[A study on relationship between plasma heat stress protein 70 and cytokine in patients with heat apoplexy].

OBJECTIVE: To study the change of levels of plasma heat stress protein 70 (HSP70) and cytokine and their interrelationship in pathogenicity of heat apoplexy. METHODS: Plasma HSP70 was determined with Western blot, and plasma interleukin-2 (IL-2), soluble interleukin-2 receptor (sIL-2R) and interleukin-6 (IL-6) were determined with enzyme-linked immunosorbent assay (ELISA). RESULTS: Plasma level of HSP70 was 4 211.2 +/- 1,286.2 (integral optical density) in average in the patients of heat apoplexy group, 4,137.8 +/- 1, 207.5 in those of severe heat apoplexy group, and 6,043.5 +/- 1,354.8 in control group, with very significant difference (P < 0.01). Plasma IL-2 level was (64.3 +/- 32.3) pg/ml and (200.0 +/- 50.0) pg/ml and sIL-2R (54.9 +/- 33.3) U/ml and (167.5 +/- 89.0) U/ml in the patients with heat apoplexy and in controls, respectively, with very significant difference. But, there was no significant difference in IL-6 between varied groups. CONCLUSIONS: The above-mentioned results suggest that establishment of heat acclimatization correlates to the interaction of network of neuroendocrine-cytokine-HSP, in which decrease in plasma IL-2 was one of the important factors causing reduction of HSP70 expression. Balance of network regulation was broken by pathological factors, finally resulting in heat apoplexy.

[Lung injury caused by passive smoking and its effects on cytokines in rats].

OBJECTIVE: A rat model with chronic bronchitis was replicated by passive inhalation of cigarette smoking fume to study its long-term effects on lung injury and nitric oxide (NO), interluekin-6 (IL-6), interleukin-8 (IL-8). METHODS: Levels of nitrogen dioxide (NO(2)) and nitrogen trioxide (NO(3)) were measured with spectrophotometry in rats indicating their level of nitric oxide (NO). Levels of IL-6 and IL-8 were determined by enzyme-linked immunosorbent assay (ELISA). RESULTS: Levels of NO in serum, bronchial alveolar lavage fluid (BALF) and lung tissue in the smoking group were significantly lower than those in the normal controls (P < 0.01). But, levels of IL-6 and IL-8 were higher in the smoking group than those in the controls. CONCLUSION: Long-term passive smoking could cause injury of lung tissue to certain extent, reduction in secretion of NO in endothelial cells and damage to pulmonary vessels.

[The changes in serum antibody level after immunization with HFRS vaccine].

OBJECTIVES: To observe the changes in serum antibody level after mass immunization with vaccine against hemorrhagic fever with renal syndrome (HFRS) and to evaluate its efficacy and effectiveness in the prevalent areas. METHODS: Healthy people aged 16 to 60 years in the villages were recruited as study subjects, excluding those suffered from HFRS previously, going out for more than nine months and those with contraindications, and were randomly allocated into immunization and control groups with 10,460 and 16,159 persons, respectively. Specific IgG antibody was determined with indirect immunofluorescent assay (IFA) and neutralizing antibody (NA) was determined with micro CPE method. RESULTS: Two weeks after the full-course immunization, sero-conversion rate for IFA reached 100% in those sero-negative before immunization, with a 95% confidence interval of 96.3 - 100.0%, and that for NA 44.4%, with a 95% CI of 22.0% - 69.0%. Geometric mean titer (GMT) were 72.1 and 4.6 for IFA and NA, respectively. Booster immunization was provided for them one year later. Positivity of IFA and NA was 28.6%, 83.3%, 75.0%, 53.1%, 22.6% and 14.8%, 55.6%, 35.0%, 31.3%, 26.0%, before booster immunization, two weeks, one year, one and a half years, two years after booster immunization, respectively. CONCLUSION: HFRS vaccine had good immunogenicity, but its duration of serum antibody sustenance was relatively short.

[p16, p53 and c-erbB-2 gene expression in bladder carcinoma].

