Chlorophyll fluorescence lifetime determination of waterstressed C3- and C4-plants.

This article describes the effect of water stress on the room temperature chlorophyll fluorescence lifetime of plants of wheat (C3) and maize (C4). In addition, net CO2 assimilation rate (PN), stomatal conductance and the fluorescence quenching coefficients qP and qNP at steady state conditions were recorded. The overall fluorescence decay of the control plants can be described by an average decay time of 1 ns for both plant types. Water stress did not modify this parameter in the case of wheat, whereas a shortening of the decay was observed for waterstressed maize plants (tau = 0.45 ns). This shortening in the chlorophyll fluorescence decay was accompanied by an increase in the non-photochemical quenching (qNP). The photochemical quenching (qP) and therefore the electron transport via photosystem II remains unaffected by water stress. The most pronounced effect of the stress for both plant types was a decrease in PN due to a closure of the stomata.

Subunit of an alpha-interferon-responsive transcription factor is related to interferon regulatory factor and Myb families of DNA-binding proteins.

Alpha interferon stimulates transcription by converting the positive transcriptional regulator ISGF3 from a latent to an active form. This receptor-mediated event occurs in the cytoplasm, with subsequent translocation of the activated factor to the nucleus. ISGF3 has two components, termed ISGF3 alpha and ISGF3 gamma. ISGF3 gamma serves as the DNA recognition subunit, while ISGF3 alpha, which appears to consist of three polypeptides, is a target for alpha interferon signaling and serves as a regulatory component whose activation is required to form ISGF3. ISGF3 gamma DNA-binding activity was identified as a 48-kDa polypeptide, and partial amino acid sequence has allowed isolation of cDNA clones. ISGF3 gamma translated in vitro from recombinant clones bound DNA with a specificity indistinguishable from that of ISGF3 gamma purified from HeLa cells. Sequencing of ISGF3 gamma cDNA clones revealed significant similarity to the interferon regulatory factor (IRF) family of DNA binding proteins in the amino-terminal 117 residues of ISGF3 gamma. The other IRF family proteins bind DNA with a specificity related to but distinct from that of ISGF3 gamma. We note sequence similarities between the related regions of IRF family proteins and the imperfect tryptophan repeats which constitute the DNA-binding domain of the c-myb oncoprotein. These sequence similarities suggest that ISGF3 gamma and IRF proteins and the c-myb oncoprotein use a common structural motif for DNA recognition. Recombinant ISGF3 gamma, like the natural protein, interacted with HeLa cell ISGF3 alpha to form the mature ISGF3 DNA-binding complex. We suggest that other IRF family members may participate in signaling pathways by interacting with as yet unidentified regulatory subunits analogous to ISGF3 alpha.

[Morphological alterations of lymph nodes and thymus during the early course of SIV infection of rhesus monkeys].

Rhesus monkeys (M. mulatta) were i.v. infected with SIV mac251. Three phases of lymph node changes were observed. 1: physiological follicular hyperplasia (3 and 6 weeks p.i.). 2: Alterations of germinal centers: loss of follicular mantle zone, fragmentation or sclerosis (12 and 24 weeks p.i.). 3: Partial depletion of T-lymphocytes, accumulation of plasma cells, increased numbers of syncytial giant cells, hemophgocytosis in the sinuses (about 1 year p.i.). The thymus of the juvenile animals showed first changes 12 and 24 weeks after infection with focal loss of immature (and Ki-67 positive) cortical thymocytes, leading to severe accidental involution of the thymuses one year after infection and reduced numbers of Hassalls corpuscles. These investigations show the value of this animal model for the study of morphology and pathogenesis of AIDS.

Clinical relevance of L-carnitine-supplemented total parenteral nutrition in postoperative trauma. Metabolic effects of continuous or acute carnitine administration with special reference to fat oxidation and nitrogen utilization.

Carnitine-free total parenteral nutrition (TPN) is claimed to result in a carnitine deficiency with subsequent impairment of fat oxidation. The present study was designed to evaluate the possible benefit of carnitine supplementation on postoperative fat and nitrogen utilization. Sixteen patients undergoing total esophagectomy were evenly randomized and received TPN without or with L-carnitine supplementation (74 mumol.kg-1.d-1) during 11 postoperative days. On day 11, a 4-h infusion of L-carnitine (125 mumol/kg) was performed in both groups. The effect of supplementation was evaluated by indirect calorimetry, N balance, and repeated measurements of plasma lipids and ketone bodies. Irrespective of continuous or acute supplementation, respiratory quotient and fat oxidation were similarly maintained throughout the study in both groups whereas N balance appeared to be more favorable without carnitine. We conclude that carnitine-supplemented TPN does not improve fat oxidation or promote N utilization in the postoperative phase.

[Effect of L-carnitine supplemented total parenteral nutrition on postoperative lipid and nitrogen utilization].

