Septicemic salmonellosis in two llamas.

At necropsy, Salmonella choleraesuis var kunzendorf was recovered from the lungs of a 6-year-old female llama that died within a few days of onset of illness. The llama had had fence-line contact with 40 sows at a farm. Salmonella typhimurium was isolated from a blood sample of a 6-day-old male cria that also died after a short illness.

Endogenous marinobufagenin-like immunoreactive factor and Na+, K+ ATPase inhibition during voluntary hypoventilation.

In previous studies investigators found that conditioned hypoventilatory breathing potentiated a sodium-sensitive form of hypertension in dogs that was not mediated by sympathetic nervous system arousal. Our study investigated effects of 30 minutes of voluntary hypoventilation, maintained by a respiratory gas monitor and feedback procedure, in 16 normotensive humans of both sexes on (1) plasma concentrations of endogenous digitalis-like factors (ouabain-like and marinobufagenin-like immunoreactivity), (2) activity of erythrocyte Na+, K+ -ATPase, (3) inhibitory activity of plasma Na+, K+ -ATPase, and (4) blood pressure. Increased end tidal PCO2 (41 +/- 0.78 mm Hg versus 37.6 +/- 1.03 mm Hg) was associated with (1) an increase in plasma marinobufagenin-like immunoreactivity (1.23 +/- 0.47 versus 4.96 +/- 1.19 nmol/L), (2) an inhibition of Na+, K+ -ATPase in red blood cells (3.68 +/- 0.22 versus 2.15 +/- 0.25 mmol Pi/mL-1/h-1; P < .01), (3) increase in plasma Na+, K+ -ATPase inhibitory activity (34.9 +/- 4.0% versus 48.8 +/- 2.1%, P < .02), and (4) increases in systolic (112.4 +/- 2.6 versus 107.6 +/- 1.8 mm Hg) and diastolic (73.5 +/- 2.1 versus 68.8 +/- 2.1 mm Hg) blood pressures. Plasma levels of ouabain-like immunoreactivity did not increase significantly. Incubation of erythrocytes obtained during hypoventilation with antidigoxin antibody restored the Na+, K+ -ATPase activity (3.99 +/- 0.34 mmol Pi/mL-1/h-1). Cessation of hypoventilation was associated with decreases in diastolic blood pressure (70.5 +/- 2.2 mm Hg) and restoration of Na+, K+ -ATPase activity in erythrocytes (2.99 +/- 0.43 mmol Pi/mL-1/h1).(ABSTRACT TRUNCATED AT 250 WORDS)

Surgical management of preputial injuries in bulls: 51 cases (1986-1994).

The purpose of this study was to compare success rates for preputial surgery depending on the initial diagnosis, ability to extend the penis, use of sedation and local anesthesia versus general anesthesia for surgery, and surgical technique. Medical records of 51 bulls treated surgically for preputial injury were reviewed. The mean age of the bulls was 2.5 years ranging from 1 to 5 years. Bos taurus breeds (82.3%) were more often affected than Bos indicus breeds (17.7%). The most common breeds represented in this study were Angus (45.1%), Simmental (11.8%), and brangus (9.8%). The seasonal incidence of preputial injuries was higher during the period of May-July (52.9%) and November-February (33.3%). The overall success rate was 70%. Posthioplasty was more successful than circumcision (90% to 43%) (P < 0.05). The success rate for surgeries performed under inhalation anesthesia in the surgery suite was 100% compared with a success rate of 63% for those animals operated on with injectable and local anesthesia in a rotary chute (P < 0.05). The success rate was 88% if the penis could be extended before the surgery and 36% if extension was not achieved (P < 0.05).

Comparative analyses of peritoneal fluid from calves and adult cattle.

Reference values for hematologic variables change with increasing age in cattle. Therefore, the purpose of the study reported here was to describe the peritoneal fluid constituents of clinically normal young calves, and to compare cellular concentration and distribution in blood and peritoneal fluid of young calves with those of adult cattle. Eight healthy 8-week-old male Holstein calves and 8 healthy 3- to 8-year-old Holstein cows were studied. Peritoneal fluid was collected from calves along the ventral midline, 4-cm cranial to the umbilicus. Abdominocentesis was performed in the region of the lower right flank in adult cattle. Correlation analysis, using the Pearson's correlation coefficient, and regression analysis were performed for blood and peritoneal fluid data from calves. Data from calves were compared with those of cows, using Wilcoxon's rank sum test. A P value < 0.05 was considered significant for all tests. Calves had significantly lower blood eosinophil count (P < 0.003) and plasma protein concentration (P < 0.001) than did cows. Calves had significantly higher peritoneal fluid nucleated cell (P < 0.05) and mononuclear cell (P < 0.05) counts, but lower peritoneal fluid eosinophil cell count (P < 0.003) than did cows. For calves, nucleated cell and lymphocyte cell counts in the blood had a high, positive correlation with those of peritoneal fluid. However, the prediction equation for nucleated cell count accounted for a modest proportion of variability. A prediction equation for peritoneal fluid lymphocyte cell count was established.(ABSTRACT TRUNCATED AT 250 WORDS)

Penetrance and expressivity of the chromosome 9p melanoma susceptibility locus (MLM).