OBJECTIVE: To investigate the expression of p16, p53 and c-erbB-2 proteins in bladder carcinoma and to analyze their correlation with tumor grade, clinic stage and metastasis. METHODS: Immunohistochemical method was used to detect the p16, p53 and c-erbB-2 gene expressions in 75 cases of bladder carcinoma. RESULTS: Positive rates of p16, p53 and c-erbB-2 in 75 cases of bladder cancer were 41.3%, 44.0% and 40.0%, respectively. p16 and c-erbB-2 gene expressions were related to the tumor grade and clinic stage (P < 0.05); p53 and c-erbB-2 gene expressions were related to the clinic stage and metastasis (P < 0.01). Oncogenes and/or antioncogenes altered expressions were found in 77.3% cases of bladder carcinoma, and oncogenes and/or antioncogenes with multiple altered expressions were found in 53.3% cases. CONCLUSION: The analysis of multiple oncogene and/or antioncogene is more valuable than that of single oncogene and/or antioncogene. The altered expression of oncogene c-erbB-2 and antioncogenes p16 and p53 might play a promotive role in the tumorigenesis and development of bladder tumors.

[Caspase 3 in the cytokines induced acute leukemia cell apoptosis].

OBJECTIVE: To explore the variation and significance of caspase 3 activity in the process of G-CSF and GM-CSF regulated NB4 cell apoptosis. METHODS: Cell morphology, flow cytometry and DNA fragmentation assay were used to demonstrate the apoptosis induced by G-CSF and GM-CSF. Caspase 3 activity was detected by spectrofluorometry, and its inhibition test by AC-DEVD-CHO. RESULTS: G-CSF could induce NB4 cell apoptosis, while GM-CSF could not. Caspase 3 activity was significantly higher in the apoptotic cells than in control cells. The G-CSF induced NB4 cell apoptosis was efficiently inhibited by AC-DEVD-CHO. CONCLUSIONS: G-CSF and GM-CSF exert different effects on the apoptosis of leukemic cells. G-CSF induced apoptosis of leukemic cells through activation of caspase 3.

[Quantitative determination of EAIgG by ABC-ELISA and its clinical significance].

OBJECTIVE: To explore a method for diagnosis of autoimmune hemolytic anemia (AIHA) and evaluate its clinical significance. METHODS: Erythrocyte associated IgG (EAIgG) was quantified in 60 healthy people, 44 patients with warm antibody AIHA (WAIHA), 108 non-immune anemia and 44 other immune diseases (total 256 cases) by using ABC-ELISA were estimated. RESULTS: EAIgG in WAIHA patients were significantly higher than those in other groups (P < 0.01). In the 44 WAIHA patients, 38 were Coombs positive and 6 Coombs negative. EAIgG in Coombs negative was significantly high. The dynamic changes of EAIgG in 12 WAIHA patients were observed during treatment. When patient's condition got better, Hb and RBC increased and EAIgG decreased. CONCLUSION: Quantitative detection of EAIgG is an index in the diagnosis of WAIHA, especially in AIHA with negative Coombs' test. It is also a useful factor for evaluating therapeutic effect and prognosis in WAIHA.

[Clinical study on treatment of atherosclerosis obliterans by integrated traditional Chinese and Western medicine].

OBJECTIVE: To investigate the mechanism in treating atherosclerosis obliterans (ASO) with integrated traditional Chinese and western medicine (TCM-WM). METHODS: The changes in level of vasoactive factors, such as endothelin (ET), nitric oxide (NO), apoprotein and the iconographic changes in angiography before and after treatment were observed in 40 patients of ASO. RESULTS: All patients had high levels of plasma ET and low level of NO, abnormal metabolism apoproteins, and showed the signs of trunk artery obstruction and marked decrease of collateral circulation in affected limb. After treatment, the plasma ET, NO and apoprotein levels recovered to normal range in 30 markedly effective cases, as compared with those before treatment, the difference was significant (P < 0.01) and their collateral circulation in the affected limb significantly increased, showing obvious improvement of peripheral circulation. CONCLUSION: The therapeutic mechanism of TCM-WM in treating ASO might be related with its effects on vascular endothelium, lipid metabolism adjustment, collateral circulation establishment and microcirculation improvement.

[Detection and activity analysis of gelatinases in bone and soft tissue tumors].

