[Comparison of results of chamber angle examination by ultrasound biomicroscopy and gonioscopy].

OBJECTIVE: To compare the consistency of the chamber angles examined by ultrasound biomicroscope (UBM) and gonioscope and provide gonioscopic examination with new methods and technology. METHODS: 10 patients (20 eyes) were selected for the research. The same eyes were examined by UBM and gonioscope in turns. The results from the two methods were compared and the data were analyzed with Spearman correlation test. RESULTS: The results of two methods of chamber angle examination were well consistent when the angle was wide, but the consistency was not good when it was narrow. The angle examined by a gonioscope was wider than that by UBM. When the iris root insertion was evaluated in a narrow angle, the consistency was also not good, and when the peripheral iris morphology was evaluated, there were almost inconsistent results in one-fourth of the patients. CONCLUSIONS: The results of chamber angle examined by a gonioscope show a little wider than its natural status. Especially, it is difficult to distinguish the functional closure from the adhesive closure. Under the influence of light, the determination of iris root insertion by a gonioscope has a sense of conjecture. When peripheral iris morphology is evaluated, the influence of irregular iris might result in a wrong conclusion. Therefore, the technique of gonioscopic examination should be improved.

[The role of the leukocytes in pathogenesis of secondary brain injury].

The levels of leukocyte count, neutrophil number, leukocyte adhesiveness, leukocyte releasing oxygen-free radical (O2.-) after phagocytosing bacteria and serum lipid peroxide MDA were determined in the peripheral blood of 40 patients with acute head injury within 48 hours and 22 normal adults. These levels of patients were significantly higher than those of the controls (P < 0.05), and increased with the severity of craniocerebral injury elevation. The leukocyte count and neutrophil number of those patients who died were significantly higher than these of those survived patients (P < 0.05). It suggested that leukocytes aggravated the secondary brain injury after craniocerebral injury by increasing of leukocyte adhesiveness, and generating and releasing oxygen-free radical, and measurement of leukocyte count and neutrophil number in the peripheral blood could evaluate the prognosis of the patient with craniocerebral injury.

[Effect of long-term experimental stress on blood glucose level in mice with diabetes-prone: the age difference and its mechanism].

OBJECTIVE: To compare the effects of long-term experimental stressors on blood glucose level and insulin secretion in the aged and young mice with diabetes-prone and to explore the mechanism. METHODS: Diabetes-prone was induced by streptozotocin (STZ) in 40 1.5-month-old mice and 40 15-month-old mice. The young and the aged STZ mice were divided randomly into the stress group and the control group, respectively. Each group contained 20 mice. Both the aged and the young stress mice were exposed to multiple stressors (restrain, rotation, crowding) for 6 weeks. Fasting blood glucose (FBG), fasting plasma insulin (FINS), pancreas lipid peroxidase (LPO) and the metabolic products of nitric oxide (NO2-, NO3-) levels and superoxide dismutase (SOD) activity were measured. The grade of insulitis in STZ mice was also evaluated. RESULTS: After 6-week stress, FBG level, pancreatic tissue LPO and NO2-, NO3- contents were significantly higher while FINS level and SOD activity lower in the aged stress mice than those in the youngs. The insulitis malignization in the aged stress group was severer than that in the young stress group. There were correlativities among the above variables. CONCLUSION: The effects of long-term stress in promoting hyperglycemia exacerbating insulitis, and inhibiting insulin secretion are more significant in the aged STZ mice than in the young ones. This difference may result from the excessive NO and the peroxide injury to the pancreas.

[A SEM observation on taxonomic and pharmacognostical identification of genus murraya produced in China].

OBJECTIVE: To Observe the morphological characteristics of the leaf epidermis of 9 Chinese species of genus Murraya and prepare and index for their differentiation. METHOD: SEM(scanning electron microscope). RESULT: Sect. Murraya was found having with blurred dividing lines on epidermis without ropy emblazonry, while sect. Bergera appeared the other way round. CONCLUSION: M. kwangsiensis var. macrophylla should be ground under Sect. Murraya as a new species, while M. tetramera should be separated from M. euchrestifolia; SEM has been proved very useful in the identification, taxonomic and pharmacognostical study of Murraya.

[Studies on individual characters of juvenile delinquents in Fujian province].

