Difluorothromboxane A2 and stereoisomers: stable derivatives of thromboxane A2 with differential effects on platelets and blood vessels.

The present study reports on the selective effects on human platelets and canine saphenous veins of four stable difluorinated analogues and thromboxane A2 (TXA2), in which the characteristic 2,6-dioxa[3.1.1]bicycloheptane structure of TXA2 has been retained. The four compounds differ in their stereochemistry of the 5,6 double bond and/or the 15-hydroxyl group. Only 10,10-difluoro-TXA2 (compound I) with the natural stereochemistry of TXA2 was an agonist in both platelets and canine saphenous veins (EC50 = 36 +/- 3.6 nM and 3.7 +/- 0.8 nM, respectively). (15R)-10,10-Difluoro-TXA2 (compound II), (5E)-10,10-difluoro-TXA2 (compound III), and (5E,15R)-10,10-difluoro-TXA2 (compound IV) were antagonists of platelet aggregation stimulated by compound I (Kd = 98 +/- 46 nM, 140 +/- 42 nM, and 1450 +/- 350 nM, respectively). However, compounds II, III, and IV stimulated contraction of canine saphenous veins (EC50 = 36 +/- 4.4 nM, 31 +/- 6.8 nM, and 321 +/- 50 nM, respectively). All four compounds could displace the TXA2/prostaglandin H2 antagonist 9,11-dimethylmethano-11,12-methano-16-(3- 125I-4-hydroxyphenyl)-13,14-dihydro-13-aza-15 alpha beta-omega-tetranor-TXA2 from its platelet receptor (Kd values = 100 +/- 30 nM, compound I; 280 +/- 60 nM, compound II; 230 +/- 70 nM, compound III; and 1410 +/- 1020 nM, compound IV). These results support the existence of two subtypes of TXA2/prostaglandin H2 receptors and emphasize the importance of the stereochemical requirements of these TXA2 analogues for interaction with these receptors. These stable fluorinated TXA2 analogues should prove useful tools for the further characterization of these and other TXA2/prostaglandin H2 receptors.

Human erythroleukemia cells express functional thromboxane A2/prostaglandin H2 receptors.

The human erythroleukemia (HEL) cell line is a cultured hematopoietic cell line reported to express platelet membrane glycoproteins and alpha-2 adrenergic receptors. The present studies were designed to determine if functional thromboxane A2 (TXA2)/prostaglandin H2 (PGH2) receptors exist in HEL cells. Radioligand binding assays were performed using [125I]PTA-OH, a TXA2/PGH2 receptor antagonist. Scatchard analysis revealed one class of binding sites for 1-PTA-OH with a Kd = 122 +/- 18 nM and maximum binding = 1.7 +/- 0.3 x 10(5) sites/cell. Competition for [125I]PTA-OH binding with the TXA2/PGH2 receptor agonists SQ26655 and U46619 revealed one class of binding sites for SQ26655 with a Kd = 17 nM and two classes of binding sites for for U46619 with a Kd = 45 nM for the high-affinity site and a Kd = 450 nM for the low-affinity site. Competition for [125I]PTA-OH by the steroisomeric TXA2/PGH2 receptor antagonists L657925 and L657926 revealed two classes of binding sites for the more potent L657925 with a Kd = 8 nM for the high-affinity site and a Kd = 400 nM for the low-affinity site whereas L657926 bound to one class of sites with a Kd = 380 nM. Stimulation of the TXA2/PGH2 receptor by SQ26655 and U46619 resulted in concentration-dependent increases in [Ca++], as measured by FURA-2 fluorescence, with EC50 values of 28 +/- 2 and 149 +/- 32 nM, respectively. I-PTA-OH, L657925 and L657926 antagonized this response to U46619 with IC50 values similar in rank potency to that seen in the binding studies.(ABSTRACT TRUNCATED AT 250 WORDS)

Characterization of an 125I-labeled thromboxane A2/prostaglandin H2 receptor agonist.

