Class II MHC molecules are spontaneously internalized in acidic endosomes by activated B cells.

The antibody response to protein antigens requires specific cooperation between B and T cells. In order to deliver the helper signal, T cells must recognize, in the context of Class II MHC, processed antigen on the membrane of B cells. Processed antigen is in the form of peptides bound in a given site of the Class II MHC molecule; in order to address the question of where, in the B cell, the complex of Class II MHC and processed antigen is formed, we studied the subcellular localization of these two molecules. Since the formation of this complex is the crucial step in antigen processing and presentation, the answer to this question is central to the whole problem of the physiology of antigen handling by B cells. To collect information pertinent to the question, we have compared, in B cells, the intracellular traffic of Class II MHC and of monovalent and divalent anti-immunoglobulin antibodies used as protein ligands of the membrane immunoglobulins. We have done so by two-color immunofluorescence microscopy, and we have detected extensive confluence of Class II MHC molecules with the immunoglobulin ligand, both mono- and bi-valent, in the endosomes of LPS-activated murine B cells. Whereas the ligand clearly reaches the endosomes by internalization from the cell membrane, the Class II MHC molecules could reach the same location either by endocytosis from the membrane or through targeting to the endosomes of newly synthesized Class II MHC molecules. We have collected quantitative evidence for endocytosis of Class II MHC by following, with the fluorescence activated cell sorter, the quenching of the fluorescence of fluoresceinated Fab' anti Class II MHC in LPS-activated murine B cells; this quenching indicates the entry of the label into an acidic intracellular compartment. Together with the results of others, obtained with different methods, our observations support the concept that, at least in mature activated B cells, Class II MHC molecules reach the organelles where they meet processed protein antigens, mainly through the endocytic route. Since activated B cells endocytose their membrane Class II MHC, and not their membrane Class I, our results contribute to the understanding of how B cells present antigens, that have bound to their membrane immunoglobulins, to Class II-restricted helper T cells and not to Class I-restricted cytolytic T cells.

Differential endocytosis of IgM and IgD in murine B cell lines.

The internalization of surface immunoglobulin (Ig) by B lymphocytes is the first step in the antigen-presenting function performed by these cells. Mature B cells coexpress on their surface IgM and IgD. At this time, there is controversy over whether these two isotypes serve different functions in the antigen-presenting process. The results presented here show that the intracellular pattern of distribution of IgM and IgD after internalization is strikingly different in the B cell lines studied. These findings support the hypothesis that the role of the two Ig classes in the antigen-presenting function may be different.

Studies of a new fatty acid analog (DMIVN) in hypertensive rats and the effect of verapamil using ARG microimaging.

Studies of myocardial utilization of fatty acids and analogs has focused on coronary heart disease. This study addresses the topic of radioiodinated fatty acid utilization in hypertensive-cardiomyopathy. The new fatty acid analog 19-iodo-3,3-dimethyl-18 nonadecenoic acid (DMIVN) was studied by autoradiographic microimaging (ARG) in salt-sensitive (S) hypertensive (salt-fed) and in salt-sensitive (S) normotensive (low-salt diet), Dahl-strain rats. A salt fed, S-strain group was treated with verapamil and the results were compared to those in a hypertensive, non-treated group. The distribution of DMIVN in the hearts of normotensive rats was uniform. In the myocardium of hypertensive rats nonuniform DMIVN concentration was seen in the subendocardial and mid-layers of the left ventricle (LV). Verapamil given to salt-fed rats prevented hypertension and uniform DMIVN uptake similar to normotensive controls was seen. The data suggest that DMIVN may be suitable for the detection of hypertension induced myocardial changes and for assessing therapy. The distribution and clearance characteristics of DMIVN indicate that DMIVN may be a useful agent for SPECT imaging in man.

Options in camera technology for the bone scan: role of SPECT.

Planar imaging with current generation gamma cameras and tomographic imaging with current generation rotating camera single photon emission computed tomography (SPECT) systems provide excellent spatial resolution and good fidelity between image quantitation and activity levels. Their use in imaging the localization and clearance of skeletal imaging agents such as the Tc-99m labeled diphosphonates, Ga-67 citrate, and In-111 leukocytes offers a sensitive and accurate means to detect occult disease and to investigate skeletal blood flow, metabolism, and pathology. SPECT progressively plays an increasingly important role in imaging studies of joint disease, infection, avascular necrosis, bone growth, developmental abnormalities and the effects of weight bearing, exercise, and other forms of treatment on skeletal repair processes.

Tn4201, a beta-lactamase transposon in Staphylococcus aureus.

We report the preliminary characterization of a 6.7-kilobase transposon, Tn4201, encoding resistance to penicillin associated with the conjugative plasmid pCRG1600 in Staphylococcus aureus. Tn4201 is capable of inverting in orientation (frequently observed in derivatives of pCRG1600) and is expressed equally well in either orientation. The element is also capable of movement to alternative plasmid and chromosomal sites. Tn4201 exhibits rec-independent transposition in an apparent site-specific manner and may play an important role in the resistance of staphylococci to beta-lactam compounds.

