A multicentre comparison of piroxicam and indomethacin in acute soft tissue sports injuries.

One hundred and five patients suffering from sport-associated soft tissue injuries were included in a randomized controlled trial comparing the efficacy and side-effects of piroxicam and indomethacin. Patients were treated for 7 days. Both groups showed a similar degree of improvement in joint tenderness, swelling and pain. Eighty-eight per cent of patients on piroxicam showed a marked or moderate improvement in their injury at the end of the trial and 79% of indomethacin-treated patients. Approximately 50% of patients in both groups resumed full activity within 7 days. One patient, in the indomethacin group, was withdrawn from treatment due to a side-effect.

Effects of dietary copper depletion on acute and delayed inflammatory responses in mice.

Acute and delayed hypersensitivity reactions were compared in mice fed diets low in copper (less than 20 mumol copper kg-1) or adequate (more than 300 mumol copper kg-1) for seven to nine weeks after weaning. In the copper depleted animals there was a significant enhancement (P less than 0.01) of histamine-induced paw oedema measured 15 minutes after challenge. The variance of responses within the two dietary groups was similar. Delayed contact hypersensitivity reactions to oxozalone were also significantly increased (P less than 0.01) in the deficient mice. In this study there were some signs of differences in variance heterogeneity but they were statistically insignificant. Delayed-type hypersensitivity to sheep erythrocytes was similarly but less markedly (P less than 0.05) affected. Group differences just failed to reach statistical significance after logarithmic transformation (P less than 0.07) or non-parametric analysis (P less than 0.06). At challenge, the copper deficient mice had a significant (P less than 0.05) plasma hypercholesterolaemia. A few individuals also showed signs of spleen enlargement and mild haemoglobinaemia but otherwise the animals appeared clinically normal and were similar in weight to the controls. Plasma (P less than 0.001), erythrocyte (P less than 0.01) and liver (P less than 0.01) copper concentrations were significantly reduced in the copper-depressed mice, although the concentrations of copper and zinc in the spleen were increased (P less than 0.05) in the same group. Whole blood superoxide dismutase activity was also significantly decreased (P less than 0.01) but erythrocyte glutathione peroxidase activities were similar in both groups, as were blood and liver zinc concentrations.(ABSTRACT TRUNCATED AT 250 WORDS)

Veratridine-stimulated central synapses in culture: a quantitative ultrastructural analysis.

Synapses in explant cultures of fetal rat neocortex at day 18 in vitro were stimulated by veratridine (10(-4)M) for 20 min. The cultures were subsequently processed for electron microscopy and the synapses were analyzed by quantitative techniques, incorporating set mathematical treatment. The mean values of area, perimeter, and form factor of the presynaptic elements significantly increased following veratridine stimulation, compared to the values of control synapses. The length of the postsynaptic thickening also increased, while synaptic curvature did not change significantly in the veratridine group. A fivefold reduction was observed in the mean number of synaptic vesicles per presynaptic element and in the vesicle-terminal area ratio, following veratridine stimulation. The cytoplasm-terminal area ratio and the occurrence of vacuoles/cisternae significantly increased after veratridine application. Planar measurement of membranes (boundary length) of different presynaptic organelles revealed that the total membrane did not change significantly in the veratridine group. The data indicated an increase in volume and swelling of the pre- and postsynaptic elements, considerable depletion of synaptic vesicles, and preservation of the total presynaptic membrane following veratridine stimulation in nerve tissue culture.

An ultrastructural study of the development of afferent and efferent synapses on outer hair cells of the guinea pig organ of Corti.

The guinea pig organ of Corti was studied using transmission electron microscopy, the second turn of the cochlea being examined at various ages between 20 days before birth and 30 days postnatal. Outer hair cells were examined at each of these ages. At all ages studied, the efferent (presynaptic) terminals are large and are packed with synaptic vesicles, whereas the afferent (postsynaptic) terminals are generally smaller, with a relatively small number of vesicles. During development, the subsynaptic cistern changes from a fragmented, diffuse profile extending over 50-70% of the length of the efferent contact zones, to a continuous, compact structure spanning neighbouring synapses. Synaptic vesicles in the efferent terminals are predominantly rounded in early development, flattened vesicles appearing postnatally. The synaptic bodies at afferent synapses do not change noticeably during development. Quantitative analysis revealed that the area of efferent terminals and the length of their active zone increase with increasing age, the same parameters decreasing in afferent terminals. Synaptic vesicles in the efferent terminals decrease in diameter, but remain constant in afferent terminals, with increasing age. The number of hair cell membrane invaginations decreases as development proceeds.

