Human lymphocyte affinity for sheep erythrocytes in young and aged healthy subjects.

The kinetics of sheep erythrocyte rosette formation by human peripheral blood lymphocytes from young-adult and aged individuals were studied in 20 paired experiments. No difference in the rate and saturation plateau of formation could be found despite a slight increase in early rosette-forming cells. We conclude that the membrane affinity for sheep erythrocytes is unaffected by ageing and that the proportion of the circulating early E-RFC subpopulation is increased in healthy older subjects.

Circulating immune complexes in various thyroid diseases.

In a study of 171 patients with various thyroid diseases, circulating immune complexes (CIC), measured by a C1q solid phase radioassay, were detected in 26% of the patients as compared to 8% of the control subjects. CIC were found in 33--55% of the patients with a well defined thyroid autoimmune disorder (Hashimoto's goitre, asymptomatic thyroiditis, spontaneous myxoedema and Graves' disease) and also in the same proportion of patients with diffuse goitre. CIC were correlated to the presence of serum antibodies to microsomal thyroid antigen but not to their titre. No relationship was observed between CIC and the age or sex of the patients and the presence of exophthalmos, or between CIC and the different thyroid function tests or serum anti-thyroglobulin antibodies. CIC were found in untreated patients as well as in those treated with prednisone, methimazole or thyroxine.

Stability of E-rosettes in aged humans: effect of cytochalasin B and colchicine.

A greater stability of E-rosettes and a reduced rate of capping of sheep erythrocytes are observed in elderly people. The lack of qualitative changes in the sensitivity of these processes to colchicine and cytochalasin B suggests that the function of the microfilaments and the microtubules is not primarily affected by ageing. The density and the affinity of the lymphocytes for sheep erythrocytes do not decrease in aged subjects. So the defective rearrangement of the E-receptors cannot be accounted for by some receptor alteration but points to a possible hindrance of their cross-linking in the membrane or their submembrane connection to the cytoskeleton.

Levamisole adds granulocyte toxicity to 5FU-based chemotherapies in adjuvant treatment of Dukes B-C colorectal cancer. A preliminary report.

41 patients (pilot study-I) and 50 patients (multicenter study II) were randomized to receive as systemic chemotherapy for 6 courses with 5 FU alone (A) [440 (I)-450 (II) mg/m2 IV bolus, 5/21 days] or folinic acid followed by 5 FU (B) (respectively 200 and 370 mg/m2 IV bolus, 5/21 days). In the multicenter trial, oral levamisole at the dose of 150 mg/day (3/14 days) was added to chemotherapy for one year. Ten patients in study I and 19 patients in study II also received a post-operative course of intra-portal chemotherapy. Toxicity was evaluated respectively on 232 (I) and 276 (II) courses. Clinical limiting toxicities were stomatitis and diarrhea. In protocol II, a significant enhancement of grades 3-4 granulocyte toxicity was seen (17.3% of courses in II vs only 3.4% in I; p < 0.001). This was especially recorded in the group treated with 5-FU alone (26% of courses in A vs 11% in B; p < 0.001). Levamisole was therefore stopped in 12 cases (10 cases in A; 2 cases in B).
