Fatty acid fingerprints were determined gas chromatographically for Strepcococcus mutans NCTC 1082 which had been grown in batch brain heart infusion at a series of nine temperatures ranging from 29.0 to 40.0 degrees C. The major acids at all temperatures were n-palmitic and octadecenoic acids. Other acids detected at all temperatures included n-myristic, palmitoleic, n-stearic, and eicosenoic acids. An increase in temperature resulted in a decrease in the proportion of unsaturated to saturated fatty acids, indicating the importance of accurate temperature control in such gas-liquid chromatographic, chemotaxonomic studies.

Streptococcus mutans NCTC 10832 was grown on synthetic medium in a chemostat, using various major carbon sources, viz, sorbitol, fructose and sucrose. Freeze-dried cells were methylated and the methyl esters analysed by GLC. The fatty acid profiles obtained showed small quantitative change with different sugars, sucrose concentrations and types of growth.
The acetoin and diacetyl contents of culture supernatants of Voges-Proskauer-positive "viridans" streptotocci, Klebsiella pneumoniae and Staphylococcus aureus, were determined by a gas liquid chromatographic procedure, in which supernatants were extracted with diethyl ether and diacetyl was measured on columns of 10% (wt/wt) polyethylene glycol 400 (PEG 400) at 73 C. Acetoin was converted to diacetyl, before analysis, by a simple oxidation procedure with ferric chloride and without a distillation step. Streptococcal culture supernatants were shown by this method to contain only acetoin; supernatants of K. pneumoniae and S. aureus contained both acetoin and diacetyl.