The effect of copper deficiency on the resistance of mice to infection with Pasteurella haemolytica.

Mice adjudged copper deficient on the basis of significantly decreased blood and tissue copper content and superoxide dismutase activity, but generally showing no clinical signs of deficiency, were infected intraperitoneally with Pasteurella haemolytica. In 3 separate experiments the LD50 for deficient animals was significantly depressed below that of copper-sufficient controls. Furthermore, in mice surviving challenge at doses near to the LD50, spleen weights were significantly increased and body temperatures depressed in the copper-deficient animals. These results indicate that subclinical copper deficiency in the mouse, is associated with an increased susceptibility to experimental infections with P. haemolytica.

Intestinal enzyme activity in lambs chronically infected with Trichostrongylus colubriformis: effect of anthelmintic treatment.

After twenty weeks of continuous dosing with Trichostrongylus colubriformis larvae substantial, but declining, numbers of worms had persisted in most of the lambs examined, although there were wide inter-individual variations. Mucosal lesions were found in the proximal small intestines of all the infected animals, their severity being directly related to worm burden. Representative brush border enzyme activities analysed in intestinal mucosal extracts from the same lambs showed differing responses. Alkaline phosphatase and glycyl-L-leucine dipeptidase were significantly depleted, whereas maltase activity was only marginally reduced, and leucine aminopeptidase activity was normal. Mucosal acetylcholinesterase activity was significantly elevated in the parasitised animals and, interestingly in view of the postulated role of this enzyme in nematode pathogenicity, the level of activity was directly correlated with individual worm burdens. Intestinal trypsin and chymotrypsin activities were unaffected and the level of superoxide dismutase, an enzyme associated with the inflammatory response, was normal. There were also no consistent changes in the mucosal activities of several enzymes including lactic dehydrogenase, creatine phosphokinase, aldolase, and glutamic oxaloacetate transaminase, whose leakage from damaged or necrotic tissues has been well defined in terms of the concomitant increase in their activity in the circulation. Lambs treated orally with fenbendazole five and/or ten weeks before slaughter either in the presence or absence of continued larval intake, had negligible worm burdens, and showed little evidence of intestinal damage at post mortem. Brush border enzyme levels, with the exception of alkaline phosphatase and, in two cases dipeptidase, were normal in these animals. The activity of alkaline phosphatase was approximately double that in the continuously infected, untreated lambs, but remained markedly lower than in the uninfected controls. The activities of the other enzymes studied, including acetylcholinesterase, were within the control range. In summary, in chronic trichostrongylosis even relatively low nematode burdens were associated with marked pathological and biochemical damage in the intestine with both lesion severity and mucosal acetylcholinesterase activity being directly related to worm numbers. Although morphological integrity was completely restored after anthelmintic treatment, the persistent low activity of brush border alkaline phosphatase coupled with the enzymological findings in untreated, infected animals suggests that recovery of the full functional capability of the intestinal mucosa may take longer.

Nigral and cerebellar synaptic terminals in the intermediate and deep layers of the cat superior colliculus revealed by lesioning studies.

