Clinical utility of the alizarin red S stain on permanent preparations to detect calcium-containing compounds in synovial fluid.

The alizarin red S stain for permanent cytologic preparations is a valuable test that is complementary to compensated polarized light microscopic examination to detect calcium crystals. Alizarin red S has the greatest sensitivity for detection of calcium pyrophosphate crystals because crystals are stained regardless of how weakly or strongly birefringent they may be. Alizarin red S stain does not distinguish between amorphous types of calcium compounds; therefore, the different types of calcium compounds can be distinguished only when typical crystal morphologic features are present. Diagnostic importance can be attached to intracellular material that is stainable. In contrast, the diagnostic value of stainable, amorphous, extracellular material is unreliable because it is difficult to distinguish this extracellular material from contaminants frequently found in clinical specimens. Alizarin red S does not stain monosodium urate or corticosteroid crystals. Air-dried cytospin smears are helpful because they may frequently demonstrate more crystals than the wet-mount preparation. Furthermore, special stains can be performed subsequently on air-dried cytospin smears if necessary.

Cutaneous histiocytic malignancy. Immunohistochemical re-examination of cases previously diagnosed as cutaneous "histiocytic lymphoma" and "malignant histiocytosis".

True histiocytic lymphoma (THL) and malignant histiocytosis (MH) have been defined by clinical and histologic findings and enzyme histochemistry. We reviewed cases previously diagnosed as cutaneous histiocytic lymphoma (HL) and MH with cutaneous lesions. These cases had been classified as "histiocytic" on the basis of previous enzyme histochemistry profiles of frozen tissue. Cutaneous tumor cells were reevaluated using a panel of immunohistochemical stains in formalin-fixed, paraffin-embedded tissue in correlation with histopathologic examination. The antibodies used in this study were directed against CD45 (leukocyte common antigen [LCA]), CD20 (L26) for B cells, CD3 and CD45RO (UCHL-1) for T cells, CD68 (KP-1) and lysozyme for histiocytes, as well as CD30 (BerH2) for Ki-1 positive cells. On re-evaluation, the seven cases originally classified as HL were reclassified as one case of THL with neoplastic cells positive for CD68 (KP-1) and lysozyme, two cases with immunohistochemical features of Ki-l lymphoma (including one of T-cell lineage), three cases of T-cell lymphoma, and one case of B-cell lymphoma, all associated with variable degrees of reactive histiocytosis. The four cases originally classified as MH were reclassified as two cases of MH and two cases of uncertain lineage. Although rare, histiocytic malignancies do exist. However, the diagnosis of histiocytic malignancy should be made only after careful correlation of atypical tumor cells in histopathologic sections and sections stained immunohistochemically. Erroneous classification of reactive histiocytes as neoplastic histiocytes using only enzyme histochemistry in frozen sections is a pitfall to be avoided.

Growth of mycorrhizal fungi in dixenic cultures with bacteria in media of different composition.

Studies were carried out on the effect of bacteria: Arthrobacter globiformis, Bacillus subtilis and Pseudomonas fluorescens on biomass production by three important ectomycorrhizal fungi: Laccaria laccata, Hebeloma crustuliniforme and Rhizopogon vinicolor in media of different composition. It was shown that bacteria stimulated, inhibited or did not affect significantly the biomass production by mycorrhizal fungi. 3-factor ANOVA have shown that although effect of bacteria was statistically significant (p < or = 0.05), composition of medium and its pH affected mycelial growth stronger than bacteria.

[The role of substance P and somatostatin in acupuncture and moxibustion-induced postsynaptic inhibition].

AIM OF INVESTIGATION: Our previous work indicated that acupuncture and moxibustion (heating acupoints with a special lamp) could produce inhibitory effects on the tail flick reflex and the nociceptive response of dorsal horn neurons. In the present study the role of somatostatin (SS) and substance P (SP) in acupuncture and moxibustion-induced postsynaptic inhibition was further investigated. METHODS: The experiments were performed on male adult Wistar rats. The antidromic action potential (AAP) induced by cervical cord stimulation was extracellulary recorded at lumbar cord (L3-4). The latency of AAP was used to represent the excitability of postsynaptic projecting neurons. The effects of acupuncture (0.5 ms, 3.3 Hz, 2 mA) for five minutes or moxibustion (the temperature up to 45-46 degrees C) for six minutes at Huantiao points on the latency of AAP were observed. And then the effects of topical administration of SS or SP antiserum on either acupuncture or moxibustion-induced postsynaptic inhibition were observed. RESULTS: The latency of AAP was markedly prolonged by acupuncture and moxibustion. The maximal prolongation was 0.196 +/- 0.071 ms (n = 12, P < 0.02) and 0.176 +/- 0.062 ms (n = 11, P < 0.02) respectively. After topical administration of SS antiserum (1:40, 10 microliters), the latency of AAP was slightly prolonged by either acupuncture or moxibustion. The maximal prolongation (0.041 +/- 0.029 ms and 0.016 +/- 0.020 ms) was significantly reduced by SS antiserum (P < 0.05). While pretreated with SP antiserum, the latency was still prolonged by moxibustion (0.142 +/- 0.067 ms), but not by acupuncture (-0.003 +/- 0.046 ms). CONCLUSION: It is referred that postsynaptic inhibition may be involved in both acupuncture analgesia and moxibustion analgesia. The former is predominately mediated by SP and partially by SS, while the latter mainly by SS but not by SP.

