[Comparative study in the guinea pig, cat and dog of hypothalamo-infundibular nervous fibers immunoreactive to an anti-LH-RH immune serum].

In the median eminence of guinea pig, cat and dog some axons of the hypothalamo-infundibular tract are immunoreactive with anti LH-RH. In these three species, the axons have a similar topography and area of peripheric terminations. These one occur at the capillaries of Mantelplexus. It was also shown, in the cat, that these areas of termination exhibit a strong monoamine fluorescence.

[Immunocytochemical study using semi-thin sections of the phenomena of early differentiation in several cell populations of the human fetal adenohypophysis].

Immunocytological investigations have been performed on semi-thin sections of human fetal pituitaries ranging in fetal age from 6 to 26 weeks. Corticotrophs can be revealed by anti-ACTH (1-24), anti-ACTH (17-39) and anti-beta MSH but not by anti-alpha MSH immunesera from the 8th week. Somatotrophs are revealed with anti-human STH from 9th week. Differentiating cells containing only alpha subunits of glycoproteic hormones are present from 8th to 12th week. At 13th week beta subunits of TSH can be revealed immunocytologically in thyreotrophs. Beta subunits of LH or FSH can be detected in same gonadotrophic cells only from 15th week.

[Cyto-immunological study of the Pleurodele pituitary gland].

In the pituitary of an Amphibian (Urodela), various cell types were localized by cytoimmunological techniques: corticotrophs located in a rostral zone near the median eminence, ventral orangeophilic cells secreting a prolactin-like hormone and erythrosinophilic cells in a more dorso-caudal situation secreting growth hormone. Alpha-MSH and beta-MSH produceng cell were identified in the intermediate lobe; these cells were also labelled with antibodies against 1-24 ACTH and 17-39 ACTH. These data are in accordance with results obtained after an experimental study of the prolactin cells.

[Cyto-immunological studies of the gonadotropic cells of the human fetus].

The immunicytological analysis of PAE fixed semi-thin sections of pituitary cells (of human fetuses aged 6 to 26 weeks) using immunesera directed against the human total glycoproteic hormones, HCG, LH, FSH and TSH or made specific for the LH, FSH, and TSH beta subunits shows: -- the presence up to the 12th week of only the subunit of the glycoproteic hormones revealed indifférently by our anti total hormones immunesera; -- the apparition of the betaTSH subunits at the 13th week, characterizing the differentiation of the thyrotrophs; -- the apparition of betaLH and betaFSH subunits on the one and same gonadotropic cell population at the 15th week.

[Immuno-cytologic study of LH-RH cells and fibers in the human fetus].

The application of immunofluorescence and immunoenzymology techniques with an anti LH RH I.S. permitted to reveal neurons and fibers in the human fetal hypothalamus from the 13th week up to birth. The morphology and dispersion areas of the neurons in the mediobassal, premammillary and anterior hypothalamus (lamina terminalis and its surroundings and the septum) as well as the direction and the destination of the immuno-reactive fibers of the hypothalamo infundibular tract have been described. These fibers are in contact with the capillaries of the Mantelplexus before and after the apparition of the intra-eminential loops at the 16th week. The general topography of the LH RH peptidergic system in the human fetus can be compared with the topography of other adult primates. The role of this system in the maturation and the control of gonadotropic cells is evoked.

Immunocytochemistry of pituitary corticotroph cells in terrestrial turtle (Testudo mauritanica).

The immunocytochemical and histochemical characters of the corticotroph cells of the Turtle adenohypophysis have been studied. These cells are localised in the rostral part of the gland and are revealed by Is anti ACTH (1-24) and (17-39). They are also colored with lead hematoxyline and PAS-Orange G. The corticotroph nature of these cells is confirmed by the study of their modifications after treatment with amphenone and ACTH. The Is anti ACTH also reveal most of the cells of the pars intermedia; while the Is anti beta-MSH reveals only these cells and some scatter cells of the pars distalis.

[Immunocytochemical study of corticotropic cells in the adenohypophysis of the chick embryo].

Corticotrophin cells of the pituitary have been studied immunocytologically with I.S. anti-A.C.T.H. (1-24) and I.S. anti-A.C.T.H. (17-39). These sera show differentiated cells as soon as the 8th of incubation. These cells remain located within the anterior part of the adenohypophysis. They are not "revelated" by I.S. anti-beta-M.S.H. and I.S. anti-beta-LPH.

[Corticotrope and melanotrope cells in cat, fox, rat and human fetus adenohypophysis: studies with induced fluorescence, cytoimmunologic technics and lead hematoxyline].

Formaldehyde-induced fluorescence, acid-catalyzed or not, methods, and immunocytology with anti-ACTH (1-24), anti-ACTH(17-39), ANTI-BETA-LPH immunserums were applied on the same preparations of cat, fox, rat and human foetus pituitaries. The superpositions of results showed that the pars intermedia and pars distalis corticomelano-lipotrophic cells of fox and cat pituitary, those of human foetal pituitary, and the purely corticotrophics cells of the rat pars distalis contained a fluorogenic probably granular compound. Moreover, the granules of the same cell types were electively revealed on our lymphilized material by plombic hematoxylin. Only the anti-beta-LPH and/or anti-beta-MSH fixing celpls exhibited hypercyanophilic, PAS-positive and bleu alcian-positive caracteristics.

Ultrastructural features of "solid cell nest" of the human thyroid gland: a study of 8 cases.

The ultrastructural features of solid cell nests (SCN), made of squamous cells, and associated calcitonin cells (C cells), of the thyroid gland were studied in only a few cases in humans. A study was performed on 8 paraffin-embedded SCN, postembedded in Epon, to look for their ultrastructural features. Immunohistochemical analysis using calcitonin antibody was performed on semithin sections of SCN to explore the presence of C cells. Three cases (37.5%) of SCN were positive for calcitonin, and electron-dense secretory granules were observed in the cytoplasm. In two of these cases, an increased number of C cells in the adjacent thyroid parenchyma was observed. The presence of ciliated and lymphoid cells, in addition to intracytoplasmic microvacuolar and microfollicular (microglandular) structures, was noticed. Ciliated cells have already been reported in embryonic rests of human and animals, but ultrastructurally for the first time in human SCN. The presence of microfollicular structures, intracytoplasmic microvacuolar, secretory granules features, and ciliated cells, in addition to lymphoid cell, suggests the existence of a common ultimobranchial stem cell for C cells or for one or more cell types of the thyroid gland.

[HLA antigen expression of human cornea. Effect of organ culture preservation].

The aim of this study is to examine the variation of HLA expression on organ-cultured human corneas. Using an indirect immunofluorescence technique and monoclonal antibodies anti-HLA-A, B, C and anti-HLA-DR histocompatibility antigens were detected. In fresh corneas class I antigens were detected on epithelium and stroma but not on endothelium. Class II antigens were not detected on any corneal layer but were present on Langerhans cells within epithelium and stroma. The expression of HLA-A, B, C antigens within the corneal epithelium and the corneal stroma was not altered by 28 days of organ culture. HLA-DR antigens were present on Langerhans cells on corneas that had been in organ culture up to 14 days (present 6/9). After 14-28 days in culture Langerhans cells were not found on most corneas (present 3/12). Corneal preservation in organ culture at 31 degrees C affects expression of HLA antigens particularly of HLA-DR antigens. Combination of HLA matching donor-recipient with an 10-15 days organ cultured corneal button in high-risk patients seems to be of interest.
