[Nucleo-cytoplasm ratio, DNA content of superficial and intermediate vaginal cells during the female genital cycle].

A quantimetric study of vaginal superficial cells is done every day of the sexual cycle on five normal women (no pill) with a cervical smear of type 2 (Papanicolaou classification). Our results show that nucelo-cytoplasmic ratio for 98% of the cells varies between 0.0040 and 0.0210. This level is independant of age and cycle duration and is proposed as reference for later studies of women under pill or with cervico-vaginal dysplasia and cancer.

[Influence of temperature on the motility of human spermatozoa].

The mean swimming speed of a suspension of human sperms has been estimated by applying a kinetic theory and using an image analysing computer. This technique allowed the study of the evolution of motility in terms of temperature: the global swimming speed significantly increases from 22 degree C to 31 degrees C, and then remains rather constant. However the percentage increases are very varying according to samples.

Alpha-glucosidase as a specific epididymal enzyme marker. Its validity for the etiologic diagnosis of azoospermia.

Azoospermic semen was obtained from 39 vasectomized men and 93 patients consulting for infertility. The latter underwent bioclinical investigations including measurement of plasma FSH and testicular biopsies. Carnitine content and alpha-glucosidase activity in semen samples were measured. The activity of alpha-glucosidase was determined systematically by a semiquantitative microtechnique and was verified by a spectrophotometric assay. A positive correlation was observed between carnitine and alpha-glucosidase activity. Both parameters were severely diminished in semen from the vasectomized men and the patients suffering from a complete obstruction of the genital tract. Enzyme activity was the most discriminant parameter in terms of sensitivity and specificity. The measurement of alpha-glucosidase in semen is a simple and sensitive method for determining the origin of azoospermia when used in conjunction with assays for plasma FSH.

[Seasonal variations of cellular composition of human semen (author's transl)].

Sperm samples taken fortnightly from 14 young europeans over a period of 8 months show evidence of a seasonal variation in the concentration of spermatozoa and in the frequency of abnormal sperm (teratospermy). Both factors are highest in winter and lowest in springtime. A study of 3,000 spermograms established during consultations for sterility over the last 2 years did not clearly reveal the same trends, however, a statistical analysis of the results leads to the conclusion that several hundred examinations per month would be needed to show a significant seasonal variation in such a heterogeneous sample.

Nuclear texture during the cell cycle in acute lymphoid leukemia.

Description of morphological and textural changes during cell cycle were studied in 31 diploid Acute Lymphoid Leukemia ALL cases. Feulgen stained nuclei were measured by image analysis. Cells with 2C, 3C and 4C content of DNA were considered as being in G1, S and G2 phase of cell cycle respectively. The main observed transformations associated with cell cycle were continuous increases in nuclear size and in quantities of black/white chromatin clumps and points. An overall correct classification of 92.5% was achieved by multivariate data analysis. However, significant differences between A L L cases were noticed.

Applications of laser scanning confocal microscopy to the observation of human oocytes and embryos.

Laser scanning confocal microscopy (LSCM) is a powerful and useful tool in developmental biology. The diverse applications of LSCM in biomedical field has led to advances in the microscopes themselves and the synthesis of novel specific probes for the observation of biological structures and the hypothesis of physiological process. LSCM was used to visualize the cellular actin cortex together with the chromatin in human arrested preimplantation embryos and in unfertilized oocytes obtained by in vitro fertilization. In embryos, we observed a majority of multinucleated blastomeres and some with fragmented nuclei or anucleated. LSCM also showed nuclei displaying the chromatin condensation and fragmentation patterns suggesting the phenomenon of apoptosis. Moreover, in arrested fragmented embryos of poor morphology, unequal sized blastomeres often showed cellular fragments without nuclei, compatible with aspects of apoptotic bodies.