Double fluorescent vital assay of phagocytosis by cultured retinal pigment epithelial cells.

PURPOSE: The goal of this study was to develop the first vital assay system for in vitro analysis of phagocytosis of rod outer segments (ROS) by normal retinal pigment epithelial (RPE) cells and for study of the phagocytic defect in RPE of the Royal College of Surgeons (RCS) rat with inherited retinal degeneration. Required features included ability to directly visualize and quantitate the phagocytic process in living RPE cultures, and capability for subsequent quantitative analysis after fixation of the cells at any chosen time after incubation with ROS. METHODS: A double fluorescent method was designed, based on the process of phagosome-lysosome fusion. For vital staining of lysosomes, confluent cultures of rat RPE cells were incubated with sulforhodamine (SR), a red fluorescent lysosomotropic dye. SR-stained cultures were challenged with isolated rat ROS tagged with fluorescein isothiocyanate (FITC), a green fluorescent probe. RESULTS: This method was used to observe all phases of the phagocytic process in the living cells and the kinetics of ROS binding, ingestion, and phagosome-lysosome fusion were determined. Control studies showed no differences in binding, ingestion, or digestion of unstained versus FITC-stained ROS. Additionally, the phagocytic defect in dystrophic RCS rat RPE cells was confirmed using this technique. CONCLUSIONS: This relatively simple new method is useful in that it uses inexpensive, readily available reagents, it enables real-time analysis of phagocytosis experiments, and it does not require termination of the cultures for analysis of phagocytic ability.

[Quantitative studies on discrimination of simple geometric patterns and related visual illusions].

Using quantitative psychophysical measurements, the abilities in discriminating some simple geometric patterns and the related visual illusions (Zoellner, Muller-Lyer, Ponzo and Delboeuf illusions) were investigated under luminance contrast (LC) and isoluminance color contrast (ICC). The results showed that the subjects possessed identical abilities in discriminating parallelity, length and area, and illusory distortion perceived for these geometric properties were of the same strength, no matter the figures were displayed by LC or ICC. The contour- and border illusions, however, vanished at ICC. The minimum contrast inducing contour and border illusions (1.8% and 5.3%, respectively) was considerably lower than that inducing parallelity illusion (15%). These imply that simple geometric properties and contours (or borders) of visual images may be separately transmitted by different visual pathways.

[Effects of luminance contrast and color contrast between components of illusory figure on the illusion magnitude].

Using quantitative psychophysical measurements, the effects of luminance contrast (LC) and equiluminance color contrast (ECC) between components of illusory figures on the magnitudes of orientation illusion (Zoellner illusion), length illusion (Müller-Lyer illusion and Ponzo illusion) and area illusion (Delboeuf illusion) were investigated. The results showed that, compared with normal illusory magnitude, the illusory magnitude of the subjects decreased obviously when LC or ECC existed between the components of the illusory figures. The decrease was especially obvious in the orientation illusion. The minimum contrasts for inducing contour and border illusions were only 1.8% and 5.3% under LC. They increased to 66.0% and 46.4% respectively when both luminance and color contrasts were introduced into the figures. These observations provided evidence for the existence of mutual inhibition between the luminance, color and form channels of the visual system.

Chronic idiopathic thrombocytopenic purpura. Splenic pathologic features and their clinical correlation.

