Rotavirus RNA replication requires a single-stranded 3' end for efficient minus-strand synthesis.

The segmented double-stranded (ds) RNA genome of the rotaviruses is replicated asymmetrically, with viral mRNA serving as the template for the synthesis of minus-strand RNA. Previous studies with cell-free replication systems have shown that the highly conserved termini of rotavirus gene 8 and 9 mRNAs contain cis-acting signals that promote the synthesis of dsRNA. Based on the location of the cis-acting signals and computer modeling of their secondary structure, the ends of the gene 8 or 9 mRNAs are proposed to interact in cis to form a modified panhandle structure that promotes the synthesis of dsRNA. In this structure, the last 11 to 12 nucleotides of the RNA, including the cis-acting signal that is essential for RNA replication, extend as a single-stranded tail from the panhandled region, and the 5' untranslated region folds to form a stem-loop motif. To understand the importance of the predicted secondary structure in minus-strand synthesis, mutations were introduced into viral RNAs which affected the 3' tail and the 5' stem-loop. Analysis of the RNAs with a cell-free replication system showed that, in contrast to mutations which altered the structure of the 5' stem-loop, mutations which caused complete or near-complete complementarity between the 5' end and the 3' tail significantly inhibited (>/=10-fold) minus-strand synthesis. Likewise, incubation of wild-type RNAs with oligonucleotides which were complementary to the 3' tail inhibited replication. Despite their replication-defective phenotype, mutant RNAs with complementary 5' and 3' termini were shown to competitively interfere with the replication of wild-type mRNA and to bind the viral RNA polymerase VP1 as efficiently as wild-type RNA. These results indicate that the single-strand nature of the 3' end of rotavirus mRNA is essential for efficient dsRNA synthesis and that the specific binding of the RNA polymerase to the mRNA template is required but not sufficient for the synthesis of minus-strand RNA.

Interaction of glycogen synthase kinase 3beta with the DF3/MUC1 carcinoma-associated antigen and beta-catenin.

The DF3/MUC1 mucin-like glycoprotein is highly overexpressed in human carcinomas. Recent studies have demonstrated that the cytoplasmic domain of MUC1 interacts with beta-catenin. Here we show that MUC1 associates with glycogen synthase kinase 3beta (GSK3beta). GSK3beta binds directly to an STDRSPYE site in MUC1 and phosphorylates the serine adjacent to proline. Phosphorylation of MUC1 by GSK3beta decreases binding of MUC1 to beta-catenin in vitro and in vivo. GSK3beta-mediated phosphorylation of MUC1 had no apparent effect on beta-catenin levels or the transcriptional coactivation function of beta-catenin. The results, however, demonstrate that MUC1 expression decreases binding of beta-catenin to the E-cadherin cell adhesion molecule. Negative regulation of the beta-catenin-MUC1 interaction by GSK3beta is associated with restoration of the complex between beta-catenin and E-cadherin. These findings indicate that GSK3beta decreases the interaction of MUC1 with beta-catenin and that overexpression of MUC1 in the absence of GSK3beta activity inhibits formation of the E-cadherin-beta-catenin complex.

Early captopril treatment prevents hypertrophydependent gene expression in hearts of SHR.

Treatment of spontaneously hypertensive rats (SHR) with captopril (100 mg . kg-1 . day-1) throughout development and during the first 16 wk of life leads to a reduction in blood pressure and left ventricular hypertrophy. Blood pressures and hypertrophy are reduced in these animals (vs. untreated SHR) for up to 24 wk after discontinuation of the drug. We used conventional blot hybridization and Western analysis to examine hypertrophy-dependent gene expression during this period. Ventricular expression of the atrial natriuretic peptide gene was reduced by >90% at 16 wk of age in the captopril-treated SHR. Expression increased in the 24 wk after discontinuation of treatment, but remained well below that of the untreated SHR. A similar reduction in ventricular c-myc gene expression was seen with captopril treatment. Neither renal expression of the atrial natriuretic peptide gene nor ventricular expression of the c-fos gene was affected by captopril. This study demonstrates that captopril treatment during a critical period of development in the SHR leads to a sustained reduction in hypertrophy-dependent myocardial gene expression, which does not revert to levels seen in the untreated SHR after discontinuation of the drug.

Estrogen receptor activation function 1 works by binding p160 coactivator proteins.