Stable synthetic mimetics of thromboxane (TX) A2 and prostaglandin (PG) H2 have been synthesized and reported to stimulate platelets and vascular smooth muscle. The synthetic agonists induce aggregation of isolated platelets and contraction of vascular tissue. The tritiated agonists [3H]U46619 and [3H]U44069 have been used in radioligand binding studies to characterize platelet and vascular smooth muscle TXA2/PGH2 receptors, but have limited usefulness due to their low specific activities and variable specific binding. In an attempt to overcome these problems, we have synthesized a stable, high affinity, 125I-radiolabeled TXA2/PGH2 receptor agonist, [1S-(1 alpha, 2 beta (5Z), 3 alpha(1E,3S*), 4 alpha)]-7-[3-(3-hydroxy-4-(4'-iodophenoxy)-1-butenyl)-7-oxabicyclo - [2.2.1]heptan-2-yl]-5-heptenoic acid (I-BOP). I-BOP induced shape change, increased intracellular free calcium concentrations and aggregated isolated human platelets (EC50 = 0.21 +/- 0.05 nM, n = 3; 4.1 +/- 1.1 nM, n = 4; 10.8 +/- 3 nM, n = 9, respectively). The kinetically determined Kd was 1.02 +/- 0.33 nM (kobs = 0.19 +/- 0.05 min-1, k-1 = 0.097 +/- 0.02 min-1, k1 = 0.119 +/- 0.03 min-1 M, n = 4). Equilibrium binding studies of [125I]BOP to isolated human platelets indicated one class of high affinity sites, Kd = 2.2 +/- 0.3 nM and a maximum binding of 0.028 +/- 0.002 x 10(-12) mol/10(7) platelets (1699 +/- 162 sites/platelet, n = 9).(ABSTRACT TRUNCATED AT 250 WORDS)

The affinities of prostaglandin H2 and thromboxane A2 for their receptor are similar in washed human platelets.

Both thromboxane A2 (TXA2) and its precursor prostaglandin H2 (PGH2) are labile and share a common receptor. The affinities of these two compounds for their putative common receptor are unknown. We compared the potencies of TXA2 and PGH2 to aggregate human platelets and bind to the TXA2/PGH2 receptor. TXA2 was more potent than PGH2 in initiating aggregation in platelet-rich plasma, EC50 of 66 +/- 15 nM and 2.5 +/- 1.3 microM, respectively. In washed platelets, however, PGH2 was more potent than TXA2 with EC50 values of 45 +/- 2 nM and 163 +/- 21 nM, respectively. The affinity of these two compounds in washed platelets was determined in radioligand competition binding assays employing [125I]-PTA-OH. The Kd values for PGH2 and TXA2 were 43 nM and 125 nM, respectively. The results demonstrate that the affinity of PGH2 for the platelet TXA2/PGH2 receptor is greater than previously thought. The data raise the possibility that PGH2 may significantly contribute to the responses attributed to TXA2 in vivo.

Different pharmacologic activities for 13-azapinane thromboxane A2 analogs in platelets and blood vessels.

A series of 16 13-azapinane thromboxane A2 analogs were synthesized and their pharmacological dissociation constants (Kd) determined for the thromboxane A2/prostaglandin H2 (TXA2/PGH2) receptor in both washed human platelets and canine saphenous veins. Twelve of the analogs were antagonists of both U46619-induced platelet aggregation and saphenous vein contraction. The rank order potencies of these analogs were significantly different in platelets compared to saphenous veins. Four of the derivatives were antagonists in the platelets but possessed agonist activity in the vessels. The potency of the analogs and the type of activity (agonist or antagonist) were found to be sensitive to the substitution pattern of the aromatic ring on the bottom side chain. These results further support the notion that platelet TXA2/PGH2 receptors are distinct from that of vascular receptors.

Subcellular localization of a thromboxane A2/prostaglandin H2 receptor antagonist binding site in human platelets.