A susceptibility locus for familial melanoma has been localized to the short arm of chromosome 9. Penetrance of melanoma was estimated by calculating the Kaplan-Meier function and fitting a log normal hazard function in 124 gene carriers in three 9p-linked kindreds. The penetrance of the gene for melanoma was estimated to be 53% by age 80. Additionally, nevus counts, skin type, and sun exposure histories were gathered for 119 individuals in two kindreds. Gene carriers were found to have higher nevus counts and nevus densities than non-gene carriers. Among gene carriers, individuals with melanoma were found to have more sun exposure within each skin type than gene carriers without melanoma. These analyses suggest that the 9p melanoma susceptibility is related to total number of nevi and that it interacts with other genetic and environmental factors to produce melanoma.

Localization of the 9p melanoma susceptibility locus (MLM) to a 2-cM region between D9S736 and D9S171.

A familial melanoma susceptibility locus (MLM) was identified on the short arm of chromosome 9 in a set of Utah and Texas kindreds. Subsequent confirmation was reported for a set of 26 Australian kindreds, a set of 7 Dutch kindreds, and a set of 13 NIH melanoma kindreds. The original localization report placed the locus near the IFNA-D9S126 interval on chromosome 9p. We report further localization using D9S171 and a newly developed marker, D9S736, both of which lie in the IFNA-D9S126 interval. Analysis of this set of kindreds places the melanoma susceptibility locus in a 2-cM region that is proximal to D9S736 and distal to D9S171.

Persistent dorsal displacement of the soft palate in two young bulls.

Dorsal displacement of the soft palate was diagnosed by means of endoscopy and radiography in a 10-month-old Chianina/Angus bull and an 11-month-old Limousin bull. Both bulls produced respiratory noise that was audible in all phases of respiration. The Limousin bull also had exercise intolerance and became dyspneic when excited. The Chianina/Angus bull was treated with anti-inflammatory medication and rest. The respiratory noise resolved over a 4-month period, and the Chianina/Angus bull has been breeding successfully on pasture. Sternothyroideus and sternohyoideus myectomy was performed in the Limousin bull. Immediate postoperative improvement was noticed clinically and endoscopically. The bull had normal respiration 4 months after surgery and was successfully breeding on pasture. Although rare, dorsal displacement of the soft palate should be considered when evaluating cattle with respiratory noise or exercise intolerance.

Hypervariable epitope constructs as a means of accounting for epitope variability.

Epitope variability is one of the greatest obstacles to development of synthetic peptide vaccines. Based on a recently described hypervariable epitope (aa 414-434) on the envelope glycoprotein (gp130) to simian immunodeficiency virus (SIVmac142), we have developed a novel approach to account for epitope variability. We have prepared, in a single synthesis, a cocktail of peptides, designated a hypervariable epitope construct (HEC), which collectively represent all the in vivo variability seen in an epitope. The HEC represents permutations of amino acid substitutions found in the epitope and has been able to induce antibodies with enhanced binding to native SIV and broad immunoreactivity to related epitope analogues.

Behavior analysis and the search for the origins of hypertension.

Hypertension is a disorder of sodium regulation that develops over time in a context of the interactions of the individual with the environment. Experimental hypertension can be induced in laboratory animals and normotensive humans via increases in sodium intake under conditions of aversive behavioral control. Readiness for avoidance contingencies includes a breathing pattern characterized by subnormal rate and normal tidal volume. Studies with humans have shown that this inhibitory breathing pattern is associated with increased plasma acidity, increased renal sodium reabsorption, increased secretion of digitalis-like hormones that inhibit sodium-pump activity, and increased vasoconstriction and blood pressure. Behavioral research is needed that defines the necessary and sufficient conditions for inhibitory breathing and its role in the development of hypertension.

Effects of chemical restraint on the endoscopic appearance of laryngeal and pharyngeal anatomy and sensation in adult cattle.

