Raised object on a planar surface stroked across the fingerpad: responses of cutaneous mechanoreceptors to shape and orientation.

The representations of orientation and shape were studied in the responses of cutaneous mechanoreceptors to an isolated, raised object on a planar surface stroked across the fingerpad. The objects were the top portions of a sphere with a 5-mm radius, and two toroids each with a radius of 5 mm along one axis and differing radii of 1 or 3 mm along the orthogonal axis. The velocity and direction of stroking were fixed while the orientation of the object in the horizontal plane was varied. Each object was stroked along a series of laterally shifted, parallel, linear trajectories over the receptive fields of slowly adapting, type I (SA), and rapidly adapting, type I (RA) mechanoreceptive afferents innervating the fingerpad of the monkey. "Spatial event plots" (SEPs) of the occurrence of action potentials, as a function of the location of each object on the receptive field, were interpreted as the responses of a spatially distributed population of fibers. That portion of the plot evoked by the curved object (the SEPc) provided a representation of the shape and orientation of the two-dimensional outline of the object in the horizontal plane in contact with the skin. For both SAs and RAs, the major vector of the SEPc, obtained by a principal components analysis, was linearly related to the physical orientation of the major axis of each toroid. The spatial distribution of discharge rates [spatial rate surface profiles (SRSs), after plotting mean instantaneous frequency versus spatial locus within the SEPc] represented object shape in a third dimension, normal to the skin surface. The shape of the SA SRSs, well fitted by Gaussian equations, better represented object shape than that of the RA SRSs. A cross-sectional profile along the minor axis [spatial rate profile (SRP)] was approximately triangular for SAs. After normalization for differences in peak height, the falling slopes of the SA SRPs increased, and the base widths decreased with curvature of the object's minor axis. These curvature-related differences in slopes and widths were invariant with changes in object orientation. It is hypothesized that circularity in object shape is coded by the constancy of slopes of SA SRPs between peak and base and that the constancy of differences in the widths and falling slopes evoked by different raised objects encodes, respectively, the differences in their sizes and shapes regardless of differences in their orientation on the skin.

[Role of hyperinsulinemia in pathogenesis of polycystic ovary syndrome and treatment by reduction of insulin secretion].

OBJECTIVE: To investigate the role of insulin in the polycystic ovary syndrome (PCOS) pathogenesis and application of Metformin in the treatment of PCOS. METHODS: Serum androgen, luteinizing hormone (LH) and sex hormone-binding globulin(SHBG) concentration during fasting and serum 17 alpha-hydroxyprogesterone (17-OHP), LH level in response to gonadotropin-releasing hormone agonist (GnRH-a) stimulation were determined. Oral glucose-tolerance tests(OGTT) before and after oral administration of metoformin for 8-12 weeks were performed in 12 obese and 11 lean women with PCOS. RESULTS: After oral administration of metformin for 8-12 weeks, fasting insulin concentration in obese group and area under curve (AUC) after OGTT in lean group decreased significantly. There were significant decrease in basal 17 alpha-hydroprogesterone, androstenedione, testosterone concentration, and increase in serum SHBG concentration in obese and lean groups. Basal LH and the response of serum 17-OHP, LH to GnRHa were not significantly changed. CONCLUSIONS: Hyperinsulinemia may play an important role in hyperandrogenism GnRH-a of PCOS, and metformin may be used in the treatment of PCOS.

[The effects of FSH,LH and insulin on steroids production by granulosa cells from polycystic ovaries syndrome].

OBJECTIVES: To investigate steroids production of granulosa cells obtained from normal ovaries and polycystic ovaries from polycystic ovary syndrome (PCOS) patients under FSH, LH and insulin stimulation. METHODS: Granulosa cells of 33 folicles from 18 pairs of normal ovaries and about 100 folicles from 8 pairs of PCOS were cultured. Estrodial (E2) and progesterone (P) accumulation in the medium was determined under different incubated conditions by RIA. RESULTS: Granulosa cells from PCOS produced more E2(P < 0.05-0.01) under basal, FSH and insulin stimulation and more P (P < 0.05-0.01) under FSH, LH and insulin stimulation than controls. Insulin markedly augmented FSH and LH-stimulated steroid accumulation by two groups of granulosa cells. Granulosa cells from PCOS with peripheral insulin resistance also responded to insulin. CONCLUSION: Enzymes for steroids synthesis in granulosa cells from PCOS may be activated in vivo, and hyperinsulinisma and elevated LH levels in PCOS may be contributable.

[Clinical observation on preventing and treating coronary atherosclerotic heart disease with tongmai jiangzhi oral liquid].