Ashenk's Character Questionnaire was adopted to study the relationship between juvenile delinquency and individual characters. 621 delinquents at the age of 13-19 and 1371 school students (as control group) were questioned, and results were statistically analyzed. For male delinquents of 16-19, scores of extroversion, psychosomatic and nervosity were significantly higher than for control group (P < 0.01). For male delinquents of 13-15, scores of psychosomatic and nervosity were significantly higher than for control group (P < 0.01). For female delinquents of 16-19, score of nervosity was lower than for control group (P < 0.05). Different types of delinquency had no statistical relations with characters. The study reveals that characters of psychosomatic and nervosity might be related with male delinquent crimes, and that stabilization with female delinquent crimes.

[Molecular cloning and sequencing of outer capsid protein gene of rice dwarf virus and its expression in Escherichia coli].

The S2 full-length cDNA of rice dwarf phytoreovirus which enocodes the viral outer capsid protein was cloned and its complete nucleotide sequence was determined. The results showed that S2 is 3512 bp long with a large open reading frame which encodes a protein of 1116 amino acids. It shares 94.6% and 95.4% identity with RDV of Japanese H isolate in terms of nucleotide and amino acid sequences, respectively, and it also shows some homology with VP2 of rotavirus at the level of amino acid sequence. The search of deduced RDV S2 amino acid sequence in Blast network found that there were 4 leucine-rich motifs in P2 protein, and ten amino acids within the hydrophibic region at amino-terminus could form an alpha-helix. Predicted secondary structure of S2 cDNA indicated that a hairpin and a stem loop are present in the 5'-end within 50 bp, and a stem loop in the 3'-end within 50 bp. RDV S2 partial and full-length sequences were then cloned into expression vector pET-11d & pTrcHisC. SDS-PAGE and Western blot proved that amino- and carborn-termini of P2 were successfully expressed in E. coli.

[Autoantibodies to the heterogeneous nuclear ribonucleoproteins and the autoimmune diseases].

Autoantibodies to the heterogeneous nuclear ribonucleoproteins (hnRNP) have a great role in the diagnosis of rheumatic diseases. hnRNP-A/B proteins are the main autoantigens in Rheumatoid arthritis (RA), systemic lupus erythematosus (SLE), and mixed connective tissue disease (MCTD). Anti-RA33 autoantibodies which target the hnRNP proteins A2, B1, B2 (the 'RA33 complex') help to the diagnosis of RA at early stage. Therefore, anti-hnRNP autoantibodies are not only valuable diagnostic markers but may also allow deep insights into the pathogenetic mechanisms of rheumatic diseases.

[Study on carbazole alkaloids of Murraya microphylla].

Four carbazole alkaloids were isolated from the methanol extract of Murraya microphylla. Their structures were identified by spectral analysis and chemical evidence, which were identified as koenigine, bis-6-hydroxy-7-methoxygirinimbine, girinimbine and mukonicine. All the compounds were discovered from this plant for the first time, and bis-6-hydroxy-7-methoxygirinimbine was a new compound.

[A new adaptive filtering method for micro-particle image processing].

Based on the local gray level histogram of macro-particle image which is more special than other images, a new adaptive noise filtering method for macro-particle image processing is developed and described in this paper. It is made up of median filter and special order statistic (OS) filters (alpha-mean filter and inverse alpha-mean filter). This adaptive filter not only has satisfactory performance in both of noise-removal and edge-preservation of macro-particle image, but also is easy and quick to implement. An example is presented in this paper.

[The Value of CT Scanning in the Diagnosis of Temporomandibular Joint Dysfunction]

OBJECTIVE:To evaluate the role of CT in the diagnosis of temporomandibular joint dysfunction MATERIAL: 48 temporomandibular joints of 26 patients suspecting osseous changes were examined by CT, with 1mm of layer thickness. All patients had Sch llers position projection as comparison. RESULTS:CT is more sensitive to detect the osseous changes of TMJ than Sch llers position projection, but not well demonstrating sclerosis of TMJ. The Sch llers position projection can only shows the lesion of lateral 1/3 of condyle, but CT including total condyle. CONCLUSION: CT is an important and sensitive tool in detecting osseous changes of temporomandibular joint.

