Venom of an aggressive African Theraphosidae (Scodra griseipes): milking the venom, a study of its toxicity and its characterization.
Two milking procedures, electric stimulation and lure-biting, are described and compared. The amount of crude Scodra griseipes venom collected in 3 years by lure-biting was 1670 mg and 1380 mg, respectively, for young females and adult males. The crude venom appears pure on the basis of UV-Vis spectra, which show no hemocyanin absorption bands. The toxicity and induced symptoms of these venoms in mice were evaluated by direct bites and also by LD50 [LD50 (young female venom, i.v.) = 8.1 mg/kg, LD50 (adult male venom, i.v.) > 9.5 mg/kg]. The venom from both sexes was compared by protein content, UV-Vis and 1H-NMR spectroscopies, polyacrylamide gel electrophoresis [SDS-PAGE] and size exclusion high performance liquid chromatography [SE-HPLC]. Differences were observed.
The components of the venom of a spider Scodra griseipes. 1. Analysis of low molecular weight products using gas chromatography/mass spectrometry.
Low molecular weight venom constituents of the spider Scodra griseipes have been derivatized using bis(trimethylsilyl)-trifluoroacetamide and pentafluoropropionic anhydride. Examination of these derivatives by coupled gas chromatography/mass spectrometry using electron ionization or ammonia- or isobutane-chemical ionization allows their structural identification. Comparisons have also been made with retention time and mass spectra of the pure compounds. Four acids: phosphoric, lactic, citric, gamma-aminobutyric (neurotransmitter) and four basic biogenic amines: putrescine, cadaverine, spermidine and spermine have been found.
The components of the venom of a spider Scodra griseipes. 2. Structural information on biogenic amines using tandem mass spectrometry.
Biogenic amines, which are components of tarantula Scodra griseipes venom, are studied using tandem mass spectrometry. Their heptafluoropropionic (PFP) derivatives are ionized under electron impact and isobutane-chemical ionization. Daughter- and parent-ion mass spectra under collisionally activated dissociation conditions provide a more detailed study of such derivatives. Discrimination of di(PFP)amines and tri and tetra(PFP)amines is possible because of the presence of diagnostic ions induced by collision.
Analysis of peptides and proteins by capillary electrophoresis/mass spectrometry using acidic buffers and coated capillaries.
The direct coupling of capillary electrophoresis (CE) and mass spectrometry, combined with ionspray ionization using a coaxial capillary arrangement, is described. The CE/mass spectrometer interface is shown to be effective for the analysis of native and tryptic peptides and of proteins of high molecular weight such as bovine serum apotransferrin (approximately 78 kDa). Adsorption of cationic analytes under acidic buffer conditions is minimized through the use of a non-covalent coated capillary possessing an overall positive charge. Since the direction of the electroosmotic flow is thus reversed, compared to that in conventional CE separation on uncoated capillaries, migration of cations is achieved by applying a negative voltage (typically -30 kV) at the injector end of the capillary. In addition to the inherent advantage of providing pre-formed cationic species for mass spectral detection, this arrangement permits analysis of proteins of high isoelectric points even at low pH. The ability to conduct electrophoresis of globular proteins under acidic conditions also provides a means of monitoring their conformational changes, as reflected both by the variation of migration times and by concurrent changes in the multiply charged ion envelopes.
Relative cardiac cost and physical, mental and psychological work load among a group of post-operative care personnel.
Continuous registration of heart rate and careful minute-by-minute observation of all physical, mental and psychological job-related events was carried out in a group of nine post-operative care personnel. The relative cardiac cost (RCC) correlated significantly with the observed work events. The magnitude of participation of physical, mental and psychological work elements in the changes in RCC varied in the investigated subjects according to the type and magnitude of the different work events. However, collective data on the group indicated that changes in RCC were related mainly to physical effort and to a lesser, albeit significant, extent to psychological reaction. The results indicate the possibility of using the heart rate in monitoring working conditions and, probably, in guiding their improvement.
[Experiences with psychosocial management of patients with multiple sclerosis in a discussion group].
Patients with multiple sclerosis must come to terms with their unpredictable chronic disease. Frequently, they want to establish contacts and exchange information with others sharing the same problems. Discussion groups of outpatients offer many opportunities to further the process of coping with the disease as an interlocking web of defensive and coping patterns. The patients can learn from each other to assess their disabilities and capabilities more realistically and to deal more openly with their fears, hopes and disappointments.

[Noninvasive diagnosis of occlusive processes of the brachiocephalic arteries: Doppler sonography compared to ophthalmodynamometry and ophthalmodynamography].
97 patients with suspected cerebrovascular disease were examined using ophthalmodynamometry/ophthalmodynamography and dopplersonography. In 49 of the patients we found haemodynamic changes typical of brachio-cephalic artery occlusion processes. In 36 patients angiography was performed either as a preparation for vascular surgery or for further clarification. Ophthalmodynamometry/ophthalmodynamography examination yielded diagnosis in 87%, dopplersonography and combined use of ophthalmodynamometry/ophthalmodynamography and dopplersonography in 94% of the brachio-cephalic artery occlusion processes.

[Occupational angioneurotic disorders provoked by vinyl monochloride and vibration].
Microcirculatory disorders of the extremities may have an occupational origin, which it is desirable to specify. Exposure to vinyl monochloride results in a chemical nuisance which also proved to be carcinogenic in man. Strict preventive measures are therefore enforced in the industry and angioneurotic disorders should no longer be observed. Use of vibrating tools results in a microvascular response to a physical nuisance which is difficult to prevent completely. In both cases the physiopathology is not well-known, but compensation is possible as occupational diseases. Repeated trauma of the ulnar artery, and/or its superficial palmar branch are, on the contrary, the main explanation for hypothenar hammer syndrome, which is easily prevented by mere adjustment of the work position and occupational movements: the conditions for medico-legal compensation for this particular syndrome, which is still not listed among occupational diseases, will be in effect in the very near future.