OBJECTIVE: To observe the clinical effect of Tongmai Jiangzhi Oral Liquid (TJOL) on blood lipid, lipid peroxidation, prostaglandin metabolism in treating coronary heart disease and angina pectoris with hyperlipidemia. METHODS: Forty-one cases in treated group and 24 in control group were treated with TJOL and Capsulae Oenoth Erae Biennisolei (COEB) for one month respectively. Serum lipid, apolipoprotein, plasma malonyldialdehyde (MDA), superoxide dismutase (SOD), thromboxane B2(TXB2), 6-keto-prostaglandin F1 alpha and blood glutathione peroxidase (GSH-Px) were measured before and after treatment. RESULTS: After treatment with TJOL, the total effective rate was 87.80% in relieving angina and 50.00% in improving ECG. Other data showed that the levels of serum TC, TG, apoB, plasma MDA, TXB2 and TXB2/6-keto-prostaglandin F1 alpha ratio lowered and levels of apoA, GSH-Px, serum HDL-C, apoA1/apoB increased (P < 0.05-P < 0.001). CONCLUSIONS: TJOL could effectively lower blood lipid, reduce injury caused by free radicals, regulate prostaglandin metabolism and treat coronary heart disease. By this study, an effective prescription of Chinese herbal medicine was offered for prevention and treatment of coronary heart disease.

[Influence of ligustrazine-contained peritoneal dialysate on efficiency of dialysis].

OBJECTIVE: To observe the influence of ligustrazine on efficiency of peritoneal dialysis. METHODS: A prospective, self-controlled study was adopted, 24 stable continuous ambulatory peritoneal dialysis (CAPD) patients were divided into 4 groups according to their peritoneal membrane solute transport types identified by peritoneal equilibration test (PET), the high transport group (H group, 4 cases), the high-average transport group (Ha group, 11 cases), the low-average transport group (La group, 8 cases) and the low transport group (L group, 1 case). After treated with standard dialysis for 2 weeks, patients were treated by peritoneal dialysis with ligustrazine (240 mg/d) contained dialysate for another 2 weeks, and changes on mean net ultrafiltration volume (UF) per exchange, and urea kinetics (KT/V) per week of patients were observed. RESULTS: After ligustrazine treatment, the UF and KT/V raised significantly, P < 0.01. In comparison of groups with different transportation, except change on UF of the H group was insignificant, increment of both parameters before and after treatment were significant (P < 0.01 or 0.05). CONCLUSION: By adding ligustrazine in standard dialysate can enhance the efficiency of peritoneal dialysis, raise the UF, so is worth further study.

[Visceral voice training for laryngectomy after hypopharyngectomy and visceral transplant].

OBJECTIVE: To establish a way to visceral voice after stomach or colon transplantation for patients with hypopharyngeal or cervical esophageal cancer. METHODS: Esophageal voice training was used and modified according to the compliance of the patients. Twenty laryngectomies with replacement of the hypopharynx and esophagus by stomach(13 cases), colon(6 cases) and jejunum (1 case) were trained for voice rehabilitation at the Department of Head and Neck Surgery. RESULTS: The patients with an artificial esophagus from vicera were easy to gain an influx of certain volume of gas into their artificial esophagus (stomach or colon) and to learn to speak. But on the whole the quality of voice was not so satisfactory. In this series nineteen out of 20 patients (95%) could express their idea by speech after a training course of three weeks. CONCLUSION: The literature has emphasized role of cricopharyngeus muscle in the training of esophageal voice. Owing to the fact that this series of patients who had had their cricopharyngeus removed, could easily get their voice rehabilitated, it seems this muscle played no major role in the voice rehabilitation.

[Internalization and replication of Edwardsiella tarda in HEp-2 cells].

It is demonstrated that Edwardsiella tarda possesses the ability to invade cultured epithelial HEp-2 cells by both lysis-counting assay and thin-section electron microscopy. Among fifteen strains of E. tarda, 6 strains internalized into HEp-2 cells and located mainly in vesicles. After internalization, the bacteria replicated in the host cells and then released into medium. Pretreatment of the HEp-2 cells with various concentration of cytochalasin D (0.1-5.0 micrograms/ml) or cytochalasin B (2.5-10.0 micrograms/ml) significantly reduced the amount of internalized bacteria in a dose-dependent manner. While pretreatment of HEp-2 cells with 0.25-100 mumol/L colchicine did not show any effect on the invasiveness of E. tarda. This strongly indicates that microfilaments are required for the internalization of E. tarda into HEp-2 cells and that microtubules are not involved in the entrance of E. tarda into HEp-2 cells.

[Karyotype analysis on Iris lactea ssp. chinensis].

