[Palatography including lingual and occlusal surface of teeth, palatogram pattern of Chinese sounds].

In order to develop a method that can determine not only the contact pattern of the tongue with the palate but also with the teeth during speech. Static palatograph recording system was used in this study. Palatograms of six normal dentulous adults were made by the recording system during pronounce the following Chinese phonetic alphabets: /S/, /X/, /D/, /J/, /N/, /K/, /R/, /L/. The contact pattern that is the contact area of tongue to palate and tongue to teeth in the palatograms were identified and analysed. The result indicated that the palatograms of the six normal adults are different which can be divided into four types: Type1/S, X/; Type2/D, N, J/; Type 3/K/; Type 4/R, L/.

[The immunogenicity of tooth allografts in mice].

The mice used in this study were of the in-bred strains C57BL/6 and Balb/c which differ at the major histocompatibility complex (MHC). The immunogenicity of tooth allografts was studied in vitro by means of detecting the response of Antibody-Complement Mediated Cytotoxity (ACMC) and Cell Mediated Cytotoxity (CMC) of the host. We found that there was significant difference for both ACMC and CMC activity between the allogenic and isogenic tooth transplantation (P < 0.01). The results indicate that tooth allografts are immunogens and do evoke the immune response of recipients.

[Expression of metastasis-suppressor gene nm23H1 product in nasopharyngeal carcinoma].

Using the specific monoclonal antibody of nm23H1 gene product and the immunohistochemical technique, we studied the expression of DNPK/nm23H1 and its correlation with lymph-node metastasis in nasopharyngeal carchnoma (NPC). The results revealed that, in a total of 31 cases of NPC, 13 showed positive expression with a positive rate of 41.9%. Among 21 cases of NPC with no lymph-node metastasis, 11 (52.3%) showed positive expression while only 20% (2/10) of those with lymph-node metastasis were positive, and there was significant difference between them (P < 0.01). Meantime the expression was very low or not detectable in the cancerous cells of the match lymph-node metastasis and all 10 cases showed negative expression. Statistical significant difference was found between the group of NPC with no metastasis and the group of lymph-node metastasis (P < 0.01). The above results indicated that high expression of DNPK/nm23H1 existed in the group of NPC with no metastasis while low expression existed in those with metastasis and lymph-node metastasis. It suggests that the expression of nm23H1 is inversely associated with lymph-node metastasis. The DNPK/nm23H1 may play a role in polymerization of microtubulin protein and participate in the process of cancer metastasis.

[GS-MS analysis of essential oils from five species of Asarum].

This paper reports the result of GS-MS analysis of the essential oils from five species of Asarum, namely, A. heterotropoides var. mandshuricum (cultivated), A. sieboldii (cultivated), A. caudigerellum (from Sichuan), A. sieboldii(from Shandong) and A. sieboldii(wild). Ninety-two constituents were detected, of which 73 compounds were identified.

[The development of ECG trans-telephone popular monitoring system].

In this essay a new kind of ECG telemetry and monitoring system based on public telephone network is presented, which is able to transfer four channels of ECG of two patients to monitoring center sited at hospital synchronizingly. At the same time doctors may make diagnosis and give instruction for treatment. The system has the functions of real time sample, reviewing, freezing, store, windows, and printing, etc.

Serum apolipoprotein levels of psoriatic patients with normal serum lipid levels.

OBJECTIVE: Recent studies indicated that incidence of occlusive cardiovascular diseases increased in psoriasis. Changes of serum lipid metabolism may be the cause. Apolipoproteins play a central role in lipoprotein metabolism in serum. The measurement of serum apo levels may reveal the characteristics of lipoprotein metabolism in serum in patients with psoriasis and may help to find an answer to the cause of the high incidence of occlusive cardiovascular diseases in psoriasis. METHOD: The apo levels of 31 patients with psoriasis vulgaris and 31 sex- and age-matched healthy individuals with normal serum lipid levels were measured by radial immunodiffusion assay. RESULTS: The apo C III (t = 2.44, P < 0.05) and apo E (t = 2.12, P < 0.05) of psoriatic group elevated significantly. The apo C III (t = 2.77, P < 0.01) of patients less than 40-year-old and apoE (t = 2.91, P < 0.01) of male patients elevated significantly. CONCLUSION: Abnormal metabolism of lipoprotein in serum exist in psoriasis, especially in male patients and/or patients less than 40 years old.

