Expression of type X collagen and matrix calcification in three-dimensional cultures of immortalized temperature-sensitive chondrocytes derived from adult human articular cartilage.

Chondrocytes isolated from normal adult human articular cartilage were infected with a retroviral vector encoding a temperature-sensitive mutant of the simian virus 40 large tumor antigen and a linked geneticin (G418)-resistance marker. G418-resistant colonies were then isolated, ring-cloned, and expanded in serum-containing media. Several immortalized chondrocyte cell lines were established from the clones that survived, some of which have been maintained in continuous culture for over 2 years. Despite serial subcultures and maintenance as monolayers, these cells retain expression of markers specific for cells of the lineage, namely type II collagen and aggrecan, detected immunocytochemically. We also examined the phenotype of three of these immortalized cell lines (designated HAC [human articular chondrocyte]) using a pellet culture system, and in this report, we present evidence that a prototype of these lines (HAC-F cells) expresses markers normally associated with hypertrophic chondrocytes. When HAC-F cells were cultivated in centrifuge tubes, for periods of up to 63 days, at 39 degrees C with mild and intermittent centrifugation they continued to express both lineage markers; total type II collagen/pellet remained stable, whereas there was a temporal decrease in cartilage-specific glycosaminoglycans content. In addition, in the presence of ascorbate but in the absence of a phosphate donor or inorganic phosphate supplement, the cells also begin to express a hypertrophic phenotype characterized by type X collagen synthesis and extensive mineralization of the extracellular matrix in late stage cultures. The mRNA encoding type X collagen was detected in the cell pellets by reverse transcriptase polymerase chain reaction as early as day 2, and anti-type X collagen immunoreactivity was subsequently localized in the matrix. The mineral was characterized by energy-dispersive X-ray microanalysis as containing calcium (Ca) and phosphorus (P) with a Ca:P peak height ratio close to that of mineralized bone tissue. The unexpected phenotype of this human chondrocyte cell line provides an interesting opportunity for studying chondrocyte maturation in vitro.

Effects of high calcium intake on fat digestion and bile acid excretion in feces of veal calves.

We tested the hypothesis that apparent digestibility of fat by veal calves is determined by the participation of bile acids in the process of fat absorption and is, therefore, negatively associated with bile acid excretion in feces. Veal calves were fed milk replacers that contained whey protein and either a low (5.2 g of calcium/kg of air-dried diet) or high (12.4 g of calcium/kg of air-dried diet) concentration of calcium. The high calcium milk replacer contained extra calcium in the form of calcium formate. Final body weight was not significantly influenced by diet after the milk replacers had been fed for 27 wk. Feces were quantitatively collected during wk 23 of the trial. The high calcium milk replacer reduced apparent fat digestibility by 5.6 percentage units and increased bile acid excretion in feces by 90% compared with the low calcium milk replacer. The extra calcium intake decreased apparent absorption of magnesium and phosphorus. We proposed that a high intake of calcium by veal calves would increase the amount of insoluble calcium, magnesium, and phosphate complexes in the intestinal lumen, which, because of the binding of bile acids, would exclude bile acids from the process of fat digestion and inhibit reabsorption of bile acids. As a result, fat digestion is impaired, and bile acid excretion in feces is enhanced. The complex formation in the ileal lumen also explains why high calcium intake reduces magnesium and phosphorus absorption in veal calves.

[Analysis of X-ray images of giant-cell tumor of bone of rare locations].

OBJECTIVE: To analyse X-ray images of giant-cell tumor of rare locations. METHODS: To retrospectively review the X-ray films, CT scans and arteriograms of 48 cases of surgically and/or pathologically confirmed giant-cell tumor of bone of rare locations. RESULTS: Of the 48 cases, expansive cystic destructive changes of the bone with soap-bubble like appearence was found in 30 cases, osteolytic destruction in 10 cases, soft tissue mass outside of bone with sinking, flattening and local expanding of bone in 4 cases, invading into neighboring bone in 4 cases, hyperplasia and sclerosis of bone in 3 cases and staining of abnormal blood vessels and tumors in 3 cases. CONCLUSION: For the diagnosis of giant-cell tumor of bone of rare locations, X-ray plain film is of great value. CT is even better than plain film; arteriogram is helpful for diagnosis and treatment planning.

