Antiviral activity of new triterpenic derivatives.

Twenty-seven triterpenic derivatives of glycyrrhetinic, oleanolic, ursolic and maslinic acid were tested for antiviral and cytotoxic properties. Twelve of them showed apparent antiviral activity against at least one of the tested viruses. The most active compounds were PAS-T45 and 48 which exerted a protection index of 4 on vaccinia virus.

Antimicrobial activity of 5-substituted-3-amino-1,2,4-triazoles.

Some dyes derived from 3-amino-5-(ortho, meta, and para-pyridyl)-1,2,4-triazoles were tested for antimicrobial activity. 13 compounds showed anti-staphylococcal activity and one had anti-candida activity. Some substances had antimicrobial activity only after light irradiation.

[Incidence of urinary bladder cancer in an industrialized area of Spain].

AIM: To identify all incident cases of bladder cancer in the county of Vallès Occidental (Spain), describe their histopathological characteristics, and make comparisons with other Spanish and European areas. METHOD: The study was carried out from the Corporació Parc Taulí (Sabadell). All new cases of bladder cancer in residents of the county Vallès Occidental, a highly industrialised area of Catalonia (Spain), were included between 1992 and 1994. Incidence rates of bladder cancer were adjusted and were compared with adjusted incidence rates reported by registries in other Spanish and European countries. RESULTS: 485 new cases were identified. Transitional cell carcinomas predominated (95.5%). The majority of tumours were diagnosed in their initial stages, 75.9% being superficial and 62.6% well to moderately differentiated. Bladder cancer was more common in men than in women, but women presented tumours of worse prognosis. The mean age at diagnosis was also higher in women than men (71 vs. 66 years, p = 0.03). The adjusted incidence rate in men (52.2 cases/100,000) was among the highest of the observed areas, whereas for women (5.4 cases/100,000) was relatively low. CONCLUSIONS: The incidence of bladder cancer among men in Vallès Occidental is among the highest in Europe, and intermediate for women. The high male/female ratio seen in all Spanish areas could be attributed to the fact that women in Spain have been less exposed than men to the risk factors, or their exposure occurred more recently.

[Temporary disability: analysis strategies].

OBJECTIVES: To apply different regression models to estimate rate ratios for temporary sick-leave (TSL) which may occur several times in the same individual during a period, and the frequency is not constant for the observation period. SUBJECTS AND METHODS: All workers employed more than 30 days between January 1st of 1992 and June 1st of 1995 were included into the population study. The following period was 1,259 days and the total number of workers included in the study was 2,306. During that period 2,649 TSL episodes were notified, which meant 85,947 lost days. Poisson regression, Generalised Estimating Equations (GEE) and Andersen-Gill modification of Cox regression modify by Wei (WLW) were applied. RESULTS: The highest TSL incidence rates were seen in women, lesser than 30 years old, cleaners, maintenance workers and auxiliary nurses, and those involved in shiftwork. This profile was not modified after applying GEE and WLW regression models, although confidence intervals were widened. CONCLUSIONS: TSL data does not fit Poisson regression assumptions, but GEE and WLW regression models do not appear as alternatives. Other conditional regression models would need to be explored to suitably analyse this data.

[Social class differences in bladder cancer in Catalonia].

OBJECTIVE: To assess the presence of socio-economic differences in bladder cancer in a heavily industrialised area of Catalonia, Spain and to evaluate risk factors associated with the presence of these differences. METHODS: 218 cases and 344 population controls resident in the area of Vallès Occidental, Barcelona, were identified prospectively and interviewed during 1993-1995. Socio-economic level was evaluated through attained education and social class on the basis of the last and longest held occupation. Logistic regression was used to derive adjusted odds ratios and 95% confidence intervals. RESULTS: There was no association between education and bladder cancer risk. Social class, evaluated on the basis of longest held job, was weakly related with bladder cancer, with the highest risk occurring in the highest social classes. This pattern was more pronounced when the analysis was done using the last occupation. Occupational exposures modified minimally these results. Among controls, the overall prevalence of smoking did not differ between social class but the prevalence of heavy-smokers (> 48.76 packs-year) was highest among subjects in social class V (35.5%) and lowest in social class I (10%). Adjusting for smoking enlarged social class differences. CONCLUSION: Similarly to the pattern in other European countries, no pronounced differences by social class were observed for bladder cancer in this area of Catalonia. The evaluation of social class patterns was crucially affected by the type of socioeconomic information examined. Measures of life-time experience should be preferred rather than socio-economic level at the time of the disease. Smoking seems to be one of the main mediators for the occurrence of bladder cancer differentials but other, yet unidentified risk factors, must be associated with bladder cancer risk, especially among the highest social classes.

Immunohistochemical study of pepsinogen C expression in cutaneous malignant melanoma: association with clinicopathological parameters.

BACKGROUND: The aim of this study was to evaluate the pepsinogen C expression in malignant cutaneous melanomas and analyze its possible relationship to clinical and pathological parameters. Pepsinogen C is an aspartyl proteinase primarily involved in the digestion of proteins in the stomach and represents one of the main androgen-inducible proteins in breast cancer cells. METHOD: Tumoral pepsinogen C expression was retrospectively analyzed in 35 paraffin-embedded tissues from patients with primary malignant cutaneous melanoma and in 10 samples from 10 benign lesions (4 dermal melanocytic nevi, 4 compound melanocytic nevi and 2 dysplastic melanocytic nevi), using immunohistochemical methods. RESULTS: The benign lesions were consistently negative for pepsinogen C, whereas 20 of the 35 malignant melanomas (57%) showed positive immunostaining for pepsinogen C. The percentage of pepsinogen C-positive tumors was significantly higher in men than in women (p=0.01) and in epithelioid melanomas than in fusocellular or mixed type melanomas (p=0.003). In addition, the percentage of pepsinogen-C positive tumors was positively and significantly correlated with lesion thickness (p=0.003), Clark's level of invasion (p=0.028) and tumor stage (p<0.001). CONCLUSION: Pepsinogen C could be a new prognosticator of unfavorable outcome in cutaneous malignant melanoma.

Efficacy of Lantigen B in the prevention of bacterial respiratory infections.

BACKGROUND: Respiratory tract infections are among the most common diseases both in adults and children. METHODS: This multicentre study, was carried out in 212 patients (118 adults with chronic respiratory diseases and 94 children with recurrent respiratory infections) at six Italian study centres. It was performed to establish the efficacy of Lantigen B for the prevention of bacterial complications and/or reduction of associated symptoms. Lantigen B is an oral product based on bacterial lysates of six different inactivated strains commonly involved in respiratory tract infections. RESULTS: The results of prophylactic treatment with Lantigen B in the population investigated were as follows: - the mean incidence of recurrence in the pediatric patients over the 6-month observation period of the study was 1.643 in the placebo group and 1.211 in the Lantigen B group (a relative reduction of 35.7%); - the mean incidence of recurrence in the adult patients over the 6-month observation period of the study was 0.73 in the placebo group and 0.56 in the Lantigen B group (a relative reduction of 30.4%). The improvement in other important parameters, such as the number of days with a high temperature, cough, and catarrh in the children and objective clinical findings at the lymph nodes of the neck, chest, and pharyngotonsillar region in the adults, provides further evidence of the efficacy of Lantigen B. CONCLUSIONS: This study demonstrates Lantigen B's effectiveness in the prevention of bacterial complications and suggests that it can be used in patients who are particularly at risk of infection (children, the elderly, diabetics, and immunocompromised patients) or those in whom an infection might aggravate a clinical picture that is already inherently complicated (diabetics again, but also patients with heart, kidney, or liver disease).
