Immunocytochemical identification of spore proteins in two microsporidia, with emphasis on extrusion apparatus.

Microsporidia can form small spores with a unique invasive apparatus featuring a long polar tube whose extrusion allows entry of infectious sporoplasm into a host cell. The reactivity of mouse polyclonal antibodies raised against sporal proteins from two microsporidian species belonging to different genera (Glugea atherinae and Encephalitozoon cuniculi) was studied by western blotting and indirect immunofluorescence. Whole protein antisera provided a few cross-reactions relatable to some proteins of the spore envelope or polar tube. Ultrastructural immunocytochemistry with murine antibodies against protein bands separated by sodium dodecylsulphate polyacrylamide gel electrophoresis allowed the assignment of several proteins to the polar tube (34, 75 and 170 kDa in Glugea, 35, 55 and 150 kDa in Encephalitozoon). Antigenic similarities were detected for the Glugea 34 kDa and Encephalitozoon 35 kDa polar tube proteins. Species-specific proteins were shown to be located in either the lamellar polaroplast of Glugea or the spore envelope of Encephalitozoon.

DNA polymorphisms associated with the factor VIII:C gene in the Portuguese population.

We have determined the allele frequencies of seven restriction fragment length polymorphisms within or close to factor VIII:C gene in the Portuguese population. The allele frequency of all studied intra- and extragenic biallelic polymorphisms does not differ significantly from those found in other European or Asian populations. On the contrary, the distribution of the different alleles of the TaqI RFLP at the DXS52 locus revealed similarity only with Algerians. We observed a correspondence between the TaqI and BclI alleles at this locus.

[Antihypertensive effects of intravenous nicardipine in arterial hypertension in the elderly].

In order to evaluate the antihypertensive action of intravenous (i.v.) nicardipine, a calcium channel blocker, we included 28 patients (20 women and 8 men) aged from 71 to 93 years (mean age: 80.4 yrs) poorly controlled by their normal antihypertensive treatment, which had not consisted of a calcium channel blocker. These patients had past histories of a variety of cardiovascular disorders: valve disease (n = 4), disorders of cardiac rhythm (n = 3), paced or unpaced disorders of cardiac conduction (n = 8), and cerebrovascular accident (n = 12). On inclusion, their systolic blood pressure (SBP) was greater than or equal to 180 mmHg and/or their diastolic blood pressure (DBP) greater than or equal to 100 mmHg. Blood pressure recordings (SBP, DBP, MBP) and heart rate (HR) were simultaneously taken every 3 minutes for a period of 140 minutes by an automatic apparatus and a mercury manometer, before and after i.v. administration of nicardipine at 3 increasing dosages, respectively 1.25 mg (20 th min), 2.5 mg (32 th min) and 5 mg (44 th min), each injected over a period of 6 minutes. With a cumulative dose of 8.75 mg nicardipine i.v., the SBP decreased significantly from 192.6 to 138.8 mmHg (p less than 0.001); similarly, the DBP fell from 93.9 to 65.8 mmHg (p less than 0.001) and the MBP from 126.2 to 90.1 mmHg (Hg manometric measurement). In addition, after the final dose of nicardipine, the blood pressure progressively rose to reach levels, by the end of the trial, of 172.1 mmHg (SBP) and 91.1 mmHg (DBP).(ABSTRACT TRUNCATED AT 250 WORDS)

The Bucharest multifactorial prevention trial of coronary heart disease. General methodology and risk factor correction after five year follow-up (1971-1977).

Both the intervention and the control group included each 5000 men aged 40-60 years, randomly selected by their home address in five districts of the city of Bucharest. The intervention group underwent an "at entry" examination for risk factor detection (high serum cholesterol, high blood pressure, overweight, diabetes, minor ECG abnormalities, family history) and subsequently a five-year multifactorial intervention aimed to reduce the risk factors. Both groups were followed up in this lapse of time for major end-points: myocardial infarction, stroke, sudden death. The qualitative analysis of the results used ten evolution indices based on a quantal counting and lead to a classification of risk factors which allowed the setting up of a strategy for their correction. The quantitative analysis showed the following decreases between the first and the last examination in the intervention group: for serum cholesterol greater than or equal to 250 mg/dl -17%; for cigarettes/day greater than or equal to 15-53%; for overweight greater than or equal to 30% - 13.57%; for high blood pressure -8%; for the overall risk computed by multiple regression -33.8%.

Bone injuries due to tetany or convulsions during hemodialysis.

Two patients with long standing uremia sustained bone injuries due to tetany in one, and convulsions secondary to hemodialysis-induced alkalosis and hypomagnesemia in the other. In the former, there were multiple fractures involving the scapula, clavicles and femoral necks. In the latter, there was a slipped capital femoral epiphysis. Caution should be exercised against tetany or convulsive episodes which may cause bone injuries during hemodiaylsis in severely osteoporotic patients. In patients with progressive renal osteodystrophy, prevention of severe osteopenia by timely kidney transplant or subtotal parathyroidectomy is an important preventative measure.

Molecular characterization of an unassigned Israeli Newcastle disease virus isolate.

Detection of Newcastle disease virus (NDV) and avian pathotyping of NDV isolates are extremely important because the appearance of virulent virus has significant economic consequences in terms of vaccination, eradication, and the ability to export poultry products. By using nucleotide and amino acid (aa) homology analysis, we could demonstrate that a NDV broiler isolate is a velogenic virus. This analysis was done after mean death time and intracerebral pathogenicity index tests gave inconsistent results. By establishing a nucleotide sequence dendrogram, we found that the disputed Ber-Tuvia was clearly in the same group as the known Herev-Laet, a velogenic isolate. The difference between Ber-Tuvia 92 and the Herev-Laet velogenic isolate was 6% as opposed to > 16% of the meso- and lentogenic isolates. The Ber-Tuvia isolate contains the Arg/Arg and Lys/Arg aa at positions 112, 113 and 115, 116, respectively, in the fusion protein cleavage aa sequence, which is typical for virulent NDV isolates.