This study was aimed to investigate the expression of gelatinases (MMP-2 and MMP-9) in mesenchymal tumors and the roles they act during invasion and metastasis. 45 cases of bone and soft tissue tumors with various pathologic categories were subjected to immunohistochemistry and gelatin zymography analysis. The results showed that the positive immuno-staining ratio and the activity of both MMP-2 and MMP-9 were higher in malignant tumors than in benign tumors and normal tissue. In malignant tumors, the positive stained cells usually gathered in the marginal area of tumor foci or the invading area, or formed distinctive positive-stained cell clusters, indicating their relationship with the aggressive biologic behavior of this kind of malignant tumor.

[Effects of bacterial protective liquid on radiation biology effect of mouse bone marrow cells].

To study the relationship between bacterial protective liquid (BPL) and the biological effect of mouse bone marrow cells by gamma-ray irradiation, apoptosis and the number of CD4+, CD8+ and NK cells were analyzed by flow cytometry(FCM), the results showed that BPL decreased the apoptosis and increased the proliferation of cells(P < 0.01). The theorem of action was related with raising the percentage vitality of NK cells and the quantity of CD4+ and CD8+. This study proved that BPL might have protective effect against radiation injury.

[Domoic acid and glutamate receptors].

Domoic acid is a kind of neurotoxin produced by diatoms and accumulated in food chain, which is harmful to the human health. The N-methyl-D-aspartate receptor and kainic acid/alpha-amino-3-hydroxy-5-methylisoxazole-4-propionate receptor could be activated by domoic acid. The concentration of the intracellular Ca2+ could be elevated, causing a series of physiological and biochemical changes. The possible mechanisms of neurotoxicity induced by domoic acid through glutamate receptors are discussed.

[Pathologic changes of lymphatic capillaries after injection of U14 cells in rat tongue perineoplastic area].

OBJECTIVE: 1. To establish a model of neck lymphatic metastasis of cancer by injecting tumor cells into the rat tongue; 2. To observe the relationship between pathologic changes of perineoplastic lymphatic in different stages of implanted tumor growth and lymphatic metastasis of tumor cells. METHODS: 60 Kunming rats with injection of 5 x 10(6) U14 tumor cells into each rat tongue were chosen and randomized into 6 groups. In every 5 days, specimens of rat tongues with implanted tumor and neck lymph nodes with possible metastatic tumor cells were removed from each group of rats. Cryosections of rat tongue were processed with a modified enzyme-histochemical staining technique, using 5'-nucleotidase with HE to demonstrate lymphatic capillaries. Attention was paid to pathologic change of lymphatic capillaries in perineoplastic area of rat tongue in different growth stages of implanted tumor. Routing wax sections of lymph nodes processed with HE staining were examined under light microscopy to determine whether there were any metastasis tumor cells. RESULTS: 1. Metastasis to neck lymph nodes could be observed 15 days later after injection of U14 tumor cells into rat tongue. The situation of metastasis of lymph nodes varied greatly from tumor cells in bundle within marginal sinus to complete occupation of lymph nodes by U14 cells. 2. Tumor cells penetrated the lymphatic capillary by passing singly or in clumps between endothelial cells or through some destroyed endothelia. A great number of lymphatic with dilated cavity was found nearby the tumor tissue which facilitated tumor cells to enter the lymphatic cavity. 3. There were much more amount of lymphatic capillaries with dilated cavity around tumor mass comparing with that of normal tongue of the control group rats, and the number of lymphatic capillaries with opening cavity tend to increase with growth of implanted tumor. CONCLUSION: Direct and objective view of lymphatic capillaries can be obtained by means of the modified enzyme-histochemical staining technique, using 5'-nucleotidase with HE to demonstrate lymphatic capillaries; A great number of lymphatic capillaries with dented cavity in normal physical state are forced to be dilated during process of tumor growth, thus in turn increase the possibility for tumor cells to penetrate the lymphatic capillary by passing singly or in clumps between endothelial cells.

[The function of benzol peroxide in the induction of Syrian golden hamster tongue carcinoma by chemical carcinogen].

