T cell activation and retargeting using staphylococcal enterotoxin B and bispecific antibody: an effective in vivo antitumor strategy.

The aim of this work was to test for cure and immunity in a micrometastatic tumor model using in vivo T cell activation with staphylococcal enterotoxin B (SEB) and retargeting with antitumor x anti-CD3 F(ab')2 bispecific antibodies (bsAb). All studies were performed in C3H/HeN mice using syngeneic tumor cell lines. For survival studies, mice were injected intravenously on day 0 with CL62 (a p97-transfected clone of the K1735 murine melanoma tumor). Day-3 treatments included saline (control), SEB (50 gamma g intraperitoneal) with or without bsAb (5 micrograms i.v.). Cured mice, surviving beyond 60 days, were rechallenged with subcutaneous CL62, K1735, or a nonmelanoma control, AG104. SEB activation studies were performed with pulmonary tumor-infiltrating lymphocytes isolated from 10-day established CL62 tumors. Maximal tumor-infiltrating lymphocyte cytotoxicity was demonstrated 24 h following SEB injection, therefore bsAb treatments were administered 24 h after SEB. When survival was examined at 60 days, there were significantly more survivors in the group receiving SEB plus bsAb (70%) compared to the group receiving SEB alone (30%), and the controls (0%) (P = 0.02 and P < 0.01, respectively). Mice cured of CL62 using SEB alone or with bsAb demonstrated equal immunity to CL62, however, mice treated with SEB plus bsAb were more often immune to the p97-parental cell line, K1735(P = 0.001). Ag104 consistently grew in all mice. Results of these studies demonstrate that SEB plus bsAb can be effective, not only in curing tumors but also in providing protective immunity against targeted and non-targeted tumor antigens.

Regulation of melatonin production by catecholamines and adenosine in a photoreceptive pineal organ. An in vitro study in the pike and the trout.

The pineal organ of fish contains photoreceptor cells with structural and functional analogies to retinal photoreceptors. In these cells, the light/dark (LD) cycle influences the production of melatonin by controlling the activity of one of its synthetizing enzymes, serotonin N-acetyltransferase (NAT). The daily rhythm in NAT activity is generated endogenously in the pike but not in the trout pineal. We report here that in addition to the LD information, chemical factors are also involved in the control of melatonin production. Adenosine and two of its analogs stimulated or inhibited NAT activity and melatonin release in cultured pike and trout pineals, depending on the experimental conditions. It is believed that the nucleoside, produced locally, exerts a modulatory role on the neurohormonal output via still enigmatic mechanisms, involving a transmembranous carrier. Nocturnal melatonin production in cultured pike pineals was inhibited by alpha-adrenergic agonists and stimulated by a beta-adrenergic agonist. No effect could be induced in trout pineals cultured under similar conditions. Because melatonin production by pineal photoreceptors is apparently regulated by both light and chemical inputs, we propose they might be multieffector cells.

Airborne manganese particulates and methylcyclopentadienyl manganese tricarbonyl (MMT) at selected outdoor sites in Montreal.

This study aims to assess the atmospheric concentrations of methylcyclopentadienyl manganese tricarbonyl (MMT), respirable manganese (MNR) and total manganese (MnT) in certain specific microenvironments and to provide an estimation of human exposure to MnR. Sampling was carried out in five microenvironments: a gas station, an underground car park, downtown Montreal, near an expressway and near an oil refinery. The samples were collected using Gil-Air portable pumps during three days and were analyzed by instrumental neutron activation analysis (INAA). The mean concentrations of MnR, MnT and MMT were 0.036 microgram m-3, 0.103 microgram m-3 and 0.005 microgram m-3 respectively. The MnR/MnT ratios vary from 25% to 43% (mean 35%) while the MMT/MnT ratios averaged about 5%. Furthermore, the mean concentration of the MnR measured near the expressway (0.053 microgram m-3) is similar to the United States Environmental Protection Agency (U.S. EPA) reference concentration (RfC = 0.05 microgram m-3). The average daily environmental exposure dose to MNR is estimated at 0.010 microgram kg-1 d-1 and its contribution to the multimedia exposure (air, food and water) is low. The overall results show a lack of potential exposure to MMT and substantial concentrations of MnR near an expressway.

[Fertilization in mammals].

The mechanisms involved in gamete recognition and gamete fusion are identical to those allowing the fixation of somatic cells to a basal lamina and cell or viral fusion. Therefore specificity of the fertilization process only derives from the cytological originality of both gametes : acrosomal reaction and egg activation are two typical examples. Mitochondria, ARNs, proteins and metabolic reserves are brought by the oocyte. Sperm contributions to zygote development are essentially an oscillatory calcium protein, oscilline, and, the sperm proximal centrosome. In mammals, once the pronuclei are formed, paternal and maternal genomes never mix. A better knowledge of the female genital tract milieu, allowing long or very long sperm survival, is primordial to improve in vitro fertilization procedure.
