A critical appraisal of the immunohistochemical detection of the c-myc oncogene product in colorectal cancer.
Expression of c-myc was studied immunohistochemically in 100 colorectal carcinomas, using a monoclonal antibody, Myc 1-6E10, which is purported to recognize the oncoprotein (p62c-myc) in paraffin-embedded material. In normal epithelium, maturing crypt cells and terminally differentiated surface cells were positive, and proliferating basal crypt cells negative. All carcinomas stained positively, but intensity was independent of histological differentiation, Dukes' stage, DNA ploidy and survival. Staining was predominantly cytoplasmic despite the suspected nuclear location of p62c-myc and there was considerable staining of fibroblasts. When staining was compared in frozen and paraffin-embedded sections fixed in different ways, different patterns were observed. Acetone-fixed frozen sections exhibited weak nuclear and cytoplasmic staining or were negative. In formol-saline fixed frozen sections, there was stronger predominantly nuclear staining. In paraffin-embedded sections staining was predominantly cytoplasmic. This study suggests that c-myc expression is enhanced in the majority of colorectal carcinomas and although independent of clinical behaviour, may be a common event in malignant transformation. However, since staining is affected by fixation and processing, data obtained using Myc 1-6E10 on routinely processed specimens should be interpreted with caution.

Transection of the retrohepatic vena cava in a case of total situs inversus following blunt trauma.
Total transection of the retrohepatic inferior vena cava from blunt trauma is a rare and usually fatal injury. Such an injury in a patient with total situs inversus and previous surgical correction of great vessel transposition is described. A possible mechanism for the injury is suggested.
Effect of various levels of avilamycin on the performance of growing-finishing swine.
A series of 12 trials involving 1,710 crossbred pigs was conducted at eight geographical locations in the United States to determine the effect of avilamycin on average daily gain (ADG), average daily feed (ADF) and feed-to-gain ratio (F/G) of growing-finishing swine. Eight of 12 trials evaluated avilamycin concentrations at 0, 5, 10, 20, 40 and 60 ppm, while an additional four trials evaluated avilamycin concentrations at 0, 10, 20 and 40 ppm in swine grower and finisher diets fed ad libitum. All trials were conducted using a randomized complete block design with data from the 12 trials pooled for statistical analysis. Pigs fed 5, 10, 20, 40 or 60 ppm avilamycin had increased (P less than .05) ADG over control pigs. No differences were detected for ADF between control and avilamycin-fed pigs. Pigs fed 10, 20, 40 or 60 ppm avilamycin had improved (P less than .05) F/G over control animals. Average daily gain, ADF and F/G for pigs fed 0, 5, 10, 20, 40 or 60 ppm avilamycin were: 749, 763, 767, 769, 771 and 771 g; 2.38, 2.40 2.39, 2.41, 2.38 and 2.38 kg; and 3.17, 3.15, 3.12, 3.13, 3.09 and 3.09, respectively. Linear plateau procedures showed that the effective dose range of avilamycin for the growing-finishing phase is 5 to 10 ppm for improving ADG and 10 to 60 ppm for improving F/G.
Processing yields and meat flavor of broilers fed a mixture of narasin and nicarbazin as an anticoccidial agent.
Processed yields (percent hot carcass) and cooked meat flavor of broilers fed 100 ppm of an anticoccidial agent (a mixture of 50 ppm narasin and 50 ppm nicarbazin) were compared with yields of birds fed a ration without the anticoccidial agent. Broilers were processed at 7 wk of age (49 days) after a 4-day withdrawal from the anticoccidial agent for the treated birds. The flavor of meat was evaluated by a 12-member sensory panel. Meat was either deep fat-fried or oven roasted. Sensory evaluations were made on freshly cooked samples and on cooked meat refrigerated for 24 h and reheated. The anticoccidial agent did not produce a difference (P greater than .05) in the hot carcass yields of the broilers as compared with control birds fed the nonmedicated diet. Analyses of triangle test data for flavor evaluations by two statistical methods indicated that there were no detectable differences (P greater than .05) in flavor between broilers fed the anticoccidial agent in the diet and those fed the control diet.

Presynaptic serotonin receptors regulate [3H]serotonin release from rat spinal cord synaptosomes.
Superfused rat spinal cord synaptosomes were studied to determine if inhibitory serotonin (5-HT) receptors (autoreceptors) exist on spinal serotonergic nerve terminals. Exogenous 5-HT (1-50 nM) produced a concentration-dependent inhibition of K+-induced [3H]5-HT release but did not affect basal [3H]5-HT release. A 32-44% inhibition was produced by 30 nM 5-HT. The inhibitory effect of 30 nM 5-HT was effectively antagonized by 100 nM metitepine, a 5-HT autoreceptor antagonist. The results provide evidence for the existence of 5-HT autoreceptors in rat spinal cord tissue.
Testicular torsion in the armed services: twelve year review of 179 cases.
All cases of testicular torsion occurring in the Armed Services between 1972 and 1983 have been reviewed. All cases were post-pubertal. The incidence in older age groups is higher than previously reported. No cases were seen in undescended testes. No seasonal variation in presentation was detected. Recurrent scrotal pain before acute presentation occurred in 29 per cent of cases. The testicular salvage rate was 75 per cent. The salvage rate in patients with pain for greater than 24 h was 46 per cent. Patients with a history of intermittent scrotal pain should be explored as a matter of urgency. Operation should still be considered in patients who have had pain for greater than 24 h.
Further probes into the molecular sites of damage to cerebral adenylate cyclase following postischemic reperfusion.
A variety of pharmacological agents were used as experimental probes to determine with greater precision the site(s) of damage to cerebral adenylate cyclase as a consequence of postischemic reperfusion in the gerbil. A paradigm of 60-min bilateral ischemia followed by 40-min reperfusion results in a decreased sensitivity of the catalytic site of adenylate cyclase to Mn2+. Likewise, the GTP-transducer site (guanine nucleotide regulatory or G protein) revealed depressed responses to GTP in the absence or presence of norepinephrine, dopamine agonists, substance P, yohimbine, and cholera and pertussis toxins. Moreover, a crude preparation of GTPase disclosed that damage elicited by postischemic reperfusion was directed to the higher-affinity form of this enzyme, which is associated with the overall function of the guanine nucleotide regulatory protein. Injury to adenylate cyclase was unrelated either to the ability of adrenergic ligands to bind to associated receptor sites or to the capacity of the brain to generate visual evoked potentials in response to visual stimuli.
Inguinal hernia repair: which suture?
Two hundred and fifty six consecutive inguinal hernia repairs in one hospital over a 15 month period have been reviewed. There was a significant association (P less than 0.00001) between the length of history and postoperative complication rate. The use of braided suture was associated with an apparent increase in sepsis and recurrence rate (P less than 0.1). Synchronous bilateral repair was not associated with an increase in complications. It is concluded that the use of braided suture material in an inguinal hernia repair may result in an unacceptable level of sepsis and recurrence. It is also concluded that attempts to reduce the interval between the onset of an inguinal hernia and operative repair will result in lower complication rates.

