[Recalcification of an 8 x 4 x 4.5 cm large brown tumor with spontaneous fracture following parathyroidectomy. Case report and course].

Primary hyperparathyroidism is typically accompanied by a vast array of symptoms and the occurrence of so-called brown tumors often localized in the metaphysis of bone. These tumors are radiologically and histologically hard to distinguish from giant cell tumors. Usually they recalcify after parathyroidectomy, except in case of genuine cysts. We report on an extended brown tumor of the tibia located just below the knee joint of a 45-year-old woman. It was a large as 8 x 4 x 4.5 cm and had been noticed after a spontaneous fracture. Complete recalcification occurred after parathyroidectomy and the bone recuperated fully after 6 months.

Active phenoxymethylpenicillin (pcV) in the mucous membranes of the oral bucca and maxillary sinus and in diffusion chambers implanted in rabbits. A methodological study.

The intention of prescribing antibiotic treatment must be to achieve a concentration of the agent within the infected tissue well above th MIC or the bacteria causing the infection. As a basis for further clinical studies on antibiotic concentrations in maxillary sinus mucosa in humans, determinations of biologically active penicillin V in the mucous membranes of the maxillary sinus and the buccal mucose as well as in tissue fluids and tissue from subcutaneously implanted tissue cages in rabbits were made. Several experimental conditions were studied, with the purpose of establishing optimal conditions with regard to tissue sampling, blood contamination, prediffusion time, etc. Penicillin V was shown to have a good penetration into mucosal tissue in the rabbit, even at low perfusion pressure.

p53 analysis in gallbladder cancer: comparison of gene analysis versus immunohistochemistry.

BACKGROUND: The survival of gallbladder cancer (GC) patients is generally poor. Both after resection and palliative procedures, additive therapy is often administered to increase outcome. The effect of cytotoxic therapies depends on a functioning p53 gene. The aim of this study was to investigate whether p53 immunohistochemistry (IHC) or p53 gene sequencing are of any survival influence in GC. PATIENTS AND METHODS: In 61 GC patients, 19 were resected and in 42 operative explorations were performed. Seven resected and 14 palliated patients received adjuvant chemotherapy. IHC p53 staining and DNA sequencing was investigated. RESULTS: p53 sequencing detected 20 mutations and no relationship was found between IHC and sequencing results. After resection chemotherapy increased survival in those having a normal p53 gene compared to those having a mutation (p=0.008). p53 status did not show an influence in resected patients without chemotherapy or in palliative-treated patients regardless of their therapy. CONCLUSION: p53 protein overexpression does not correlate with p53 gene mutation. Response to chemotherapy in resected GC patients may depend on a functioning p53 gene.

Cellular and genetic properties of two melanoma cell lines established from the same tumor.

We report here the detailed cellular and genetic analysis of two new human melanoma cell lines established from the same tumor biopsy. Although derived from the same tumor biopsy, the two lines differed in their method of in vitro culture establishment with one line (HA-A) grown first in agar culture and then transferred to liquid culture, while the second cell line (HA-L) was established directly from cells grown in liquid culture. Detailed characterization of both cell lines was performed using techniques to analyze their cytogenetic, surface marker, morphology (light and electron microscopy), growth (in agar and in nude mice), and drug sensitivity profiles in comparison to the original tumor biopsy. Our results revealed no significant biologic or genetic difference between the two cell lines established by different culture techniques. Our results suggest that in some cases agar culture prior to liquid culture may be useful in establishment of human tumor cell lines.

The calcium-binding protein calretinin-22k is detectable in the serum and specific cells of cancer patients.

BACKGROUND: Calretinin-22k (CR-22k), an alternatively spliced form of calretinin (CR) belongs to the EF-hand family of calcium-binding proteins and is expressed in several colon adenocarcinoma cell lines (e.g. WiDr, HT-29). MATERIALS AND METHODS: Serum samples of cancer patients were screened with a sandwich ELISA technique using the CR-specific antiserum 7696. Highly positive samples were analyzed by Western blots and immunohistochemistry. RESULTS: CR-22k was detected in the serum of several patients and values were as high as 0.19 microgram/ml. Western blot analysis confirmed the identity of the bound protein as CR-22k. The highest concentrations were detected in patients with colon or breast cancer, but also in a patient with ischemic necrosis of the gut. CR immunoreactivity was localized to epithelial cells, nerve fibres, cells of the connective tissue and to mesothelial cells. CONCLUSIONS: Our results establish that CR-22k is detectable in the serum of cancer patients under specific pathological conditions.
