Controlled reperfusion after myocardial ischemia in a canine model monitored by two-dimensional phosphorus 31 chemical shift spectroscopic imaging.

Phosphorus 31 magnetic resonance spectroscopy at 2 T was used to monitor high-energy phosphate metabolism over a 3-week period in a canine model of myocardial infarction and reperfusion. Twenty animals were divided into two groups: group 1 (n = 11) received intravenous nitroglycerin beginning at the onset of coronary occlusion; group 2 (n = 9) received a 105-minute infusion of superoxide dismutase (SOD) beginning at the onset of reperfusion. A metabolic protective effect was observed (vs controls) with both agents, manifested by a reduction in the degree of pH decline from baseline values and preservation of the adenosine triphosphate/total phosphate ratio during occlusion and reperfusion. Further, both treatments, compared with controls, produced a lower infarct/zone at risk ratio: controls, 1.5 +/- 1.2; nitroglycerin, 0.52 +/- 0.50; and SOD, 0.64 +/- 0.40. The technique of 31P magnetic resonance spectroscopy demonstrated its use for the noninvasive assessment of myocardial metabolism in response to therapeutic intervention.

Unbalanced translocation in a mother and her son in one of two 5;10 translocation families.

We present two families with different distal long arm 5;10 translocations. In one family the propositus and his mother inherited the same derived chromosome 10 from the maternal grandfather who has a balanced t(5;10)(q35.3;q26.13). The phenotype of both the affected patients is milder and only partially overlaps with that of previous cases of distal 10q deletion. Other previously reported cases of transmitted imbalance are also remarkable for mild phenotype, occurrence of deletions rather than duplications and a strong bias toward maternal as opposed to paternal transmission. In the second family, the propositus inherited a derived chromosome 10 from his mother who carries a balanced (t(5;10)(q35.1;q26.3) translocation; his clinical manifestations are consistent with an emerging phenotype for distal 5q duplications.

Rapid in situ harvesting and cytogenetic analysis of perinatal tissue samples.

Thirty perinatal solid tissue samples were used in a pilot study to test the efficacy of collagenase disaggregation onto coverslips, open system culture under low O2 conditions, pre-harvest incubation in bromodeoxyuridine (BrdU) and colcemid, and in situ automated harvesting. Following the success of the pilot study, the new method was applied to a further 126 consecutive diagnostic samples making a total of 156. The method reduced average tissue culture times from 17 to 8.7 days (range 2-17), improved success rates from 76 to 88 per cent, and simultaneously increased the resolution of cytogenetic analysis.

Determination of cysteine plus half-cystine in protein and peptide hydrolysates: use of dithiodiglycolic acid and phenylisothiocyanate derivatization.

Cysteine and half-cystine in proteins and peptides can be determined by acid hydrolysis in the presence of dithiodiglycolic acid, derivatization with phenyl isothiocyanate, and HPLC separation of the phenylthiocarbamyl amino acids. The determination requires no additional sample handling, oxidation, reduction, alkylation, or desalting. Samples containing cystine are treated in the same manner as those containing cysteine. Derivatization of cysteine or half-cystine by dithiodiglycolic acid has no effect on any other amino acids. Recovery of the PTC-cysteine derivative is greater than 90% over the linear range of 5 pmol to 1 nmol. Precision of the cysteine plus half-cystine determination is equivalent to that observed for all other amino acids. Structure work on the derivative indicates that it is a mixed disulfide containing cysteine and thioglycolic acid.

Overview of the Department of Energy's Off-Site Radiation Exposure Review Project (ORERP).

The Off-Site Radiation Exposure Review Project (ORERP) was established by the U.S. Department of Energy to (1) collect, preserve, and disseminate historical data related to radioactive fallout and health effects from nuclear testing, and (2) reconstrut, insofar as possible, the exposures to the off-site public from nuclear testing at the Nevada Test Site and doses to individuals resulting from these exposures. The goals, methods, and example results of the ORERP are presented.

Insensitivity of Tc-99m pertechnetate for detecting metastases of differentiated thyroid carcinoma.

To evaluate the use of Tc-99m pertechnetate whole body scanning for the detection of metastases of differentiated thyroid carcinoma, the authors performed sequential Tc-99m pertechnetate and I-131 scans in five patients with known or suspected metastatic thyroid cancer. All five patients had abnormal I-131 uptake, but only two patients had abnormal Tc-99m pertechnetate uptake. A total of 33 abnormal foci were located with I-131; Tc-99m pertechnetate detected only 3 of these foci, and did not demonstrate any foci that were not apparent with I-131. Despite the theoretical advantages of Tc-99m pertechnetate, it cannot be recommended as a substitute for I-131 for locating thyroid cancer metastases.

Malignant gonadal stromal tumor: case report and review of the literature.

Of the 80 cases of gonadal stromal tumor reported in the literature 10 were malignant. We herein report another malignant case. The role of chemotherapy and radiation therapy in malignant gonadal stromal tumor has not been defined. While the only sue evidence of malignancy is metastatic disease we recommend that if macroscopic and microscopic examination of the primary tumor suggests malignancy a retroperitoneal node dissection should be performed.

Identification and quantification of steroids in the serum of rainbow trout during spermiation and oocyte maturation.

17 alpha-Hydroxy-20 beta-dihydroprogesterone and 17 alpha-hydroxyprogesterone were found in higher concentrations in serum from female Salmo gairdneri undergoing final oocyte maturation immediately before ovulation than in serum from spermiating male trout. Other steroids (11-deoxycorticosterone, 11-deoxycortisol and progesterone) which have been implicated in oocyte maturation and/or ovulation in lower vertebrates were not identified at such high concentrations and the differences between the serum of both sexes were not so great. These results confirm that 17 alpha-hydroxy-20 beta-dihydroprogesterone and 17 alpha-hydroxyprogesterone, the most potent inducers of trout oocyte maturation in vitro, are present in the blood when oocyte maturation occurs. The concentration of testosterone was found to be higher in serum from female than from male trout indicating that testosterone is unlikely to be the principal androgen in trout. High concentrations of 11-oxotestosterone in male and barely detectable levels in female fish support the hypothesis that 11-oxotestosterone is an important androgen in the regulation of testicular activity.