The presence of degenerating nigral and cerebellar synaptic terminals in the intermediate and deep layers of the cat superior colliculus was demonstrated by electron microscopy following lesions of the substantia nigra or brachium conjunctivum. The superior colliculus was taken for analysis 4-5 days after operation. Nigral terminals underwent a dark type of degeneration following kainic acid lesion of the pars reticulata of the substantia nigra. The majority of nigral degenerating terminals and axons were found in the stratum griseum intermediale with a few in the stratum griseum profundum. Two kinds of cerebellar terminals were distinguished by general appearances such as size, type of synaptic contact and type of synaptic vesicle and by the pattern of degenerative changes following electrical lesion of the brachium conjunctivum. Large elongated synaptic terminals 4-7 microns in diameter, were found mainly in the stratum griseum profundum. They often had double termination with conventional dendrites and with vesicles containing dendrites. This kind of terminal had a filamentous type of degeneration. A second type of degenerating cerebellar terminal, characterized by an electron-lucent type of degeneration, was predominantly located in the stratum griseum intermediale. These terminals were circular, about 4 microns in diameter, and did not have synaptic contact with vesicle-containing profiles. The finding of the two types of degenerating terminal after lesion of the brachium conjunctivum can be considered as evidence of the coexistence of at least two kinds of cerebellar terminals in the superior colliculus. The presence of nigral and cerebellar terminals in the intermediate and deep layers of the superior colliculus implicates the involvement of the substantia nigra and cerebellum in control of collicular visuomotor function.

A morphometric study of cultured rat cerebral synapses exposed to different cationic media.

Quantitative techniques have been applied to compare the effects of high-K+, Mg2+ and Li+ media on the ultrastructure of cultured synapses alongside Na+-incubated controls. The explant cultures were prepared from 18-day-old embryonic rat cerebral cortices and maintained for 19 days in vitro. K+ -Stimulation for 25 min resulted in an increase in the mean perimeter and area of presynaptic terminals. Of these, the perimeter increase was the more pronounced, as indicated by a decrease in the value of the two-dimensional form factor. Reductions were also observed in the number of synaptic vesicles per presynaptic terminal, in the vesicle-terminal area ratio and in the synaptic vesicle density in an area adjacent to the presynaptic membrane, the latter two parameters being in positive linear correlation. The frequency of presynaptic cisternal/vacuolar profiles increased, and the synaptic curvature shifted in a positive direction. Synaptic length did not change following K+-exposure. Qualitative assessment indicated the presence of a network subjacent to the post-synaptic thickening and swelling of the postsynaptic ending after K+-stimulation. Incubation and fixation in Mg2+-media of two concentrations resulted in an increase in the number and area ratio of synaptic vesicles per terminal, and an elevation in the synaptic vesicle density in the higher Mg2+ concentration medium. Li+-treatment reduced the number of synaptic vesicles per terminal, the vesicle-terminal area ratio, and the vesicle density in the vicinity of the presynaptic membrane, while the synaptic curvature shifted in the positive direction. These changes were less pronounced than those characteristic of synapses in the K+ medium.

Quantitative ultrastructural changes in rat cortical synapses during early-, mid- and late-adulthood.

Quantitative ultrastructural analysis of rat parietal cortex was undertaken to determine the nature of the synaptic changes occurring in the molecular layer over a series of ages in early- (3 months), mid- (6 and 10 months) and late- (17 months) adulthood. The total number of synapses remained constant until 10 months of age, but decreased significantly by 17 months. Asymmetrical synapses on dendritic shafts were lost earlier (by 6 months) than asymmetrical synapses on dendritic spines (by 17 months). Symmetrical axodendritic synapses remained constant throughout adulthood. Analysis of synaptic terminal parameters revealed the following. Both individual and total presynaptic terminal areas decreased over the age range studied. Individual and total postsynaptic terminal areas, however, remained constant over the 3--17-month period. Positive correlations were obtained for the relationships between presynaptic terminal area and both age and synaptic vesicle number. The presynaptic terminal area was largest and contained the greatest number of vesicles at 3 months of age. This age was, in addition, characterized by the least numbers of mitochondria in the presynaptic terminal and spine apparatus in the postsynaptic terminal. The vacuolar and tubular cisternae of the presynaptic terminal were considerably reduced at 17 months. These data suggest that in the molecular layer of the cerebral cortex the period of adulthood is characterized by a diversity of synaptic changes. The 3-month age may reflect the end of the developmental phase and may be marked by changes in synaptic functional activity. The asymmetrical axodendritic synapses may constitute an intermediate form of synapse, capable of being transformed into axospinous synapses as dendritic spines continue to be formed in the adult.