In this paper, the chromosome number, karyotype and chromosome volume of Iris lactea ssp. chinensis were reported. The normal diploid was 2n = 32. Karyotype formula was based on Levan's publication in 1964, k (2n) = 32 = 22 m + 8 sm + 2T. According to the method of Kuo SR, the species chromosome based on the relative length was 2n = 32 = 6L + 4 M2 + 18 M1 + 4 S which belong to "2A" type according to the Stebbins' karyotype classification. The AS.K% was 62.5% from Arano's method. The total volume of chromosome groups was 34.95 micron 3.

Functional coupling of a human retinal metabotropic glutamate receptor (hmGluR6) to bovine rod transducin and rat Go in an in vitro reconstitution system.

The cDNA encoding hmGluR6, appended with a 15-amino acid antibody epitope (1D4), was transiently transfected in COS-7 cells. The receptor was purified from COS cell membranes using an antibody affinity column. The purified receptor was then reconstituted into lipid vesicles, and its ability to activate either transducin, the rod photoreceptor-specific GTP-binding protein, or the alpha subunit of Go was assayed in vitro using a guanosine 5'-3-O-(thio)triphosphate binding assay. Activation of both transducin and Go was observed. The rate of Go activation was 18-fold greater than the rate of transducin activation. This indicates that the coupling of mGluR6 to Go is more efficient and suggests that Go may be involved in coupling to mGluR6 in ON-bipolar cells.

Tissue-specific regulation of G-protein-coupled inwardly rectifying K+ channel expression by muscarinic receptor activation in ovo.

We investigated the effects of muscarinic acetylcholine receptor stimulation on the expression levels of the G-protein-coupled inwardly rectifying K+ channel (GIRK) subunits using solution hybridization and immunoblot analyses. We report here that treatment of chick embryos in ovo with muscarinic agonist causes decreases in mRNA levels encoding GIRK1 and GIRK4 in atria but does not alter GIRK1 expression in ventricles. In addition, GIRK1 protein levels also demonstrate a decrease in atria upon muscarinic acetylcholine receptor stimulation. Numerous receptors couple to the activation of the GIRK family of inwardly rectifying K+ channels; thus, these decreases represent a novel mechanism for regulating physiological responses to chronic agonist exposure.

Association of vascular endothelial growth factor expression with tumor angiogenesis, survival and thymidine phosphorylase/platelet-derived endothelial cell growth factor expression in human colorectal cancer.

To examine the association of vascular endothelial growth factor (VEGF) expression with tumor angiogenesis, survival and thymidine phosphorylase/platelet-derived endothelial cell growth factor (dThdPase/PD-ECGF) expression in human colorectal cancer, immunohistochemical studies were performed on 136 cases of resected colorectal cancer specimens using antibodies for VEGF, KDR, CD34 and dThdPase/PD-ECGF. Fifty-nine cases (43%) were evaluated as positive for VEGF staining and 71 cases (52%) were evaluated as positive for dThdPase/PD-ECGF staining. The expression of VEGF correlated significantly with vessel counts and the expression of dThdPase/PD-ECGF (P = 0.01 and 0.01, respectively). Cox proportional hazards model analysis showed that vessel counts and VEGF expression were significant and independent prognostic factors, but that KDR expression was not.

Stimulus-secretion coupling in porcine adrenal chromaffin cells: effect of dexamethasone.

Recent studies from this laboratory established that dexamethasone (DEX) potentiates Ca2+ current via voltage-gated Ca2+ channels (VGCC), and as a consequence potentiates agonist-induced cytosolic Ca2+ transients in rat adrenal chromaffin cells. The present study examined whether DEX can also modulate VGCC activity and agonist-induced cytosolic Ca2+ transients in porcine adrenal medullary chromaffin (PAMC) cells, and if so whether this results in alterations in catecholamine secretion. Forty-eight-hr exposure to 1 microM DEX significantly increased peak Ca2+ current (delta + 138%; n = 6; P < 0.05) in PAMC cells. DEX treatment also significantly potentiated the increase in cytosolic Ca2+ in response to membrane depolarization with KCl (delta + 20%; n = 29; P < 0.05), but did not affect the amplitude of Ca2+ transients elicited by nicotine or acetylcholine. Despite the potentiation of intracellular Ca2+, DEX treatment had no effect on KCl-induced secretion of either norepinephrine or epinephrine. These data demonstrate that as in the rat chromaffin cell, DEX can also increase VGCC activity in PAMC cells. However, the subsequent potentiation of selected agonist-induced increases in intracellular Ca2+ does not appear to be sufficient to alter catecholamine secretion.