The effect of blockage of macrophage on tumor-suppressive activity of polyactin B.

OBJECTIVE: To investigate the tumor-suppressive activity of polyactin B (PB) and the life-span of tumor bearing mice treated with PB and the anti-tumor mechanisms of PB by blockage of macrophage in vivo. METHODS: Thirty-two S180 sarcoma-bearing (s.c.) BALB/c inbred mice were divided into 4 groups, i.e., control group (8 mice), PB group (8), Trypan blue (TrB) group (8) and PB + TrB group (8). Five days after PB injection (i.p.), the tumor size of the mice was measured, and the ratio of TNA to TTA in HE section of the tumor maximal area was calculated and the pathological features of the tumors were observed. Forty S180 sarcoma-bearing (i.p.) ICR mice, which were randomly divided into 4 groups as above, were used for observation of the life-span. RESULTS: PB could strongly inhibit the growth of S180 sarcoma with the tumors regressing completely in 3 of 8 mice, prolong the life-span of mice bearing S180 sarcoma, increase the infiltration of macrophages and lymphocytes in the periphery and inside of the tumors. In addition, there were more prominent hemorrhagic necrosis, neutrophil infiltration and microthrombi in small vessels around the necrotic areas in the tumors of PB treated group. The tumor-suppressive effect of PB disappeared after macrophage system blockage with TrB. In PB + TrB, TrB, control and PB groups, the size of tumors reduced, and the life-span of mice increased in successive order. CONCLUSIONS: (1) PB is a strong activator of macrophage. (2) In vivo the tumor-suppressive effect of PB is initially mediated by PB activated macrophage system. Then other immunocompetent cells, e.g., T-cells, NK-cells, etc, activated directly and/or indirectly by PB, may play a very important role in tumor suppression. (3) PB-activated macrophages are more sensitive to TrB blockage. Therefore, using TrB + PB to block macrophage system is more effective than TrB alone. (4) Blockage of macrophage function might influence the life-span of the mice.

Adrenal sparing surgery for renal cell carcinoma.

OBJECTIVE: To investigate the feasibility of adrenal sparing surgery for selected renal cell carcinoma (RCC), since the necessity of routine adrenalectomy during radical nephrectomy for RCC has been challenged in recent years. METHODS: 178 patients underwent perifascial nephrectomy in a 16-year period. Of these patients, 96 had ipsilateral adrenal gland preserved during nephrectomy and 82 underwent concomitant adrenalectomy. 75 excised adrenal specimens were examined pathologically. Disease specific survival rates were assessed according to the pathological stage of the tumors. RESULTS: Of the 75 patients, 53 presented a macroscopically normal adrenal gland without pathohistological changes. The other 22 patients were suspected to have adrenal metastasis intraoperatively, while only 5 of them were confirmed to have adrenal involvement by histopathology. Two patients in the adrenal gland preservation group developed ipsilateral adrenal recurrence and synchronous or metachronous contralateral adrenal metastasis during follow-up, although both were documented to have a normal-appearing adrenal gland intraoperatively. Five patients with adrenal metastasis and 2 patients with adrenal recurrence had large renal tumors. The survival difference among subgroups of patients undergoing adrenalectomy or with adrenal gland left in situ was not statistically significant. CONCLUSION: Adrenal sparing surgery could be done for patients with small renal tumors along with macroscopically normal ipsilateral adrenal glands.

[Relationship between helicobacter pylori infection and cirrhosis of liver].

Twenty-five (71.4%) of 35 cirrhotic patients were infected with Helicobacter pylori (Hp), compared with 21 (65.6%) of 32 patients with gastric or duodenal diseases (P > 0.05). The Hp infection rate in patients with liver lesion was higher than that of patients without liver lesion (79.3% and 33.3%, respectively). Ten (83.3%) of 12 patients with hepatic encephalopathy were infected with Hp, compared with 15 (65.2%) of 23 patients without hepatic encephalopathy. The cirrhotic patients have a high Hp infection rate and those infected are at higher risk for hepatic encephalopathy.

