A cauliflower mosaic virus promoter directs expression of kanamycin resistance in morphogenic transformed plant cells.

The promoter region of the CaMV inclusion body protein gene was modified for use in chimeric gene fusions. The modified promoter was used to construct a selectable marker for plant transformation based on the Tn 5 kanamycin resistance gene. This chimeric selectable marker was introduced into plant cells using oncogenic and deoncogenized strains of Agrobacterium tumefaciens. Both types of transformation produced kanamycin-resistant cell lines. The resistant cell lines derived from the deoncogenized strains were used to regenerate shoots. A second type of selection based on the ability of octopine synthase to detoxify aminoethyl cysteine was also used to select transformants in both oncogenic and nononcogenic transformation.

Quantitative determination of clobazam in serum and urine by gas chromatography, thin layer chromatography and fluorometry.

The procedures available for determination of clobazam (Frisium, Hoechst) are gas chromatography, fluorometry, and thin-layer chromatography. The study presents detailed descriptions of analytical procedures appropriate for routine determinations in serum and urine, and results from human trials. Moreover, the physicochemical properties of clobazam, viz., solubility, distribution, and protein binding are given.

Physico-chemical and anlytical studies of penbutolol.

In the present paper physico-chemical and analytical studies on penbutolol sulfate (Hoe 893d) are reported. In addition to an interpretation of ultraviolet and fluorometric spectra, data are given regarding the dissociation constant, solubility, distribution, and protein binding. The substance and its major metabolte, as well as their glucuronides, are detectable by means of a selective fluormetric method. Moreover, mention is made of the results of a human pharmacokinetic study.

[A new fluorometric method for the determination of drugs in biological material (author's transl)].

A description is given of a new method for fluorometric determination of drugs in biological material. The assay is based on photochemical transformation of non-fluorescent compounds in fluorophors by short-wave UV light. The pharmacokinetics of the 2-[4-(4'-chlorophenoxy)-phenoxy]-propionic acid (HCG 004) serves as an example to demonstrate the usefulness of this method; reference is also made to analogously successful studies of other drugs.

Rapid fluorimetric procedure for the analysis of fendosal in plasma and data following oral dosing.

A simple, rapid, and sensitive procedure is described for the determination of Fendosal in plasma. After extraction of Fendosal from buffered plasma, the drug is determined by measurement of the fluorescence induced by irradiation with short-wave ultra-violet (UV) light. The complete procedure can be completed in less than 2 h; one technician can perform up to 50 assays in one working day. The limit of detection corresponds to 0.1 microgram ml-1 plasma. Drug concentration and induced fluorescence were linearly related over the concentration range 0--8 micrograms ml-1.

Early lead exposure and neonatal jaundice: relation to neurobehavioral performance at 15 years of age.

A cohort of children who attended first grade in 1983 was identified in a Danish community with low-level lead pollution. Two groups with high and low postnatal lead exposure were generated on the basis of the dentin-lead concentration in shed deciduous incisors. At age 8 years, examination of 162 children matched according to gender and socioeconomic status had shown lead-related deficits in verbal intelligence and visuomotor coordination. Re-examination was now carried out in 141 children at age 15 years using the Wechsler Intelligence Scale for Children (WISC), Bender Visual Motor Gestalt, Trail Making, and Visual Gestalts. In general, no lead-related effects could be detected in the group. However, in children with a history of neonatal jaundice, increased lead exposure was associated with mild neurobehavioral deficits, as indicated by lower verbal IQ scores and decreased visuomotor coordination. This finding suggested that moderate neonatal hyperbilirubinemia may have precipitated an increased sensitivity to subsequent exposure to lead.

Isolation and characterization of the human pulmonary surfactant apoprotein gene.

Pulmonary surfactant is a phospholipid-protein complex which serves to lower the surface tension at the air-liquid interface in the alveoli of the mammalian lung and is essential for normal respiration. Inadequate levels of surfactant at birth, a frequent situation in premature infants, results in respiratory failure. In all species examined, surfactant is composed primarily of dipalmitoylphosphatidylcholine and two major protein species of relative molecular mass (Mr) 32,000 (32K) and 10K (refs 2-5). Reconstitution in vitro of purified 32K pulmonary surfactant apoprotein (PSAP) with synthetic lipids forms a lipoprotein complex that lowers surface tension by spreading to create a thin interfacial film. Here we describe the cloning of the human PSAP gene and complementary DNA, and discuss features of the unusual encoded protein.