We studied 83 patients who had chronic idiopathic thrombocytopenic purpura refractory to steroid therapy and who underwent splenectomy. The positive rates of antiplatelet and antinuclear antibodies were 40% (30/75 patients tested) and 41% (25/61 patients tested), respectively. The morphologic characteristics of splenic pathologic findings included prominent secondary follicles and foamy macrophages, present in 23 (28%) and 56 (67%) of patients, respectively. Extramedullary hematopoiesis was noted in 50 (60%) of the patients. Eight patients (10%) presented with periarterial fibrosis, and three of them had systemic lupus erythematosus. The 23 patients (28%) with prominent secondary follicles were younger than the other patients, were preponderantly female, and had a higher rate of antiplatelet antibody production, and all had an initial response with a greater increase in platelets postoperatively. Eight patients (10%) had neither prominent secondary follicles nor foamy macrophages and lower splenic weights; their initial response compared with that of other patients was significantly poorer. The thrombocytopenia in these patients might have a different mechanism.

[Experimental study on effect of jiawei yupingfeng san on microcirculation and plasma superoxide dismutase activity in old mice].

The effects of Jiawei Yupingfeng San on microcirculation and plasma SOD of old mice were observed. The results showed that the prescription could markedly improve old mice's blood flow of microcirculation, increase their speed of blood flow and raise their plasma TSOD and Cu, Zn-SOD activity, and there was very significant difference compared with control (P < 0.001). The results suggested that this prescription had the effects of improving the quality of life and delaying the aging of organism.

Immature teratomas of different origin carried by a pregnant mother and her fetus.

We describe the case of a pregnant mother and her fetus who both carried teratomas during the pregnancy. The fetus was diagnosed at 38 weeks' gestation to have an intracranial mass, which was later determined to be an immature teratoma. During a cesarean section delivery, an ovarian tumor was found in the 27-year-old mother that was also diagnosed to be an immature teratoma. Because of the similar histology of the tumors carried by both mother and child, a single clonal origin was suspected. Using polymerase chain reaction (PCR) and electrophoresis of highly polymorphic DNA satellite sequences, we determined that the origin of the intracranial teratoma carried by the child was independent of the mother's tumor. We also examined the p53 tumor suppressor gene in constitutional cells from both mother and child for the possible presence of a cancer-predisposing inherited mutation, but none was found. To our knowledge, this is the first report of the simultaneous occurrence of independent malignant immature teratomas in a mother and child during pregnancy.

[Temporal phase complementarity between the lateral geniculate neurons sharing a common receptive field].

Bi-unit recordings were sometimes obtained in the lateral geniculate nucleus of cat with a single microelectrode. The neuron pairs consisted of an on-center and an off-center cell sharing a common receptive field and both cells showed either transient or sustained responses to light stimulation. When a sinusoidally modulated light spot was presented at their receptive field center, the on-center and off-center cells responded half a cycle (approximately 180 degrees) out of phase with one another at all intensities tested. At a low temporal frequency (5 Hz), peak responses occurred in response to the maximum (in on-center cells) or minimum (in off-center cells) luminances of light modulation with an identical delay. The complementary phase property makes the cell couple work in a "push-pull" manner and enables transient cells, which show half-cycle rectification as they work alone, to transmit modulated light signals of complete cycle.

Production of B-group vitamins by Azospirillum spp. grown in media of different pH at different temperatures.

Studies were carried out on B-group vitamin (thiamine, biotin, nicotinic acid, riboflavin, pantothenic acid) production by 3 strains of Azospirillum (one derived from coniferous ectomycorrhizae and two--from sporocarps of ectomycorrhizal fungi) grown in media of different pH (5.5, 6.5, 7.5) at different temperatures (10 degrees C, 20 degrees C, 26 degrees C). Riboflavin was produced in largest amounts by all the strains studied; biotin was not detected in culture filtrates at all. Qualitative-quantitative composition of vitamins in post culture liquids of azospirilla depended on the temperature of growth, pH of the medium and on the strain studied. Thiamine was synthesized in largest quantities at pH 5.5 by all strains of Azospirillum--independently of the temperature of growth. In media of higher pH this vitamin was detected in considerably smaller amounts or was not detected at all. The smallest quantities--and the smallest numbers of vitamins produced were observed at temperature 10 degrees C and pH 5.5.

Kinetic characterization of reductant dependent processes of iron mobilization from endocytic vesicles.