[Ultrastructural morphometric studies of fat storing cells in human chronic viral hepatitis].

OBJECTIVE: To study the dynamic changes of fat storing cells (FSC) in human chronic viral hepatitis. METHODS: Liver biopsies from 5 chronic persistent hepatitis (CPH), 5 mild chronic active hepatitis (CAH), 6 moderate CAH, 5 severe CAH cases and 8 normal liver tissues were used to investigate the number, appearance and organellae of FSC in non-fibrotic areas by cell counting under light microscope with semi-thick sections and morphometric study under electron microscope. RESULTS: The number of FSC in non-fibrotic areas of CAH groups were decreased significantly compared with the normal group and CPH group; the percentages of transitional cells tended to increase along with the increase in degree of severity of liver fibrosis, some of the FSC were transformed into fibroblasts. CONCLUSIONS: During the progress of liver fibrosis caused by CAH, FSC were constantly transformed into fibroblasts which tended to migrate to the fibrous septum of the hepatic lobules.

[The effect of tissue inhibitor of metalloproteinase-2 gene (TIMP-2 gene) on gastric carcinoma cells in vitro and in vivo].

OBJECTIVE: To study the effect of TIMP-2 gene on the invasive behavior of gastric carcinoma cells and to explore the possibility of blocking tumor metastasis. METHODS: TIMP-2 gene was transferred into human gastric cancer cell line SGC-7901 by lipofection technology. The ability of tumor cell invasion was detected by Boyden chamber in vitro and tumor invasion animal model in vivo. Type IV collagen and type IV collagenase were detected by immunohistochemical technique. RESULTS: The invasive ability of gastric cancer cells SGC-7901 transfected with TIMP-2 gene was significantly inhibited in vivo and in vitro. The expression of type IV collagenase in the untransfected group was higher than in the transfected group. CONCLUSIONS: TIMP-2 gene transfection can reduce the ability of gastric cancer cell invasion in vitro and in vivo.

Effects of beta-amyloid peptide on transient outward potassium current of acutely dissociated hippocampal neurons in CA1 sector in rats.

OBJECTIVE: To explore the mechanism of beta-amyloid peptide (beta-AP) in Alzheimer's disease at ionic channel level. METHODS: Hippocampal CA1 neurons of 7-21 days' rats were acutely dissociated and the effects of beta-AP on transient outward potassium current were observed by a whole-cell recording patch clamp technique. RESULTS: beta-AP can significantly block transient potassium current in dose-dependent, time-dependent and partly voltage-dependent manners. CONCLUSION: beta-AP may decrease the membrane conductance of K+ channels in hippocampal neurons, playing an important role in the pathophysiological mechanism of Alzheimer's disease.

[Assessment of effect of Shexiang Baoxin pill for labour angina pectoris patients with computerized tomography myocardial imaging].

OBJECTIVE: To assess the effect of Shexiang Baoxin Pill (SXBXP) for labour angina pectoris. METHODS: Effect of 66 patients treated with conventional treatment or conventional treatment plus SXBXP were observed and compared with 99mTC-MIBI single photon emission computerized tomography (SPECT) target cardiograph to estimate ischemia myocardial area. RESULTS: The total effective rate was much higher in patients treated with SXBXP than that in patients treated with conventional treatment only (87.8% vs 56%, P < 0.05). The effective rate on ECG in the two groups was 70.7% and 52.0% respectively, without significant statistical difference between them(P > 0.05). While significant difference was observed in ischemia area of myocardial image between the two groups after treatment, 18.2 +/- 8.2% vs 23.8 +/- 9.8%, P < 0.05. Moreover, the recurrence of angina pectoris and cardiac event were also less in the SXBXP group. CONCLUSION: The SXBXP was effective for labour angina pectoris.

[The effect of antioxidant sodium ferulate on human lymphocytes apoptosis induced by H2O2].