Channel activators regulate ATP-sensitive potassium channel (KIR6.1) expression in chick cardiomyocytes.

ATP-sensitive potassium channels (K(ATP)) are widely expressed and yet little is known about the mechanisms regulating their expression. Here we report that expression of chick heart Kir6.1 is regulated by channel activators. Activation of K(ATP) with either ATP depletion or pinacidil, up-regulated Kir6.1 mRNA 1.8- to 2.4-fold in cultured ventricular myocytes as measured by competitive PCR. Pinacidil treatment also increased Kir6.1 protein as detected using an antibody to Kir6.1. Glibenclamide, a K(ATP) inhibitor, completely blocked the pinacidil-induced increase in Kir6.1 levels. It appears that Kir6.1 is up-regulated by an unknown signal transduction pathway initiated by K(ATP) opening.

Folding of the conserved domain but not of flanking regions in the integrin beta2 subunit requires association with the alpha subunit.

We have used immunoprecipitation with mAbs to probe folding during biosynthesis of the beta2 integrin subunit of lymphocyte function-associated antigen 1 (LFA-1; CD11a/CD18) before and after association with the alphaL subunit. An evolutionarily conserved region is present in the beta2 subunit between amino acid residues 102 and 344. mAbs to one subregion before the conserved region, and two subregions after the conserved domain, immunoprecipitated both the unassociated beta'2 precursor and mature alphaL/beta2 complex, suggesting portions of these subregions are folded before association with alphaL. An activating mAb to the C-terminal cysteine-rich region, KIM127, preferentially bound to the unassociated beta subunit, suggesting that it may bind to an epitope that is in an alphabeta interface in unactivated LFA-1. By contrast, mAbs to five different epitopes in the conserved region did not react with unassociated beta'2 precursor, suggesting that this region folds after alphaL association and is intimately associated with the alphaL subunit in the alphaL/beta2 complex. mAbs to two different epitopes that involve the border between the conserved region and the C-terminal segment, were fully or partially reactive with the beta'2 precursor, suggesting that this region is partially folded before association with alphaL. The findings suggest that the conserved region is a distinct folding and hence structural unit, and is intimately associated with the alpha subunit.

Effect of dexamethasone on voltage-gated Ca2+ channels and cytosolic Ca2+ in rat chromaffin cells.

The present study examined whether the synthetic glucocorticoid dexamethasone (DEX) can modulate voltage-gated Ca2+ channel (VGCC) activity, and as a consequence agonist-induced increases in cytosolic Ca2+, in cultured rat adrenal medullary chromaffin (RAMC) cells. Exposure to 1 microM DEX for 48 h significantly increased peak VGCC current (delta +140%). DEX treatment also significantly potentiated the increases in cytosolic Ca2+ in response to submaximal stimulatory concentrations of KCl (delta +64%) and nicotine (delta +32%). The Ca2+ channel agonist BAY K-8644 increased both VGCC current (delta +109%) and potentiated the KCl-stimulated increase in cytosolic Ca2+ (delta +35%) to a comparable extent to that seen with DEX. These data suggest that DEX treatment increases VGCC activity, and that this increased Ca2+ influx leads to potentiation of agonist-induced increases in cytosolic Ca2+ in RAMC cells.

Modulation of hybrid bass retinal gap junctional channel gating by nitric oxide.

1. To elucidate the role of the nitric oxide (NO) transmitter system in the regulation of gap junctional channel gating, we have examined the effects of the NO donor sodium nitroprusside (SNP) on the electrical synapses of hybrid bass H2-type horizontal cells. 2. SNP reversibly reduced the macroscopic junctional conductance without significantly changing voltage sensitivity. 3. Kinetic analyses showed that SNP made the voltage-dependent decay of junctional currents more rapid. 4. Single-channel data showed that SNP reduced channel open probability by reducing channel open frequency. 5. The action of SNP can be prevented or largely reduced by the NO scavenger, haemoglobin. NO release by SNP solutions was detected directly by a NO sensor. 6. NO appears to modulate the gap junctional conductance by activating the cGMP-cGMP-dependent protein kinase G (PKG) pathway. A membrane-permeable cGMP analogue, 8-Br-cGMP, mimics the action of SNP. A soluble guanylate cyclase inhibitor (LY-83583) and a highly specific cGMP-dependent protein kinase inhibitor (RKRARKE) blocked the action of NO. 3-Isobutyl-1-methylxanthine (IBMX), a non-specific phosphodiesterase inhibitor, potentiated the effect of SNP. 7. [Ca2+]i image studies showed that NO donors did not change [Ca2+]i in horizontal cells, suggesting that the regulation of junctional channels by NO is [Ca2+]i independent.