Effects of 2 drugs commonly used for chemical restraint of cattle were evaluated for their effect on laryngeal and pharyngeal anatomy, function, and response to stimuli. Eighteen adult Jersey cows, free of respiratory tract disease, were studied. Cows were assigned at random to 1 of 3 treatment groups. Endoscopic evaluations were performed before and at a predetermined time interval after administration of each drug. Responses to stimuli were evaluated by stimulating 7 preselected sites (epiglottis, left and right arytenoid cartilages, left and right vocal folds, and left and right dorsolateral pharyngeal walls) with a closed, transendoscopic biopsy probe. Xylazine HCl (0.05 mg/kg of body weight, i.v.) was administered to group-1 cows (n = 6), and endoscopy was repeated 5 minutes after administration of the drug. Xylazine (0.07 mg/kg, i.v.) was administered to group-2 cows (n = 6), and endoscopy was repeated 5 minutes after administration of the drug. Acepromazine maleate (0.035 mg/kg, i.v.) was administered to group-3 cows (n = 6), and endoscopy was repeated 10 minutes after administration of the drug. Responses to stimuli were scored as brisk (0), moderate (1), slow (2), and absent (3). Scores for responses to stimuli were compared, using the Wilcoxon signed-rank test for data within groups, and a general linear models procedure, using the Kruskal-Wallis test between groups. Interobserver agreement rates were generated for each group. A value of P < 0.05 was considered significant. Xylazine profoundly changed laryngeal sensitivity and function at both dosages. The corniculate processes of the arytenoid cartilages were observed to be in a markedly adducted position after sedation.(ABSTRACT TRUNCATED AT 250 WORDS)

Comparison of peritoneal fluid analysis before and after exploratory celiotomy and omentopexy in cattle.

The effect of right paralumbar fossa exploratory celiotomy and omentopexy on peritoneal fluid constituents was studied in 22 adult dairy cows. Six cows were eliminated on the basis of physical examination findings (n = 2), surgical findings (n = 2), or inability to obtain a sufficient volume of peritoneal fluid (n = 2). Sixteen cattle had normal results of CBC and serum biochemical analysis, and a minimum of 1 ml of peritoneal fluid was obtained by abdominocentesis. Abdominocentesis was repeated on days 1, 2, and 6 after surgery. Statistical analysis for repeated measures was performed, using a significance level of P < 0.05. Stage of gestation was evaluated for interaction with time. Mean total nucleated cell count was 3,200 cells/microliters before surgery, was significantly increased 2 days after surgery (16,336 cells/microliters), and continued to increase through day 6 (20,542 cells/microliters). Mean polymorphonuclear cell count was 1,312 cells/microliters before surgery and was significantly higher at 2 (11,043 cells/microliters) and 6 (10,619 cells/microliters) days after surgery. Mean lymphocyte count was 254 cells/microliters before surgery and was significantly increased 2 days (1,911 cells/microliters) after surgery. By day 6, lymphocyte numbers were similar to preoperative values. Mean mononuclear cell count was 770 cells/microliters before surgery and was significantly increased on days 1 (3,084 cells/microliters), 2 (3,285 cells/microliters), and 6 (2,349 cells/microliters) after surgery. Mean eosinophil numbers were 1,388 cells/microliters before surgery and were significantly increased on day 6 (6,347 cells/microliters) only. Interaction between time and stage of gestation was found only for specific gravity and total protein concentration.(ABSTRACT TRUNCATED AT 250 WORDS)

Biological studies of a nitroso compound that releases nitric oxide upon illumination.

2-Methyl-2-nitrosopropane (MNP) has long been known to undergo photochemical and thermal decomposition, generating di-tert-butyl nitroxide, in organic solvent. The present study was undertaken to demonstrate that MNP can be used as a caged-nitric oxide (NO), which can liberate NO upon illumination. Photolysis of MNP leads to the generation of tert-butyl radical and NO, as detected by spin-trapping/ESR spectroscopy and by oxyhemoglobin/visible spectroscopy, respectively. Using soluble guanylate cyclase in neuroblastoma N1E-115 cells as an NO target, we found that MNP in the presence of light caused a dose- and time-dependent increase in cGMP. Finally, illumination of a solution of MNP was also found to induce relaxation of preconstricted isolated rat pulmonary artery rings. These studies demonstrated that MNP can be useful biochemical research tool for delivering NO in a controlled manner, by using light.

Endoscopic evaluation of the nasopharynx, pharynx, and larynx of Jersey cows.

Endoscopy of the nasopharynx, pharynx, and larynx was performed in each of 25 adult Jersey cows, age and body weight of which ranged from 2 to 6 years and 300 to 365 kg, respectively. The endoscopic appearance of normal anatomic structures of the proximal portion of the airway were described. Observations specific to female dairy cattle were: the nasal septum, which tapered caudodorsally in the distal third of the nasal passage; the ability to observe both ethmoturbinates from the same viewing side; presence of a pharyngeal septum; the nasopharyngeal opening of the auditory tubes dorsolateral to the pharyngeal septum; and the appearance of the larynx--a triangular epiglottis with round borders and prominent corniculate process of the arytenoid cartilages. Tracheoscopy was performed in 13 cows. Of 11 cows for which the soft palate could be observed immediately after withdrawing the endoscope, 7 had dorsal displacement of the soft palate.

