[Ribozyme targeted the point-mutation of activated oncogene inhibits its expression in vivo].

We demonstrated previously that the hammerhead ribozyme designed by us against the activated oncogene T24-ras cleaved the target mRNA specifically and efficiently in vitro. To study its properties in vivo, we cloned the DNA fragment encoding the ribozyme into the eukaryotic expression vector pSMG and transfected it into the T24-ras gene transformed NIH3T3 cell lines. The intracellular cleavage of T24-ras mRNA was evaluated on either the cytological level or the molecular level. The malignant cells expressing ribozyme were partially reversed in morphology as evidenced by slower growth speed, partial recovery of contact-inhibition, reduced frequency of colony forming in soft agar and close-to-normal behaviour in agglutination teat. The primer extension experiment verified that the ribozyme had efficiently cleaved the transcripts of T24-ras gene at the target site in vivo.

[Invasive fungal infection in 3447 autopsy cases].

OBJECTIVE: To evaluate the prevalence of invasive fungal infection at autopsy. METHOD: To retrospectively analyse 3447 autopsy cases in Peking Union Medical College Hospital from 1953 to 1993. RESULTS: There were 85 cases of invasive fungal infection at autopsy among 3447 cases. The prevalence steadily increased, especially during the recent 20 years. Among the 85 cases, only 5 patients were diagnosed clinically (5.9%). The primary diseases were mainly leukemia, cancer, and sepsis. Lung was involved in 70% of the cases, and 85% of the pathogens were fungi. CONCLUSIONS: Fungus has become a major pathogen in nosocomial infection. The prophylactic use of antifungal agents is recommended in high-risk patients because of the difficulty to establish the clinical diagnosis of invasive fungal infection.

Changes of distribution of platelet activating factor in the lung of rats with hypoxic pulmonary hypertension.

OBJECTIVE: To investigate the role of platelet activating factor (PAF) in the hypoxic pulmonary hypertension. MATERIAL AND METHODS: Fifteen of 30 male Wistar rats were exposed to hypoxia for 3 weeks, and another 15 rats served as controls. The pulmonary arterial pressure was examined by catheterization. The sections of rat lung were treated by the avidin-biotin-peroxidase complex method to expose the location of PAF. RESULTS: The rats developed pulmonary hypertension and right ventricular hypertrophy after hypoxic exposure. Under the light microscope, PAF is distributed on the vascular and alveolar walls of normal lung, and the content of PAF in the lung of rats with hypoxic pulmonary hypertension are remarkably higher than those of normoxic controls. CONCLUSIONS: PAF plays not only a physiological role in the rat lung, but also a pathophysiologic role in hypoxic pulmonary hypertension.

[The detection and clinical significance of nitric oxide synthetase activity in placenta tissue].

OBJECTIVE: To study the role of nitric oxide (NO) secreted by placenta tissue in pregnancy. METHODS: The activity of nitric oxide synthetase (NOS), the concentration of malondialdehyde (MDA) and the activity of superoxide dismutase (SOD) in placental tissue were measured in 37 normal pregnant women at various gestational ages and in 6 patients with pregnancy induced hypertension (PIH). RESULTS: The activity of NOS decreased with the advancement of pregnancy. It was higher in normal mid and late pregnancy than that in PIH. In normal pregnancy, there was positive correlation between the activity of NOS and the activity of SOD (r = 0.3588, P < 0.05), and birth weight as well (r = 0.4340, P < 0.05). Negative correlations between the activity of NOS and the concentration of MDA (r = 0.4355, P < 0.05), and gestational age (r = -0.4742, P < 0.05) were found. CONCLUSION: NOS secreted by placenta may play an important role in regulating the process of pregnancy and the pathogenesis of PIH.

[Obesity may have more pronounced effect on the occurrence of insulin resistance than hypertension].

