[Study on the inhibition of nude mice transplantation tumor growth by telomerase ribozyme].

OBJECTIVE: To explore whether telomerase ribozyme could inhibit the growth of nude mice transplantation tumor. METHODS: A xenograft human-nude mouse model was constructed. A total of 24 mice were divided into 4 groups, namely, the saline control(Group 1), the blank plasmid control(Group 2), the 20 microg/mouse/day p(XJ-neo-teloRZ)(Group 3) and the 30 microg/mouse/day p(XJ-neo-teloRZ)(Group 4). All plasmids were packaged by lipofectamine and used for subcutaneous, continuous infusions over 14 days. Tumor measurements and observations of animal behavior were recorded daily. On completion of the study, the mice were killed by cervical dislocation and their organs were removed, weighed, and stored in formaldehyde for histological examination. The telomerase activities of tumor tissues of each group were also detected. RESULTS: The telomerase ribozyme effectively inhibited the telomerase activities of tumor tissues and promoted the apoptosis of tumor cells. The in vivo studies showed a significant decrease in tumor size in mice treated with p(XJ-neo-teloRZ) when compared to the mice treated with controls. Furthermore, the in vivo effect of p(XJ-neo-teloRZ) was dose dependent. CONCLUSION: Telomerase ribozyme is a powerful telomerase inhibition agent; probably it could be effective in tumor gene therapy.

[The study of the distribution of Chinese surnames and the diversity of genetic population structure in the Song dynasty].

Surname in China, as a cultural genetic factor, is a very huge and remarkable human population study resource. The transmission feature of surname has been proved to be similar to that of the genetic markers on Y chromosome of males. It, therefore, has been applied in the population genetic studies since 1965's. In the present study, we have analyzed a group of Chinese surname data of the Song dynasty (AD. 960-1179). The distribution of the present surname data is corresponded with the Karlin-McGregor's neutral allele model. The surname abundance alpha and the migration rate parameter v in the Song dynasty in 16 provincial regions reflected degrees of migration and admixture at that time. Surname phylogenetic dendrogram analysis demonstrated that the Han Chinese had been segregated into North and South two parts in Song dynasty of 1000 years ago. Wuyi and Nanling Mountains divided Northern and Southern Han populations into the two major parts of Han Chinese. The results also showed that the surname distribution of Song dynasty 1000 years ago was quite similar to the present model. The 100 high frequency surnames in Song dynasty were well corresponded with the current surname distribution. This phenomenon suggests that Chinese surnames have been steadily transmitted from ancient to the present. The present study improved that the Chinese surname is a very important genetic resource for the studies of structure of the Chinese populations, migration, and affinities among different populations.

[Studies of improving the frequency of wheat transformation by biolistic bombardment] [In Process Citation]

In the present study immature embryos of spring wheat Zhong-60634 were bombarded with gold particles coated with pAHC25 containing Bar and GUS genes. A total of 17 bialaphos-tolerant plants were obtained from 342 immature embryos which were first cultured for 3-4 days and then bombarded and selected. Integration of the introduced genes into the genome of transgenic wheat plants was shown by PCR and Southern analysis and transformation frequency was 0.88%. Furthermore, the present study showed that calli regeneration ability was improved while the embryo culture medium was supplemented with 2 mg/L dicamba instead of 2 mg/L 2,4-D. Meanwhile, in order to keep the calli differentiation ability calli should be transferred to differentiation medium supplemented with 3 mg/L bialaphos from calli selection medium with 5 mg/L bialaphos within a month.

[A study of plasma terminal SC5b-9 complement complex in patients with pregnancy induced hypertension].

