The characterization of cDNA clones coding for wheat storage proteins.

Poly(A)+ RNA isolated from the developing wheat endosperm var. Chinese Spring, has been used as template for the construction of a cDNA library. Within the library, clones have been identified by in vitro translation of hybrid-selected mRNA which encode alpha/beta gliadin related sequences and gamma-gliadin related sequences. The DNA sequence of one such clone has been determined and it shows homology with that of a clone encoding a barley storage protein, B-hordein. The sequence includes a tandem DNA repeat which is discussed in relation to the generation of diversity within the gliadins.

[Continuous intra-arterial perfusion of prostaglandin E1 in inoperable stage-IV arteriopathies].

Inoperable stage IV arteriopathies have been treated by intra-arterial PGE1 infusion since January 1978. Inoperability was established in all cases by angiography or by attempted exploration and angiography during operation. Strict criteria for therapy were applied with the result that treatment was administered to only 100 patients among 3,335 cases undergoing vascular surgery. Dosage was 0.1 to 0.2 ng of PGE1 per kilo of body weight and per minute, mean duration of treatment being 34 days. Of the 100 limb extremities probably requiring wide amputations, 47 were saved from immediate amputation, the majority of patients in whom amputation could not be avoided being diabetic. Follow-up in July 1982, 1 to 4 years after treatment, showed that 24 extremities were still intact. Best results were noted in patients with endangiitis obliterans, and long-term results no longer demonstrated differences in patients with or without diabetic arteriopathy. Intra-arterial therapy can be repeated if necessary.

Conservative treatment of inoperable arterial occlusions of the lower extremities with intra-arterial prostaglandin E1.

Prostaglandin E1 has a strong vasodilator effect, it inhibits the aggregation of platelets and improves the microcirculation. The hormone is synthesized from arachidonic acid and 80-90 per cent is eliminated in the first transit of the pulmonary circulation. From 1 January 1978 to 30 June 1981 100 patients suffering from inoperable occlusion of the arteries of the lower limbs and facing amputation were treated with intra-arterial PGE1 by prolonged perfusion over an average period of 34 days. PGE1 0.1-0.2 ng kg-1 body weight min-1 was administered. Forty-seven of 100 legs were saved; 22 of 29 patients suffering from diabetes requiring insulin had to undergo amputation, but only 1 of 18 patients suffering from thromboangiitis obliterans had to have an amputation. The results of treatment in the 52 cases of uncomplicated arteriosclerosis lie in between, with 18 amputations. The complications that followed treatment were haemorrhage, arterial thrombosis, infection and mycotic aneurysms.

[Use of computer technic in cardiotocographic childbirth monitoring].

Reference is made in this paper to cardiotocographic monitoring of labour by the principle of selective alarm recording, using trend memory scope, TMS-2, and a modified cardiotocographic recorder, FTS-101.--Upper or lower limits of individually preselected thresholds of foetal heart rate as well as upper limits of amplitude, frequency, basal tonus, and Montevideo units of uterine contractions were chosen as criteria by which to signal alarm. High-economy monitoring and savings on recording paper are some of the advantages provided by the system. Automatic alarm function as well as cardiotocographic recording twelve minutes prior to, during, and any time after an alarm signal proved to be sufficient for proper documentation of findings.

Molecular cloning and analysis of cDNA sequences derived from poly A+ RNA from barley endosperm: identification of B hordein related clones.

A collection of over 130 cDNA clones has been constructed in the bacterial plasmids pPH207 and pBR322 using as template the poly A+ RNA from membrane-bound polysomes of barley endosperm (cv. Sundance). Fifty four B hordein cDNA clones have been identified by cross-hybridization analysis and in vitro translation of plasmid-selected mRNAs. Hybridization of 11 of the B hordein cDNA clones to Northern blots of size-fractionated RNA indicated that the B hordein mRNA is ca. 1300 nucleotides long. One cDNA clone, pHvE-c16, has been partially sequenced and shown by comparison with C-terminal and other peptide sequences to be related to B1 hordein polypeptides. The results obtained from the analysis of the B hordein cDNA clones support the idea that the Hor 2 locus, which specifies the B hordeins, is complex and codes for a family of related mRNA species.

