Regulation of parathyroid hormone-related peptide (PTHrP) gene transcription: cell- and tissue-specific promoter utilization mediated by multiple positive and negative cis-acting DNA elements.

The gene encoding PTH-related peptide (PTHrP) is expressed in a wide variety of normal and neoplastic tissues. Increased PTHrP gene expression in and secretion of PTHrP by specific tumors directly contributes to the development of malignancy-associated hypercalcemia in vivo. To define the genetic elements important for the control of PTHrP gene transcription, we used the reverse transcription polymerase chain reaction to delineate the control of promoter utilization and the splicing patterns of the exons encoding 5'-untranslated sequences. The majority of normal and neoplastic human tissues contained PTHrP mRNA transcripts initiating from both the up-stream (P1) and down-stream (P2) human PTHrP promoters. Furthermore, the downstream promoter was preferentially used by a factor of more than 30-fold. P1-initiated transcripts contained RNA species both with and without exon 2 (E2) sequences, except in the pancreas, adrenal, and stomach, where E2-containing sequences predominated. The transcriptional activities of P1, P2, and P1 + P2 were assessed by transfection of the corresponding PTHrP-chloramphenicol acetyltransferase (CAT) fusion genes into heterologous cell lines. Fusion genes containing P2 sequences were more transcriptionally active than fusion genes containing P1 sequences. The transcriptional activities of P1 + P2 in their natural tandem orientation were additive in rat keratinocytes and human JEG choriocarcinoma cells. In contrast, the activity of P1 + P2 was less than that of P2 alone in hamster BHK fibroblasts and InR1-G9 cells, and human HeLa cells. Analysis of the transcriptional properties of 5'-deleted human PTHrP-CAT constructs revealed the presence of multiple positive and negative DNA sequences (within both P1 and P2) functionally important for human PTHrP gene transcription. Distinct positive and negative DNA elements were also identified from analysis of 5'-deleted rat PTHrP-CAT fusion genes. The results of these experiments provide evidence for cell- and tissue-specific utilization of 1) distinct human PTHrP transcription start sites and specific patterns of 5'-exon splicing and 2) multiple positive and negative DNA control elements, important for the regulation of human and rat PTHrP gene transcription.

Quantitative computer analysis of the sounds of isolated motor unit action potentials.

To develop quantitative measures of motor unit action potential (MUAP) sounds, we correlated Fourier domain features of isolated, individual MUAPs with classic time domain measurements. There were moderate correlations between amplitude or duration measurements, and absolute low-frequency (39 to 234 Hz) energy and amplitude of the peak frequency, and high correlations between the composite time domain feature of amplitude x duration, and total energy, amplitude of the peak frequency, and absolute low-frequency energy. Polyphasic potentials have multiple peaks in the magnitude component of the Fourier transform. Phase information appears to convey the "crisp" sound of MUAPs close to the recording electrode. The clinical description of "large" or "small" MUAPs by sound is likely based on absolute low-frequency energy, and incorporates both amplitude and duration information. We conclude that features of isolated MUAPs may be analyzed in the Fourier domain, and that they correlate closely with traditionally used measures of known diagnostic significance. The sound of the EMG used by clinical electromyographers is amenable to quantitative analysis.

[Genetic profile of LIBP/1 inbred strain derived from the Kunming outbred stock of the mouse].

To make the genetic profile of the LIBP/1 inbred strain obtained from Kunming mice, the most widely used outbred stock in China, 26 loci were examined. The genotypes of four kinds of coat color genes were a/a, B/B, c/c and D/D. The results of testing 21 biochemical marker genes showed Akp-1b, Amy-1a, Car-2a, Ce-2a, Es-1b, Es-3a, Es-10a, Es-11a, Gpd-1a, Gpi-1a, Gus-1b, Hbbs, Idh-1a, Ldr-1a, Mod-1a, Mup-1b, Pep-3b, Pgm-a, Sep-1b, Tam-1c, and Trfb. The H-2 gene loci were Kb and Db.

Recombinant PDGF from lower vertebrates: receptor binding and immunochemical analysis with metabolically labeled growth factor.

