Variability of the response to pentoxifylline in vitro in infertile normozoospermic and asthenozoospermic patients.

The in vitro effect on the motility of spermatozoa of the methylxanthine pentoxifylline was studied in 30 patients consulting for infertility (13 normozoospermic and 17 asthenozoospermic). After separation by centrifugation on Percoll gradient, the spermatozoa were incubated for 30 min in pentoxifylline (3.6 mM), then the pentoxifylline was removed by washing and centrifugation. The residual effect of pentoxifylline was studied by computer-assisted videomicrographic analysis of the sperm motion parameters. In both groups of patients a decrease in the percentage of motile spermatozoa after exposure to pentoxifylline was observed, as compared with a control group. The effect on the motion parameters varied according to the time of observation (30, 120, 240 min). In the asthenozoospermic patients, the curvilinear velocity (VCL) was not modified in a statistically significant manner. In the normozoospermics, the VCL was increased after exposure to pentoxifylline at 120 min (p = .03) and 240 min (p = 10(-5)); similarly, the amplitude of the lateral head displacement (ALH) was greater in this group at 120 and 240 min (p = .01 for maximum ALH). However, a heterogeneity was noted in the response between individuals, which required that the statistical analyses take into account these differences.

Multidrug-resistant tuberculosis in prison inmates, Azerbaijan.

In a tuberculosis (TB) program in the Central Penitentiary Hospital of Azerbaijan, we analyzed 65 isolates of Mycobacterium tuberculosis by IS6110-based restriction fragment-length polymorphism (RFLP) and spoligotyping. From 11 clusters associated with 33 patients, 31 isolates had an IS6110-based banding pattern characteristic of the Beijing genotype of M. tuberculosis. In addition, 15 M. tuberculosis isolates with similar RFLP patterns constituted a single group by spoligotyping, matching the Beijing genotype. Multidrug resistance, always involving isoniazid and rifampin, was seen in 34 (52.3%) of 65 isolates, with 28 belonging to the Beijing genotype.

[Construction and validation of visual analogue scale for risk assessment].

This paper describes the factor structure and validity of a visual analogue scale designed to rate risk proneness: EVAR (EVAluation of Risk). Risk behaviors and sensation seeking are related, therefore we correlated EVAR (24 100 mm items) with Zuckerman Sensation Seeking Scale (SSS) (40 conventional items) in a population of 199 volunteers. Subjects had an age range of 18-68 years (mean age of 31), and 49 were females. Factor analysis using a principal component solution and an orthogonal rotation of the factor matrix were computed. Five factors were selected, accounting for 46 per cent of the total variation: F1 "self control", F2 "danger seeking", F3 "energy", F4 "impulsiveness", F5 "invincibility". F1, F2 and F3 from EVAR were correlated with SSS factors "general", "danger seeking", "experience seeking" and "boredom susceptibility". F4 was correlated with "general", "boredom susceptibility" and "disinhibition" whereas F5 was not correlated with any of SSS factors. This visual analogue scale is likely to reveal change in subjects, regarding risk proneness, when submitted to various stressors (sleep deprivation, fatigue...).

[Prevention of type 1 diabetes: interventions in mice and in humans].

Prevention of type 1 diabetes mellitus is a major challenge in health care research, but until the present day we have not succeeded in achieving this goal. Thanks to the availability of good animal models of type 1 diabetes, our insight in the pathogenesis has grown, and new possibilities of intervention have emerged. In the coming years, major advances in prevention of type 1 diabetes are to be expected.

Structural and functional mapping of immunoglobulin V-regions.

V-regions of immunoglobulins chains contain 3 types of positions, which are equally represented: invariant and sub-group characteristic, which account for the "framework" and hypervariable positions, responsible for antigen recognition. The 3 types of positions are grouped and fall within a very few discrete stretches. Sub-group characteristic segments containing one of the 2 cysteyl residues of the V-regions may be isolated by high voltage paper electrophoresis and provide a basis to type for sub-groups in the VK and in the VH human systems. This allowed to characterize a large set of human myeloma proteins that were used in a series of competitive hybridizations which indicated that sub-groups had no influence on preferential reassociations, which occurred in 80% of the cases. This preference seems to rely mostly on individual structural differences, which may be linked to heterogeneity at the framework level. Distinction between framework heterogeneity and hypervariable regions heterogeneity may be approached by raising antibodies against a mouse myeloma protein, MOPC 173, of known sequence, by means of syngeneic and allogeneic immunizations, using Balb/c and A/J mice. Junction of distinct portions of immunoglobulin chains such as the V and the C regions raises the possibility that some recognition signals may operate at the DNA level. Since rotational symmetry regions in the DNA are known to act as such signals, it is discussed whether such regions can be expected from the amino acid sequence data, especially in the vicinity of the "switch" peptide.

[Problems posed by oocyte donation organization].

Ten years after the first pregnancy in France and 3 years after the setting of the law of Bioethic oocyte donation procedure is coming up against many difficulties (search of donors, cost, efficacy). We will try to consider all of them to prepare the way for an official recognition of this specific procedure of AMP.