[Hemodynamic parameters determined by impedance cardiography in hyperthyroidism].

The impedance-cardiography renders possible the recognition of central haemodynamic parameters on an unbloody way. For the judgment of the method 18 female patients with hyperthyreosis were examined before and during the thyreostatic treatment. At the same time the systolic time intervals were estimated and the results from the two methods were compared. Apart from the abbreviation of PEP and the decrease of the contractility index PEP/LVET the increase of the cardiac index and the reduction of the resistance index indicate the thyreotoxic effect on the cardiovascular system.

The expression of a plant genome in hnRNA and mRNA.

Representation of genomic kinetic sequence classes and sequence complexities were investigated in nuclear and polysomal RNA of the higher plant Petroselinum sativum (parsley). Two different methods indicated that most if not all polysomal poly(A) -RNA is transcribed from unique sequences. As measured by saturation hybridization in root callus and young leaves 8.7% and 6.2%, respectively, of unique DNA were transcribed in mRNA corresponding to 13.700 and 10.000 average sized genes. Unique nuclear DNA hybridized with an excess of polysomal poly(A)mRNA to the same extent as with total polysomal RNA. 3H-cDNA - poly(A)mRNA hybridization kinetics revealed the presence of two abundance classes with 9.200 and about 30 different mRNAs in leaves and two abundance classes with 10.500 and 960 different mRNAs in callus cells. The existence of plant poly(A)hnRNA was proven both by its fast kinetics of appearance, its length distribution larger than mRNA, and its sequence complexity a few times that of polysomal RNA.

[Cardiotocography -- proposal of system solution (author's transl)].

A system solution with new technical details and new demands on space distribution is proposed for the use of antenatal cardiotocography and cardiotocographic monitoring of labour. Bedside equipment will be reduced and widely shifted to a centralised control room by introduction of the system. All installations are of high flexibility to leave room for future developments in medical engineering and for necessary services. -- Large-scale use of the new technique with capacity utilisation of the hardware involved will be ensured by appropriate upgrading of labour room personnel as well as by techno-organisational improvement of work and minimisation of expenses on diagnosis and monitoring. The measures proposed work according to expectation and lead to good perinatological results.

The role of the head nurse in primary nursing.

Primary nursing brings about changing roles, responsibilities, and communication patterns for all members of the health care team. The head nurse, as she introduces new staff to primary nursing and its day-to-day applications, is in an ideal position to generate enthusiasm for the goals of the concept. In attempting to implement this new care pattern, she must continually promote the philosophy behind it by her support and recognition of its principles. In the long run, it is the positive attitude and high motivation of the head nurse that will determine the success or failure of the program wherever it is introduced.

[Surgical indications and results of thoracic sympathectomy].

Between the 1st of July 1971 and the 31st of December 1974, among 1,459 operations on vascular surgery, we carried out 77 thoracic sympathectomies in 46 patients. In most patients there was an oblitering angiopathie of digital type, stage II to IV, confirmed by angiography. Two patients had mixed type obstructions, two others had hyperhidrosis, one patient suffered from cleroderma, another had a thoracic outlet syndrome with digital arterial obstruction. In all cases, the transaxillary approach was that which caused the least complications and which permitted satisfactory exposure of the sympathetic nerve. The cosmetic results were good. The second and third thoracic ganglia were resected and the planes of cleavage marked with silver clips. In advanced stage III and stage IV, we noted 92 p. 100 improvements i.e. return to stages I or II. The operative mortality was nil. The most serious complication was a case of hemothorax which required later pulmonary decortication. We noted in two cases, a Horner's syndrome.