Indirect ultrastructural evidence for lithium uptake by cultured rat cerebral cells through the sodium channel.

Cerebral cortices, prepared from 18-day-old rat embryos, were grown in explant cultures for 18 days. They were then incubated for 25 min in media of 3 types of cationic composition with and without tetrodotoxin and/or veratridine. Qualitative and quantitative ultrastructural observations revealed a considerable swelling of neuronal elements following veratridine exposure in both Na+ and Li+ media; this swelling was prevented by tetrodotoxin. By contrast, veratridine failed to induce swelling of neuronal profiles in a choline+ medium. The results indirectly indicate that Li+ ions enter cultured rat cerebral cells through the Na+ channels.

Synaptic remodelling and astrocytic hypertrophy in rat cerebral cortex from early to late adulthood.

Ultrastructural observation of the molecular layer of the parietal cortex of rats, aged 3, 6, 10 and 17 months, revealed various atypical synaptic profiles besides typical synapses. The atypical synapses were frequently in the vicinity of hypertrophied astrocytic profiles, and were sometimes completely surrounded by astrocytic processes. The presynaptic terminal contained either no vesicles or a few small distorted vesicles. Vacant postsynaptic terminals were occasionally seen. The total surface area of astrocytic profiles and the numbers of atypical synapses increased significantly between 3 and 10 months. The astrocytic acquisition of degenerating terminals was repeatedly observed over this period. Since there was no decrease in total synaptic number at this age, the astrocytic phenomenon may represent a stage in a continuous cycle of synaptic loss and replacement in the normal brain. By 17 months, when total synapse numbers decrease, synaptic replacement may be less than optimal.

Lithium entry into neural cells via sodium channels: a morphometric approach.

Rat cerebral cortical explants prepared from 18-day-old embryos were grown for 18 days in vitro. Cultures were exposed to Na+, Li+ and choline+ media, respectively, in the presence or absence of tetrodotoxin and/or veratridine, and processed for electron-microscopy. Veratridine (50 microM) induced an increase in summated and mean areas of neuronal profiles in Na+- and Li+-media but not in the choline+-medium: the summated perimeters did not change. The mean value of the neuronal form factor was significantly elevated following exposure to veratridine in a Na+- or Li+-dependent manner, indicating that the shape of the sectioned neuronal elements shifted towards an (ideal) circle. Qualitative assessment revealed an increased electron-lucency of the cytoplasm of neuronal profiles in Na+- and Li+-media containing veratridine. The veratridine-induced neuronal changes were inhibited by simultaneous addition of tetrodotoxin (1 microM) to the media. In the case of the glial cells, the values of the summated area and form factor did not change in the various experimental groups. The area of the extracellular space per unit area of sections significantly decreased in the Na+- and Li+-media following veratridine exposure; this did not occur in the choline+-medium. The results indicate a considerable swelling of the neuronal elements, reflecting cation, Cl- and water uptake following prolonged sodium channel activation in the presence of Na+ or Li+ ions. The quantitative ultrastructural data strongly suggests an entry of Li+ ions into cultured rat cerebral cells via sodium channels.

Junctions in rat neocortical explants cultured in TTX-, GABA-, and Mg++-environments.

The occurrence of various cell-to-cell contacts and membrane specializations was quantitated in neocortical explant cultures prepared from 18-day-old rat embryos and exposed continuously to tetrodotoxin (TTX), gamma-aminobutyric acid (GABA) and an elevated level of magnesium ions (Mg++), respectively. Chronic TTX treatment resulted in a decrease in the number of synapses, paired neuronal membrane thickenings and nerve terminals with synaptic vesicles; the area of the neuronal compartment also decreased. By contrast, the gap junctions between glial cells increased, although the glial paired membrane thickenings decreased in number per unit area. Long-term GABA and Mg++ exposures did not alter significantly the occurrence of any of the cell contacts and membrane specializations analyzed when compared to control values. The results suggest an inhibitory effect of TTX on neuronal maturation and synapse formation in explant cultures of rat neocortex; this may lead secondarily to an increased demand for glial cell-to-cell communication.