OBJECTIVE: To study the relationship between complement SC5b-9 complex and damage of vascular endothelial in pregnancy induced hypertension (PIH). METHODS: Plasma SC5b-9, Serum circulate immunocomplex (CIC), fibronectin (Fn) levels were determined with ELISA in 47 cases of PIH (study group 1) and 15 cases with potential risk factors of PIH (study group 2) and 40 cases of normal pregnancies (control group). RESULTS: SC5b-9 level increased and CIC level decreased significantly in study group 1 and study group 2 than that in control group (P < 0.01). The Fn level increased in moderate and severe PIH cases than that of mild PIH and control group (P < 0.01). The positive rates of SC5b-9 in study group 1 and study group 2 were significantly higher than those of the control group (P < 0.01). The positive rate of Fn in moderate and severe PIH cases significantly higher than those of mild PIH cases and control group (P < 0.01). CONCLUSION: Complement was activated by classical pathway. Terminal SC5b-9 complement complex play an important roles in the pathogenesis of PIH and has close relationship with vascular endthelial damage.

Hepatic radioembolization with Yttrium-90 glass microspheres for treatment of primary liver cancer.

OBJECTIVE: To study the clinical results of hepatic radioembolization with Yttrium-90 (90Y) glass microspheres in the treatment of primary liver cancer. METHODS: Seventeen patients with liver cancer were treated with glass microspheres from August 1996 to May 1998. Hepatic radioembolization with 90Y and lipiodol-ultrafluid was used. Percutaneous port-catheter system (PCS) implantations via femoral artery were performed in 12 patients. RESULTS: In the 17 patients, their mean ratio of absorbed doses between tumor and normal liver was 2.4:1. CT showed a significant reduction in tumor size in 11 of the 17 patients. Average survival was 19.5 months. The indwelling catheters of all the 12 patients were patent and no catheter tip locations were found. CONCLUSIONS: 90Y glass microsphere is one of the best radioisotopes. Not only good responses to the therapy of 90Y glass microspheres can be achieved in patients with metastatic liver cancer, but also in those with primary liver cancer, specially the localized or hypervascular mass. The patients with massive arterioportal shunt should not be limited to this form of radiation therapy. The percutaneous PCS implantation via the femoral artery is a new passageway for the treatment of primary liver cancer with 90Y glass microspheres and other interventional therapy.

Very low and intermediate density lipoprotein fractions from apolipoprotein E gene-knockout mice induce cholesteryl ester accumulation in J774 macrophages.

OBJECTIVE: To investigate the atherogenic role of very low density lipoprotein (VLDL) and intermediate density lipoprotein (IDL) fraction isolated from plasma of apolipoprotein E (apoE)-knockout mice. METHODS: Containing both VLDL and IDL (apoEko-VLDL/IDL), lipoprotein fraction was isolated from plasma of apoE-knockout mice by ultracentrifugation, and its interaction with J774 macrophages was examined together with its physicochemical properties including lipid content, electrophoresis mobility and apolipoprotein components. RESULTS: ApoEko-VLDL/IDL could be actively taken up by J774 macrophages, resulting in marked cholesterol ester accumulation in the cells (55 +/- 9 nmol/mg cell protein), which was a 7-fold increase over the corresponding lever induced by low density lipoprotein and significantly higher than non-loaded group (P < 0.01). CONCLUSION: ApoEko-VLDL/IDL induced significant cholesterol ester accumulation through an apoE independent pathway. This pathway may explain, partly the mechanism of cholesterol deposition in arterial wall in apoE-knockout mice.

[Observation on left ventricular remodeling in acute myocardial infarction].