OBJECTIVE: To compare the extent of insulin resistance in hypertensive and in obesity patients and verify their role on the occurrence of insulin resistance. METHODS: Insulin sensitivity was assessed by using euglycemic insulin clamp technique (EICT) in normal subjects (NS, n = 36), obesity (n = 23), hypertensive non-obesity patients (HT+nonobesity, n = 51) and hypertensive and obesity patients (HT+obesity, n = 54) Glucose metabolism ratio (M) served as an index of insulin sensitivity. RESULTS: The height sequence of M value was: NS > HT+nonobesity > obesity > HT+obesity (ie, 9.2 +/- 1.0 > 7.6 +/- 2.0 > 6.7 +/- 1.5 > 5.2 +/- 1.8 mg.kg-1.min-1, P < 0.05). The incidence rate of decreased insulin sensitivity among the four groups was: HT+obesity > obesity > HT+nonobesity > NS (87.0%, 69.6%, 41.1% and 2.8%, P < 0.05). After adjusting for age, sex and BMI, M value was greater in normotensives (n = 59) than in hypertension patients (n = 105) (8.0 versus 6.6 mg.kg-1min-1, P < 0.01). Multivariates analysis showed that BMI and SBP are the two most important determinants for M value, but the effect of 1 kg/m2 BMI is equivalent to that of 20 mmHg SBP. CONCLUSION: Decreased insulin sensitivity may be the common linkage between obesity and hypertension. Obesity may have more pronounced effect on insulin sensitivity than hypertension.

[Effects of selective nitric oxide synthase inhibitor in sheep with endotoxic shock].

OBJECTIVE: To investigate the effects of S-methylisothiourea sulfate (SMT), a selective inducible nitric oxide synthase (iNOS) inhibitor, on hyperdynamic endotoxic shock sheep. METHODS: Endotoxic shock was induced by Escherichia coli endotoxin in both control (n = 8) and SMT groups (n = 8). SMT was given intravenously. Hemodynamic data, oxygen delivery derived parameters and intramucosal pH (pHi) were measured. RESULTS: The control group had a hyperdynamic state, similar to that of human septic shock. In the SMT group, blood pressure was maintained at baseline, and cardiac index (CI) was lower than that in the control group (P < 0.05). Oxygen extraction ratio (O2 ext) was increased up to 40% +/- 5% and was much higher than that of the control group (P < 0.01). Pulmonary artery pressure (PAP) was higher than that of the control group (P < 0.01), and pHi decreased gradually similarly to the control group. CONCLUSION: SMT restored the blood pressure and increased O2 extespecially in the gut, but decreased CI and oxygen delivery and increased PAP. So over inhibition of iNOS should be cautiously considered.

[Radiation therapy of esophageal carcinoma: a report on 180 cases].

One hundred eighty patients with pathologically proved esophageal carcinoma accepted for surgery, for various resons, were given radical radiation therapy. The length of the lesions ranged from 2 to 9 cm. They were treated either by the isocenter technique alone or A-P plus isocenter irradiation to a total dose of 50-70 Gy/5-7 wk. The 1-, 3- and 5-year survival rate was 64.4%, 34.4% and 23.3%, respectively. The 3- and 5-year survival rates showed that lesions in the upper third responded to radiotherapy better than did those in the middle and lower thirds (P < 0.05). The 5-year survival of patients with upper third lesions treated with radiation alone (44.5%) was slightly better than that of surgery. The result of lesions in the middle third was slightly inferior, and that of lower third was even more inferior to that of surgery. The result of radiation alone for operable esophageal carcinoma was comparable to that of surgery.

[Effect of electro-acupuncture and transcutaneous electric nerve stimulation on experimental diabetes and its neuropathy].

In this paper, the experimental diabetic rats induced by streptozotocin (STZ, i.p. 50 mg/kg) were divided into three groups, electro-acupuncture (EA group, n = 8), transcutaneous electric nerve stimulation (TENS group, n = 8), at bilateral Shenshu and Zusanli points for 20 minutes once every 2-3 days for five weeks, and without any treatment (DM group, n = 6) respectively. As compared with the DM group, the increased plasma glucose levels was lowered significantly in EA group (P < 0.05) and slightly in TENS group (P > 0.05) by the end of the sixth week. And the symtoms of polyphagia, polydipsia and polyuria were attenuated in EA group. The motor nerve conduction velocity slowing was prevented or corrected after a course of four and six weeks treatment in EA and TENS group respectively. In the DM and TENS groups the pain threshold was lowered at 6-20 days after injection of STZ, but elevated in EA group, their differences were of significance (P < 0.05). In general, the efficiency of EA treatment on experimental diabetes and its neuropathy was better than that of TENS therapy.

