Away from the edge: SAD phasing from the sulfur anomalous signal measured in-house with chromium radiation.

Anomalous scattering with soft X-ray radiation opens new possibilities in phasing for macromolecular crystallography. Anomalous scattering from S atoms collected on an in-house chromium radiation source (lambda = 2.29 A) was used to phase the X-ray diffraction data of thaumatin (22 kDa) and trypsin (24 kDa) crystals. The contribution to the anomalous term, Deltaf" = 1.14 e(-), from sulfur for Cr Kalpha radiation is doubled compared with that for Cu Kalpha radiation, Deltaf" = 0.56 e(-). The direct-methods programs RANTAN or SHELXD successfully found sulfur positions using data sets with resolution limited to 3.5 A. The statistical phasing program SHARP was used to produce the electron-density maps using the sulfur anomalous signal alone at low resolution ( approximately 3.5 A). An interpretable electron-density map for each structure was obtained solely from the phases derived from single-wavelength anomalous dispersion (SAD) data obtained using Cr Kalpha radiation. Much fewer data (that is, lower redundancy) are required for this sulfur SAD phasing procedure compared with the highly redundant data reported in the sulfur SAD phasing procedure with Cu Kalpha radiation. Cr Kalpha radiation can also improve the strength of anomalous scattering of many other intrinsic elements in macromolecules, such as calcium, zinc and phosphorus, because of the increased Deltaf". Furthermore, the anomalous scattering of selenium is increased substantially from 1.14 e(-) with Cu Kalpha radiation to 2.28 e(-) with Cr Kalpha radiation. In order to measure the small Bijvoet differences accurately, several devices were developed for the experiment, including an Osmic Confocal MaxFlux optic optimized for Cr Kalpha radiation, a helium path and a beam stop. In the cases studied here, radiation damage to the samples and reduction of anomalous signal were observed in some long exposure time data sets. Therefore, an adequate data-collection strategy to maximize the completeness in a short scan range was used in subsequent data collections. The results show that the anomalous signal of S atoms can be collected quickly. Since the absorption of solvent and the loop may no longer be negligible with Cr Kalpha radiation, the orientation of the crystal and exposure time were taken into account in order to minimize the effects of radiation damage and absorption. This experimental study shows that using Cr Kalpha radiation from an in-house rotating-anode X-ray generator can provide sufficient phasing power from sulfur anomalous signals to routinely phase protein diffraction data.

Expression of HO-1 in chronic renal insufficiency. Rat kidney and implication.

The expression, activity and clinical implication of heme oxygenase-1 (HO-1) in the chronic renal insufficiency (CRI) rat kidney and its mechanism were investigated. The 5/6 nephrectomized rats were assigned to sham operation group, CRI group and Hemin group. At the 8th week after second operation, blood pressure, urinary protein, serum creatinine (Scr) and BUN were measured. Renal pathologic changes were observed. The activity of HO and contents of erythropoietin (EPO) in serum and renal tissue were determined. Immunohistochemistry was used to detect the expression and distribution of HO-1 in the CRI rat kidney. As compared with CRI group, the urinary protein, blood pressure, Scr and BUN in Hemin group were reduced significantly (P < 0.05). The glomerular mesangial proliferation, inflammatory cellular infiltration of renal interstitium and interstitial fibrosis were ameliorated significantly. Immunohistochemistry and measurement of HO-1 activity revealed that the expression and activity of HO-1 was decreased in renal tissues and increased in serum in CRI group as compared with normal rats. HO-1 distributed mainly in tubular epithelial cells. The EPO contents in Hemin group were significantly higher than in CRI group. Through up-regulating the EPO level in serum and renal tissues, HO-1 retards the progression of CRI.

[Clinical characters and CT findings of steatohepatitis in highland area].

