In a continued search for leukaemomycin-blocked mutants of three leukaemomycin-producing strains IMET JA 3933, IMET JA 5142 and IMET JA 5570 of Streptomyces griseus, 32 mutants producing aerial mycelium and spores were detected. Furthermore, in all mutants cosynthetic capability has been observed. This report describes characterization of leukaemomycin-blocked mutants obtained by mutagenic treatment experiments using NTG and combined UV-/X-rays. According to the biosynthetic capability for anthracyclinones or other pigments the mutants could be divided into six classes. The first class contains 14 leukaemomycin-blocked mutants unable to synthesize anthracyclinones. Besides two classes of mutants (12)synthesizing well-known anthracyclinones as epsilon-rhodomycinone, 7-deoxy-epsilon-rhodomycinone, 11-deoxy-derivatives of daunomycinone, three new classes of mutants (6) synthesizing reddish-brown, brown and blue-violet pigments on solid media with structures not elucidated as yet, will be described.

The properties of folic acid gamma-glutamyl hydrolase (conjugase) [EC 3.4.12.10] in rat bile and plasma were investigated. Conjugase activity in bile showed two pH optima; one at pH 4.5-5.0 and one at pH 6.7-7.5. The enzyme activity in plasma had a broad pH--profile with an optimum at pH 6.2-7.5. Conjugase activity from both bile and plasma was inhibited in the presence of the sulfhydryl reagent, p-hydroxymercuriphenylsulfonate, and stimulated in the presence of 2-mercaptoethanol. Conjugase activity in bile was inhibited by Zn2+ at pH 7.5 but not at pH 4.5 and was much more stable to heat at pH 4.5. No separation of the biliary conjugase activity measured at the two different pH values was obtained by Sephadex G-150 chromatography. Secretion of biliary conjugase was essentially constant over a 6-hour period when activity was assayed at pH 4.5 or pH 7.5. The enzyme in bile converted pteroyltriglutamate to a mixture of about 5% glutamate an 95% gamma-glutamylglutamate at either pH, whereas the enzyme in plasma produced 23% glutamate and 77% gamma-glutamylglutamate. The possible contribution of biliary conjugase to intestinal absorption of folate polyglutamates is discussed.

The folate-binding protein of rat liver mitochondria [Zamierowski, M. & Wagner, C. (1977) J. Biol. Chem. 252, 933-938] has been purified to homogeneity by a combination of gel filtration, DEAE-cellulose, and affinity chromatography. This protein was assayed by its ability to bind tetrahydro[3H]folic acid in vitro. the purified protein contains tightly bound flavin and has a molecular weight of about 90,000 as determined by sodium dodecyl sulfate electrophoresis. This protein also displays dimethylglycine dehydrogenase [N,N-dimethylglycine: (acceptor) oxidoreductase (demethylating), EC 1.5.99.2] activity which copurifies with the folate-binding activity. It is suggested that the role of the tetrahydrofolic acid is to accept the formaldehyde produced during the course of the reaction.
Assessed the utility of the cognitive constructs of Locus of Control and Psychological Differentiation as co-determiners of success on a biofeedback task. Eighty male and female psychology undergraduate students were divided into four groups based on median splits of scores on Levenson's Internal Locus of Control scale and on Witkin's Group Embedded Figures Test. Thus, four groups of subjects were identified: field-independent internals (Fl-I; n = 17), field-dependent externals (FD-E; n = 23), field-dependent internals (FD-I; n = 19), and field-independent externals (Fl-E; n = 21). The first two groups consisted of individuals whose locus of control orientation was congruent with their perceptual style and the last two contained the incongruent subjects. Fifteen subjects were randomly selected from each of the four groups resulting in a total sample of 60 subjects. All subjects were told to decrease their GSR using biofeedback during each of five, 2-minute trials. A 1-minute rest period was provided between each trial. Analysis of variance revealed that congruent subjects decreased their GSR significantly (p less than .01) more than incongruent subjects. Scheffe's test revealed that all subjects were able to learn to significantly decrease GSR. Additional research designed to examine the utility of the congruence-incongruence dimensions in biofeedback seems warranted.

A very through ocular examination was made in 48 hypertensive patients treated for 18 months with acebutolol in a daily dose of between 400 and 2400 mg. Abnormal symptoms in the form of a prickling sensation were seen in two cases. Microscopic punctate lesions of the conjunctive were seen in 9 subjects, either with the Bengal Pink or fluorescein tests. These very slight microscopic lesions were found in general only in the region of the palpebral slits, a zone normaly exposed to microtrauma, which decreases the value of this finding. The results of a Schirmer test were considered to be abnormal in six patients. These ocular abnormalities were generally mild and it could not be shown conclusively that they were related to treatment with acebutolol. Ocular surveillance, with examinations repeated every six months, indicated the absence of any harmful effect on the eye of prolonged used of acebutolol in the treatment of hypertension.