The subcellular localization of a binding site for the competitive thromboxane A2/prostaglandin H2 (TXA2/PGH2) antagonist, 9,11-dimethylmethano-11,12-methano-16-(3-iodo-4- hydroxyphenyl)-13,14-dihydro-13-aza-15 alpha beta-omega-tetranor TXA2 ([125I]-PTA-OH), was determined. Subcellular fractions of platelets were prepared by glycerol lysis or nitrogen cavitation, and were characterized by the use of enzymatic markers specific for plasma membranes, endoplasmic reticulum (dense tubular system), mitochondria, granules, and cytosolic constituents. The Kd and density of binding sites in the subcellular fractions were determined by Scatchard analysis of equilibrium binding data. The Kd and Bmax for [125I]-PTA-OH determined in the lysates were 49 +/- 11 nM and 4.1 +/- 1.7 pmol/mg protein respectively (N = 6). The Kd values were not significantly different in any of the fractions assayed. The binding sites were coenriched (4.5 +/- 0.66 fold) with the enzymatic markers for plasma membranes (3.7 +/- 0.5 fold) and dense tubular system (2.4 +/- 0.4 fold). The binding sites were not coenriched with markers for cytoplasmic constituents, mitochondria, or granules. The ability of the TXA2/PGH2 mimetic U46619 to compete with [125I]-PTA-OH for the binding site was also determined for the various subcellular fractions. The IC50 for U46619 was 5.4 +/- 1.2 microM in the lysate, and was not significantly different in the subcellular fractions. These data suggest that the binding site is the TXA2/PGH2 receptor described previously. These data are consistent with the notion that the putative TXA2/PGH2 receptor is localized in the plasma membranes and/or the dense tubular system.

Polymyxin B increases TXA2/PGH2 agonist and antagonist affinities in human platelet membranes.

The effects of polymyxin B, a cyclic polycationic peptide antibiotic, on the binding of thromboxane A2/prostaglandin H2 (TXA2/PGH2) receptor agonists and antagonists to the putative human platelet TXA2/PGH2 receptor have been determined. Crude membranes were prepared from washed human platelets and the binding of [125I]-PTA-OH, a TXA2/PGH2 receptor antagonist, was used as a ligand to characterize the effects of polymyxin B on agonist and antagonist binding. Polymyxin B produced a concentration-dependent decrease in the IC50 values for the TXA2/PGH2 agonist, U46619, in radioligand competition binding assays. Polymyxin B also decreased the IC50 values of the TXA2/PGH2 agonist ONO 11113 from 75 +/- 11 nM to 38 +/- 2.5 nM and the TXA2/PGH2 receptor antagonist SQ29548 from 60 +/- 14 nM to 20 +/- 2.3 nM. The Kd for [125I]-PTA-OH was also reduced by polymyxin B from 107 +/- 10 nM to 76 +/- 6 nM but it had no effect on the maximum number of binding sites (Bmax). Polymyxin B had no significant effect on the binding affinity of U46619 in intact washed human platelets. These results raise the possibility that polymyxin B can alter the conformational state of the TXA2/PGH2 receptor when it is in a crude membrane fraction but not in intact platelets.

Desensitization of platelet thromboxane A2/prostaglandin H2 receptors by the mimetic U46619.

Thromboxane A2 (TXA2) and prostaglandin H2 (PGH2) activate platelets through membrane receptors. This study sought to determine if changes occur in the TXA2/PGH2 receptor during its desensitization induced by exposure to its agonist 11 alpha,9 alpha-(epoxymethano)prosta-5Z,13E-dienoic acid (U46619). Washed human platelets were incubated for 30 min with U46619 in the presence of an antiaggregatory agent, Iloprost. The platelets were washed and the aggregation response to U46619 was determined. The EC50 increased from 372 +/- 94 nM in the control group to 826 +/- 143 nM for the U46619-pretreated group (n = 7). This desensitization was specific inasmuch as the aggregation responses to thrombin and the calcium ionophore A23187 were not affected by U46619 pretreatment. Desensitization was accompanied by a decrease in the number of binding sites for [3H]U46619 from 789 +/- 189 in the control to 386 +/- 120 sites per platelet in the U46619-treated group (n = 5) and a decrease in the number of binding sites for the TXA2/PGH2 receptor antagonist, [125I]9,11-dimethylmethano-11,12-methano-16(3-iodo-4-hydroxyphenyl )-13,14- dihydro-13-aza-15-alpha beta-omega-tetranor-TXA2 from 3988 +/- 957 to 2443 +/- 553 (n = 8). The Kd for U46619 was 37 +/- 10 nM in the control group and 23 +/- 11 nM for the U46619-treated group (n = 5). The Kd for I-9,11-dimethylmethano-11,12-methano-16(3-iodo-4-hydroxyphenyl)-13 ,14-dihydro- 13-aza-15-alpha beta-omega-tetranor-TXA2 changed from 58 +/- 12 nM in the control to 44 +/- 9 nM in the U46619-treated group (n = 8).(ABSTRACT TRUNCATED AT 250 WORDS)

Decrease in agonist affinity for human platelet thromboxane A2/prostaglandin H2 receptors induced by a platelet-derived supernatant.

