New assays were compared with tritiated thymidine (3HTdR) and trypan blue dye exclusion for evaluating hepatocyte viability on cytodex 3 beads and in microcapsules, the matrices used in bioartificial liver support systems. They were the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) 5 min qualitative assay and the 3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4 -sulfophenyl)-2H -tetrazolium, inner salt (MTS) 1 h quantitative assay. Both tetrazolium salts are cleaved in active mitochondria, the reaction occurring, therefore, in living cells only. After bathing at 39 degrees C with MTT for 5 min, porcine or rat hepatocytes on cytodex 3 beads were detached by collagenase while those in microcapsules were released by citrate treatment or passage through a fine needle. Cell viability was determined directly by microscope. The MTT 5 min metabolic inclusion test and MTS 1 h quantitative assay results correlated closely with those obtained by the 3HTdR, trypan blue dye exclusion, and fluorescein diacetate (FDA) methods. Both the new assays are sensitive, accurate, simple, and time-saving.

It is important to create a dynamic link method to link distributed patient data across multiple hospitals on an "as needed" basis because the pre-defined links (an item of data has a character or group of characters that indicates the storage of another item of data) are difficult to be managed, or can only be established in part, or are not necessary to be pre-defined in many cases, especially in linking the descriptive data such as history data with the corresponding examination data across multiple hospitals. A method of linking electrocardiogram (ECG) with clinical data dynamically in a Common Object Request Broker Architecture (CORBA) environment has been achieved and verified in a real computing environment to approach to this goal. By this method, distributed patient data can be linked dynamically by a CORBA-based query engine and temporal mapping no matter where they are located on the Internet. The necessary temporal information is provided by either computing or human being. Since multiple time-axes for different databases are involved in, some temporal reasoning methods (such as mapping occurrences across temporal contexts and determining bounds for absolute occurrences, etc.) are applied to this study, and a series of temporal mappings including the first mapping, the secondary mapping, the contextual mapping, the extended mapping, the previous mapping and the next mapping are created. In comparison with the pre-defined link, the major strengths of this method are the dynamic link on an "as needed" basis, no limitation of institutional boundaries, easy creation, simplifying the data storage, and the high flexibility, etc.
Aberrant expression of the platelet-derived growth factor alpha receptor (PDGF alpha R) has been linked to developmental abnormalities in vertebrate models, and has been implicated in multiple disease states in humans. To identify cis-acting regulatory elements that dictate expression of this receptor, we generated transgenic mice bearing the reporter gene beta-galactosidase (lacZ) under the control of a 6-kb promoter sequence. Expression of lacZ was monitored throughout embryonic development, with special focus on nervous tissue, skeleton, and several organ systems wherein PDGF alpha R expression is thought to play a pivotal role. In several independent transgenic mouse strains, lacZ expression recapitulated predominant features of PDGF alpha R gene expression during mouse development. These results demonstrate that critical tissue-specific regulatory elements for PDGF alpha R expression are located within a 6-kb upstream region of the PDGF alpha R gene.
OBJECTIVES: To study the secondary pathological changes in the central nervous system after injury and the role of dynorphin A1-13 in hypoxi-ischemic brain injury in neonatal rats. METHODS: Changes of concentrations of dynorphin A1-13 immunoreactives in some brain areas at different times after brain injury and the effect of injecting 8 microliters dynorphin A1-13 anti-serum into the medulla pool on pathological process of brain injury were observed in perinatal cerebral hypoxia ischemic rat models prepared by permanent ligation of right common carotid artery combined with a temporary systematic hypoxia at 37 degrees C in 7 day old Wistar rats. RESULTS: Concentrations of dynorphin A1-13 in cortex, hypothalamus and hippocampus increased significantly after the injury. Microinjection of anti-dynorphin A1-13 serum into the medulla pool 1 h preinjury might markedly reduce brain edema and improve the physiological condition. CONCLUSIONS: Dynorphin A1-13 plays a role in the pathophysiological process of brain hypoxia-ischemia and excessive amount of dynorphin A1-13 had some detrimental effect on the process.