OBJECTIVE: To explore the relationship between the CT findings of steatohepatitis and the special environmental factors in highland area, such as anoxia and life styles. METHODS: CT scan was performed for 64 patients, who were 44 males and 20 females in Tibet with a 3500-meter elevation. RESULTS: Hypodensity in liver parenchyma and the invertion of liver density and spleen density were found in all the 64 patients. According to the degree and scope in their pathological changes, these patients were divided into three groups: severe 14, moderate 26, and mild 24.And diffused 43, sectional 14, regional 7 by CT findings. Among the 44 drinkers, 34 with more than 10 years were diffused by CT scan, the other 10 drinkers under 10 years, diffused 8, and sectional 2. In addition, the 27 steatohepatitis patients were all diffused by CT who were obese and had drinked more than 10 years. CONCLUSION: anoxia and drinking excessively in highland area are relevant to fatty degeneration in liver.

[Tuberculosis of pharynx and larynx: a report of 32 cases].

OBJECTIVE: To evaluate the trends and the clinical changes in tuberculosis of pharynx and larynx. METHODS: The clinical data of 32 patients with tuberculosis of pharynx and larynx from Jan. 1982 to Dec. 2000 in Daping hospital were studied retrospectively. RESULTS: (1) The local manifestations were mainly single lesion that commonly involved the vocal cord (10 cases). (2) The lesions appearances were mainly the proliferation such as mass (11 cases) or granulation(8 cases). (3) anti-tuberculosis is the main treatment, the operation is the second. Twelve patients cured in clinic, six patients received operation and cured without any complications. Fourteen patients condition controlled. CONCLUSION: The classical manifestations with tuberculosis of pharynx and larynx were not exited, the new clinical manifestations were associated with local lesion in nowadays.

[Influence of different temperatures on internalization of epidermal growth factor receptor complexes in Tca-8113 cells].

OBJECTIVE: To investigate the effect of different temperatures on internalization of epidermal growth factor receptor complexes in human tongue squamous carcinoma cells(Tca-8113). METHODS: Human epidermal growth factor r (hEGF) was labeled with 125I according to Chloramine-T method. The quantity of surface binding and internalization of 125I-hEGF in Tca-8113 cells at 4 degrees C and at 37 degrees C was compared respectively with radiobinding assay. RESULTS: At 37 degrees C, the amount of EGFR complexes internalization exceeded that of surface binding after 10 min. At 4 degrees C, the level of interlization maintained very low. CONCLUSION: The study indicated that the surface binding of 125I-hEGF at 4 degrees C was steadier than that at 37 degrees C. The study on phenomenon of EGFR complexes internalization provide the experimental basis for selecting anti-EGFR monoclonal antibody in clinic.

[Transforming the sintered ostrich cancellous bone to multiphasic calcium phosphate ceramic].

OBJECTIVE: To transform the sintered ostrich cancellous bone(ostrich true bone ceramic, OTBC) to multiphasic calcium phosphate ceramic and study its components and characterization. METHODS: The OTBC blocks were soaked in different concentration of sodium pyrophosphate (Na(4)P(2)O(7).H(2)O,NP) solution and heated to 1100 degrees C to transform its constitution from HAP into multiphasic calcium phosphate ceramics. Then the surface configuration of the material was observed by SEM and the following properties of the material:porosity,bending strength,mineral composition and element ratio were analyzed. RESULTS: The prepared material was a kind of porous calcium phosphate ceramic composed of beta-TCP, HAP and NaCaPO(4). With increase of the NP concentration,the content of HAP decreased while beta-TCP and NaCaPO(4) increased. The average bending strength of the material was (1.95+/-0.46)Mpa,the Ca/P element ratio was 1.511 and the average porosity was(60.71+/-6.9)%. The pore size was uneven. CONCLUSION: The OTBC could be transformed into beta-TCP /HAP /NaCaPO(4) multiphasic calcium phosphate ceramic by heating with NP. With characteristic porous structure, the prepared OTBC could become a new type of bone graft.

[The effects of banxia houpu decoction on a chronic mild stress model of depression].