Technetium Tc 99m plasmin in the diagnosis of inflammatory disease.

The localization characteristics of technetium Tc 99m plasmin were studied in experimental animals to investigate the use of 99mTc-plasmin for imaging inflammatory processes. At various times after abscess induction using turpentine in rats, the in vivo distribution properties of 99mTc-plasmin, gallium citrate Ga 67, 125I-fibrinogen, and 99mTc-human serum albumin (HSA) were studied by gamma-camera imaging. The in vivo binding of each radiopharmaceutical was also tested in rat and human plasma clots. Region-of-interest analyses of gamma-camera images showed relatively poor 99mTc-plasmin localization at sites of abscess formation. The ratio of abscess-to-control activity of this radiopharmaceutical did not exceed that of 67Ga, 125I-fibrinogen, or 99mTc-HSA. In vitro assays of each of the radiopharmaceuticals in plasma clots showed 99mTc-plasmin and 125I-fibrinogen to have the best localization characteristics.

Chemical breakdown of technetium-99m DTPA during nebulization.

Aerosols of 99mTc diethylenetriaminepentaacetic acid ([99mTc]DTPA) used for measuring lung permeability and lung ventilation require a radioaerosol delivery system to produce an aerosol with reproducible size and radiochemical purity. To test how well nebulizers meet this requirement, radiochemical purity of aerosols produced with a jet and an ultrasonic nebulizer was evaluated. The activity median aerodynamic diameter (AMAD) and geometric standard deviation (sigma g) of radioaerosols were 0.46 micron (sigma g = 1.6) for the jet nebulizer and 0.70 micron (sigma g = 1.7) for the ultrasonic nebulizer. Paper and liquid chromatographic assays were obtained on the [99mTc]DTPA aerosol solute produced with each nebulizer. The results of these tests showed major differences in radiochemical purity. Aerosols produced in the jet nebulizer consistently showed greater than 90% of the radioactivity bound to the DTPA ligand whereas aerosols produced in the ultrasonic nebulizer showed less than 10% of the radioactivity bound to DTPA. The results support the need to test radiochemical purity of aerosols before using an aerosol nebulizer for pulmonary imaging and clearance studies.

Effects of ACTH on seizure susceptibility in the developing brain.

Adrenocorticotropic hormone (ACTH) has been frequently used as an anticonvulsant drug in some childhood seizure disorders. Despite its widespread use, few studies have evaluated the effects of ACTH on seizure susceptibility in the developing animal or the long-term consequences of ACTH treatment on brain development. In this study, ACTH was given either for 2 days or 14 days prior to kindling in 15-, 22-, and 30-day-old rats. Morphological changes in the brain were studied using routine light microscopy and dendrite branch counting following Golgi staining. Both acute and chronic ACTH treatment inhibited the rate of kindling in all three age groups. There were no differences in brain morphology between the controls and the ACTH-treated rats killed shortly after kindling. Rats treated with ACTH and killed as adults, however, had significantly more dendrite branches than did controls. In the immature brain, ACTH treatment reduces seizure susceptibility and has no long-term deleterious effects on neuronal growth.

Segregant-defective heterokaryons of Candida albicans.

Heterokaryons (hets) of the asexual, pathogenic yeast Candida albicans obtained by fusing protoplasts of complementing auxotrophic strains generate large numbers of parental-type auxotrophic monokaryons by random assortment of single nuclei into blastospores, and smaller numbers of monokaryons bearing hybrid nuclei formed through either karyogamy or the transfer of genetic material from one het nucleus to another. Het populations grown at 30 degrees C or 37 degrees C contain high frequencies (approx. 5%-10%) of two kinds of stable variants peculiar specifically for segregation of parental-type monokaryons: NS variants produce inviable auxotrophic monokaryons of one or both parental classes while AT variants yield parental-type monokaryons which grow very slowly. Variant frequencies are not affected by the wild-type strain background of hets, or the auxotrophies used to force heterokaryosis. However, both kinds of variants are induced by growth at 25 degrees C or by treatments with certain chemical or physical metabolic inhibitors. Evidence is presented that variant nuclei of independent origins carry different nutritionally irreparable recessive lethal (NS) or debilitating (AT) defects acquired in the course of actual or potential internuclear transfers of genetic material within het cells. The high incidence of variants, therefore, indicates considerable intrinsic genetic instability among het nuclei. Significances of these observations for parasexual genetic analyses of C. albicans and other yeasts through protoplast fusions are considered.

Bronchial and alveolar absorption of inhaled 99mTc-DTPA.