OBJECTIVE: To study the incidence of thrombosis of small pulmonary arteries and arterioles during the exacerbation stage of chronic cor pulmonale. METHODS: 49 autopsy cases died from the exacerbation of chronic cor pulmonale were chosen as the study group, while other 103 autopsy cases without chronic cor pulmonale and disseminated intravascular coagulation (DIC) as control group. Morphologic and morphometric studies were taken on lung tissues and other organs with focus on the thrombi in small pulmonary arteries and arterioles. RESULTS: 44 cases of the study group had multiple thrombi in small pulmonary arteries and arterioles, the incidence is 89.8%, and in 9 of them, thrombi in proximal pulmonary arteries co-existed, the incidence is 18.4%, 80% of the thrombi existed in pulmonary arteriole. In control group, only 3 cases had thrombi in small pulmonary arteries and arteriole, the incidence is 2.9%. All thrombi adhered to endangium, implying that they were in situ thrombi. No intravascular thrombosis were found in other organs. chi(2) test showed that the incidence of thrombosis and the number of thrombi in small pulmonary arteries and arteriole in cases with chronic cor pulmonale were significantly higher than that of the control group (P < 0.01). CONCLUSIONS: Multiple in situ thrombosis in small pulmonary arteries and arterioles is a prominent and common pathological change during the exacerbation stage of chronic cor pulmonale. The study suggests a new diagnostic and therapeutic concept and provides a morphological and theoretical basis for the clinical application of anticoagulants or even the thrombolytic agents for the cases of chronic cor pulmonale in the exacerbation stage.
OBJECTIVE: To verify from the aspects of immune senescence and free radical metabolism that "removing blood stasis as well as tonifying kidney is an ideal way to delay aging" and further investigate the anti-aging mechanism of this therapy. METHODS: In the first period, the old mice were separately fed with the herbal decoction of the recipe of tonifying kidney, or of removing blood stasis, or of tonifying kidney and removing blood stasis for four weeks, and then IL-2 production of splenic lymphocytes, IL-1 production of peritoneal macrophages, SOD, CAT and GSH-Px activity in erythrocytes, and LPO contents in liver and brain were compared between the young control group and the old control group, or the old control group and each of the old medication groups. In the second period, the adult mice inhaled ozone or the splenic lymphocytes from adult mice were cultured in the medium containing peroxide hydrogen. The effects of free radicals on immune system and the reversion of the herbal decoction of the recipe of removing blood stasis as well as tonifying kidney were observed. RESULTS: The activities of IL-1, IL-2, SOD, CAT and GSH-PX in old mice were significantly lower than those in young mice, and LPO contents were higher. The three herbal decoctions all have reversing effects on each of these parameters except that the decoction of removing blood stasis has inhibitory effects on IL-1 and IL-2 production. The effect of the decoction of removing blood stasis as well as tonifying kidney was the best among the three decoctions, and it can also reverse the impaired production of IL-2 by ozone in vivo and peroxide hydrogen in vitro. CONCLUSIONS: The therapeutic method of removing blood stasis as well as tonifying kidney is an ideal way to delay aging, and its effects to improve immune senescence is correlated with the decrease of free radical damages.
OBJECTIVE: To make clinical analysis and pathological study on 40-case Gullain-Barré syndrome with sural nerve biopsy. METHODS: A total of 90 cases of GBS were retrospectively analyzed of which sural nerve biopsies were performed in 40 cases. The type of GBS was determined according to Asbury and Ropper criteria, and its correlation with pathological findings was studied. RESULTS: In the 40 biopsy cases, demyelination was the predominant pathological change in 38 cases. Mild axonal degeneration was also seen in 13 of the 38 cases. Prominent axonal degeneration was seen in only 2 of 13 cases. The increased monocyte infiltration and varied capillary permeability were observed in 19 of 25 cases. Onion-bulb like changes were seen in 6 cases. CONCLUSION: We studied the correlation between paresthesia and the pathological findings of sural nerve biopsy and concluded that sural nerve biopsy is very useful in the diagnosis of GBS and its variant.
Debilitating diseases such as osteo- and rheumatoid arthritis, trauma and bone cancer can be treated by using prostheses. The wear resistance of materials for joint replacement can affect the service-quality and service-life of prostheses. Materials for joint replacement and friction and wear were briefly reviewed in this paper.
SD rats were fed with 1% cholesterol and 0.2% cholate for eight weeks. Sodium alginate and carrageenan, two kinds of soluble algae polysaccharide, were added to the fodder for the trial groups and none added for the control one. Results showed that 5% sodium alginate could lower lipid levels in the blood and liver and platelet aggregation (P < 0.05) in rats, but not affect their food intake and growth. Addition with 5% or 10% carrageenan had no obvious effects on lowering blood lipid and liver total cholesterol (P > 0.05), could not lower the ratio of liver weight to body weight, and had no stable (or sure) effect on lowering platelet aggregation. Ten percent of carrageenan could decrease their mean body weight by 12.17% in rats (P > 0.05), but a few rats suffered from slight diarrhea.
