Gas-chromatographic analysis of 4-prenyl-1,2-diphenyl-3-5-pyrazolidinedione (feprazone) at therapeutic levles in human plasma.

A specific and sensitive procedure for the determination of prenazone at therapeutic levels in human plasma has been developed. The method involves extraction of the drug from acidified plasma into chloroform. After further purification by back extraction the residue is examined on a gas-liquid chromatograph fitted with a flame ionisation detector. Tetraphenylethylene is used as an internal standard for quantitation by the relative peak height technique. Plasma levels encountered after oral ingestion of therapeutic doses are reported.

Determination of trichloroethanol at therapeutic and overdose levels in blood and urine by electron capture gas chromatography.

A specific and sensitive gas chromatographic method for the determination of trichloroethanol, the active metabolite of chloral hydrate, in blood and urine is reported. A simple dilution of the sample with an ethanolic solution of internal standard followed by gas chromatography with electron capture detection is described. The method has been used to determine plasma levels after therapeutic dosing with chloral preparations.

A double-blind cross-over trail of Prenazone and aspirin in the management of rheumatoid arthritis.

A double-blind cross-over trial of Prenazone 600 mg and Aspirin 4 g daily was carried out on 20 patients suffering with rheumatoid arthritis. The analgesic and anti-inflammmatory activity was indistinguishable from that of aspirin under the conditions of the trail. Twelve patients expressed a general preference for Prenazone and six for aspirin. Prenazone appeared to be well tolerated and free from serious side-effects. These results suggest that it will be a useful drug in the management of rheumatoid arthritis.