During episodes of trauma carnitine-free total parenteral nutrition (TPN) may result in a reduction of the total body carnitine pool, leading to a diminished rate of fat oxidation. Sixteen patients undergoing esophagectomy were equally and randomly divided and received isonitrogenous (0.2 gN/kg.day) and isocaloric (35 kcal/kg.day TPN over 11 days without and with L-carnitine supplementation (12 mg/kg.day). Compared with healthy controls, the total body carnitine pool was significantly reduced in both groups prior to the operation. Without supplementation carnitine concentrations were maintained, while daily provision of carnitine resulted in an elevation of total carnitine mainly due to an increase of the free fraction. Without supplementation the cumulative urinary carnitine losses were 11.5 +/- 6.3 mmol corresponding to 15.5% +/- 8.5% of the estimated total body carnitine pool. Patients receiving carnitine revealed a positive carnitine balance in the immediate postoperative phase, 11.1% +/- 19.0% of the infused carnitine being retained. After 11 days of treatment comparable values for respiratory quotient, plasma triglycerides, free fatty acids, ketone bodies, and cumulative nitrogen balance were observed. It is concluded that in the patient population studied here carnitine supplementation during postoperative TPN did not improve fat oxidation or nitrogen balance.

Smoking and invasive cervical cancer risk. Results from a case-control study.

The association between cigarette smoking and risk of invasive cervical cancer was investigated in a case-control study conducted in four hospitals in Leipzig, German Democratic Republic in 1983-1985. Analyses were based on 225 women aged 64 years or younger with newly diagnosed invasive cervical cancer and 435 age-matched controls with orthopedic or skin diseases. The crude relative risk of invasive cervical cancer was significantly elevated for women who had ever smoked (relative risk (RR) = 1.5, 95% confidence interval (CI) = 1.1-2.1). Adjustment for number of sexual partners reduced the risk to RR = 1.2 (95% CI = 0.8-1.6). Furthermore, the risk for smokers depended significantly on number of sexual partners (p less than 0.05) and was increased only among women with no partner or one partner (RR = 2.7, 95% CI = 1.2-6.1).

Use of oral contraceptives and risk of invasive cervical cancer in previously screened women.

Within the context of a larger hospital-based case-control study carried out to assess the efficacy of cervical cancer screening, the possible association between oral contraceptives and risk of invasive cervical cancer has been studied as well. Because in the GDR cytological screening is an integral part of the gynaecological basic care, only a few women reported oral contraceptive use but had no Pap-smears in that study. Thus, the analysis was confined to those 129 cases and 275 controls who had at least one screening Pap-smear in their history and were below the age 55. The significantly increased relative risks for users decreased after adjustment for factors of sexual behaviour and interval since last Pap-smear but remained statistically significant or at borderline significance for some categories of usage. This concerns, in particular, long-term use (7+ years) and early onset of use (less than or equal to 24 years) with relative risks of 1.8 and 3.0, respectively.

[Overlap behaviour as a cell-biology characteristic of cultivated ovarian cancer cells].

The in vitro growth behaviour of ovarian cancer cells is investigated regarding its ability to characterize this tumor biologically. Out of 60 patients suffering from ovarian cancer with different extension and histologic types 79 in vitro cell cultures were cultivated from either tumor biopsies, ascites or pleura effusions. The overlap index of each culture is calculated by the ratio of actually observed to as a maximum expected nuclear overlaps and is compared to parameters being acknowledged characterization of ovarian carcinomas. Correlations were found between overlap behaviour and other typical parameters such as histologic type, grading, and tumor staging. Beside some other parameters nuclear overlap index seems to be appropriate for characterization of neoplastic ovarian tumors.

Protein transport in intact, purified pea etioplasts.

We have developed a method to isolate intact, purified pea etioplasts. These etioplasts were capable of recognizing, transporting, and processing the precursor form of the small subunit of the ribulose-1,5-bisphosphate carboxylase, a protein which is not detectable at this developmental stage. Transport of proteins was completely dependent on ATP and could not be substituted for or stimulated by light. The transported precursor protein was processed to its proper molecular weight. The mature form of the small subunit was assembled with the large subunit of the ribulose-1,5-bisphosphate carboxylase already present at this stage to form an oligomer. Protein transport was completely abolished using the phosphatase inhibitor sodium fluoride. This is the first time protein transport has been demonstrated in isolated, purified etioplasts.

Study on the effect of a high fat diet on diaphragm and liver glycogen and glycerides in the rat.