The reductant dependence of iron mobilization from isolated rabbit reticulocyte endosomes containing diferric transferrin is reported. The kinetic effects of acidification by a H(+)-ATPase are eliminated by incubating the endosomes at pH 6.0 in the presence of 15 microM FCCP to acidify the intravesicular milieu and to dissociate 59Fe(III) from transferrin. In the absence of reductants, iron is not released from the vesicles, and iron leakage is negligible. The second-order dependence of rate constants and amounts of 59Fe mobilized from endosomes using ascorbate, ferrocyanide, or NADH are consistent with reversible mechanisms. The estimated apparent first-order rate constant for mobilization by ascorbate is (2.7 +/- 0.4) x 10(-3) s-1 in contrast to (3.2 +/- 0.1) x 10(-4) s-1 for NADH and (3.5 +/- 0.6) x 10(-4) s-1 for ferrocyanide. These results support models where multiple reactions are involved in complex processes leading to iron transfer and membrane translocation. A type II NADH dehydrogenase (diaphorase) is present on the endosome outer membrane. The kinetics of extravesicular ferricyanide reduction indicate a bimolecular-bimolecular steady-state mechanism with substrate inhibition. Ferricyanide inhibition of 59Fe mobilization is not detected. Significant differences between mobilization and ferricyanide reduction kinetics indicate that the diaphorase is not involved in 59Fe(III) reduction. Sequential additions of NADH followed by ascorbate or vice versa indicate a minimum of two sites of 59Fe(III) residence; one site available to reducing equivalents from ascorbate and a different site available to NADH. Sequential additions using ferrocyanide and the other reductants suggest interactions among sites available for reduction. Inhibition of ascorbate-mediated mobilization by DCCD and enhancement of ferrocyanide and NADH-mediated mobilization suggest a role for a moiety with characteristics of a proton pore similar to that of the H(+)-ATPase. These data provide significant constraints on models of iron reduction, translocation, and mobilization by endocytic vesicles.

Leukemic manifestation in a case of alpha-chain disease with multiple polypoid intestinal lymphocytic lymphoma.

We describe a female aborigine from Taiwan with alpha-chain disease associated with multiple polypoid intestinal lymphocytic lymphoma and leukemic manifestation. Initially, the patient experienced intermittent diarrhea, abdominal pain, and leukemic manifestation. No evidence of bone marrow involvement was found. Alpha-chain protein was demonstrated in the serum. Gastroendoscopy and a series of radiographs of the small intestine revealed multiple polypoid tumors involving the entire length of the small intestine. Duodenal biopsy showed diffuse lymphocytic lymphoma. Immunohistochemical staining of tumor samples revealed features typical of alpha-chain disease. Cytogenetic analysis showed the same abnormal karyotypes of neoplastic clones in intestinal tumor cells and in circulating leukemic cells. The data suggest that alpha-chain disease can present initially with intestinal multiple polypoid lymphocytic lymphoma and leukemic manifestation without evidence of bone marrow involvement. The data also support the homing theory of lymphomas from mucosa-associated lymphoid tissue.

Is persistent polyclonal B lymphocytosis caused by Epstein-Barr virus? A study with polymerase chain reaction and in situ hybridization.

Previous reports of patients with persistent polyclonal B lymphocytosis have found associations with female sex, cigarette smoking, HLA-DR7 phenotype, and moderate elevation of peripheral blood polyclonal B lymphocytes. The presence of binucleated cells and atypical lymphocytes in the peripheral blood of these patients was highly suggestive of a viral infection, such as with the Epstein-Barr virus. We report a 47-year-old asymptomatic woman who was incidentally found to have persistent polyclonal B lymphocytosis and serum IgG against virus capsid antigen (VCA) and Epstein-Barr virus (EBV)-associated nuclear antigen (EBNA) of EBV. The presence of EBV was investigated in the peripheral blood lymphocytes from this patient by in situ hybridization and polymerase chain reaction methods. EBV DNA was demonstrated in the lymphocyte fraction by polymerase chain reaction, and it was further located in lymphoid cells by in situ hybridization. These results indicate that persistent polyclonal B lymphocytosis is strongly associated with EBV.

Evidence for coincident mutations in human lymphoblast clones selected for functional loss of a thymidine kinase gene.

A mitotic "recombination-competent state" inducible by x-irradiation is thought to exist in yeast. We sought evidence for such a process in mammalian cells by examining the occurrence of mutations at unlinked loci in clones derived from a human lymphoblast cell line. A total of 169 independent clones that arose spontaneously or after exposure to x-rays or ethyl methanesulfonate were selected for new somatic mutations at the thymidine kinase gene on chromosome 17q. They were subsequently screened for coincident mutations by use of variable-number-of-tandem-repeat probes located on different chromosomes. Three coincident mutations were positively identified by Southern analysis on chromosomes 7 and 14; they included one that produced a new allele and two that caused loss of allele heterozygosity. Densitometric analysis of the latter two indicated the presence of two copies of the remaining allele. Several possible coincident genetic events were also observed on chromosome 17. These findings revealed a coincident mutant fraction of about 10(-2)/cell, whereas the expected mutation fraction at these loci is less than 10(-4)/cell. These results may thus provide the first molecular evidence that a "global" mutational process capable of inducing genetic instability exists in mammalian cells.