OBJECTIVE: After an acute myocardial infarction (AMI), there will be regional dilation and global remodeling of the infarcted left ventricle. This study was aimed to observe the features of the ventricular remodeling after AMI. METHODS: Based on the time of onset of the symptoms, 37 patients (pts) with AMI were divided into 4 groups. The time of onset of AMI for group 1 (16 pts), group 2 (7 pts), group 3 (11 pts) and group 4 (3 pts) was < or = 3 hours (hrs), > 3 hrs - < or = 6 hrs, > 6 hrs - < or = 12 hrs and > 12 hrs respectively. The size of the left ventricle in all the patients were analysed with resting myocardial gated SPECT (GSPECT). Tc-99m MIBI was infused at admission and data of GSPECT were collected 2 hours later following the infusion. End-diastolic volume (EDV), end-systolic volume (ESV) and LVEF were shown by the system. Stroke volume (SV) was then calculated. RESULTS: EDV for group 1 to 4 was (98.5 +/- 31.9) ml, (99.4 +/- 55.6) ml, (128.1 +/- 55.1) ml and (140.0 +/- 25.3) ml, respectively. ESV for group 1 to 4 was (57.0 +/- 30.9) ml, (55.1 +/- 40.3) ml, (77.4 +/- 39.8) ml and (81.3 +/- 26.0) ml, respectively. SV for group 1 to 4 was (57.0 +/- 30.9) ml, (55.1 +/- 40.3) ml, (77.4 +/- 39.8) ml and (81.3 +/- 26.0) ml, respectively. LVEF for group 1 to 4 was (44.6 +/- 13.1) ml, (49.7 +/- 13.1) ml, (42.8 +/- 13.5) ml, and (42.7 +/- 8.6) ml, respectively. In pts with anterior infarction, EDV, ESV and SV after 6 hrs of the onset of AMI were greater than those in pts with in 6 hrs of the onset. It was the same in pts with inferior infarction. CONCLUSION: It is suggested that the left ventricular size increased in 3 hrs after AMI. Following the initial dilation of left ventricle, EDV, ESV and SV did not change significantly with in 6 hrs, but then increased gradually after 6 hrs from the onset of the symptoms. The early and significant dilation of left ventricle after AMI may be one of the risk factors of severe cardiac events.

[The association between plasma homocysteine and the number of coronary artery with stenosis].

OBJECTIVE: Previous studies have shown that elevated plasma homocysteine is a risk factor for arterosclerotic disease and is associated with increased severity of carotid-artery stenosis and peripheral arterial disease. In this study, we examine the precise relation of homocysteine to the number of coronary artery with stenosis. METHODS: One hundred and seventeen subjects who underwent coronary angiography from June to October 1998 in our hospital were recruited in this study. Cases (n = 101) with >or= 50% stenosis in at least one of the three coronary vessels were angiographically defined as having coronary heart disease (CHD). While subjects (n = 16) had 0% stenosis in all the three coronary vessels were considered as controls. CHD cases were divided into single-vessel (n = 29), double-vessel (n = 35) and triple-vessel group (n = 37). Plasma homocysteine was measured within 1 week before angiography in all subjects. RESULTS: Mean plasma homocysteine level was significantly higher in coronary patients than in controls [(17.56 +/- 1.14) micromol/L versus (10.59 +/- 0.43) micromol/L, P = 0.02]. Homocysteine level in the triple-vessel group or double-vessel group was significantly higher compared with that in the controls [(19.49 +/- 1.98) micromol/L versus (10.59 +/- 0.43) micromol/L, P = 0.006 and (17.41 +/- 2.03) micromol/L versus (10.59 +/- 0.43) micromol/L, P = 0.036, respectively]. The level of homocysteine in the stingle-vessel group tended to be slightly higher as compared with that in the controls [(15.2 +/- 61.18) micromol/L versus (10.59 +/- 0.43) micromol/L, P = 0.16]. CONCLUSION: Homocysteine level is elevated in patients with coronary heart disease. Higher level of homocysteine is associated with increased number of coronary arteries with stenosis.

[Cellular immune function influence on postoperative patients with hepatocellular carcinoma and the clinical effect of TILs treatment].