[Clinical Application of CT assisted Double Contrast Arthrography of Temporomandibular Joint]

OBJECTIVE:To introduce double-contrast arthrography of TMJ with reformatted sagittal and coronal CT imaging and to evaluate the value of its application in the diagnosis of temporomandibular joint dysfunction. METHODS: 14 patients in whom diagnosis can not be established with conventional arthrography were examined by double contrast arthrography with reformatted sagittal and coronal CT imaging. RESULTS: In double-contrast arthrography with CT imaging, 1 case was normal;1 case had anterior displacement with reduction;2 cases were anterior displacement without reduction, and 8 cases were with disk adhesion. CONCLUSION:The double contrast arthrography with reformatted sagittal and coronal CT imaging was a very important alternative to conventional arthrography, and has special value to demonstrate adhesion of TMJ.

[A Comparative Study of MRI and CT Double-Contrast Arthrography in Diagnosis of Disc Adhesion of TMJ]

OBJECTIVE:To compare the results of CT-assisted double-contrast arthrography and MRI in the diagnosis of adhesion of TMJ. MATERIAL:18 patients with adhesion of TMJ were examined by CT-assisted double-contrast arthrography and MRI, and their results were analysed including position, degree and with or without osseous changes. RESULTS:18 cases of adhesion were demonstrated on CT-assisted double-contrast arthrography, and that only 6 cases of them were diagnosed on MRI.CONCLUSION: CT-assisted double-contrast arthrography is more sensitive and accurate than MRI in demonstrating the disk adhesion of TMJ.

[Multi-channel probing for emission spectra of metals stimulated by LD pumping laser].

Emission spectra of several metals stimulated at a tiny spot by an YAG laser pumped by a LD were obtained analyzed by means of a CCD optical multi-channel analyzer. The emission spectra collected and analyzed by multi-channel with high signal to noise ratio and high dynamic range were obtained, and the wavelengths accord well with the standard spectra of these metals. Application of this method was discussed.

[Determination of diclofenac sodium and codeine phosphate in shuerfen tablets by CPA-matrix method].

The contents of diclofenac sodium and codeine phosphate in shuerfen tablets may be determined simultaneously by CPA-matrix method without separation of its components. The program was edited by BASIC. The average recoveries and RSD were 99.85%, 0.46% for diclofenac sodium and 99.80%, 0.50% codeine phosphate, respectively. This method is simple and rapid, and the result is accurate.

Modulation of aromatase expression in the breast tissue by ERR alpha-1 orphan receptor.

We have previously identified a silencer element (S1) that is situated between promoters I.3 and II of the human aromatase gene and that down-regulates the action of these promoters. We recently applied the yeast one-hybrid approach to screen a human breast tissue hybrid cDNA expression library for genes encoding the proteins binding to the silencer region. Most proteins identified from this approach belong to the nuclear receptor superfamily. Fifty % of the positive clones encode for ERR alpha-1, and other positive clones include EAR-2, EAR-3 (COUP-TF1), RAR gamma, and p120E4F. Because ERR alpha-1 was found to be the major protein interacting with S1, we decided to examine the regulatory action of ERR alpha-1 on promoter I.3 of the human aromatase gene. Using a reporter plasmid that includes the aromatase genomic fragment containing promoter I.3 and S1, ERR alpha-1 was found to have a positive regulatory function in breast cancer SK-BR-3 cells. Gel mobility shift assays have confirmed that ERR alpha-1 binds to S1 in a dose-dependent manner, and DNase I footprinting analysis has revealed that ERR alpha-1 binds to a region, 5'-AAGGTCAGAAAT-3', which is within S1 and between 96 and 107 bp relative to the transcriptional start site of promoter I.3. In addition, despite the fact that the nuclear receptor SF1 was shown previously to bind to the same site and to mediate a cAMP response in ovary, our yeast one-hybrid screening did not find any SF-1 clones. Gel mobility shift assays further revealed that SF-1 can bind to the silencer element with an affinity comparable with ERR alpha-1. Because our reverse transcription-PCR analysis was not able to detect SF1 mRNA in breast cancer tissue or in SK-BR-3 cells, it is thought that SF1 protein is not expressed in breast cancer tissue. Two ERR alpha-1 RNA variants with differences at the 5'-end have been reported. Our reverse transcription-PCR analysis identified the shorter variant in 28 of 32 breast tumor specimens and the longer variant in only 1 specimen. In addition, the shorter variant was detected in breast cancer SK-BR-3 cells as well as in a breast tumor fibroblast line WS3TF. The results suggest that ERR alpha-1 is one of the nuclear proteins interacting with S1 in breast cancer tissue. It is thought that the silencer element in the human aromatase gene may function differently in different tissues because of distinct expression patterns of transcription factors.
