Relationship of auscultatory fourth heart sound to the quantitated left atrial filling fraction.

The fourth heart sound (S4) has been associated with vigorous atrial contraction. However, the presence of S4 has not been previously correlated with quantitated left atrial filling fraction. In this study, the presence of an auscultatory S4 was compared with the Doppler echocardiographically quantitated left atrial filling fraction in 41 consecutive patients in whom S4 was judged to be present or absent according to the consensus of cardiologists. Left atrial filling fraction in patients with S4 was 49% and was significantly greater than 33% found in patients without S4 (p less than 0.005). Using atrial filling fraction of 35% as a dividing line, S4 had a sensitivity of 84% and a specificity of 75% for determining left atrial filling fraction greater than or equal to 35%. Furthermore, of 25 patients with S4, 21 (84%) had atrial filling fraction greater than or equal to or less than 35%, and of 16 patients without S4, 12 (75%) had atrial filling fraction less than 35%. Thus, the presence or the absence of S4 is quantitatively related to the left atrial filling fraction and appears to predict atrial filling fraction greater than or equal to or less than 35% reasonably well.

Design, synthesis, antineoplastic activity, and chemical properties of bis(carbamate) derivatives of 4,5-bis(hydroxymethyl)imidazole.

A series of bis(carbamate) derivatives of 1,2-substituted 4,5-bis(hydroxymethyl)imidazoles were prepared and evaluated against murine P388 lymphocytic leukemia. Electron-withdrawing substituents at either N-1 or C-2 gave rise to inactive compounds. However, electron-donating substituents gave active compounds and the 2-(methylthio)-1-methyl derivative 2i (carmethizole), as the bis(N-methylcarbamate), was found to be very active. The derivative 2i, referred to by the name carmethizole, was also shown to be active against the MX-1 mammary xenograft, the human amelanotic melanoma cell line (LOX) xenograft, the M5076 sarcoma, and L1210 lymphocytic leukemia. The solution stability, water solubility, pKa, and log P of carmethizole are also reported.

Liquid membrane phenomenon in diuretics.

Surface activity and critical micelle concentrations are reported for two diuretic drugs, furosemide and triamterene. The drugs generate a liquid membrane on a supporting membrane. Transport of chloride, sodium, and potassium ions through the liquid membranes generated by the drugs was studied. The data suggest that the phenomenon of liquid membrane formation may contribute to the diuretic action.

Cytotoxicity of ethyl methanesulfonate in mice spermatogonia.

Enumeration of different types of spermatogonia, following a single i.p. administration of different doses of ethyl methanesulfonate in mice, showed a survival of A1-A4 and in spermatogonia is markedly reduced due to cell killing while the remaining types of spermatogonia were affected marginally. The cell killing effect was dose-dependent, and replenishment of these cells was observed by the end of one cycle of the seminiferous epithelium comprising of 8.5 days.

Effects on the spermatogonia of mice following treatment with sodium bisulfite.

The effect of sodium bisulfite on differentiating spermatogonia has been investigated. Adult mice were given either a single intraperitoneal injection (500, 600, 700, 800, 900 and 1000 mg/kg body weight) or repeated intraperitoneal injections (200 and 400 mg/kg body weight) of sodium bisulfite. In the latter case, the doses were administered 20, 30 and 40 times during 28, 42 and 56 days respectively. Different types of spermatogonia were enumerated from paraffin sections of tests stained with periodic acid--Schiff and Ehrlich's hematoxylin. No mortality was observed up to 700 mg/kg dose within 24 hours. At 1000 mg/kg dose 80% of the mice died within 24 hours post treatment. Cytotoxicity data showed that sodium bisulfite, at any of the dosage level tested after acute or repeated administration did not alter the population of various types of spermatogonia.