Effects of caffeine on the metabolic and catecholamine responses to exercise in 5 and 28 degrees C.

The influence of caffeine on the metabolic and catecholamine responses to mild exercise in a cold and a warm environment was studied in eight healthy males. The subjects performed 60 min of cycling at 50% VO2max in a cold environment (5 degrees C and 70% relative humidity) and a warm environment (28 degrees C and 50% relative humidity) 30 min after ingesting caffeine (5 mg.kg-1 body weight) or placebo (dextrose). Caffeine ingestion prior to exercise in the warm environment resulted in increased plasma epinephrine, with no effect on plasma norepinephrine. Neither lipid nor carbohydrate metabolism was altered by caffeine in the warm trial. Exercise in the cold environment (placebo) produced increased VO2 and carbohydrate metabolism, decreased lipid metabolism, and no difference in plasma catecholamines compared with the warm-placebo trial. Responses to the combination of caffeine ingestion and the cold environment did not differ from cold-placebo responses in VO2 during the cycling bout. However, in the cold-caffeine trial plasma epinephrine was elevated. In addition, fat oxidation and serum free fatty acids were elevated in the cold-caffeine condition. Carbohydrate oxidation was depressed, while serum glucose and blood lactate were elevated in this trial compared to cold-placebo. Thus, caffeine increases plasma epinephrine; cold increases oxygen consumption and carbohydrate metabolism, while decreasing lipid metabolism; and the combination of caffeine and cold during exercise increases plasma epinephrine and lipid metabolism, but decreases carbohydrate metabolism.

Small-intestinal volvulus in cattle: 35 cases (1967-1992).

Medical records of 35 cattle with small-intestinal volvulus were reviewed. Surgical correction was performed on 32 cattle, and 17 of these cattle were discharged from the hospital. Mean duration of clinical signs for survivors was not significantly different from that for nonsurvivors, and the most commonly recorded clinical signs were abdominal pain, anorexia, lethargy, abdominal distention, and dehydration. Physical examination of cattle with intestinal volvulus revealed tachycardia, tachypnea, and normothermia. Rectal examination findings included distended small intestine, scant feces or mucus, and tight bands coursing dorsoventrally in the middle portion of the abdomen. Clinicopathologic testing revealed azotemia, hypocalcemia, hyperglycemia, and leukocytosis with a left shift. Nonsurvivors had significantly lower mean preoperative venous blood pH and mean base excess and higher mean serum potassium concentration than did survivors. A diagnosis of volvulus of the entire small intestine was made during surgery in 25 cattle, whereas volvulus of the distal jejunum and ileum was diagnosed during surgery in 7 cattle. Survival rate following surgical correction of volvulus of the entire small intestine (44%) was not significantly different from survival rate following surgical correction of volvulus of the distal jejunum and ileum (86%). However, survival rate for dairy cattle (63%) was significantly higher than survival rate for beef cattle (22%). To determine potential risk factors for the development of small-intestinal volvulus, epidemiologic data from cattle admitted to veterinary teaching hospitals throughout North America were collected by searching records entered into the Veterinary Medical Data Base.(ABSTRACT TRUNCATED AT 250 WORDS)

Familial breast cancer risks. Effects of prostate and other cancers.

BACKGROUND: Previous studies have provided conflicting results concerning the effects of a family history of prostate and other cancers on breast cancer risks. METHODS: Three groups of families were studied to determine the effects of a family history of prostate cancer on breast cancer risks. Also considered were the effects of a family history of melanoma, colon, lung, ovarian, and endometrial cancers. Included were 422 first-degree female relatives of 114 patients with bilateral breast cancer, 320 first-degree relatives of 88 male patients with breast cancer, and 633 relatives of 186 unselected female patients. For estimating relative risks, observed numbers of breast cancers in relatives were compared with expected numbers based on population incidence data. Multiple logistic-regression also provided odds ratios of relatives affected with breast cancer. RESULTS: A family history of prostate cancer increased the breast cancer risks in each of the groups, compared with families without prostate cancer. Ovarian cancer in a family increased the breast cancer risks only in the bilateral breast cancer group, and endometrial cancer increased the risks in the bilateral and unselected groups. These increases in risk appeared to be the consequence of families with multiple cancers, including those with hereditary breast-ovarian cancers and the cancer family syndrome. A family history of melanoma, lung, or colon cancer did not increase breast cancer risks. CONCLUSIONS: A family history of prostate cancer, as well as endometrial and ovarian cancer, significantly increases the risk of breast cancer.