The aluminum hydroxide gel, propolis and oil adjuvant inactivated vaccines were developed by silecting good immunity, representing principal epidemic serotype I GVF44 of Gardnerella vaginalis of fox in civil. The results showed that the aluminum hydroxide gel vaccine in safety was better than propolis and oil adjuvant vaccines, the most optimal inactivated concentration of formalin is 0.1%. Efficient immunity doses are 4 billon bacteria/1 ml. There were on any bad influences to occur for fox after the aluminum hydroxide gel of three times immunity doses were injected. The inoculated foxes could resist challenge of a hundred ID50 virulent strains after the vaccine was injected to foxes for 21 d. Immunity period of the vaccine is 6 months storage period is 10 months at 4-10 degrees C. The field experiments confirmed that the vaccine possessed accuracy, stability and reliability.
In order to investigate the effect of repair of paratendon in tendon healing, two different ways were performed to repair the transected extensor tendons of chick's toe. End to end suture of the extensor tenon was performed in group 1 while the paratendon was also repaired simultaneously in addition to suture of the tendon in group 2. Gross observation and histological examination were undertaken in the 3rd and 6th week after operation. The result showed, in group 1, extensive adhesion and irregular proliferation of fibroblasts was found in the 3rd week, severe adhesion and irregular arrangement of fibroblasts with less collagen fiber was found in the 6th week; while in group 2, smooth and regular "fusiform structure" was formed, slight adhesion and regular proliferation of fibroblasts were found in the 3rd week, adhesion disappeared and the structure of paratendon and tondon recovered in the 6th week. It was concluded that repair of extensor tendon and paratendon simultaneously could promote the intrinsic tendon healing and prevent tendon adhesion.
Thumb and finger reconstruction by the method of pedal digit transplantation had been successfully performed in 541 casee from 1977 to 1996, which contained 404 cases of thumbs and 78 cases of fingers. The thumb reconstruction was mainly the simple transplantation of distal phalanx (42 cases) and the compound transplantation of hallucal nail-cutaneous flap with iliac bone segment (16 cases) for the defect of thumbs in degree 1 and 2. The combined transplantation of hallucal nail-cutaneous flap with the joint and tendons of the second toe (34 cases) and the transplantation of the distal part of the second toe (182 cases) for the defect of degree 3 and 4. The combined transplantation of the second pedal digit with its metatarsalphalangeal joint (189 caese) for the defect in degree 5 and 6. The finger reconstruction was performed by anastomosis of the arteries of the digit with those of the fingers for 29 cases with the defect in degree 2 and 3, 60 cases with the defect in degree 4 and 5, and 17 cases with the defect in degree 6. One-hundred and four cases of versels vasiation were found in this group (19 cases with the pedal dorsal artery, 13 cases with the greater saphenous vein and 72 cases with the first dorsal metatarsal artery). The main point of the operation and the treatment of the vessel variations were discussed.
One eyeball of experimental rabbits was enucleated in routine procedure, and then implanted the biocative glass ceramic (BGC). No rejection was observed during the postoperation period. After three months of implantation, they were examined with ultrasound, and a venous-flow-like spectra was detected in the ceramic implant. After six months of implantation, the implans were examined histologically; the ingrowth fibrovascular tissue filled the 90% gaps of the implant. The results suggest that the bioactive glass ceramic implants made in China possess acceptable histocompatibility.
To analyse the anaerobic infection of maxillofacial surgery and estimate the efficacy of antianaerobic therapy, 45 patients were divided into two groups, tinidazole group and metronidazole group. Bacterial culture was positive before treating in all cases. There were Bacillus Melaniogenicus, Veillonella, Peptococcus and Peptostreptococus, etc. There was excellent efficacy in the treatment of maxillofacial anaerobic infection by tinidazole intravenously. After treatment, the result of bacterial examination was negative. The healing rate was 96.4% in 28 cases which used tinidazol, but 82.4% in control group which used metronidazole. The value of white blood cell and the function of liver and kidney pro- and post-treatment were not significantly different (P < 0.05) by comparision.
To improve the function of filtering blebs after trabeculectomy, we divided randomly 44 patients (60 eyes) with moderate or late glaucoma into mitomycin C and control groups (30 eyes per group). Two small surgical sponges were soaked with 0.2 mg.ml-1 mitomycin C solution. One sponge was put on the top of the superficial scleral flap, the other was placed underneath the scleral flap and contacted with the bed of the scleral flap. After 5 minutes the wound was then irrigated with normal saline. All patients were followed up from 1 to 12 months (7.9 +/- 4.0 months). We found that: (1) functional filtering blebs were 92.9% in the mitomycin C group and 55.2% in the control group (P < 0.01), (2) the decreasing percentage of intraocular pressure (IOP) in the mitomycin C group was much more than that in the control group (P < 0.05), (3) the successful rates of the operation in MMC group were higher than that of the control group, and the visual field was effectively maintained in MMC group.
