[Cloning and expression of human TNFR (P55)-IgG Fc fusion protein in eukaryotic cells].

OBJECTIVE: To express soluble biologically active TNF receptor protein in eukaryotic cells, which can inhibit cytotoxic activity of TNF. METHODS: PCR amplified the extra-cellular region of TNF receptor P55 and IgG Fc gene. Then the two were linked through an oligomer encoding a thrombin-sensitive peptide linker and cloned into eukaryotic expression vector pcDNA3.1 (+). The eukaryotic expression plasmid pcDNA3. 1/TI was then transfected to the mammalian cell line COS7 and BHK. Using the G418 system, BHK cell clones were selected and can continuously secrete biological protein in large amount. RESULTS: The expressed protein was fused with IgG Fc and secreted into the cell culture supernatant. It has good antigenicity and binding ability to TNF. It can also inhibit the cytotoxic activity of TNF on L929 cell. CONCLUSION: The TNFR-IgG Fc fusion protein expressed in eukaryotic cells has biological activities of human TNF receptor P55.

Germ cell apoptosis induced by ureaplasma urealyticum infection.

AIM: To study the effect of Ureaplasma urealyticum (UU) infection on germ cell apoptosis of male rats. METHODS: Male rats were infected artificially with UU serotype 8 (T960). Morphological changes of germ cells in the seminiferous tubules and the lumen of the epididymides were observed under the light microscope. Fluorescence-conjugated polyclonal antibodies to Fas and Fas ligand (FasL) were used to localize Fas and FasL. TUNEL staining of germ cells and Sertoli cells was performed by the AKPase method. TUNEL-positive rate (% positive cells) and TUNEL-positive area (area occupied by stained cells) were analysed by KS400 Image Analysis System. The DNA laddering analysis was performed by agarose gels electrophoresis. RESULTS: In those rats infected with UU: (1) Exfoliated germ cells were dramatically increased. Many multinucleated giant cells were found in the seminiferous tubules and the lumen of the epididymides. (2) The number of TUNEL-positive cells and the TUNEL-positive area were significantly increased. (3) The expression of Fas and FasL in germ cells and Sertoli cells was up-regulated. (4) Discrete bands of fragmented DNA were found in the testicular cells. CONCLUSION: In male rats, germ cell apoptosis was increased in UU infection.

Clean-up and disposal process of polluted sediments from urban rivers.

In this paper, the discussion is concentrated on the properties of the polluted sediments and the combination of clean-up and disposal process for the upper layer heavily polluted sediments with good flowability. Based on the systematic analyses of various clean-up processes, a suitable engineering process has been evaluated and recommended. The process has been applied to the river reclamation in Yangpu District of Shanghai City, China. An improved centrifuge is used for dewatering the dredged sludge, which plays an important role in the combination of clean-up and disposal process. The assessment of the engineering process shows its environmental and technical economy feasibility, which is much better than that of traditional dredging-disposal processes.

[Response of structural plasticity of Tilia amurensis sapling crowns to different light conditions].

This paper studied the crown structure of Tilia amurensis saplings, e.g. the shape of crown, and the distribution of branch and leaf. It is shown that the crown structure of the saplings was modified plastically in response to different light conditions. The differentiation of the first order branches was promoted by strong radiation, due to the bole growth being inhibited under high light condition, whereas the bifurcation density of the first order branches whose growth was inhibited by shading was promoted in under-canopy environment. The quantitative leaf densities of the saplings obviously decreased with the decrease of light level, while their leaves tented to distribute in upper crown. The interception of the saplings was enhanced by the distribution pattern of branches and leaves in crown. Under adequate shade condition, the saplings had the strategy of shade-avoidance of pioneer species in upward growth, and had the strategy of light-forage of moderate shade tolerance in horizontal branch growth. The saplings expressed typically shade-tolerant under weak light condition. Such a change of crown structure was an advantageous adaptation for enhancing light interception.

[Effect of soybean protein and high calcium intake on the concentration of serum lipids in hypercholesterolemic rats].

OBJECTIVE: To investigate the effects of dietary soybean protein or/and high calcium intake on the concentration of serum total cholesterol (TC) and serum lipids in hypercholesterolemic rats. METHODS: Following 14 days of inducing the hypercholesterolemia model successfully, 24 female rats were fed 4 experimental diets contained 20% of protein each from either casein or soybean protein and either a normal or high concentration of calcium (0.59% and 1.12%) in a 2 x 2 factorial design. RESULTS: After the experimental diets had been fed for 21 days, TC was (10.56 +/- 2.74, 7.99 +/- 2.19, 5.48 +/- 0.51 and 6.81 +/- 0.98) mmol/L in the rats fed diets containing casein and normal concentration of calcium, casein and high concentration of calcium, soybean, protein and normal concentration of calcium, and soybean protein and high concentration of calcium, respectively. According, Serum apolipoprotein B100(ApoB100) was (0.46 +/- 0.14, 0.31 +/- 0.12, 0.17 +/- 0.07, and 0.21 +/- 0.05) g/L, respectively. Comparing with the rats in casein group, the TC and ApoB100 of the rats in other 3 groups were significantly low (P < 0.05). Whereas, the changes of TG and HDL-C were not significant in these rats. CONCLUSIONS: Dietary soybean protein or/and high calcium intake lower TC in hypercholesterolemic rats and there is an interactive effect of these two dietary factors on lowering TC. Further researches are needed for explaining the possible mechanism of soybean protein and high calcium intake on lowering TC.