We used a baculovirus vector/insect host cell system to express cDNA clones of PDGF A genes from mouse and frog (Xenopus laevis). The insect host cells process PDGF A subunits from either frogs or mice into biologically active AA homodimers with yields in the range of 0.5-1.0 mg/liter of culture medium. The recombinant PDGFs can be metabolically labeled with 35S-cysteine for use in radioreceptor and radioimmunoassays. Neutralizing polyclonal antisera can be raised against the mouse and frog PDGFs. These antisera are markedly species-specific in action. However, in radioreceptor binding assays and bioassays for mitogenic activity, human, mouse and frog PDGF AA homodimers occupy and activate murine PDGF receptors with equal efficiency.

Effects of ammonium chloride and sulfate on acid-base status and calcium metabolism of dry Jersey cows.

Eight nonlactating, nonpregnant Jersey cows were used in a crossover experiment with two 28-d periods. The control diet consisted of corn silage plus a concentrate mix (68:32, DM basis). The treatment diet was the same, except that NH4Cl and (NH4)2SO4 (98 g of each/d per cow) were added to the concentrate. Cows fed the treatment diet had lower blood pH, higher ionized Ca in blood, and more urinary excretion of Ca, titratable acid, and ammonium than cows fed the control diet. For cows fed the treatment diet, ionized Ca in blood was greater after equal amounts of Na2-EDTA were infused to both treatment groups, and treatment cows recovered faster after infusion of Na2-EDTA was stopped than did control cows. The treatment diet induced mild metabolic acidosis and increased the cows' ability to maintain normal blood Ca concentrations; it potentially could reduce incidence of milk fever.

Effects of diet magnesium on acid-base status and calcium metabolism of dry cows fed acidogenic salts.

The objective was to study effects of dietary Mg on acid-base status and Ca metabolism of Holstein cows fed acidogenic diets with relatively high Ca concentrations. Eight nonlactating, nonpregnant Holstein cows were used in a switchback experiment with three 28-d periods. The normal Mg (.2%, dry basis) diet consisted of corn silage plus a concentrate mix supplemented with NH4Cl (126 g/d per cow) and (NH4)2SO4 (126 g/d per cow). The high Mg (.37%, dry basis) diet had MgSO4 substituted for an equivalent amount of S supplied by (NH4)2SO4 in the normal Mg diet. Cation-anion differences of the two diets were -302 (normal Mg) and -289 (high Mg) meq/kg of dietary DM. Compared with cows fed the normal Mg diet, those fed high Mg tended to have higher blood pH and plasma concentrations of total Ca but lower plasma concentrations of P and lower urinary excretion of ammonium and net acid. Cows fed the high Mg diet also tended to increase Mg excretion with a decrease in urinary excretion of Ca. Metabolic responses to intravenous infusion of Na2-EDTA were similar among cows fed either diet. Results indicate that increasing Mg intake of cows fed acidogenic salts was of no advantage with regard to Ca metabolism.

[Studies on polypeptides. IX. Synthesis and antigen specificity of pre-S region fragments of hepatitis B surface antigen].

Seven peptide fragments of HBsAg Pre-S region have been synthesized by liquid phase and solid phase methods. All peptide fragments were linked to protein carrier by carbodiimide and glutaraldehyde methods. The antigen specificity of the peptide--protein conjugates was assayed by anti-HBsAg polyclonal antibodies and anti--"a" monoclonal antibody. P1 and P5 were found to have higher antigen specificity.

[Effects of various recombinant human hematopoietic growth factors on proliferation of blast cells in acute myeloid leukemia in vitro].

In vitro proliferative response of the blast cells from 21 AML patients to hematopoietic growth factors (IL-3, GM-CSF, G-CSF and MCSF) was investigated. Proliferation of AML cells in the majority of cases was induced or promoted by one or more CSFs, among which the stimulation of IL-3 was the most effective. Spontaneous proliferation of the blast cells was also observed in half of the cases and could be inhibited as well as promoted by some CSFs. It is suggested that in vitro proliferation of AML cells varies from patient to patient and that CSF plays important roles in leukemogenesis.

