Studies with timed-pregnant squirrel monkeys (Saimiri sciureus).

Timed-pregnant monkeys were produced in a large nonhabituated colony of Saimiri sciureus of Bolivian origin. In a colony of 373 females and 40 males, 277 females (74%) were considered to be inseminated, based on microscopic observation of sperm and/or detection of a coagulum (plug) in the vagina. Forty-six full-term progeny were delivered. The mean gestational period was 152.5 days (SD = 3.9 days). For continuous cohabitation, the median time to insemination was five days, with 75% of inseminations occurring within eight days. Pregnancy evaluation (mouse bioassay) indicated a high level of resorptions within the first 50 days of gestation. This may help explain the low birth rates reported for other nonhaibuated colonies.

Pregnancy diagnosis in squirrel monkeys: hemagglutination test, radioimmunoassay, and bioassay of chorionic gonadotropin.

The Nonhuman Primate Pregnancy Test may be useful for diagnosis of pregnancy in squirrel monkeys between 40 and 60 days in pregnancy. However, single determinations have an inherent 10% risk of false negative responses caused by low chorionic gonadotropin levels; thus, initial negative test responses should be followed within one week by an independent confirmatory test. Preliminary results with this hemagglutination inhibition test compare favorably with bioassay and may be useful, in conjunction with conventional uterine palpation, for diagnosis of pregnancy in squirrel monkeys.

Reproductive counseling in patients who have had a spontaneous abortion.

A detailed morphologic study was made of spontaneously aborted embryos and fetuses from 472 patients. These patients were followed through 638 subsequent pregnancies. The patients were divided into four groups, depending on their previous obstetric history. The outcome of the subsequent pregnancies was correlated with the previous obstetric history of the patient and the developmental status of the aborted specimen. Patients who had never had a live child in the previous obstetric history were identified as a high-risk group. Patients who had had live births and pregnancy loss previously were found to be at significant risk of a subsequent premature infant following the late abortion of a fetus.

A chronic gastrostomy and test system for evaluation of gastric secretions in rhesus monkeys.

We have developed a test system for the evaluation of gastric secretion in the rhesus monkey on a chronic basis that does not require the usual restraining techniques. The system is characterized by ease of animal maintenance and handling, reliability in introducing test substances into the stomach, and long term postsurgical stability. The system comprises a surgical preparation whereby the gastrostomy tube is inserted permanently in the stomach, is passed subcutaneously up the back, and is exteriorized at the top of the skull, and a training procedure that conditions the monkeys so as to facilitate easy handling during the administration of test substances and the collection of gastric controls. Values for pH, volume, total acid, and acid concentration appear reasonably consistent under base-line (no test substance) conditions as well as after the intragastric administration of water or the subcutaneous injection of Histalog.

Down syndrome in British Columbia, 1952-73: incidence and mean maternal age.

Records of children with Down syndrome (DS) at the BC Health Surveillance Registry were linked to their Birth Registrations to derive maternal ages. Incidence and maternal-age specific rates were calculated for 1952-73. Mean maternal age has declined both for normal and DS children, the latter to a marked degree, so that in 1972-73 80% were born to women under 35 years. Using maternal age of 40 and over as an indication for amniocentesis would only detect 10% of DS children. The crude incidence rate (mean 1.28/1000 livebirths) has not changed appreciably over the study period except for 1969 in which a statistically significant peak occurred. The standarized rate showed an increasing trend but it is not clear whether this was a true biological increase or resulted from better ascertainment.

Quantitative analysis of individual bile acids by gas-liquid chromatography: an improved method.

The quantitative analysis of individual bile acids by gas-liquid chromatography has been improved by column oven temperature programming and by a new liquid phase, SP-2401. The method is fast; bile acids are well resolved; retention times are reproducible; detector responses are linear and sensitive to 0.1 mug: and there is little adsorption onto the liquid phase. The method has been successfully used for bile, and it has the potential for use on serum.