OBJECTIVE: To testify the function of promoter(benzol peroxide) in the induced tongue carcinogenesis of Syrian golden hamster as well as the function of carcinogen (DMBA). METHODS: In this study, group 1: 20% benzoyl peroxide in acetone solution was brushed to the right tongue border of Syrian golden hamster, followed by 0.5% 7, 12-dimethybenzanthrancene(DMBA) in acetone, twice a week, lasting 20 weeks. Group 2: 0.5% 7,12-dimethybenzanthrancene(DMBA) in acetone was painted on the right tongue border of golden Syrian hamster in the manner of group 1. Group 3: 20% benzoyl peroxide in acetone solution was used with mentioned methods. RESULTS: Group 2: showed 90% (27/30) carcinogenesis-rate in tongue of Syrian golden hamster. While it was 100% (30/30) of induced tongue carcinoma and 3/30 lymph node metastasis was found in group 1. No tongue carcinogenesis was found in group 3. The former two groups had the manifestations of hyperplasia of epithelium cell, carcinoma in situ, invasive and cervical lymph node metastatic carcinoma in the process of experiment, but it was 2 weeks shorter in group 1 than that of group 2. Both groups had the same features under eye and light microscope. The samples were high-differentiated squamous cell carcinoma histologically through HE stain. CONCLUSION: This study indicated that benzoyl peroxide could increase carcinogenesis of golden hamster tongue which induced by DMBA. Consideration of promoting agent should be taken in carcinogenesis as well as carcinogen.

[Treatment of hemangioma in oral and maxillofacial region with pingyangmycin injection].

OBJECTIVE: To investigate the indication and result in hemangioma in oral and maxillofacial region with Pingyangmycin injection. METHODS: 1282 patients (554 males and 728 females) with hemangioma in oral and maxillofacial region have been treated in our hospital during the 10-year period from May 1990 to March 1999. The patients range between 0.5-84 years old. Lip, cheek and tongue were usually involved. 1211 patients were treated with injection of Pingyangmycin(about 0.89 mg/ml) and dexamethasone (about 0.55 mg/ml), 1 time/1-2 weeks. RESULTS: The curative rates of cavernous hemangioma and strawberry hemangioma were 93.87% and 84.90%, but that of plexiform hemangioma was 22.22% and those of portwine stain and central hemangioma of the jaws were 0%, respectively. Serious complications associated with Pingyangmycin injection, such as pulmonary fibrosis were not seen. CONCLUSION: Injection of Pingyangmycin is selective primary method for cavernous and strawberry hemangioma treatment.

[Simultaneous intrusion and retraction of the anterior teeth using a three-piece base arch].

OBJECTIVE: To evaluate the effects of the three-piece base arch on overbite correction of Class II malocclusion. METHODS: 20 patients with high angle, flared incisors were treated using a three-piece base arch appliance. The intrusion force of four upper incisors was adjusted to approximately 50 g. The line of force action was 2 mm distally to the resistant center(RC) and the retraction force was 20 g, the right and left posterior segments were joined by a palatal bar. Cephalograms were taken before treatment (T1) and six months after treatment (T2). RESULTS: The upper molars moved mesially 0.60 +/- 0.35 mm and the distance of the vertical extrusion was 0.80 +/- 0.52 mm. The distances of the upper central incisor retraction and intrusion were -4.20 +/- 2.12 mm and 3.10 +/- 0.54 mm respectively. The RC of the central incisor retracted -4.12 +/- 1.96 mm and intruded 3.20 +/- 0.66 mm. The axial inclination of the upper incisor-palatal plane changed from 123.21 degrees +/- 4.26 degrees to 116.00 degrees +/- 3.96 degrees. CONCLUSION: The three-piece segmented approach can effectively intrude and retract the upper anterior teeth for flared incisors and deep overbite.

[Dynamic study of hepatocellular pathological change and uptake rate of 125I-insulin during experimental hepatocarcinogenesis].