OBJECTIVE: To investigate the cellular immune function influence on postoperative patients with hepatocellular carcinoma (HCC) and clinical effect of TILs, rIL-2 and cyclophosphamide treatment. METHODS: Eight postoperative HCC patients were treated with TILs, rIL-2 and cyclophosphamide. Six patients were infused from the hepatic artery, and the other 2 infused from the portal vein. RESULTS: The IL-2 NK activity, Tsubsets, CD4(+)/CD8(+) level in the peripheral blood of all patients increased and the IL-6, TNF level of most patients decreased (P > 0.05, P < 0.01). For 6 - 18 month follow-up after management, 6 patients with radical tumor resection showed no recurrence. One patient having palliative resection showed no remnant tumor enlargement. One patient with increased TNF level for 8 IU died from hepatic failure 8 months after chemotherapy. CONCLUSIONS: The TILs, rIL-2 and cyclophosphamide treatment of postoperative HCC patients is effective in improving antitumorgenic immune function, preventing tumor recurrence, and restraining remnant tumor growth.

[Primary shortening followed by bone lengthening in situ for the treatment of gunshot bone defect].

OBJECTIVE: To study the treatment of gunshot bone defect and the biological basis of primary shortening followed by callus distraction in situ. METHODS: The canine with gunshot bone defect were treated by two methods, primary shortening followed by callus distraction or delayed bone graft. The local blood flow and pathomorphological change were observed and compared with each other. RESULTS: In the experiment, the bone lengthening was stopped at the 56th day with (2.82 +/-e 0.94) cm (19.3%) gain in the group of bone lengthening, and the time of bone healing was much shorter in the group of bone lengthening than in the group of delayed graft. There was no significant difference between the groups in the time of soft tissue healing and local blood flow. CONCLUSIONS: The technique of primary bone lengthening is simple and has no side effect on tissue healing and local blood flow.

[Prevalence of blindness among adults aged 50 years or above in Shunyi county of Beijing].

OBJECTIVE: To estimate the prevalence of blindness among older adults aged >or= 50 years in Shunyi county of Beijing, China. METHODS: Cluster sampling was used in randomly selecting individuals aged >or= 50 years in 28 villages in Shunyi county in fall, 1996. The survey was preceded by a pilot study where operational methods were refined and quality assurance evaluation was carried out. RESULTS: 5,555 individuals were enumerated. The response rate was 91.5%. In this population, the prevalence of blindness was 1.7% defined as pinhole visual acuity < 0.05 in both eyes; or 2.8% were blind, defined as presenting visual acuity < 6/60 in both eyes. Blindness was associated with older age and female gender. The principal cause of blindness in eyes was cataract. CONCLUSIONS: The prevalence of blindness was decreased 35.8% after ten year active eye-care program in the survey area. However, cataract blindness continues as a significant problem among elderly, especially females, in this population sample of rural area.

[The patho-morphology and biological behavior of repeatedly recurrent liposarcoma in retroperitoneum].

OBJECTIVE: To investigate the patho-morphological features and biological behavior of repeatedly recurrent liposarcomas in retroperitoneum as well as the differential diagnosis with other malignant mesenchymal tumors. METHODS: Totally 10 patients accompanied with detailed clinical data. Except HE staining, immunohistochemical assays for S-100 actin, vimentin and myoglobin were also used for 32 specimens. RESULTS: 6/8 cases of the primary and 13/24 cases of the recurrent tumors of this group were diagnosed as sclerosing liposarcoma with spindle cell proliferation arranged in fascicles and whorls. Ten retroperitoneal tumor recurred within 20 months to 10 years, and among them, distant metastasis obtained only in one case. The histologic subtypes of recurrent and metastasized tumors became varied in 5 cases. CONCLUSION: Sclerosing liposarcoma was the most common histological subtype, see in retroperitoneal liposarcoma, local recurrence frequent, but distant metastasis rare. Surgical removal of the recurrent tumors might prolong the survival expectation of these patients.

[Observations on prostate histology in 50 young and middle aged men. Its implication on clinical pathological diagnosis].