[The correlation between gastric intramucosal pH and oxygen delivery in patients with septic shock].

We studied the correlation of intramucosal pH (pHi), oxygen delivery (DO2) and oxygen consumption (VO2) in 21 patients with septic shock, and found that pHi decreased dramatically (P < 0.05) in nonsurvivors while it had no significant change in survivors. In survivors, there was a positive correlation between pHi and DO2 (r = 0.71, P < 0.001), pHi and VO2 (r = 0.63, P < 0.001) when DO2 was below the critical level of 641 ml/min/m2. We concluded that the change of pHi is related with the prognosis of septic shock. The correlation between pHi and DO2 proves the oxygen supply dependency of VO2. The hypoxia in tissue level is the main factor which results in a high mortality of septic shock patients. pHi is a reliable and useful parameter not only for prognosis, but also for titrating the therapy in septic shock if pHi is monitored serially together with DO2.

Relationship between plasma membrane Ca(2+)-ATPase activity and acrosome reaction in guinea pig sperm.

The results obtained by biochemical measurement demonstrated for the first time that significant decrease of the plasma membrane Ca(2+)-ATPase activity occurred during capacitation and acrosome reaction of guinea pig sperm. Ethacrynic acid, one kind of Ca(2+)-ATPase antagonists, inhibited the plasma membrane Ca(2+)-ATPase activity, but calmodulin (50 micrograms/mL) and trifluoperazine (200-500 mumol/L) did not, suggesting that calmodulin is not involved in ATP-driven Ca2+ efflux from sperm. However, calmodulin is involved in the control of Ca2+ influx. TFP, one kind of calmodulin antagonists, accelerated the acrosome reaction and Ca2+ uptake into sperm cells significantly. Ca(2+)-ATPase antagonists, quercetin, sodium orthovandate, furosemide and ethacrynic acid promoted the acrosome reaction, but inhibited Ca2+ uptake, which cannot be explained by their inhibitory effects on the plasma membrane Ca(2+)-ATPase activity. It is speculated that this phenomenon might be caused by simultaneous inhibitions of the activities of Ca(2+)-ATPase present in the plasma membrane, the outer acrosome membrane and the outer mitochondrion membrane resulting in Ca2+ accumulation in the cytoplasm, which in turn blocks further Ca2+ entry through some negative feedback mechanism(s). The inhibitory effect of Ca(2+)-ATPase antagonist on glycolytic activity may also be the reason for Ca2+ accumulation in cytoplasm and inhibition of Ca2+ uptake.

[Combined effects of high temperature and carbon monoxide on heat stress protein levels in rat brain and viscera].

Combined effects of high temperature and carbon monoxide (CO) on the amounts of heat stress protein 60 (HSP 60) and 70 (HSP 70) in rat brain and viscera were studied by western dot blot technique. Results showed that (1) amounts of HSP 60 in rat brain, spleen and kidney, as well as its rectal temperature, increased with CO concentrations, while those in heart decreased; and both of them showed synergistic effects on those in spleen and kidney, antagonistic ones in brain and liver, and synergistic ones of lowering them in heart; (2) amounts of HSP 70 changed with CO concentrations in five rat viscera, with synergism in brain, and antagonism in other organs; and (3) amounts of HSP 60 and 70 in most rat viscera and their body temperature were the most sensitive to 693.6 mg/m3 of CO.

[A study on plasma non-species specific antibody in employees working in a automobile engine testing workshop].

To investigate the existence of the non-species specific antibody in plasma of the employees working in an automobile engine testing workshop, and to use it as a scanning marker of various hazards, the heat-stress protein antigen method and western blot technique were used. This study showed that employees working in the automoblile engine testing workshop were affected by various hazards, such as noise, toxic chemicals (carbon monoxide, lead fume, benzene, and so on), and there existed non-species specific antibodies against protein 103,900 and 54,200 of rat liver in their plasma, which were postulated as the specific products produced by exposure to occupational hazards, such as noise, carbon monoxide, et al.

[Diagnostic significance of CT in the orbital inflammatory lesions].