Estrogen receptor-alpha contains two transactivation functions, a weak constitutive activation function (AF-1) and a hormone-dependent activation function (AF-2). AF-2 works by recruiting a large coactivator complex, composed of one or more p160s, CREB-binding protein (CBP)/p300, and P/CAF (p300 and CBP-associated factor), via direct contacts with the p160s. We report here that independent AF-1 activity also requires p160 contacts. Unlike AF-2, which binds signature NR boxes in the center of the p160 molecule, AF-1 binds to sequences near the p160 C terminus. We propose that the ability of AF-1 and AF-2 to interact with separate surfaces of the same coactivator is important for the ability of these transactivation functions to synergize.

Evaluation of a safety educational programme for the prevention of pesticide poisoning.

The causal factors of pesticide poisoning in spraymen usually include sloppy handling, leakage of sprayers and a lack of personal protection, as well as of a knowledge about pesticide toxicity. This paper outlines the results of a safety educational programme to prevent pesticide poisoning among Chinese village spraymen in 1991 and 1992. The programme consisted of two parts: an interview of 3,286 trained subjects who were surveyed before and after the education courses, and a survey on the prevalence of pesticide poisoning among spraymen in 10 villages in 1991 and 1992, respectively. The results showed a general improvement in the knowledge about pesticide toxicity and safe use among the surveyed subjects. The prevalence of pesticide poisoning among the surveyed spraymen decreased from 1.05% in 1991 to 0.25% in 1992. Moreover, a 68.2% reduction of pesticide poisoning cases was observed in 10 villages in 1992. The success of the safety educational programme proved that education is an effective measure for preventing pesticide poisoning.

A study on continuous low-flow perfusion with low-potassium dextran for donor isolated lung preservation.

OBJECTIVE: To test the validity of continuous low-flow perfusion with low-potassium dextran (LPD) to preserve rabbit lung. METHODS: Isolated rabbit lungs were preserved for eight hours either in Ringer's solution by simple storage (Group I) or in continuous low-flow perfusion with LPD (Group II). After preservation, lung functions were assessed to compare these two methods. RESULTS: The water gain in Group I was higher than that in Group II. During reperfusion, the functional test values for the immersed lungs were lower than those for the perfused lungs. The lipid peroxidation product (MDA) was significantly decreased in perfused lungs during reperfusion. CONCLUSIONS: Low-flow perfusion with LPD is better than immersion for the lung preservation.

Microencapsulation of rat islets prolongs xenograft survival in diabetic mice.

OBJECTIVE: To protect the transplanted islets from the host's immune system by means of immunoexclusion membranes. METHODS: Rat islets were isolated from Wistar rat pancreas by ductal collagenase distention, stationary digestion, and finally with the aid of dextran gradient separation. Then the islets were encapsulated in alginate-polylysine-alginate (APA) semipermeable membranes. RESULTS: In vitro studies demonstrated that encapsulated islets secreted insulin in response to glucose challenge for at least 8 weeks, which was similar to free islets. In vivo studies showed that 15 streptozotocin (STZ)-induced diabetic mice were transplanted intraperitoneally with 1000 encapsulated islets without immunosuppression. Diabetes was reversed within 3 days, and the mice remained normoglycemic for up to 160 days, with a mean xenograft survival time of 126 days. The encapsulated islets had a significantly greater effect than unencapsulated islets, which functioned for less than 8 days. CONCLUSIONS: Encapsulation of pancreatic islets in semipermeable membranes can effectively prolong xenograft survival without immunosuppression in an animal model.

A clinical trial of tang shen ning for treatment of diabetic nephropathy.

This paper reports the clinical trial of Tang Shen Ning ([symbol: see text], TSN) for treating diabetic nephropathy (incipient and clinical, as divided by Mogensen). The results showed that the total effective rate in treatment group (TSN + western medicine) was 90.0%, and that in the control group (simply with western medicine), 56.7%. TSN plays important roles in decreasing proteinuria and improving renal functions.

Studies on dynamics of growth and biosynthesis of artemisinin in hairy roots of Artemisia annua L.

Seven hairy root lines with the properties of fast growth and high artemisinin contents were selected from 747 hairy roots induced by transformation of Artemisia annua L. strain 025 with Agrobacterium rhizogenes ATCC15834. The differences of growth rates and artemisinin contents among the 7 selected hairy root lines were extremely significant, of which HR-9 gave the highest yield of artemisinin, reaching 33.25 mg/y.L. The differences of growth rates and artemisinin contents among hairy roots, untransformated roots and callus were also significant. There were no obvious lag phase in batch culture of hairy roots of Artemisia annua L. The exponential growth phase was 7 approximately 15 days after inculation. The growth rate reached the highest on the 11th day, and the cultures reached a stationary phase on the 20th day. The properties of artemisinin content of hairy roots were obviously "related to growth". The artemisinin content decreased slowly during the exponential phase, increased while the growth rate slowed down and remained consistent after the growth stopped. The optimum culture time for hairy roots of Artemisia annua L. was 21 days in our system.