[Cyst of temporomandibular joint with two cases of clinical and pathological analysis]

In this paper,two cases of cyst of temporomandibular joint were reported.We studied eleven cases' literature combined with our clinical and pathological findings,including B typed ultrasonography, CT image and surgical management.We analyzed this kind of cyst from etiological pathological and clinical features and diagnostic therapeutic event.It is a very rare disease with the preferential incidence in woman.We suggested B typed supersonic exam be the first exam method,surgical removal is the best regime.But final diagnosis is depend upon biopsy.

[Study on the immunophenotype of myeloid cells in myelodysplastic syndromes and its clinical implications].

OBJECTIVE: To explore the immunophenotype of myeloid cells in myelodysplastic syndromes and its clinical implications. METHODS: A panel of monoclonal antibodies was used to detect CD13, CD33, CD15 and CD14 on the membrane surfaces of myeloid cells in the bone marrow from 51 patients with myelodysplastic syndromes (MDS), 21 with aplastic anemia (AA), 21 with paroxysmal nocturnal hemoglobinuria (PNH), and 15 normal subjects, by immunoenzymatic assay. The morphology and chromosome karyotype of bone marrow cells of MDS patients were also examined. RESULTS: CD14+ cells, CD13+ cells and CD33+ cells in the bone marrow were more in MDS patients than in normal controls, AA patients and PNH patients. CD15+ cells in the bone marrow were less in MDS patients than in normal controls. The percentages of CD14, CD13 and CD33 positive cells in the bone marrow of MDS patients were related to the percentage of myeloblast, the chromosomal aberrations and the response to treatment. CONCLUSION: There was an immunophenotypic misexpression of myeloid cells in MDS patients. Immunophenotype analysis of myeloid cells might be useful for the diagnosis and directing treatment in MDS patients.

[Effects of cytokines on multidrug-resistance in K562/A02 cells].

OBJECTIVE: To explore the effects of cytokines on human leukemic cell line K562/S and its multidrug-resistant counterpart K562/A02. METHODS: The toxicities of cytokines and the IC50 (the concentration causing 50% inhibition of cell growth) of DNR were assayed by MTT method; intracellular drug concentration was measured by fluorometry; p-glycoprotein (p-gp) expression was detected by APAAP and mdr-1 mRNA was assayed by RT-PCR. RESULTS: The IC50 of DNR for K562/A02 and K562/S cells were 45.08 microg/ml and 0.607 microg/ml, respectively. Pretreating K562/A02 cells with rhu IFN (500 U/ml) or rhu IL-2 (250 U/ml) for 24 hours partially restored the sensitivity of K562/A02 cells to DNR (IC50 were 16.39 and 11.96 microg/ml, respectively) but had not effect on K562/S cells, and it elevated the intracellular DNR accumulation in K562/A02 from 2151 ng/mg x protein to 2570 and 2503ng/mg x protein, respectively. p-gp and mdr-1 mRNA were not down regulated. By contrast, rhu G-CSF and rhu GM-CSF had no effect on either K562/A02 or K562/S. CONCLUSION: rhu IFN or rhu IL-2 could partially restore the sensitivity of K562/A02 to DNR and elevate the intracellular DNR accumulation via a mechanism independent of p-gp or mdr-1 mRNA down-regulation.

[Effects of antisense oligodeoxynucleotides targeting multidrug resistance gene on resistant cell line K562/ AO2].

OBJECTIVE: To investigate the reversal effect of antisense oligodeoxynucleotide on human multidrug-resistant leukemic cell line K562/AO2. METHODS: Antisense oligodeoxynucleotides (AOD) targeting-6 approximately 9 sites of exon 2 in human multidrug resistance gene(mdr-1), one of which is sequence-strict-complied and linked with polyethyleneglycol (PEG) at 5' end (AP, 15 mer), the other lacks nucleotide complied site-1 (AP', 14mer), were synthesized. AP, AP' and verapamil were simultaneously added to human mdr-1-mRNA positive leukemia cell line K562/AO2 and, mdr-1-mRNA and p170 were detected. AS' and AP'were labelled by FITC and designated as ASF' and APF', respectively. In addition, the intracellular concentration of them was detected by FACS. RESULTS: AP' significantly enhanced the sensitivity of K562/AO2 to DOX, down-regulated the expression of mdr-1 and MRP-mRNA and p170, elevated the intracellular concentration of the two AOD, while AP had no effect. The uptake of APF' was significantly higher than that of ASF' in K562/AO2, and the fluorescence was observed in the plasma and nuclear of K562/AO2 cells. CONCLUSION: (AOD targeting mdr-1 promoted the drug sensitivity of drug-resistant tumor cells. 2 AOD had no cytotoxicity to tumor cells. 5 Low molecule PEG enhanced significantly the uptake of AOD by tumor cells.