The present work was undertaken to study the effect of nutritional obesity induced by a high fat diet on the consumption of glycogen and glycerides in rat liver and diaphragm. Groups of rats were fed for five weeks from weaning either a fat-rich-carbohydrate (CHO)-poor diet, or a CHO-rich-fat-poor diet. Basal plasma glucose and free fatty acids (FFA) were significantly increased in the animals adapted to the fat-rich diet. Half of the rats were submitted to a 48-h fast. After fast, basal plasma glucose and immunoreactive insulin (IRI) fell significantly, whereas plasma FFA levels were higher than in the group fed the CHO-rich-fat-poor diet. In the liver, glycogen concentration fell in both groups after fast, with a glycogen breakdown of 1930 +/- 244 mumole glycogen glucose/liver in the fat-fed group vs 4636 +/- 216 mumole/liver in the CHO-fed group. Glycerides fell by 750 +/- 68 mumole glyceride glycerol/liver in the fat-fed rats while remaining unchanged (increased by 82 +/- 57 mumole/liver) in the CHO-fed group. In the diaphragm glycogen concentration also fell in both groups, with a glycogen breakdown of 6.0 +/- 0.3 mumole glycogen glucose/g wet tissue in the fat-fed rats vs 15.2 +/- 1.4 mumole/g wet tissue in the CHO-fed animals. Glycerides fell by 23.1 +/- 4.0 mumole/g wet diaphragm in the CHO-fed animals. The lower breakdown of glycogen in both liver and diaphragm of fat-fed rats demonstrates a decreased utilization of glycogen during fast, with energy consumption originating in larger part from triglycerides.(ABSTRACT TRUNCATED AT 250 WORDS)

[Comparative studies of the effect on the in vitro growth of endometrial cancers of hydroxyprogesterone caproate, norethisterone acetate and D-norgestrel].

The influence of the progestogens hydroxyprogesteroncaproate , norethisteronacetate and d-norgestrel on the growth of endometrial cancer was observed on 78 cell cultures. 40 cases revealed an influence on in vitro growth by at least one progestogen, 26 times by progesterone, 21 times by norethisterone and 20 times by norgestrel. In 17 cases two or more progestogens had an effect on in vitro growth, however, in 23 cases only one progestogen revealed an effect. The possible consequences of these results for the planning of therapy are discussed.

[Studies into dependence of sensitivity of cytostatics on overlap index of ovarian cancer in cell cultures (author's transl)].

Overlap index and cytostatic sensitivity pattern were determined from 66 cell cultures of ovarian carcinoma. The average overlap index of all cell cultures tested was 42.2 per cent +/- 20.5 and proved to be equivalent to that of malignant tumours in different locations. The wide scatter was attributed to the biological individuality of ovarian carcinoma. High overlap indices usually are indicators to high sensitivity to cytostatics, whereas low overlap indices may reflect low sensitivity or even resistance to cytostatic agents. Yet, no high-accuracy prediction can be made for the real individual case. The question is raised, if both parameters are separate and independent indicators to characterise a given tumour or parameters with correlations between them.

[Comparison of the results of gestagen therapy of cancer of the corpus uteri with a preliminary study of tumor cell culture sensitivity to the preparations].

The paper deals with a comparative study on the efficacy of adjuvant gestagen therapy of 61 patients with primary endometrial carcinoma. A control group of 219 patients did not receive hormones. In gestagen group, sensitivity to drugs was determined prior to therapy in 67 cell cultures of carcinoma. This provided a basis for a study of the meaning of a test of sensitivity to drugs which was carried out prior to therapy in order to select a suitable gestagen. Survival (2 years) was significantly higher in the group in which tumor sensitivity to chosen gestagen was determined in cell culture prior to treatment. Due to selection of suitable gestagen before therapy by means of tumors sensitivity test, a more pronounced clinical effect is obtained.

[Use of gestagens to control in vitro growth of endometrial carcinoma (author's transl)].

The impact of three gestagens, hydroxyprogesterone caproate, norethisterone acetate, and D-norgestrel, on in vitro cell cultures was tested in 40 test tubes and ten cover glasses. All tested cultures were of endometrial carcinomas. Cytomorphological changes were recordable from more than half of all cultures and interpreted as sensitivity to gestagens of the carcinoma concerned. No differences were safely established regarding effectiveness of different gestagens and different concentrations. In vitro gestagen action was clearly demonstrated by determination of nuclear overlap indexes of the cover glass cultures. No answer has yet been found as to whether reduction in nuclear overlap index, following addition of gestagens, had been caused by cellular modification. - Sensitivity of endometrial carcinoma to gestagens in vitro depended clearly on the degree of morphological differentiation.

[Results obtained from gestagen treatment of patients for endometrial carcinoma, with particular reference to results obtained from in vitro tests before treatment (author's transl)].

Reported in this paper is a retrospective review of two-year survival of 280 patients with endometrial carcinoma, with 22 per cent of them having received additional progestagen therapy. Survival rates were more favourable in response to progestagen therapy, particularly in the context of advanced stages. The best results were achieved by individual progestagen therapy preceded by pretherapeutic sensitivity testing.