This study was designed to observe the hepatocellular pathological change and uptake rate of 125I-insulin during experimental hepatocarcinogenesis in rats and clarify the possible mechanism of increasing uptake of 125I-insulin. 80 SD rats were divided into 2 groups, control and experimental groups. All of the rats were given common feed, and the rats of experimental group were given extra diethylnitrosamine (DENA) 70 mg/kg each week. At 6th, 11th, 15th and 20th week after start of the experiment, 10 control and 10 experimental rats were killed 1 hour post administration of 125I-insulin by tail vein. Blood, liver, lung, spleen, kidney, muscle and bone were collected; the radioactivity was measured and calculated %ID/g. The data of 2 groups were compared and examined with t-test. All of livers were pathologically examined. The results showed all livers of control group were normal. At 6th week, the surfaces of experimental rats' livers were coarse. Hepatocellular hyperplasia was observed, 125I-insulin-uptake rate was 1.86 time as much as that of control group. At 11th week, the liver's colour became lighter than that of control group. Hyperplasia and hepatocirrhosis were observed, 125I-insulin-uptake rate was 1.50 time as much as that of control group. At 15th week, hyperplastic nodules were observed in all experimental rats' livers. Hepatomacellulae were observed in 6 rat livers. 125I-insulin-uptake rate was 1.56 time as much as that of control group. At 20th week, the livers became enlarged out of shape. There were a lot of big or small greyish white nodules in all livers. Necrosis, liquefaction and hemorrhage were observed. Hepatomacellulae were observed in all of experimental rats livers. 125I-insulin-uptake rate was 1.46 time as much as that of control group. The differences of 125I-insulin-uptake rate between experimental and control groups were significant. These results demonstrated that the liver ability of uptaking 125I-insulin increased which mainly took place during the period of hepatocellular hyperplasia, and the hepatomacellulae kept this characteristic.

[Genetical analysis on cross incubation of Beauveria bassiana].

Heterokaryon of Beauveria bassiana was formed during the cross incubation of two vegetative compatible strains with genetic markers of actidione resistance and 34 degrees C tolerance. The chromosome(s) or its fragment successive losses, recombination and segreation led to haploidization during the conidia formation period. After at least 4 generations of parasexual cycle, the genetic character of heterokayon could get to relative stable. Genetic marker and RAPD analysis indicated that the combinants showed the phenomenon of preferential selection of one parental type by unrandom chromosome lossing and the gene of the other parental strain was suppressed or lost completely. Different culture medium, in vivo or in vitro, and different mixture ratio of original strain spores could affect parasexual process and then the preferential selection. The results also demonstrated the heterosis effect of cross culture.

[Histological study of the local-regional invasion of pyriform sinus carcinoma].

OBJECTIVE: In order to offer the basis for the operation of the pyriform sinus carcinoma, the local-regional invasion patterns of the pyriform sinus carcinoma were investigated. METHODS: Twenty-six surgical specimens obtained from patients who undergone laryngopharyngectomy were subjected to a whole organ section study. RESULTS: Tumors located in the lateral wall of the pyriform sinus spread mainly towards the lateral wall of the hypopharynx. Tumors located in the inner wall of the pyriform sinus spread mainly towards the larynx and opposite side pyriform sinus, the paraglottic space and thyroid cartilage were particularly frequent invaded. It was rare of the invading of the cricoid cartilage. The invasion of the epiglottis and preepiglottic space across the midline had not been found. There were two invading routines, the paraglottic space and preepiglottic space. The invasion of the paraglottic space was through the aryepiglottic fold and the inner surface of the thyroid cartilage. The invading of the preepiglottic space was through the aryepiglottic fold and the upper part of the inner surface of the thyroid cartilage. CONCLUSIONS: The invading of the preepiglottic space was not the contraindication of the partial laryngectomy. It is practicable to preserve the laryngeal function in most of the patients while tumors located on the lateral or inner wall of the pyriform sinus. There was a tendency to the invading to the contralateral side in the inner wall of the pyriform or postcricoidarytenoid region. Attention should be paid to the submucosal spreading in the postcricoidarytenoid region.

[Intracytoplasmic injection with sperm from ejaculation, epididymis and testis in treating different male infertility with oligo-asthenozoospermia or azoospermia ].

OBJECTIVE: To retrospectively study 400 intracytoplasmic sperm injection (ICSI) cycles by using three different sources of sperm. METHOD: Different male infertility with oligo-asthenozoospermia or azoospermia was treated by ICSI using sperm from ejaculation, epididymis and testis. RESULTS: In the ejaculation group (n = 277), the fertilization rate was 69.6% and the clinical pregnancy rate 28.9%. In the 104 cycles using epididymal sperm, the fertilization and clinical pregnancy rate was 65.9% and 37.5% respectively. And the fertilization and clinical pregnancy of testis sperm (n = 19) was 65.7% and 21.1%. The difference of fertilization and clinical pregnancy rate among the three groups were not significant. CONCLUSIONS: Male infertility of different causes can be treated by ICSI. The outcome has nothing to do with the common parameter of semen except that the sperm motility is 0, but the effectiveness of ICSI should be studied by long-term observation.
