Agricultural land use and ensuing eutrophication both shape parasitic trematode communities in rural African lakes.

Land use is a major driver of biodiversity loss, but how it impacts parasite communities is scarcely documented. Crater lakes and their catchments in rural western Uganda greatly vary in their intensity of anthropogenic disturbance, thus providing an opportunity to assess the effects of land use on snail-borne parasitic trematodes. We applied state-of-the-art molecular biomonitoring to 2385 Bulinus tropicus snails from 34 lakes to detect and genotype trematode infections. The 45 trematode taxa recovered infect a wide range of final vertebrate hosts, and some can cause health burdens of significant public importance. Using constrained ordinations and generalized additive models, we found that B. tropicus reaches peak abundance in lakes with catchments partly under agriculture, whereas trematode infections increase with B. tropicus abundance and peak at intermediate aquatic productivity. Trematode diversity also increases with aquatic productivity, levelling off only in the most productive lakes. These relationships likely reflect the higher abundance and variety of final hosts sustained by more productive lakes. Finally, we found that land use affects trematode community composition, with more livestock parasites and less bird parasites occurring in agricultural catchments. Our results indicate that both land use and lake eutrophication affect the distribution of hotspots for parasitic disease transmission.

DNA: a programmable force sensor.

Direct quantification of biomolecular interaction by single-molecule force spectroscopy has evolved into a powerful tool for materials and life sciences. We introduce an approach in which the unbinding forces required to break intermolecular bonds are measured in a differential format by comparison with a known reference bond (here, a short DNA duplex). In addition to a marked increase in sensitivity and force resolution, which enabled us to resolve single-base pair mismatches, this concept allows for highly specific parallel assays. This option was exploited to overcome cross-reactions of antibodies in a protein biochip application.

Ecological and morphological differentiation among cryptic evolutionary lineages in freshwater limpets of the nominal form-group Ancylus fluviatilis (O.F. Müller, 1774).

The phylogeny and potential mode of speciation of the river limpet Ancylus fluviatilis (Basommatophora) was examined using mitochondrial DNA sequences from 16S ribosomal RNA, cytochrome c oxidase subunit I (COI) and nuclear DNA from internal transcribed spacer (ITS-1) regions from 103 populations across Europe. Four highly divergent lineages were observed within Ancylus. Clade 1, representing the nominal taxon Ancylus fluviatilis (O.F. Müller, 1774), is mainly found in central and northern Europe, Clade 2 is present in a single Portuguese population, Clade 3 is distributed on the Canary islands, North Africa and the eastern Mediterranean region, whereas Clade 4 inhabits the Northern Mediterranean coasts. Phylogenetic analyses revealed an overall consistent topology of nuclear and mitochondrial gene trees. Based on a molecular clock, we estimated that the basic radiation occurred in the late Pliocene. Although clades differ significantly in size independent shell shape, morphological differentiation of lineages is not feasible without genetic data. Environmental data related to climate (precipitation, temperature, etc.) showed a significant differentiation of clades. Clade 1 dwells in relatively colder and more stable habitats than Clades 3 and 4, whose habitats in turn differ in a low or high amount of precipitation during spring and autumn, respectively. Based on the combined data sets on mitochondrial DNA, nuclear DNA, morphological and ecological differentiation, we conclude that Ancylus represents a cryptic species complex of reproductively and genetically isolated lineages. In addition, the joint analysis suggests that ecological speciation is probable to explain current patterns.
