Synergistic activation by the glutamine-rich domains of human transcription factor Sp1.

We have examined the role of protein-protein interactions in modulating the activity of Sp1, a human transcription factor that utilizes glutamine-rich activation domains. These domains may represent a commonly used structural motif, since a nonhomologous glutamine-rich segment from the Drosophila Antennapedia protein is also a potent activator when fused to the Sp1 DNA binding domain. Sp1 is generally considered a proximal promoter factor that can only stimulate transcription when bound close to the initiation site. However, here we present evidence that distally and proximally bound Sp1 can stimulate transcription synergistically. In addition, a DNA binding-deficient mutant of Sp1 that retains glutamine-rich domains can interact with proximally bound Sp1 to superactivate transcription. Glutaraldehyde cross-linking provides direct evidence for an interaction between Sp1 monomers. Thus, Sp1-Sp1 interactions may play an important role in modulating promoter activity.

In vitro technique for studying garter snake (Thamnophis sp.) development.

Garter snake (Thamnophis sp.) embryos of different embryonic ages were explanted from pregnant females and grown in vitro for up to 35 days. The culture medium consisted of Eagle's Minimum Essential Medium with various organic and inorganic supplements and antibiotics. Cultured embryos were injected with tritiated thymidine, sacrificed, and processed for autoradiographic examination. The in vitro technique allowed direct visualization of external morphologic changes over time, which followed a timetable similar to that of embryos growing in vivo. This technique also allowed easier manipulation of embryos for determination of embryonic stage and for injection of tritiated thymidine. This is the first report of long-term culture of snake embryos which adds to the limited number of reptilian embryos that have been studied in vitro.

Issues in conducting a cancer risk assessment using epidemiologic data: arsenic as a case study.

Using human data to extrapolate cancer risks at ambient levels of airborne pollutants avoids the uncertainty due to interspecies differences, but introduces other uncertainties. The unusually extensive data relating lung cancer to occupational inhalation of arsenic provide examples of 3 uncertainties: the extrapolation from partial to full lifetime risks, from high to low doses, and between populations with differing smoking habits.

Isolation and sequence of a cDNA corresponding to a src-related gene expressed in murine hemopoietic cells.

We have isolated a murine cDNA that shares extensive homology with genes encoding the src (Rous sarcoma virus oncogene)-related family of protein-tyrosine kinases. The cDNA includes an open reading frame of 1509 base pairs, and conceptual translation predicts a protein of 56 kDa. Blot-hybridization analysis indicates that this src-related gene is expressed in normal macrophages and in cell lines representing both the myeloid and lymphoid B-cell lineages and, accordingly, is designated "bmk" (B cell/myeloid kinase). In addition, bmk mRNA levels increase following the induced differentiation of the murine myelomonocytic leukemic cell line WEHI-3B.