The clearance rate of 99mTc-DTPA deposited in the lung by inhalation has been used as an index to measure lung epithelial permeability. To determine if differences exist between alveolar and bronchial absorption of 99mTc-DTPA we measured regional clearance rates in 4 beagle dogs for 30 min after preferential bronchial and alveolar deposition. Alveolar deposition was maximized by inhalation for 2 min of small 99mTc-DTPA particles (activity median aerodynamic diameter, AMAD = 0.5 micron; geometric standard deviation, GSD = 1.6) with deep slow ventilation (VT = 350 ml; f = 9 min-1), and bronchial deposition was increased by inhalation of large particles (AMAD = 4.1 microns, GSD = 2.3) with rapid shallow ventilation (VT = 50 ml; f = 65 min-1). Respective clearance rates from basal regions, which represent mainly alveolar absorption, were: for small particles, 2.29% min-1; for large particles, 1.57% min-1 (p = 0.10). Apical regions, which contain relatively more bronchial surface than do the basal regions, showed the following clearance rates: for small particles, 1.76% min-1; for large particles, 1.31% min-1 (p less than 0.05). These results indicate that in vivo alveolar absorption of 99mTc-DTPA is more rapid than bronchial absorption. Control or verification of the site of deposition of the tracer in the lung is of importance for the interpretation of the results of the 99mTc-DTPA lung permeability assay.

Effects of hypoxic-ischemic encephalopathies on kindling in the immature brain.

The effects of hypoxia and ischemia on subsequent kindling in the immature animal were assessed using three experimental designs. In the first experiment, 15- and 30-day-old rats were exposed to either 15 or 30 min of 6% oxygen and then subsequently kindled. In the second experiment, 15- and 30-day-old rats underwent unilateral carotid ligation followed by kindling in the ipsilateral amygdala. In the third experiment, the Levine procedure was used (unilateral carotid ligation followed by exposure to 6% oxygen) and rats were subsequently kindled in the amygdala either ipsilateral or contralateral to the side of the carotid ligation. Hypoxia and carotid ligation alone failed to result in any significant difference from controls in rate of kindling. In rats that underwent the Levine procedure and were kindled in the amygdala ipsilateral to the side of the carotid ligation, there was a marked inhibition of kindling. These results suggest that although mild hypoxia or ischemia did not alter seizure susceptibility, moderate to severe hypoxia-ischemia inhibited the development of generalized seizures. The mechanism of this inhibition is not known.

The effect of progesterone on kindling: a developmental study.

Clinical epileptologists since Gowers have known that hormones may affect central nervous system excitability. Progesterone has been noted to have anticonvulsant properties in both clinical and animal studies. The effect of progesterone on central nervous excitability in the immature animal, however, has not been studied. This study evaluated the effects of progesterone on the rate of kindling in adult and immature animals. While progesterone has no effect on kindling in the adult animal, in the immature animal progesterone markedly inhibited kindling, preventing generalization of seizures. This study demonstrates that progesterone has a marked age-dependent effect on neuronal excitability.

Relationship of endocrine function to inhibition of kindling.

Both animal and clinical studies have demonstrated that progesterone has antiepileptic properties. Progesterone in the immature, but not mature brain, markedly inhibits kindling. The purpose of this study was to determine if the kindling inhibition effect of progesterone is related to systemic endocrine properties. The effect of alphaxalone and alphadalone acetate, two progesterone-like compounds devoid of significant endocrine effect, were administered to both immature and mature male rats during kindling. Alphaxalone and alphadalone acetate markedly inhibited kindling in the immature rat, but had minimal effects in the mature animal. This study demonstrates that the kindling inhibition effects of progesterone are not dependent on systemic endocrine properties.

Radionuclide skeletal imaging and single photon emission computed tomography in suspected internal derangements of the temporomandibular joint.

Fifty one subjects who had pain involving the temporomandibular joint were evaluated using multidirectional tomography, arthrography, conventional nuclear scanning, and single photon emission computed tomography (SPECT) to assess the association of arthritis of the temporomandibular joint with internal derangements related to meniscal dysfunction. Five (56%) of the nine subjects who had normal arthrograms and normal multidirectional tomograms had SPECT scans that were positive for osseous changes. Twenty-two subjects (27 temporomandibular joints) were diagnosed by arthrography to have meniscal displacement with reduction. Multidirectional tomograms of the 27 joints were positive for osseous changes in five (18%) joints, whereas SPECT scans were positive in nine (70%) joints. Twenty subjects (20 temporomandibular joints) had an arthrographic diagnosis of meniscal displacement without reduction. Multidirectional tomograms of the 20 joints were positive for osseous changes in 14 (70%) joints, and SPECT scans were positive in 16 (80%) joints (P less than 0.001 vs control group). Initial observations with SPECT indicate it is a promising imaging method for detecting and staging osseous disease of the TMJ related to meniscal dysfunction.