Platelets possess membrane receptors which mediate the aggregatory response to thromboxane A2 (TXA2) and prostaglandin H2 (PGH2). It has been observed recently that the affinities for a series of TXA2/PGH2 mimetics are decreased in crude human platelet membranes and solubilized membranes compared to intact washed platelets. The present study investigated the notion that platelets contain a substance that is released during platelet lysis that reduces the affinity of the TXA2/PGH2 receptor for agonists. The displacement of 9,11-dimethylmethano-11,12-methano-16-(3-iodo-4-hydroxyphenyl)-13, 14-dihydro-13 - aza-15 alpha beta-omega-tetranor-TXA2 ([125I]PTA-OH), a TXA2/PGH2 receptor antagonist, from its binding site in intact washed platelets by TXA2/PGH2 mimetics and antagonists was characterized in the presence or absence of the supernatant (50,000 g) obtained from sonicated platelets. In the presence of the supernatant, there was a significant (P less than 0.025) increase in the IC50 values for the TXA2/PGH2 mimetics U46619, SQ26655, and ONO11113. The increase in the IC50 for U46619 induced by the supernatant was abolished by either boiling or treating the supernatant with trypsin. The supernatant did not affect the Kd or Bmax of [125I]PTA-OH or the IC50 of the TXA2/PGH2 antagonist, SQ29548. Pretreatment of the platelets with the supernatant resulted in a significant (P less than 0.02) reduction in the aggregation response induced by U46619. Gel filtration (Sephacryl S200) of the supernatant revealed a fraction (molecular weight approximately 100,000 daltons) which significantly increased the IC50 for U46619 to displace [125I]PTA-OH from its binding site. Thus, human platelets appear to possess a protein(s) that is released into the supernatant upon sonication and inhibits the binding of TXA2/PGH2 agonists but not antagonists to their receptor. This protein may play a role in the regulation of platelet responses to the aggregatory stimuli TXA2/PGH2.

Extraction of thromboxane B2 from urine using an immobilized antibody column for subsequent analysis by gas chromatography-mass spectrometry.

This paper describes an antibody affinity (immunoaffinity) column which, in one step, extracts and sufficiently purifies urinary thromboxane B2 (TXB2) for quantitative analysis by high resolution gas chromatography-negative ion chemical ionization-selected ion monitoring-mass spectrometry (HRGC-NICI-SIM-MS). Polyclonal TXB2 antibody from rabbit was partially purified using immobilized Staphylococcus aureus Protein A. The purified IgG fraction was then immobilized using an N-hydroxysuccimidyl silica gel. The resulting matrix bound 570 ng TXB2 per ml of gel. TXB2 was quantitatively eluted with acetonitrile-water (19:1). Columns constructed from the gel could be used repeatedly since binding capacity was reconstituted using 0.01 M phosphate buffer (pH 7.4) with no apparent loss of activity. Using these columns, urinary TXB2 was sufficiently purified in one step such that in subsequent analysis by HRGC-NICI-SIM-MS interference free chromatograms were observed.

Binding of a thromboxane A2/prostaglandin H2 agonist [3H]U46619 to washed human platelets.

The binding characteristics of [3H]U46619 to washed human platelets were studied. [3H]U46619 binding to washed human platelets was saturable and displaceable. Kinetic studies yielded a Kd of 11 +/- 4 nM (n = 4). Scatchard analysis of equilibrium binding studies revealed one class of high affinity binding sites with a Kd of 20 +/- 7 nM and a Bmax of 9.1 +/- 2.3 fmole/10(7) platelets (550 +/- 141 binding sites per platelet) (n = 4). A number of compounds that act as either agonists or antagonists of the TXA2/PGH2 receptor were tested for their ability to inhibit the binding of [3H]U46619 to washed human platelets. The Kds of the agonists and antagonists were similar to their potencies to induce or inhibit platelet aggregation. These data provide some evidence that [3H]U46619 binds to the putative human platelet TXA2/PGH2 receptor.