[A high performance liquid chromatographic method for the determination of teniposide in brain tissue using electro-chemical detection].

A reversed-phase HPLC method for the determination of teniposide in brain tissue is described. Teniposide can be separated on a Hypersil ODS column with a mobile phase of methyl alcohol-water-acetic acid (56:41:3, V/V) and flow rate of 0.8 mL/min (10 MPa). Column temperature was 35 degrees C and operating potentials for electrochemical detection was 0.70 V. To 50 mg brain tissue were 1 mL 50% ammonium sulfate and 4 mL ethyl acetate. The sample was vortexed for 5 min and ultrasonically agitated for 15 min, then centrifuged at 3000 r/min for 15 min. The upper (organic) layer was collected and the lower layer reextracted with 4 mL of ethyl acetate by vigorously vortexing, then centrifuged at 3000 r/min for 15 min. The organic layer was combined with the previously collected ethyl acetate layer. This organic extract was dried under a gentle nitrogen stream, and the residue was reconstituted with 1 mL internal standard of guaifenesinum before HPLC analysis. The linear range was 0.1-10.0 mg/L, and the detection limit 0.1 mg/L. Intra-day and inter-day RSD for assaying brain tissue sample were 0.87% and 1.41%, respectively. The average recovery was 92.87% with coefficient of variation of 2.35%.

[Dual wavelength multiplication subtractive spectrophotometric determination of sulfadiazinum and trimethoprimum in compound sulfadiazine tablets].

This paper described the simultaneous determination of sulfadiazine (SD) and trimethoprim (TMP) in tablets co-Trimediazini by dual wavelength spectrophotometry multriplication subtractive method. This method was based on the measurement of both absorbance of standard solution of SD (8-14) x 10(-6)g/mL, TMP (1-2) x 10(-6)g/mL and sample solution at wavelength 242.5 nm and 228.0nm. The average recovery and RSD of SD and TMP were 100.60%, 0.33% and 98.27%, 1.58%, respectively (n=20). The method was simple, quick and results satisfactory.

[Rapid spectrochemical qualitative analysis of simple OMA system].

Simple optical multichannel analyzer (OMA) system which is formed by ordinary one dimension CCD camera, spectrograph and microcomputer was introduced in this paper. Spectrochemical analysis feasible of this OMA system was studied. The applied software programed, by us can realize wavelength calibration, spectral line identification and give out automatically the results of qualitative and semidefinite quantity analysis.

Treatment of capillary vascular malformation (port-wine stains) with photochemotherapy.

One-hundred and thirty patients (85 female, 45 male) with port-wine stains were treated with photodynamic therapy, also called photochemotherapy, which recently has become acknowledged as effective for a variety of malignant tumors. Probably based on the photochemical reaction with the generation of toxic species, photochemotherapy results in endothelial cell injury and death of abnormal capillaries under overlying epidermis. A retrospective review of 118 available patients with port-wine stains reveals that 98.3 percent responded to photochemotherapy with varying degrees of success after one-time treatment. Results were reported under a simple classification system ranging from ordinary to dilated to posttreatment type. In the ordinary group, the results evaluated as excellent, good, fair, and poor were 37.8, 53.7, 8.5, and 0 percent, respectively, before a second treatment; the treated area was an average of 9.8 (range 7 to 13) cm in diameter. In addition, hypertrophic scars, permanent hyperpigmentation, and hypopigmentation were not seen based on proper parameters. Photochemotherapy offers a potentially efficient and promising choice based on a completely different mechanism from that of selected photothermal therapy with the pulsed-dye laser.

Mechanisms of bradykinin-induced insulin secretion in clonal beta cell line RINm5F.