OBJECTIVE: To investigate the morphology and the relationship between the prostate and the adjacent structures in order to clarify its significance in histopathological diagnosis. METHODS: By means of light microscopy, and in part by electron microscopy and immunohistochemistry, the histology of 50 normal prostates were observed. RESULTS: The parenchymal component can be divided into four zones, i.e. peripheral zone, central zone, transitional zone and periurethral gland region, each characterized by its own anatomical and histological features. CONCLUSION: This 4 zone division of prostate has more clinical significance than the traditional division of the prostate. A good knowledge of the normal structure of the prostate has great practical significance in pathologic diagnosis.

[Technique of enzyme digestion adding brushing for isolating bronchial epithelial cells].

This study improved the previous techniques of harvesting rabbit bronchial epithelial cells. With 0.05% of trypsin on the epithelial side of trachea and bronchus, a mild digestion was used before a brushing protocol. The cells were identified with immunocytochemistry and electron microscope, both the cell viability and the membrane integrity of cells were evaluated. A comparison analysis of this method with the simple mechanic brushing and the simple digestion method were done. The results showed that, with this method, cells in a high purity and high viability could be obtained and the recovered cell number could be enough for experiments. It can be concluded that the method is useful and effective for isolating bronchial epithelial cells especially for small animals.

[Affection of epidermal growth factor on VIP secretion and VIPR expression in airway epithelial cells].

In order to confirm the essential role of vasoactive intestinal peptide(VIP) on protection of local airway epithelium and investigate the modulation of the protective effect of VIP, secretion of VIP and expression of VIP receptor(VIPR) were studied in bronchial epithelial cells(BEC), and alveolar macrophages (AM). The radioimmunoassay was used for determining VIP secretion and the radio-ligand affinity analysis for VIPR expression. The effect of epidermal growth factor(EGF) was observed. The results showed that: 1. Both BEC and AM secreted VIP basically without remarkable stimulus, and some vesicles with electron-dense core which usually exist specifically in neuropeptide secreting cells were found in plasma of BEC; 2. In a dose-dependent manner, EGF promoted BEC to secrete VIP(r = 0.84, P < 0.05), and it was found that VIP secretion from AM was also elevated in EGF pretreated group(P < 0.05); 3. EGF exhibited a dose-dependent up-regulatory effect on the expression of VIPR in BEC(r = 0.95, P < 0.01). These results indicate that VIP may play an essential role in the protection at local airway by autocrine or paracrine, and the protective effect can be modulated by growth factors.

[Time-effect relationship between the positional microinjection of HCY-2 gene and the neural tube teratogenesis of chick embryos].

The time-effect relationship between positional transferred novel gene(HCY-2) and neural tube teratogenesis, and their possible mechanisms were studied. An eukaryotic expressing vector which containing whole-length HCY-2 cDNA was microinjected into chick embryos in culture at days 0, 1 and 2 (approximately Hamburger-Hamilton stages 1, 6 and 12) mediated by lipofect AMINE reagent. The techniques of RT-PCR, immunohistochemical staining, scanning electron microscope(SEM) and transmission electron microscope (TEM) were used to investigate the expression and distribution of HCY-2 mRNA with its coding product and dysmorphogenesis of the neural tube at 96 h (approximately stage 22). Neural tube defects (NTDs) were discovered in every transferred gene group, only the day 1 embryos which positional site injected was the area pellucida of head, however, the rate of NTDs was the highest (35.3%). There was an obvious time-effect relationship. The phenotypes of NTD were encephalocele, anencephaly, spina bifida and microcephaly. The embryos with transferred gene could express HCY-2 mRNA and its coding product, and the HCY-2 protein mainly distributed in embryonic brain cells as compared to controls. It was found that HCY-2 gene could result in abnormal ultrastructure at the surface and inside of cells under SEM and TEM. It has been observed apoptosis at the sites with NTDs. It is concluded that HCY-2 gene may be a new genotoxic factor, which plays an important role in the mechanisms of neural tube teratogenesis during the early developing stage of embryos.