OBJECTIVE: Sodium ferulate is a potent antioxidant purified from ferula assafoetida L. The effect of sodium ferulate on apoptosis of human lymphocytes induced by H2O2 in vitro was studied. METHODS: Apoptosis was detected by light and electron microscopic, DNA electrophoresis and Flow cytometric cell cycle analysis. mitochondrial function was detected by MTT method. BCL-2 and Bax expression was detected by confocal laser scanning microscope. RESULTS: The morphological and biochemical changes characteristic of apoptosis were partially restored after treatment with sodium ferulate, Immunofluorescence assay showed a decrease in BCL-2 and an increase in Bax oncogene proteins in lymphocytes treated with H2O2 and an improvement of cell apoptosis was found after sodium ferulate treatment. CONCLUSION: Oxidative stress can induce apoptosis in lymphocytes, and this induction can be partly prevented by sodium ferulate.

[Effect of aloe-emodin on proliferation of vascular smooth muscle cells after arterial injury].

OBJECTIVE: To study the effect of Aloe-emodin (AE), an active ingredient of Rhubarb, on the kinetics of proliferation of smooth muscular cells (SMCs) cultured in vitro after rabbit iliac arterial injury. METHODS: Forty-eight hours after de-endothelialization (balloon endothelial denudation), the iliac arteries of the Japanese white rabbits were isolated and the smooth muscle cells were cultured primarily. AE was added to culture medium containing 10% fetal calf serum (FCS). The cultures were pulse-labeled with 3H-TdR and TdR uptake into VSMC were measured and the cell cycle of the cultures were analyzed by using flow cytometer. RESULTS: Compared with control, when the concentration gradient ranged from 10(-1) to 10(-5) g/L, the amount (cpm, count per minute) of 3H-TdR uptake into SMCs has significant differences (P < 0.05) and 10(-1) and 10(-2) g/L AE showed strong inhibitory effects on TdR uptake into VSMC and the percentage of inhibition [% inhibition = (cpm without AE - cpm with AE)/cpm without AE x 100%] was more than 90%. AE displayed concentration dependent inhibitory effects. The percentage of cells in G0/G1 phase was increased, but the percentage of cells in S phase was decreased in AE group, the transition of SMC cycle phase from G0 to S was blocked. CONCLUSIONS: AE is a strong inhibitor to the proliferation of SMCs and the pharmacological action of AE might reduce SMC proliferation in vivo and decrease intimal hyperplasia of restenosis.

[The diagnosis of non-pituitary space-occupied lesions in sphenoidal sinus and sellar area].

OBJECTIVE: To improve the diagnostic accuracy of non-pituitary lesions in sphenoidal sinus and sellar area. METHODS: Twenty-four cases with non-pituitary lesions in sphenoidal sinus and sellar area were analyzed. Eighteen cases were benign, including 7 sphenoidal sinus cyst, 6 craniopharyngioma, 2 chordoma, 1 nasal polyp extending into the sella, 1 neuronoma in sella, and 1 pinealoma. Six cases were malignant, including 2 malignant neuroendocrinoma in sphenoidal sinus, 1 adenoidocystic carcinoma, 1 sphenoidal sinus low differentiated carcinoma, 1 nasopharyngeal carcinoma extending into sella, 1 lung cancer metastasis to sphenoidal sinus. Clinical presentation, examination, imaging study, diagnosis and treatment were discussed. RESULTS: Among 24 cases, 18 had headache, 17 had visual symptoms. In all cases the space-occupied lesions in sphenoidal sinus and sellar area were verified by CT scan. CONCLUSIONS: 1. Headache at different levels was the commonest symptom for benign and malignant lesions. The visual loss was another common symptom. It is important to realize the relationship between visual symptom and space-occupied lesions in sphenoidal sinus and sellar area; 2. Imaging study is very important for the diagnosis of the lesions in sphenoidal sinus and sellar area; 3. It is helpful to do needle-aspirating biopsy under endoscopy to confirm the pathologic diagnosis. The benign lesions were operated on, and malignant lesions were treated by combined operation and radiation.