We investigated the mechanisms underlying bradykinin (BK)-induced rise in intracellular Ca++ concentration [Ca++]i and insulin secretion using clonal beta cell line RINm5F. Incubation with a range of concentrations of BK increased in concentration-dependent manners both insulin secretion (BK of 10 nM to 10 microM) and [Ca++]i (BK of 100 nM to 100 microM). In Ca++-containing medium, BK (1 microM) induced a biphasic [Ca++]i rise, which was characterized by a Ca++ peak and a sustained Ca++ phase. In the Ca++-free medium, BK failed to increase insulin secretion and induced only a Ca++ peak without the sustained Ca++ phase. Thapsigargin (1 microM), an inhibitor of the Ca++ pump in the endoplasmic reticulum, abolished the Ca++ peak and the sustained phase. Nimodipine (1 microM), a voltage-dependent Ca++ channel blocker, abolished the BK-induced sustained Ca++ phase and inhibited BK-induced insulin release. The BK1 receptor agonist des-Arg9-BK (1 microM) did not change either [Ca++]i or insulin secretion. Both the BK-induced insulin secretion and rise in [Ca++]i were inhibited by a selective BK2 receptor antagonist, HOE 140 (3.3-100 nM), in concentration-dependent manners but were not by a BK1 receptor antagonist des-Arg9,Leu8-BK (1 microM). Pretreatment with pertussis toxin (0.1 microg/ml) did not block the BK-induced insulin secretion or increase in [Ca++]i. U-73122 (4, 6 and 8 microM), a phospholipase C inhibitor, antagonized both the BK-induced insulin secretion and the increase in [Ca++]i in a concentration-dependent and parallel manner. BK increased intracellular concentrations of inositol-1,4,5-trisphosphate (IP3). Neither (p-amylcinnamoyl)anthranilic acid (100 microM), a phospholipase A2 inhibitor, nor N(G)-nitro-L-arginine methylester (100 microM), a nitric oxide synthase inhibitor, inhibited these effects of BK. Taken together, these findings suggested that in beta cells, BK activates BK2 receptors, which, in turn, activate a pertussis toxin-insensitive G protein. The G protein couples to phospholipase C, which promotes the formation of IP3 and diacylglycerol. IP3 releases [Ca++]i from the intracellular Ca++ store, probably the endoplasmic reticulum, which triggers Ca++ influx via voltage-dependent Ca++ channels and thus increases insulin secretion.

Adaptation of a strain of Plasmodium falciparum from a Montagnard refugee to Aotus monkeys.

A strain of Plasmodium falciparum from a Montagnard refugee was shown to produce large numbers of gametocytes in culture. Attempts were made to establish this strain in Aotus monkeys via trophozoite and sporozoite inoculation. The Montagnard S-1 strain was readily adapted to A. l. griseimembra monkeys via trophozoite inoculation. Other species of Aotus failed to support the development of high density parasitemia. None of 12 attempts to transmit the infection via sporozoites from Anopheles freeborni or An. dirus mosquitoes was successful; however, developing exoerythrocytic stages were demonstrated in hepatocytes of an A. lemurinus griseimembra monkey.

Preliminary study of the effects of metoprolol and propafenone on ventricular arrhythmia with positive ventricular late potential.

OBJECTIVE: To compare the effects of metoprolol and propafenone in patients with ventricular arrhythmia (VA) of positive ventricular late potential (VLP) and to discuss the effect of medicine on high risk VA. METHODS: 30 A total of 128 patients (78 males and 50 females) with VA of positive VLP were randomly divided into 3 groups. Groups A and B were given metoprolol and propafenone respectively, group C was given vitamin C as placebo. Twenty-four hours dynamic electrocardiogram and VLP were examined before treatment and 4 weeks after treatment. RESULTS: Propafenone could effectively control VA but could not reverse positive VLP to negative meanwhile it had proarrhythmia effect (2/40), metoprolol could effectively control VA (38/46) and reverse positive VLP to negative (39/46) with no obvious side effects. The dosage from 50 to 100 mg/day made no difference. VA and VLP had no significant changes in group C. CONCLUSIONS: Metoprolol is superior to other medicine for VA of positive VLP when the patients have no contraindication of preceptor blocking agents.