OBJECTIVE: To analyze and summarize the CT signs of orbital inflammatory lesions. METHOD: HITACH-W, or EXEL 2400 pattern of CT was applied for the orbital scanning of 52 cases with orbital inflammatory lesions proved by clinicopathology. RESULTS: The CT signs of orbital inflammatory lesions in our series included exophthalmos, orbital mass, ill-marginated mass, multiple space involvement, extraocular muscle enlargement, scleral thickening, eyelid thickening, optic nerve thickening, orbital fat density increasing, lacrimal gland enlargement, sinusitis, subperiosteal abscess, ring-enhancement in masses, etc. CONCLUSION: All these findings on CT are of important significance for the diagnosis of orbital inflammatory lesions.

[Chemical constituents of Aster albscens Hand.-Mazz].

Four compounds, stigmasterol, D-hurulon-14-en-3-ol, stigma-delta7.22-dien-3beta-palmitate and friedelin were isolated from the root of Aster albscens for the first time. Their structures were elucidated by spectral methods.

[Protective effects of radix Salviae miltiorrhizae on azathioprine hepatotoxicity in rats].

The changes of liver function and malondialdehyde (MDA) and glutathione (GSH) were studied in rats intoxicated with azathioprine (Aza) 15mg/(kg.d) and in rats treated with Aza mixed Salviae 8g/(kg.d). The results showed that in the Aza group, the levels of ALT, AKP, MDA increased and GSH decreased significantly in week 1 and week 2. But in the Salviae group the levels of ALT, AKP, MDA decreased and GSH increased significantly, approaching the normal levels, It follows that Salviae has protective effects on Aza hepatotoxicity in rats.

Signal transduction pathways in guinea pig sperm.

Trifluoperazine (TFP), the antagonist of calmodulin (CaM), significantly stimulated the capacitation and acrosome reaction of guinea pig spermatozoa at the concentration of 10-100 mumol/L, independent of the external Ca2+. Forskolin, dbcAMP and caffeine evidently promoted the occurrence of acrosome reaction of spermatozoa at early capacitation stage (5 h) in nonsynchronous system but not in synchronous system. If the spermatozoa were capacitated for 15 h in synchronous system, the above three drugs significantly stimulated acrosome reaction in a Ca(2+)-independent manner. Protein kinase C activators, i.e. phorbol 12-myristate 13-acetate (PMA) and phorbol 12,13-dibutyrate (PDB) did not influence the occurrence of acrosome reaction of spermatozoa at early capacitation stage, but significantly increased the acrosome reaction rate in capacitated spermatozoa in a Ca(2+)-independent manner. In contrast, PKC inhibitor staurosporine significantly inhibited the occurrence of acrosome reaction.

The mouse bone morphogenetic protein-4 gene. Analysis of promoter utilization in fetal rat calvarial osteoblasts and regulation by COUP-TFI orphan receptor.

Bone morphogenetic protein-4 (BMP-4) is one of a member of related polypeptides that are important in bone formation and other developmental processes. We isolated the BMP-4 gene from a mouse genomic library and characterized the exon-intron structure and one of the candidate promoters. Two alternative 5'-noncoding exons, 1A and 1B, were identified by reverse transcription polymerase chain reaction assays. Quantitative competitive polymerase chain reaction using Exon 1A, Exon 1B, and Exon 3 primers indicate the 1A-containing transcript is the primary BMP-4 mRNA expressed in bone cell cultures. Primer extension analysis supports that 1A is the major promoter utilized in bone cell cultures as well as in 9.5-day mouse embryos. 1A promoter activity indicate selective DNA regions functional in bone cells. We found potential regulatory response regions in the 1A 5'-flanking region of the BMP-4 gene for the chicken ovalbumin upstream-transcription factor I (COUP-TFI). Specific binding to the COUP-TFI response regions in the BMP-4 1A promoter was demonstrated. By co-transfection of a COUP-TFI expression plasmid with the BMP-4 1A promoter in fetal rat calvarial osteoblasts, we demonstrated that COUP-TFI inhibits the BMP-4 promoter activity. This suggests that COUP-TFI could act as a silencer for BMP-4 transcription in vivo.
