Testis weight as a percentage of body weight did not change following bilateral ligation of the efferent ducts (EDL) close to the epididymis, whereas following removal of part of the epididymis between the site of ligation and a point close to the junction between the caput and corpus (PCE), testis weight first rose linearly until Day 4 and then showed an exponential decrease between Days 4 and 28. After EDL, the perimeter of the seminiferous tubules rose for the first 7 days and then remained elevated, whereas after PCE, there was a linear decrease between Days 4 and 28. Following EDL, the percentage of altered and degenerated tubular cross-sections rose to about 30% and 10%, respectively, during the first 7 days after operation and then remained constant; after PCE, the percentage of altered tubules reached a maximum of 54% by 4 days and then fell, whereas the percentage of degenerated tubules continued to rise to 95% by 28 days. It would appear that all the effects of removal of a portion of the epididymis cannot be explained by blockage of the excurrent ducts, and a specific endocrine effect of the epididymis on the testis is proposed.
Doses (2.5 mg/kg) of rhodamine 6GO were given subcutaneously to five Charles-Rivel mice. After 15 minutes the testis and epididymis were observed with a fluoroscope and a fluorescent microscope which showed that rhodamine 6GO was present in a rhodamine-fluorescent zone (RF-zone) of the caput epididymidis. In this RF-zone, the fluorochrome was localised in the epithelial cells of the epididymal duct but not in the lumen nor the vascular system. At the edge of the RF-zone a wide band of connective tissue was present; the epithelium of the epididymal duct in this zone was higher and its tubular diameter greater than in the rest of the epididymal duct and spermatozoa were usually not present in the lumen. A possible role played by the epididymis in the endocrine control of the testicular function is suggested.
Adult Wistar rats were either partial (caput) and bilaterally epididymectomized or bilaterally efferentectomized, as controls of duct obstruction. The effects on testicular germinal epithelium were studied at 7, 9, 11, 13, 15, 17, 19, 21, 23 and 25 days after surgery. No abnormalities were detected in sham-operated animals. Epididymectomized animals showed different levels of alterations with progressive disruption of the seminiferous epithelium, emergence of multinucleated bodies and some tubules obliterated by degenerated cells and cellular debris. Half way through the experiment there were tubules lacking their epithelia, as well as the Sertoli cells. On the 25th day degeneration was so important that is affected not only the epithelium (missing in almost all tubules) but also the tubular morphology. Eventually efferentectomized animals showed a progressive alteration, but its level was much lower than that observed after partial epididymectomy, indicating a possible specific function of the caput epididymidis in the control of testicular function.
The application via i.v. jugular, in rats, of two fluorochromes (rhodamine 6GO and phosphine 3R) with a 10 min interval, allows the direct observation of nephronal corpuscles using an epifluorescent microscopical technique, as well as their eventual quantification and qualification into normal or semifunctional corpuscles. In rats previously treated by i.p. injection of gentamicin or netilmycin a decrease of the active nephronal population as well as an increase of the percentage of semifunctionality has been noticed.
The alterations of the epididymal F-zone caused by apical mesotestis bilateral cutting after scrotal exposition were studied in adult albino rats. The epididymis were removed at 2, 4, 7, 14, 21 and 28 days after surgery. With a semi-automatic program, from a IBAS 2000 image analyzer, areas, perimeters and maximum and minimum diameters were measured from both the tubular and the lumen sections of the caput epididymal F-zone. The animals with cutting of the apical mesotestis showed a significative increase with respect to controls for all the considered variables, indicating a tubular dilatation as a consequence of the surgical intervention. The mast cells present in the connective tissue of the F-zone of the control animals presented a typical fluorescent colour with Acridine Orange, while this was not observed in the animals with cutting of the apical mesotestis.
A virus isolate with the properties of a pararotavirus was found after routine analysis by RNA electrophoresis of 658 samples of diarrheic feces from hospitalized infants and young children in Mexico City. Of the patients included in this survey, which was initiated in 1977, 29% excreted rotaviruses which were detected by their characteristic RNA pattern after gel electrophoresis. The morphology and sedimentation coefficient of this pararotavirus, which was the apparent cause of a diarrhea with moderate dehydration in a 2-year-old male infant, were indistinguishable from those of rotaviruses, but its buoyant density in CsCl was slightly higher than that of simian rotavirus SA11. By electron microscopy, the viral particles showed a regular pattern of cavities or holes that constituted the 5- and 6-coordinated units of the virion with a structure characteristic of T = 12. The virion also was apparently composed of protein classes similar to those found in rotaviruses. Seroconversion in the patient and presence of anti-pararotavirus antibodies in most of the members of the family of the patient was shown by immunoelectron microscopy. Of the sera from 12 healthy adults which were examined by this technique, seven were negative, three were slightly positive, and only two were strongly positive.
