63% but was not thought to represent hyperplasia. No more advanced lesions were seen.

Inhibition of Neisseria gonorrhoeae by a bacteriocin from Pseudomonas aeruginosa.

Supernatants from broth-grown cultures of Pseudomonas aeruginosa PA 103 exhibited bactericidal activity against Neisseria gonorrhoeae. The concentration of the bactericidal substance increased significantly after induction by mitomycin C. Purification was effected by salt fractionation, chromatography on diethylaminoethyl-cellulose, and sedimentation by centrifugation at 100,000 x g for 90 min. Electron microscopy of this purified preparation revealed structures resembling R-type pyocins in both the contracted and uncontracted state. Pyocins in the contracted state were observed in association with the gonococcal cell surface. No loss of bactericidal activity was observed after treatment with proteolytic enzymes. Standard pyocin typing procedures identified the pyocin pattern as 611 131. The bactericidal activity of this pyocin was examined on various species of Neisseria. Out of 56 strains of N. gonorrhoeae from disseminated and nondisseminated infections, all were susceptible to pyocin 611 131. However, only 3 of 20 strains of N. meningitidis and 5 of 16 strains of N. lactamica were susceptible. The bactericidal activity that pyocin 611 131 has for N. gonorrhoeae and other species of Neisseria is significant because it departs from the expected specificity that heretofore has distinguished bacteriocins from most "classical" antibiotics.

Studies related to antitumor antibiotics. Part V. Reactions of mitomycin C with DNA examined by ethidium fluorescence assay.

The cytotoxic action of the antitumor antibiotic mitomycin C occurs primarily at the level of DNA. Using highly sensitive fluorescence assays which depend on the enhancement of ethidium fluorescence only when it intercalates duplex regions of DNA, three aspects of mitomycin C action on DNA have been studied: (a) cross-linking events, (b) alkylation without necessarily cross-linking, and (c) strand breakage. Cross-linking of DNA is determined by the return of fluorescence after a heat denaturation step at alkaline pH's. Under these conditions denatured DNA gives no fluorescence. The cross-linking was independently confirmed by S1-endonuclease (EC 3.1.4.-) digestion. At relatively high concentrations of mitomycin the suppression of ethidium fluorescence enhancement was shown not to be due to depurination but rather to alkylation, as a result of losses in potential intercalation sites. A linear relationship exists between binding ratio for mitomycin and loss of fluorescence. The proportional decrease in fluorescence with pH strongly suggests that the alkylation is due to the aziridine moiety of the antibiotic under these conditions. A parallel increase in the rate and overall efficiency of covalent cross-linking of DNA with lower pH suggests that the cross-linking event, to which the primary cytotoxic action has been linked, occurs sequentially with alkylation by aziridine and then by carbamate. Mitomycin C, reduced chemically, was shown to induce single strand cleavage as well as monoaklylation and covalent cross-linking in PM2 covalently closed circular DNA. The inhibition of this cleavage by superoxide dismutase (EC 1.15.1.1) and catalase (EC 1.11.1.6), and by free radical scavengers suggests that the degradation of DNA observed to accompany the cytotoxic action of mitomycin C is largely due to the free radical O2. In contrast to the behavior of the antibiotic streptonigrin, mitomycin C does not inactivate the protective enzymes superoxide dismutase or catalase. Lastly, mitomycin C is able to cross-link DNA in the absence of reduction at pH 4. This is consistent with the postulated cross-linking mechansims.

Studies of the metabolism of 5,5-diphenylhydantoin relating principally to the stereoselectivity of the hydroxylation reactions in man and the dog.

The hydroxylated metabolites of 5,5-diphenylhydantoin (DPH) in dog and human urine, after release by beta-glucuronidase, have been isolated and purified by procedures entailing only solvent partitioning without crystallization or any other procedure that could change the proportions of optical isomers. Dogs produced both the meta- and para-5-hydroxyphenyl-5-phenylhydantoins (m-HPPH and p-HPPH) from DPH, the former in approximately 6 times the amount of the latter. The m-HPPH from dog urine was dextrorotatory and had the properties of a pure optical isomer. The p-HPPH from dog urine was a mixture of optical isomers with approximately a 2:1 preponderance of the levorotatory isomer. When dog urine was heated with acid rather than treated with beta-glucuronidase, the isolated p-HPPH contained a small preponderance of the dextrorotatory isomer, probably owing to production of d-p-HPPH from the dihydrodiol metabolite. No o-HPPH was found in dog urine. When racemic p-HPPH and m-HPPH were administered to dogs, there was little if any preponderance of either optical isomer in the materials isolated from urine. Enzymatically released p-HPPH isolated from urine of human patients receiving DPH was a mixture of optical isomers with approximarely a 10:1 preponderance of the levorotatory isomer. Crystallization of this material readily yields pure l-p-HPPH. Human urine contained only very small amounts of m-HPPH, which was detectable by gas chromatography but insufficient in amount to permit isolation. Hypotheses are discussed that could account for the differences in the metabolism of DPH in man and dog. Small amounts of 2,2-diphenylhydantoic acid were found in urine of dogs but not of human patients receiving DPH.

Somatosensory evoked potentials and magnitude of perception.

With step indentations of the index finger tip in randomized order, via a mechanostimulator, the tactile receptors of human skin were adequately stimulated. Recording the EEG over the contralateral and ipsilateral cortex, the evoked potentials and their 95% confidence limits were analysed. Simultaneously the psychophysical magnitude estimations were analysed. 1. The perceptual estimations were linearly related to step amplitude. 2. The early components of the E.P. show no obvious correlation to stimulus amplitude. 3. The later components (with peak latencies of 120 msec or more) show a monotone, non-linear rising function with respect to stimulus amplitude. 4. The early waves of the evoked potentials up to about 120 msec are well localized over the contralateral postcentral hand area while the late components resemble the alpha rhythm in wave length and distribution over both hemispheres. The possible role of alpha-synchronisation in the later components is discussed.

Osteopetrosis associated with familial paraplegia: report of a family.

A clinical analysis of three members of a family with documented osteopetrosis and familial paraplegia is presented. All patients had a long history of increased bone density and slowly progressing paraparesis of both legs. A thorough review of the literature has revealed no other cases which presented with paraplegia without spinal cord compression. Although the etiologic factor or factors remain unknown, our review supports the contention that this is a distinct clinical entity.