[Pathologic grading and differential diagnosis on oligodendroglioma].

UNLABELLED: To investigate the histological characteristics of oligodendroglioma (ODG), 65 cases of ODG were observed with light microscopy and partially with immunohistochemistry. According to WHO classification of tumors, 65 cases of ODG were graded in degree of differentiation of tumor cells. Of these, 19 cases belonged to grade I, 32 grade II, and 14 grade III. CONCLUSION: Except the degree of differentiation, some other criterlons as cellular density, mitotic activities, and the presence of focally concentrated apoptotic cells are closely related to grading of ODG.

[Rapid microdose 14C-urea breath test for detection of Helicobacter pylori infection].

The purpose of this study was to develop and evaluate a rapid microdose 14C-urea breath test (14C-UBT) with a simplified protocol for detecting the infection of hilicobacter pylori (HP). 157 fasting patients who underwent endoscopy with histological examination and rapid urease test (RUT) were given a drink of 37 kBq of 14C-urea. Samples of breath carbon dioxide (1 mmol) were collected at baseline and 10, 20 and 30 min after administration by trapping in hyamine solution. 14C activity was measured by liquid scintillation counting. Results were expressed as cpm. Histolal examination and RUT were used as gold standard for the detection of HP infection. The cutoff value was selected as 200 cpm at 10 min. The results showed that the sensitivity, specificity, positive predictive value, negative predictive value and accuracy were 94.79%, 90.16%, 93.81%, 91.60% and 92.99% respectively. In this study, a 10 min, single sample, 37 kBq 14C-urea breath test for detection HP was developed. The test has good diagnostic accuracy with minimal radiation exposure and low cost. Thus, the authors considered the test to be reliable, safe, convenient and cost-effective to clinical use.

[The expression of integrin beta 3 in cycling and early pregnant endometrium and its relationship with primary unexplained infertility].

OBJECTIVES: To profile the changes in integrin beta 3 expression in cycling and early pregnant endometrium and its relationship with unexplained infertility. METHOD: Expression of integrin beta 3 on endometrium was detected in 56 normal cycling women during different menstrual phases or early pregnant women, and 9 cases with unexplained infertility during mid-luteal phase by immunohistochemistry technique. RESULTS: Integrin beta 3 was expressed only in the midluteal or pregnant endometrium(after cycle day 19). Compared with normal women, integrin beta 3 failed to express in the midluteal endometrium from women with unexplained infertility. CONCLUSIONS: Our findings suggest integrin beta 3 may play important roles in the process of implantation and decidualization. It might be a new reliable marker of uterine receptivity. Abnormal endometrial integrin beta 3 expression might be the major cause of unexplained infertility.

[Analysis of critically ill patients with bacteremia].

OBJECTIVE: To study the epidemiology of bacteremia of critically ill patients and the mortality associated with bacteremia and risk factors for death in an intensive care unit. METHOD: 75 patients with 116 episodes of bacteremia were retrospectively studied. RESULTS: The mortality rate of the patients was 43%. Staphylococci (29%), Enterococci (12%), Class I inducible beta-lactamase-producing Enterobacter (12%), extensive spectrum beta-lactamase-producing Enterobacter (10%) were the most common pathogens, among which gram-positive organisms constitute a major part. The most commonly affected organs were respiratory (77%), hepatic (53%), circulatory (53%), gastrointestinal (50%), renal (47%), central nervous (36%), and hemopoietic systems (27%). Multiple organ failure accounted for 76%. Univariate analysis revealed that the severity of underlying illness, multiple organ failure (MOF), septic shock, hepatic failure, renal failure and infection focus were the risk factors for in-hospital death (P < 0.05). Cox proportional hazard model analysis suggested central nervous system failure, septic shock, hemopoietic failure, hepatic failure and respiratory manipulation significantly affected the survival time of patients with bacteremia (P < 0.05). CONCLUSION: Gram-positive organisms are major pathogens of bacteremia in critically ill patients. Antibiotic therapy can not prevent the occurence of bacteremia, nor can it improve the prognosis.

[The effect of amlodipine, nifedipine and perindopril on insulin sensitivity and blood lipid of patients with essential hypertension].