Two cases are reported of subdural hematoma of the posterior fossa in normal-weight full-term newborn infants. The most salient factors in these cases were the lack of specific symptoms and signs indicating the nature and location of the lesion, the importance of computerized tomography for diagnosis, and the good results obtained with early surgical treatment.
Electron microscopy after negative staining of SA11-infected cell homogenates revealed that most of the viral particles are associated with membrane-like material. Many of the particles seemed to be fully enveloped in a membrane. This association could also be detected by the observed cosedimentation of viral proteins and cell membranes. Pulse-chase experiments showed that viral glycoproteins rapidly associate with membranes, whereas most of the structural proteins appearing in the soluble fraction immediately after the pulse were slowly chased into the membrane fraction. The membranes could be further fractionated into at least four fractions differing in density and containing a different distribution of viral proteins. Also, the distribution of label into each of these membrane fractions changed after long chase periods. The inhibition of glycosylation with tunicamycin yielded viral particles without an outer layer, but did not affect the described association with membranes. The possible relationship of this finding to the maturation of the virion is discussed.
The two doses, 5 and 7 mg/kg/day, were applied for periods of 14 or 28 days, with a 21 day break between two 14 days treatments in the latter case. Controls were only treated with physiological saline. A decrease in corporal weight and rectal temperature, as well as a decrease in ingestion and faeces together with urine increment were observed in the experimental animals during the first period of treatment. Normal levels were recovered during the break time, but the previous pattern reappeared during the second period of treatment. Suprarenals, liver, kidneys and spleen increased in some experimental groups when compared with controls whereas the thymus presented no significant differences. The studied organs showed signs of impairment.

A multiplex polymerase chain reaction (PCR) based on the simultaneous amplification of human papillomavirus (HPV) types 6/11, 16 and 18 in a single-step procedure was developed, using primers chosen in the E6-E7 region. The specificity and sensitivity of this technique have been proved by amplifying mixtures or various amounts of plasmid-containing HPV DNA; it allowed the detection of as few as 5-25 HPV DNA copies. Application of the multiplex PCR to 71 clinical samples showed that HPV DNA was detected in 80% (45/57 cases) of mucosal biopsies and 35% (5/14 cases) of cutaneous specimens. HPV 16 was predominant in high-grade CIN whereas HPV 6 and 11 were detected more frequently in genital condylomas and laryngeal papillomas. In cutaneous Bowen's disease HPV 16, 18 or 6/11 + 16 were detected and in squamous cell carcinomas HPV 6/11 or 16 were found. After sequence amplification with primers of one HPV type, the clinical samples displayed the same HPV types but the frequency of positive and coinfected lesions increased. Thus, multiplex PCR is a valuable technique for typing HPV DNA but coinfections may be underestimated.
UNLABELLED: In France, no case of transfusion-transmitted malaria has been reported since 1994, reflecting the efficiency of the blood donations screening. However, an insufficiently specific serological assay leads to an unjustified loss of red cell units. MATERIALS AND METHODS: A retrospective study included 55 sera previously disqualified for the presence of malarial antibodies. We used different assays detecting these antibodies: Paludix, Falciparum-Spot without or with the elimination of natural antiglobulins by two different methods. RESULTS: Using Paludix, 87% of sera were negative (P < 0.001). With the sera treated for the elimination of natural antiglobulins by neutralisation or absorption, Falciparum-Spot(TM) gave respectively 87% and 94.5% of negative results. CONCLUSION: When using assays avoiding non-specific fluorescence, the number of false positive results significantly decreased. Because the number of blood donors travelling in at-risk countries is continuously rising, a sufficiently specific method to detect malarial antibodies should save a lot of red cell units.
PURPOSE: 99mTc-MIBI, an alternative radiopharmaceutical for myocardial perfusion study has been proposed for use as a tumor imaging agent, including breast cancer, lung cancer, lymphomas, melanomas, and brain tumors. After routine radiation therapy, deteriorating clinical status or treatment failure may be due to either radiation changes or recurrent tumor. CT and MRI offer imperfect discrimination of tumor viability and radionecrosis. MATERIALS AND METHODS: Thirty-five malignant glioma patients with clinical deterioration were studied retrospectively. Tomoscintigraphy was performed 15 minutes after intravenous injection of 1110 Mbq 99mTc-MIBI. The images were obtained from a dual headed gamma camera using fan beam collimator. Transverse, coronal and sagittal views were reconstructed. RESULTS: A dramatic MIBI uptake was found in 31 patients. This uptake was correlated to tumor recurrence proven by histological fragments and/or the rapid, fatal evolution of these patients. Death occurred after the brain SPECT had been performed for those cases showing MIBI uptake, an average 5.48 months later. No MIBI uptake was found for these four remaining patients: their evolution can be currently considered to be a disease-free time. CONCLUSIONS: According to our results, the sensibility and specificity of 99mTcMIBI brain SPECT seems to be high. Moreover, this investigation is more accurate for discriminating tumor recurrence from radionecrosis than a CT scan or MRI.