[Effect of oxytocin and cholecystokinin octapeptide (CCK-8) on electroacupuncture (EA) analgesia].

The effect of oxytocin (OT) and cholecystokinin octapeptide (CCK-8) on EA analgesia was studied in rats. The increase of 20.8-39.8% and 9.0-45.0% in pain threshold was observed respectively when ICV of CCK-8 or naloxone was combined with EA, these increases were lower than that in saline-EA group significantly, while the simultaneous ICV of OT and CCK-8 or OT and naloxone in combination with EA produced the increase of 76.2-116.6% and 41.8-104.5% in pain threshold separately. These results showed that only a small part in the role of OT enhancing EA analgesia was blocked by CCK-8 and naloxone. The data suggest that the role of OT in EA was not entirely dependent upon the endogenous opiate peptides.

[Hemodynamic effects of amrinone on cardiopulmonary decompensated chronic cor pulmonale].

Amrinone was given intravenously combined with the routine treatment to 10 patients with late chronic cor pulmonale in exacerbation stage. The initial loading dose was 1.5-2 mg/kg followed by the maintenance dose of 15 micrograms/kg. min for 6 hours. Hemodynamic and blood gas monitoring showed that pulmonary artery pressure, pulmonary vascular resistance index, right atrial pressure and pulmonary capillary wedge pressure decreased significantly and maintained the level markedly below baseline after amrinone administration (the extents decreased were 0.53-1.1 kPa, 80-140 dyn.s.m2.cm-5, 0.27-0.40 kPa and 0.40-0.67 kPa respectively. P < 0.05 for each parameter) and that cardiac index and stroke volume index increased identically (the extents increased were 0.3-0.6 L/min.m2 and 2-4 ml/beat.m2 respectively, P < 0.05 for each parameter). There were no significant changes in systemic artery pressure, arterial blood gas analysis and blood platelet count after amrinone administration. We suggest that amrinone may be effective in the treatment of cardio-pulmonary decompensated chronic cor pulmonale.

[Effect of Asu-AVT on electroacupuncture (EA) analgesia].

The Asu-AVT (1,6-aminosuberic acid -8-arginine-vasotocin) in an analogue of 8-arginine-vasotocin (AVT) which is one of pineal hormones. The effect of Asu-AVT on the pain threshold and EA analgesia was studied in rats. An increase of 16.2-41.5% in pain threshold was observed within 70 min. after ivc of Asu-AVT (75ng), while the Asu-AVT injection in combination with EA produced a significant increase of 164.6-309.1% in pain threshold, which was much higher than that in the saline-EA group (p < 0.05-0.01). The effect of Atu-AVT is analogous to that of oxytocin and arginine-vasopressin. The data indicate ivc of ASu-AVT not only elevates the pain threshold, but also enhances the EA analgesia. These results suggest that the pineal hormone, AVT may play a role in the EA analgesia.

[Identification of seven snake-drugs by iso-electric point].

This paper deals with the identification of seven snake-drugs by means of iso-electric point of protein. The experimental results show that different species of snake-drugs have different iso-electric points. The experimental data are accurate and easily reproducible (CV less than 0.5%). The difference of the averages of iso-electric points of the seven snake-drugs (P less than 0.01%) is extremely significant and thus provides a scientific criterion for the identification of peeled or powdered snake-drugs.

Cytotoxic activity of trewiasine in 4 human cancer cell lines and 5 murine tumors.

Trewiasine (TWS) is a mytansinoid compound. It possessed a significant cytotoxic activity against various human cancer cell lines in vitro. U937 cells, which were more sensitive to the TWS, required TWS 1 microgram.ml-1 to inhibit cell growth over 90% (P less than 0.01). TWS also showed activities against murine tumors in vivo, such as the ascitic tumors S180, hepatoma, U14, and solid tumor Lewis lung carcinoma. Depression of leukocytes was not seen when mice were given ip TWS 10 or 50 micrograms.kg-1.d-1 x 7 d. TWS 0.1-1 micrograms.ml-1 caused no sister chromatid exchange induction in Chinese hamster cell line V79.