OBJECTIVE: To investigate the effect of amlodipine, nifedipine and perindopril on insulin sensitivity(IS) and blood lipid of patients with essential hypertension(EH). METHODS: 105 EH patients were randomly divided into 3 groups: amlodipine group; nifedipine group; perindopril group. Treatment period lasted for 4 weeks. Before and after treatment, IS was measured by euglycemic insulin clamp technique with glucose metabolism rate (M) as an index of IS. RESULTS: Amlodipine, nifedipine and perindopril significantly reduced blood pressure compared with that before treatment(P < 0.01). In the EH patients with decreased IS, the M value (mg.kg-1.min-1) after the use of amlodipine, nifedipine and perindopril was higer than that before treatment (amlodipine: 6.6 +/- 1.5 Vs 4.6 +/- 0.6, P < 0.01; nifedipine: 5.2 +/- 1.2 Vs 4.4 +/- 0.6, P < 0.05; perindopril: 6.8 +/- 1.6 Vs 4.4 +/- 0.6, P < 0.01). The increase of M value(mg.kg-1.min-1), nifedipine (1.2 +/- 0.9) were significantly lower than that of amlodipine(2.1 +/- 1.1) and perindopril(2.4 +/- 1.5), P < 0.01, respectively, but no significant difference was found between amlodipine and perindopril(P > 0.05). CONCLUSION: Amlodipine and perindopril have beneficial effect on improving insulin sensitivity in EH patients with decreased insulin sensitivity. The effect of nifedipine on insulin sensitivity is lower than that of amlodipine and perindopril. Blood lipid is not significantly changed after short treatment.

[Pathological analysis of 180 cases of cerebrovascular disorder in autopsy].

OBJECTIVE: To evaluate the ratio of each kind of causes in 180 cases of cerebrovascular disorder(CVD) in autopsy. RESULTS: In 180 cases of CVD in autopsy: (1)125 (69%) belong to brain hemorrhage including hypertension 36 cases(20%), vascular malformation 28 cases(16%), hemopoitic diseases and blood coagulation disturbances 23 cases(13%), congenital aneurysm and brain tumor 9 cases(5%) each separately. (2) 55 cases (31%) are due to brain infarction, main of them are caused by embolism 29 cases (16%) and thrombosis 17 cases(10%). Thrombosis related to atherosclerosis has 5 cases(3%). CONCLUSION: In CVD in autopsy, brain hemorrhage is more than infarction, the ratio of them is 2.3:1. More than 50% of cases is due to hypertension and vascular malformation. In infarction, embolism is more than thrombosis, the ratio of them is 1.7:1. Vascular malformation is more than congenital aneurysm, the ratio of them is 3:1.

Effect of splenectomy on CCl4-induced liver fibrosis in rats.

OBJECTIVE: To assess the effects of splenectomy on CCl4-induced liver fibrosis in rats. METHODS: Wistar rats were injected with CCl4 subcutaneously for 3 to 15 weeks to develop liver fibrosis. Two hundred and thirty-six rats were divided into 9 groups: (1) model group without splenectomy; (2) splenectomized group in which operation was performed before induction of fibrosis; (3) sham-operated group in which caparotomy was done before induction of fibrosis; (4) (5) (6) groups in which splenectomy was performed at the 6th, 9th and 12th week after induction of fibrosis, respectively; and (7) (8) (9) sham operation was also performed at the 6th, 9th and 12th week after induction of fibrosis, respectively. The spleen and liver biopsy, hepatic hydroxyproline, liver homogenate levels of procollagen type III (PC III), hyaluronic acid (HA), laminin (LN) and volume density of collagen were examined at different times. RESULTS: The results showed that the degree of hepatic damage and liver fibrosis in splenectomized group was significantly milder than that in model group and sham-operated groups. Splenectomy might delay the progress of liver fibrosis for about 3 weeks. The role of preventing rat liver fibrosis was also observed when splenectomy was performed after CCl4 induction of fibrosis for 6 and 9 weeks. CONCLUSIONS: These results suggest that splenectomy may be of a preventive role against CCl4-induced rat liver fibrosis to a certain degree.

[Enhancement of Vp16 inducing apoptosis of leukemic cells by retrovirus-mediated bcl-2 anti-sense RNA].

OBJECTIVE: To explore the effects of retrovirus-mediated bcl-2 antisense RNA on apoptosis of leukemic cells. METHODS: Retrovirus was packaged in vitro with PA317 cells and Jurkat cell was transducted with collected virus; The expressions of bcl-2 mRNA and protein were assayed by RT-PCR and Western blotting, respectively. Apoptosis was assayed by flow cytometry and DNA "ladder". RESULTS: Expression of intrinsic bcl-2 was decreased, and the sensitivity of leukemic cells to Vp16 and the apoptosis of leukemic cells line Jurkat cells were enhanced by transfected bcl-2 antisense RNA. CONCLUSION: Antisense bcl-2 enhances Vp16 inducing apoptosis of leukemic cells. The results provide a useful experimental basis for leukemia therapy.