Effect of aloe-emodin on expression of proliferating cell nuclear antigen of vascular smooth muscle cells in culture after arterial injury.

OBJECTIVE: To observe the effect of aloe-emodin on the expression of proliferating cell nuclear antigen (PCNA) in smooth muscle cells (SMCs) after arterial injury and study the molecular mechanism of inhibition of aloe-emodin on SMC proliferation. METHODS: Deendothelialization was performed at the abdominal aorta in Japanese white rabbits using a 3F Fogarty arterial embolectomy catheter. 48 hours later, the medium of abdominal aorta was isolated and primary SMCs culture was performed. Cells were synchronized to G0 by serum starvation, then aloe-emodin at a concentration of 20 micrograms/ml was added to the culture medium containing 10% [v/v] fetal calf serum. Vehicle was also added to the medium as a control. After 18 hours, the expression of PCNA at the level of mRNA and protein were examined using techniques of RT/PCR, Western blotting and inmmunocytochemistry respectively. RESULTS: Compared with the control group, the expression of PCNA mRNA and protein was prominently decreased after addition of aloe-emodin. CONCLUSION: The inhibition of aloe-emodin on SMCs proliferation may be caused by inhibiting the expression of the PCNA gene.

[Apoptosis of bone marrow cells aplastic anemia patients treated with cyclosporin A].

OBJECTIVE: To evaluate the effects of cyclosporin A (CsA) on the apoptosis of bone marrow cells of aplastic anemia (AA) patients. METHODS: The apoptosis of and Fas antigen expression on the bone marrow mononuclear cells (MNC) in 25 AA patients and 10 normal controls were assayed by TUNEL and FACS, respectively. RESULT: The percentage of CD(34)(+) cells was decreased significantly in AA patients than in normal controls (P < 0.05), and so did the percentage of CD(34)(+)Fas(+) cells (P < 0.05). The percentage of apoptotic cells in AA patients was higher than that of controls (P < 0.01). After the patients were treated with CsA for 2 months, the percentage of apoptotic cells was decreased (P < 0.01). The percentage of CD(34)(+)Fas(+) cells was positively correlated with that of apoptotic cells in AA patients. CONCLUSION: Fas is involved in the apoptosis of CD(34)(+) cells of AA patients. CsA could reduce the percentage of apoptosis of bone marrow cells in AA patients.

[The mutation of anti-CD3 antibody (HIT3a) gene and its expression].

OBJECTIVE: To improve the expression of anti-CD(3) single chain Fv (scFv) by site mutation and identify its biological activity. METHODS: Anti-CD(3) scFv gene was mutated by PCR, the target clones were screened by both the fingerprints of DNA restriction endonuclease digestion and Western blot, the antigen-binding activity of scFv was examined by FACS, competitive inhibition was performed with (125)I-labeled HIT3a and the cytotoxic effect mediated by the anti-CD(3) scFv-activated T lymphocytes was analyzed by (51)Cr-released assays. RESULTS: The DNA sequencing showed that the 6th amino acid of the anti-CD(3) antibody (HIT3a) heavy chain gene was mutated from E (GAG) to Q (CAG). The expression of mutated anti-CD(3) scFv (m2) was increased by 100 times higher than that of the parent scFv, and there was no difference in the Jurkat cell (CD(3)(+))-binding activity between the (m2) and parent scFv. The preliminary results of competitive assays showed that m2 could partially block the sites of CD(3)(+) Jurkat cells where the parent antibody bound to. Cytotoxicity assays demonstrated that CD(3)AK cells induced by IL-2 and m2 showed stronger cytotoxic effect than that of LAK cells induced by IL-2 alone in vitro. CONCLUSION: By site mutation, a high expression fragment m2 of anti-CD(3) scFv antibody was obtained. The results of some experiments indicated that m2 could bind to CD(3)(+) Jurkat cells, furthermore, by co-stimulated with IL-2, it could activate peripheral T lymphocytes and induce CD(3)AK cytotoxic effect.

[Investigation on relation between Yang Shangshan and the classified compilation of Tai su (Comprehensive Notes)].

Taisu, a book complied by Yang Shangshan by imperial decree is the earliest complete commentary and classified complation of Neijing (The Yellow Emperor's Canon of Internal Medicine) now extant. Recently, some scholars argued about the classified compilation of Taisu performed by Yang Shangshan. However, by analyzing and researching into the text and commentary of Taisu, conclusion can be drawn that the classified compilation of Taisu was indeed complied by Yang Shangshan himself. First, "youben" (certain edition) and "yiben" (an edition) in the commentary are not referring to the other editions of Taisu. Second, quotations cited from Su wen (Plain Questions) and Jiu juan (Nine Volumes) in the commentary are not used to collate Taisu. Thire, analysis made on the compilation and commentary by Yang Shangshan. Fourth, analyzing "taisu jing lun (Classical Discourses of Taisu)", the commentary of Taisu Shui lun (On water in Comprehensive Notes).

