Isolation of a lambda dv plasmid carrying the bacterial gal operon.

A lambdadvgal plasmid carrying genes for controlled plasmid replication from phage lambda and the bacterial gal operon was isolated as a deletion mutant of phage lambdagalq4, which carries the gal operon between lambda genes P and Q. The plasmid DNA was found in cell extracts as covalently closed circular molecules. The plasmid was characterized by using genetic crosses, digestion with the specific endonuclease EcoRI, sucrose gradient centrifugation, and electron microscopy. In one clone analyzed, the plasmid was a complete dimer (O(lambda)P(lambda)galO(lambda)P(lambda)gal); in a subclone derived from it, the plasmid was a partial dimer with only one copy of gal (O(lambda)P(lambda)O(lambda)P(lambda)gal). The partial dimer may be a recombination product of the complete dimer, since test crosses show that the gal and lambda sequences in the plasmid can be separated by recombination. Analyses of the EcoRI digests of plasmid DNAs indicated one cleavage site per lambda gene sequence and none in the gal operon. A lambdadvgal monomer was approximately 6.7 x 10(6) daltons and the lambda gene and gal components were 3.9 x 10(6) and 2.8 x 10(6) daltons, respectively. The lambdadvgal plasmid can be introduced into a new bacterial host by transfection at an efficiency of 10(-6) per DNA molecule.

The modification and generalization of voice loudness in a fifteen-year-old retarded girl.

A fifteen-year-old severely disturbed girl was treated for aphonia. Because of the extent of her withdrawal, the subject was conditioned in a laboratory setting and received tokens for speaking loudly enough to operate a voice-operated relay. Conditioning at first consisted of saying 100 monosyllabic words, with the possibility of reinforcement on each word. Later, the subject was required to say a polysyllabic word, and finally, five or six words per token. The subject was shaped to speak with normal loudness in the laboratory, and generalization to a reading situation in the laboratory was measured and observed to occur, at first for a few minutes, and later for a longer period. Generalization to a reading situation in the classroom did not occur, but the subject's voice loudness also increased in the classroom when several new reinforcement contingencies were put into effect there.

Biochemical method for inserting new genetic information into DNA of Simian Virus 40: circular SV40 DNA molecules containing lambda phage genes and the galactose operon of Escherichia coli.

We have developed methods for covalently joining duplex DNA molecules to one another and have used these techniques to construct circular dimers of SV40 DNA and to insert a DNA segment containing lambda phage genes and the galactose operon of E. coli into SV40 DNA. The method involves: (a) converting circular SV40 DNA to a linear form, (b) adding single-stranded homodeoxypolymeric extensions of defined composition and length to the 3' ends of one of the DNA strands with the enzyme terminal deoxynucleotidyl transferase (c) adding complementary homodeoxypolymeric extensions to the other DNA strand, (d) annealing the two DNA molecules to form a circular duplex structure, and (e) filling the gaps and sealing nicks in this structure with E. coli DNA polymerase and DNA ligase to form a covalently closed-circular DNA molecule.

Imprinting of hippocampal metabolism of choline by its availability during gestation: implications for cholinergic neurotransmission.

Choline supplementation during the second half of the gestational period in rats permanently improves visuospatial memory. Choline availability during this period also alters the turnover of choline and acetylcholine in the hippocampus in 3-4 week-old animals in a complex pattern consistent with the notion that cholinergic neurotransmission is enhanced by prenatal choline supplementation.

Indirect test weighing: a new method for measuring overnight breast milk intakes in the field.

Indirect test weighing (ITW), a new method for measuring nighttime breast milk intake, is based on separate weighings of the mother and infant at the start and end of the nighttime sleep period. All other incidental weight changes overnight are measured, so that the corrected infant's overnight weight gain and the mother's overnight weight loss are then accounted for by breast milk transfer and evaporative water loss (EWL) alone. The combined EWL is partitioned between mother and infant on the basis of metabolic body size (body weight raised to the power of 0.73). The infant's EWL is added onto the corrected overnight weight gain to give nighttime milk intake. A field validation study on 24 northern Thai infants, conducted in the subject's homes, in which ITW was compared with nighttime direct test weighing (DTW) on the same night, gives 95% confidence intervals for the method of +/- 39 g for a range of milk intakes of 77-344 g. Confidence intervals for estimates using feeding patterns alone, or multiplying 12 h intakes by a constant, range from +/- 104 to 180 g, indicating that these methods are insufficiently accurate for individual estimates.
