The 11 beta-OHSD inhibitor, carbenoxolone, enhances Na retention by aldosterone and 11-deoxycorticosterone.

Carbenoxolone sodium, CS, a liquorice derivative associated with hypertension and sodium retention, has been demonstrated to inhibit 11 beta-hydroxysteroid dehydrogenase, an enzyme that metabolizes cortisol and corticosterone to their respective inactive 11-dehydro products (cortisone and 11-dehydrocorticosterone). It has been proposed that the increased bioavailability of unmetabolized corticosterone and cortisol following 11 beta-OHSD inhibition allows these steroids to act on renal mineralocorticoid receptors to elicit the mineralocorticoid action. Here we describe how CS amplifies the antinatriuretic activity of aldosterone and deoxycorticosterone; the latter steroid is of particular importance in that it does not possess a hydroxyl group at the C-11 position in the steroid ring, indicating that another mechanism(s) in addition to 11 beta-OHSD inhibition is responsible for the amplification of the action of deoxycorticosterone.

Chromosomal assignment and regional localization of myeloperoxidase in the mouse.

Using a rat x mouse somatic cell hybrid that contains chromosome 11 as the only mouse material, we have shown that myeloperoxidase, which maps to human chromosome 17, maps to mouse chromosome 11. Regional assignment of the gene by in situ hybridization localized Mpo to the region C-E1, with a peak at band 11C. These results further confirm and extend observations on the remarkable homology between human chromosome 17 and mouse 11.

Fulminant hepatic necrosis caused by adenovirus type 5 following bone marrow transplantation.

A 34-year-old patient was transplanted from an HLA-identical sister for high grade non-Hodgkin's lymphoma in first complete remission. One month post-transplant, he developed hepatitis and haemorrhagic cystitis. He died 2 months post-transplant from fulminant hepatic failure. Adenovirus type 5 was cultured from urine, and characteristic adenovirus inclusions were seen in the liver. Striking paracrystalline arrays of adenoviruses were seen in the liver on electron microscopy. Reactivation of adenovirus infection is increasingly recognized post-BMT, but this complication of type 5 infection is unusual, and we describe in detail this second reported case.

Effect of high calcium diet and nitrendipine on the development of high blood pressure in adrenalectomised spontaneously hypertensive rats treated with aldosterone.

The present investigation was designed to study the effect of high calcium diet and nitrendipine on aldosterone-induced development of high blood pressure in young adrenalectomised spontaneously hypertensive rats (SHR). Animals raised on a control calcium diet (0.5% Ca) or high calcium diet (2.5% Ca) were adrenalectomised aged 5-6 weeks and treated with either aldosterone (1 microgram/day subcutaneously) in propylene glycol or aldosterone (1 microgram/day) plus nitrendipine (20 mg/kg/48 hr subcutaneously) in peanut oil. Controls received the aldosterone or nitrendipine vehicle and treatment lasted two weeks. The animals were given 0.9% NaCl ad libitum. Aldosterone caused a significant increase in systolic BP in adrenalectomised SHR fed the control calcium diet when compared with controls and the aldosterone-induced hypertension was blocked by the high calcium diet and nitrendipine (P less than 0.001, n = 4-7). These findings suggest that high dietary calcium has a protective effect against the development of aldosterone-induced hypertension and may enhance that of nitrendipine.

18-substituted steroids--Part 17. 2 alpha-hydroxylated liver metabolites of aldosterone identified by high-field [1H]NMR spectroscopy.

11 beta,18-Epoxy-2 alpha,3 alpha,18,21-tetrahydroxy-5 alpha,17 alpha- pregnan-20-one (2 alpha-hydroxy-3 alpha,5 alpha-tetrahydro-17-isoaldosterone) and its apo isomer have been identified by high-field NMR studies, supported by thermospray HPLC/MS, to be among the major polar metabolites formed from incubation of aldosterone with rat liver microsomal fraction. Indications that unreduced 2 alpha-hydroxy-aldosterone is also present among the metabolites have still to be confirmed.