OBJECTIVE: To study the effect of Banxia Houpu Decoction on a chronic mild stress model of depression and investigate the antidepressive mechanism. METHOD: With consumption of a 1% sucrose solution as an index, and by subjecting rats to a variety of mild stressors for a prolonged period of time, a chronic mild stress model was developed. The levels of the blood lipid were measured by blood lipid kits, the natural kill (NK) cell activity in the spleen was measured with the method of the enzyme lactate dehydrogenase (LDH) released, the superoxide dismutase (SOD) activity in red blood cell was assayed by the Autoxidation of Pyrogallol method, the NO synthase (NOS) activity in serum and tissue was measured by NOS kits, and the content of malondialdehyde(MDA) was measured by MDA kits. RESULT: Banxia Houpu Decoction significantly increased the consumption of sucrose solution, increased the level of the high density lipoprotein (HDL-C), decreased the level of the Triglyceride(TG) in serum, enhanced the activity of the NK in spleen, decreased the activity of the SOD in red blood and the activity of the NOS in serum and tissue, and reduced the content of MDA in tissue by effect on lipid Peroxidation in CNS model of depression. CONCLUSION: Banxia Houpu Decoction has antidepressant effect in different ways.

Crystal structures of unligated and CN-ligated Glycera dibranchiata monomer ferric hemoglobin components III and IV.

Erythrocytes of the marine annelid, Glycera dibranchiata, contain a mixture of monomeric and polymeric hemoglobins. There are three major monomer hemoglobin components, II, III, IV (also called GMH2, 3, and 4), that have been highly purified and well characterized. We have now crystallized GMH3 and GMH4 and determined their structures to 1.4-1.8 A resolution. The structures were determined for these two monomer hemoglobins in the oxidized (Fe3+, ferric, or met-) forms in both the unligated and cyanide-ligated states. This work differs from two published, refined structures of a Glycera dibranchiata monomer hemoglobin, which has a sequence that is substantially different from any bona fide major monomer hemoglobins (GMH2, 3, or 4). The high-resolution crystal structures (presented here) and the previous NMR structure of CO-ligated GMH4, provide a basis for interpreting structure/function details of the monomer hemoglobins. These details include: (1) the strong correlation between temperature factor and NMR dynamics for respective protein forms; (2) the unique nature of the HisE7Leu primary sequence substitutions in GMH3 and GMH4 and their impact on cyanide ion binding kinetics; (3) the LeuB10Phe difference between GMH3 and GMH4 and its impact on ligand binding; and (4) elucidation of changes in the structural details of the distal and proximal heme pockets upon cyanide binding.

Effect of calcium phosphate surface coating on bone ingrowth onto porous-surfaced titanium alloy implants in rabbit tibiae.

PURPOSE: The purpose of the present study was to determine whether calcium phosphate coating has a significant impact on bone ingrowth into a porous titanium implant. MATERIALS AND METHODS: Porous-surfaced titanium alloy Ti-6Al-4V implants were prepared with or without the addition of a thin surface layer of calcium phosphate applied by sol-gel coating. Implants were placed into the tibiae of 16 rabbits. Implanted sites were allowed to heal for 2 weeks, after which specimens were retrieved for morphometric assessment using backscatter scanning electron microscopy. RESULTS: The data collected show that there is more extensive ingrowth into the porous regions of the calcium phosphate-coated implants than into the control implants. The weighted average ingrowth for the calcium phosphate-coated implants was 2.01, whereas that for the noncoated implants was 1.49; the difference is statistically significant (P <.01). CONCLUSIONS: The addition of a thin layer of calcium phosphate to these implants appears to promote a more extensive implant-to-bone interface by allowing the neck regions to become intimately ingrown with bone even after only 2 weeks of initial healing.

Molecular dynamics simulations of calcium-free calmodulin in solution.