[The diagnosis and treatment of cerebrospinal fluid rhinorrhea].

OBJECTIVE: To summarize our experience in managing cerebrospinal fluid(CSF) rhinorrhea. METHODS: Twenty-nine cases with cerebrospinal fluid rhinorrhea from 1982 to March of 1997 were reviewed. The duration of CSF rhinorrhea varied from 3 months to 23 years. The ages of the patients ranged from 7 to 72 years. RESULTS: The causes of CSF leak were spontaneous in 18 cases, traumatic in 6 (1 had two accidents and got two different leaking sites) and iatrogenic in 5 cases. Twenty-two cases underwent surgical repair. Overall closure rate was 81.8%. The causes, symptoms and signs, sites of leakage and surgical techniques were analyzed. The classification, diagnosis and treatment of CSF rhinorrhea were discussed. CONCLUSION: Transnasal extracranial repair undertaken by otorhinolaryngologist got better closure rate(93.8%) in this group. Transnasal endoscopic repair could afford excellent view, facilitate precise tissue graft placement and get better closure rate.

Aging of human mature erythrocytes is like a process of apoptosis in enucleated cell.

Apoptosis of nucleated cells is well known, but how about the unnucleated cells is still not elucidated. In the present paper, the morphological and biochemical features of the aged erythrocytes were observed and compared with the characteristic events of apoptosis. Membrane of aged erythrocytes tends to shrink, protrude, from vesicle and lose lipid asymmetry. Aged erythrocytes were removed by phagocytosis. Both of the events are very similar to the apoptotic nucleated cells. The authors suggested that aging of erythrocytes is also a process of apoptosis.

[Discrimination of black inks by FT-Raman spectrometry].

Six brands of black inks on paper were examined by FT-Raman spectrometry. The results showed that the black inks could be discriminated by FT-Raman spectrometry. As the method is easy, quick and undestructive to samples, it is useful in black ink examination.

[Inhibitory effect of Howiinol A(GHM-10) on the synthesis of biological macromolecules in L1210 cells].

Howiinol(GHM-10), a new compound isolated from Goniothalamus howii, had been shown to have marked anticancer activity both in vitro and in vivo. In the present study, the effect of GHM-10 on the biosynthesis of macromolecules in L1210 cells was investigated. By using [3H] labeled precursor incorporation assays, we found that the biosyntheses, of DNA, RNA and protein in L1210 cells were inhibited markedly after the cells were pretreated with GHM-10 in 4-12 micrograms.ml-1 for 6 h and the inhibition of DNA synthesis was the most obvious one. We also used the methods of [3H]TdR incorporation curve, shift of UV-absorption spectrum and shift of fluorescence spectrum to investigate the mechanism of inhibitory action of GHM-10 on DNA biosynthesis. The results showed that the inhibition of DNA synthesis already became irreversible after the L1210 cells were treated with GHM-10 in 6-8 micrograms.ml-1 for 1 h, indicating that GHM-10 may induce damage on DNA molecular structure. However, when calf thymus DNA was treated with GHM-10, the UV-absorption spectrum and the fluorescence spectrum of the DNA were not changed significantly. It is suggested that GHM-10 was not able to intercalate into DNA molecules or to damage the structure of DNA directly.

[Studies on the anticancer effect of howiinol A, a new compound isolated from Goniothalamus howii].

The effect of howiinol A (GHM-10) on the growth of several cancer and normal cells were studied, using the methods of cell growth curve determination, MTT test and soft-agar colony formation assay. The results showed that GHM-10 exhibited potent inhibitory effect on cancer cells with an IC50 of 2 micrograms.ml-1 approximately. Normal cells, especially bone marrow progenitor cells, were shown to be less sensitive to GHM-10. A drug-resistant cell line, KB/VCR200, was found to be as sensitive as its parental KB cells. GHM-10 did not induce HL-60 cell differentiation. In murine transplantable tumors, GHM-10 was found to have significant therapeutic effect on mice bearing solid tumor hepatoma H22, Lewis lung carcinoma and S180 ascites.