Characterization of sodium-dependent, high-affinity serotonin uptake in rat spinal cord synaptosomes.
Synaptosomal accumulation of [3H]serotonin was used to determine if the rat spinal cord possesses a high-affinity neuronal uptake system for serotonin. Two temperature-dependent accumulation processes were found, one sodium-dependent, the second sodium-independent. Sodium-dependent [3H]serotonin accumulation was linear with sodium concentrations up to 143 mM, was associated with the purified synaptosomal fraction (P2B), and decreased 76% by osmotic lysis, 88% by sonication, and 96% by 0.1% Triton X-100. Drug inhibition studies demonstrated fluoxetine to be the most potent inhibitor of this system (IC50 0.075 microM) while desipramine (IC50 0.43 microM) and nomifensine (IC50 0.95 microM) were less potent. Kinetic analysis revealed that sodium-dependent accumulation in purified synaptosomes was saturable at low [3H]serotonin concentrations (Ku = 50 nM, Vmax = 4 pmol/mg protein/min). Sodium-independent [3H]5-HT accumulation was substantially less sensitive to fluoxetine, desipramine and nomifensine. While sodium-independent accumulation was not significantly affected by osmotic lysis, it was markedly increased by prior sonication of tissue. Also, in contrast to sodium-dependent accumulation, sodium-independent accumulation was evenly distributed in all tissue fractions, and was not saturable at low [3H]serotonin concentrations. It is concluded that sodium-dependent [3H]serotonin accumulation reflects uptake into spinal serotonergic nerve terminals while sodium-independent accumulation probably reflects a temperature-sensitive binding to membrane fragments. Comparison to brain uptake of serotonin and the necessity for using 37 degrees C sodium-free blanks rather than 0 degree C blanks in spinal cord homogenates is discussed.

The postnatal development of norepinephrine-stimulated cyclic AMP accumulation in rat spinal cord.
Norepinephrine (NE) stimulated cyclic adenosine 3',5'-monophosphate (cyclic AMP) accumulation in regional spinal cord and cortical tissue slices from postnatal rats demonstrated distinct developmental patterns. NE-concentration-response studies using a 10-min incubation period demonstrated minimal NE-stimulated cyclic AMP accumulation in whole spinal cord at PD 1-5 with maximal sensitivity on PD 12. Thereafter, sensitivity decreased to adult values at PD 30. Sensitivity changes were reflected in alterations in maximal response only since NE EC50s all averaged 10(-6) M. This agrees with no change in calculated Kd for NE with increased Vmax to PD 12 and a reduction thereafter. Studies on regional spinal cord and cerebral cortical cyclic AMP accumulation indicated peak NE sensitivity in cervical and thoracic cord occurred at PD 10, in lumbar cord at PD 15, and in cerebral cortex at PD 20. The fact that inhibition of phosphodiesterase (PPDE) produced the same percentage alteration in peak accumulation in spinal cord slices regardless of postnatal age suggests that PPDE is not the primary determinant of the ontogenic changes. The results indicate that the postnatal development of spinal NE receptors may be reflected in an increase in the responsiveness of the cyclic AMP system to NE.
Dibutyryl-cyclic GMP stimulation of Ca2+ -ATPase activity in rat brain synaptic membranes.
The effects of dibutyryl cyclic AMP (db-cAMP) and dibutyryl cyclic GMP (db-cGMP) were tested on Ca2+-ATPase, Mg2+-ATPase, and (Ca2+ + Mg2+)-ATPase activities in lysed synaptosomes prepared from whole rat brains (minus cerebellum). At concentrations from 0.1 to 2.0 mM, db-cGMP produced a selective, concentration-dependent increase in Ca2+-ATPase activity. Both db-cGMP and db-cAMP slightly reduced Mg2+-ATPase activity, whereas neither compound had concentration-dependent effects on (Ca2+ + Mg2+)-ATPase activity. These findings suggest that the Mg2+-independent, Ca2+-ATPase activity in rat brain is regulated by a cyclic GMP-dependent process. Further, the data provide evidence that the Ca2+-ATPase activity in lysed synaptosomal membranes represents an enzyme that is distinguishable from both the Mg2+ -and (Ca2+ + Mg2+)-ATPase.