A 4-ns molecular dynamics simulation of calcium-free calmodulin in solution has been performed, using Ewald summation to treat electrostatic interactions. Our simulation results were mostly consistent with solution experimental studies, including NMR, fluorescence and x-ray scattering. The secondary structures within the N- and C-terminal domains were conserved in the simulation, with trajectory structures similar to the NMR-derived model structure 1CFD. However, the relative orientations of the domains, for which there are no NMR restraints, differed in details between the simulation and the 1CFD model. The most interesting information provided by the simulations is that the dynamics of calcium-free calmodulin in solution is dominated by slow rigid body reorientations of the domains. The interdomain distance fluctuated between 29 and 39 A, and interdomain orientation angle, defined as the pseudo-dihedral formed by the four calcium binding sites, varied between -2 degrees and 108 degrees. Similarly, the domain linker region also exhibited significant fluctuations, with its length varying in the 34-45 A range and its bend angle in the 10-100 degrees range. The simulations are in accord with fluorescence results suggesting that calcium-free calmodulin is more compact and more flexible than the calcium activated form. Surprisingly, quite similar solvent accessibilities of the hydrophobic patches were seen in the calcium-free trajectory described in this work and previously generated calcium-loaded calmodulin simulations. Thus, our simulations suggest a reexamination of the standard model of the structural change of calmodulin upon calcium binding, involving exposure of the hydrophobic patches to solvent.

Molecular dynamics simulations of a calmodulin-peptide complex in solution.

We have performed an 4-ns MD simulation of calmodulin complexed with a target peptide in explicit water, under realistic conditions of constant temperature and pressure, in the presence of a physiological concentration of counterions and using Ewald summation to avoid truncation of long-range electrostatic forces. During the simulation the system tended to perform small fluctuations around a structure similar to, but somewhat looser than the starting crystal structure. The calmodulin-peptide complex was quite rigid and did not exhibit any large amplitude domain motions such as previously seen in apo- and calcium-bound calmodulin. We analyzed the calmodulin-peptide interactions by calculating buried surface areas, CHARMM interaction energies and continuum model interaction free energies. In the trajectory, the protein surface area buried by contact with the peptide is 1373 A(2) approximately evenly divided between the calmodulin N-terminal, C-terminal and central linker regions. A majority of this buried surface, 803 A(2), comes from nonpolar residues, in contrast to the protein as a whole, for which the surface is made up of mostly polar and charged groups. Our continuum calculations indicate that the largest favorable contribution to peptide binding comes from burial of molecular surface upon complex formation. Electrostatic contributions are favorable but smaller in the trajectory structures, and actually unfavorable for binding in the crystal structure. Since nonpolar groups make up most of buried surface of the protein, our calculations suggest that the hydrophobic effect is the main driving force for binding the helical peptide to calmodulin, consistent with thermodynamic analysis of experimental data. Besides the burial of nonpolar surface area, secondary contributions to peptide binding come from burial of polar surface and electrostatic interactions. In the nonpolar interactions a crucial role is played by the nine methionines of calmodulin. In the electrostatic interactions the negatively charged protein residues and positively charged peptide residues play a dominant role.

The influence of preparation conditions on the fluorescence properties of Eu(Sal)(3)Phen.

The influence of the synthesis conditions (method and sequence of adding reagents, reaction temperature, stirring rate) on the luminescence performance of the rare earth ternary complex compound europium-salicylic acid-(1,10-phenanthroline) [Eu(Sal)(3)Phen] was studied. We show that the method and sequence of adding reagents greatly affects the luminescence properties of the products. Within the range of synthesis temperatures (30-70 degrees C), the complex has a better luminescence performance at the lower temperature. However, performance decreased when the temperature decreased to 20 degrees C. Increase of stirring rate led to better performance in the range 200-700 r.p.m. Infrared (IR) spectroscopy, scanning electron microscopy (SEM) and wide-angle X-ray diffraction (WAXD) were utilized to characterize the structure and morphology of Eu(Sal)(3)Phen. The studies demonstrate that the crystallization of the product increased with increasing luminescence of the product. Therefore, it is proposed that increasing the crystallization of the product will enhance its luminescence performance.