Binding of thromboxane A2/prostaglandin H2 agonists to human platelets.

The competition of [125I]-9, 11 dimethylmethano-11, 12 methano-16-(3-iodo-4-hydroxyphenyl)-13, 14-dihydro-13-aza 15 alpha beta-omega-tetranor-thromboxane A2 ([125I]-PTA-OH), a thromboxane A2/prostaglandin H2 receptor antagonist, with a series of thromboxane A2/prostaglandin H2 (TXA2/PGH2) mimetics for binding to the putative TXA2/PGH2 receptor in washed human platelets was studied. The rank order potency for the series of mimetics to compete with [125I]-PTA-OH for binding was compared with their rank order potency for induction of platelet aggregation. The rank order potency for the mimetics to compete with [125I]-PTA-OH for binding was ONO-11113 greater than SQ-26655 greater than U44069 greater than U46619 = 9, 11-azo PGH2 greater than MB28767. This rank order potency was highly correlated with their rank order potency for inducing platelet aggregation (r = 0.992). Changes in the intra or extracellular concentrations of Na+ did not have a significant effect on the competition between U46619 and [125I]-PTA-OH for binding to the putative receptor. In summary, it appears that these TXA2/PGH2 mimetics activate human platelets through the putative TXA2/PGH2 receptor.

Platelet and vascular smooth muscle thromboxane A2/prostaglandin H2 receptors.

Thromboxane A2 (TxA2) and prostaglandin H2 (PGH2) aggregate platelets and contract vascular smooth muscle. Inasmuch as both compounds produce the same effects and presumably through the same receptor, their receptors have been referred to as TxA2/PGH2 receptors. Pharmacological studies of stable agonists and antagonists of the TxA2/PGH2 receptors have shown different rank order potencies for these compounds in platelets compared with blood vessels. These studies have provided evidence to support the hypothesis that the platelet TxA2/PGH2 receptor is different from the one found in vascular tissue. The vascular receptor has been named [TxA2/PGH2]tau and the platelet receptor has been named [TxA2/PGH2]alpha. In the past few years several radiolabeled antagonists and agonists have been developed and used in radioligand-binding studies, primarily in platelets. One of these ligands, 125I-labeled PTA-OH, a TxA2/PGH2 receptor antagonist, has been extensively used to characterize the human platelet TxA2/PGH2-binding site. It has been found to have a Kd of approximately 20 nM and a Bmax of 2500 binding sites/platelet. Through the combination of pharmacological and biochemical approaches, it should be possible to characterize platelet and vascular TxA2/PGH2 receptors.

Binding of a thromboxane A2/prostaglandin H2 receptor antagonist to guinea-pig platelets.

The binding of [125I]9,11-dimethylmethano-11,12-methano-16-(3-iodo-4-hydroxypheny l)-13,14- dihydro-13-aza-15 alpha beta-omega-tetranor-TXA2 [( 125I]PTA-OH), a thromboxane A2/prostaglandin H2 receptor antagonist, to washed guinea-pig platelets was studied. [125I]PTA-OH bound to guinea-pig platelets in a saturable and displaceable manner. The Kd for [125I]PTA-OH was 14.5 +/- 2 nM (n = 4) and the Bmax was 32 +/- 7 fmol/10(7) platelets or 1,927 +/- 422 binding sites/platelet. The IC50 value for a series of 13-azapinane TXA2 analogs to antagonize the TXA2/PGH2 mimetic U46619-induced platelet aggregation and displace [125I]PTA-OH from its binding site was determined. The IC50 values for the series of five antagonists were highly correlated (r = 0.99) in the binding assays and aggregation studies. The ability of a series of five agonists to displace [125I]PTA-OH from its binding site was compared to their ability to induce platelet aggregation. All the agonists completely displaced the ligand from its binding site but their rank order did not correlate well with their ability to induce aggregation (r = 0.37). Collectively, the data are consistent with the notion that [125I]PTA-OH binds to a putative TXA2/PGH2 receptor in guinea-pig platelets.