[Expression of hepatitis B virus antigen in brain tissue from liver cirrhosis patients with hepatitis B and its significance].

OBJECTIVE: To investigate the expression of hepatitis B virus (HBV) antigen in brain tissue from liver cirrhosis patients with hepatitis B and explore its significance. METHODS: HBsAg and HBcAg were detected in the brain tissue from 70 liver cirrhosis dead patients with hepatitis B by S-P immunohistochemical assay, and the relationship between the expression of HBV antigen in brain tissue testing and clinic and pathology was analyzed. RESULTS: 30 patients (42.89%) were positive for HBV antigen. Among them, 24 patients (34.29%) were positive for HBsAg and 18 patients (25.71%) were positive for HBcAg. HBV antigen was mainly found in cytoplasm and distributed in neurons, neurogliocytes and vascular endothelial cells. The positive cells were distributed separately, scatteredly or focally. The expression of HBV antigen was not associated with the serum level of HBV replication but associated with the occurrence of hepatic encephalopathy (HE) and the severity of brain tissue pathologic lesions of HE. CONCLUSIONS: The results indicate that HBV infection develop in brain tissue from liver cirrhosis patients with hepatitis B and HBV may replicate in it. HBV infection in brain tissue may p lay an important role in occurrence and development of HE from liver cirrhosis patients with hepatitis B.

Biorheological characteristics of skin after expansion.

This study investigated the changes in biorheological characteristics of skin after expansion. Expanders were implanted into the back of eight adult dogs, and after an eight-week expansion the expanded and the non-expanded control skins were transferred to an adjacent site. The expanded specimens and their controls were obtained at 3, 6, 12 and 24 weeks after transfer, and their biorheological characteristics and histological changes were studied. The characteristics of the stress-relaxation, the stress-strain relationship and tensile strength of the expanded and control specimens were measured. The tests demonstrated that during the initial stage after transfer, the biorheological properties of experimental specimens differed significantly from those of their controls. However, the differences between expanded and control specimens began to lessen gradually with increasing recovery time. When the recovery time reached 24 weeks or longer, the experimental skin exhibited the same mechanical properties as the control skin. Histological examination of expanded specimens showed thickened epidermis and thinned dermis. But there were no significant histological differences between expanded skins and their controls after 24 weeks.

[Effects of low energy laser on tooth movement and remodeling of alveolar bone in rabbits].

OBJECTIVE: The aim of this study was to investigate effects of low energy laser on the experimental tooth movement and the remodeling of alveolar bone in rabbits. METHODS: A total of 42 white rabbits were chosen and randomized into one control group and six experimental groups, with 6 rabbits in each group. After anesthesia with 2% pentobaritone sodium, orthodontic appliances consisting of a coil spring connected bilaterally the upper first molar with the upper incisor by using a ligature wire. The force exerted at the time of insertion was approximately 80 g. The left side served as the control side, and the right side was the experimental side treated by receiving irradiation of low energy laser. The treatment periods of different groups lasted separately for 1, 3, 5, 7, 14, 21 days respectively. The displacement extent of teeth was measured by employing the computer image analyzing system. The results were analyzed statistically. Through HE staining, the histomorphological character of tissue around first molar was also investigated, and numbers of osteoclasts were counted. RESULTS: The displacement extent of teeth on the experimental side, which was irradiated by low energy laser, was more obvious than that of the normal control side. The difference was statistically significant 1, 3, 14, 21 days after the beginning of the treatment. Through histological observation under a light microscope, the osteoclasts and osteoblasts on the experimental side remained more active than those of the control side. There is significant difference in amount of osteoclasts between the experimental and the control sides, 3, 5 or 7 days after the treatment (P < 0.01, P < 0.05). CONCLUSION: The irradiation of low energy laser promote tooth movement and remodeling of alveolar bone.

[An investigation of the optical spectra and the EPR spectrum of Ni2+:CsMgCl3].

A method which can calculate accurately the zero-field splitting parameter D and the g-factor is founded by applying the energy matrix elements, including Coulomb interaction and spin-orbit coupling of the strong-field coupling scheme for d2 (d8) electron configuration in trigonal crystal field and adopting the method of complete diagonalization. In the calculation, all spin singlet states and spin triplet states hae been considered, then the calculation result is exact. The present theory shows that g parallel > g perpendicular, D < 0 or g parallel < g perpendicular, D > 0 are due to the different distortion of ligands along C3 axis. The optical spectra and the EPR spectrum of Ni2+:CsMgCl3 with D3d symmary are calculated by using the theory. The agreement of theory with the experiment shows that the present theory is reasonable.
