[Purification and identification of human leukocyte myeloperoxidase].

Myeloperoxidase (MPO) plays an important role in the oxygen-dependent microbicidal mechanism of polymorphonuclear neutrophils. The purpose of our study was to investigate the therapeutic potency of human MPO preparations. This paper is to present our work on MPO purification and its identification. White blood cells, isolated freshly from normal donors, were lysed with cetyltrimethylammonium bromide to liberate myeloperoxidase. The purified MPO was obtained by 65% (NH4)2SO4 precipitation followed by separation over the Sephadex G150 column. These pure MPO preparations were green in color and had an A430/A280 nm of 0.68. The enzymatic activity was of 29.77 u/mg. The pure normal MPO was composed of a 59,000 MW peptide, a 13,500 MW peptide, and a 38,000 MW peptide when subjected to SDS-PAGE under reducing conditions. The study of the therapeutic effect of the pure MPO on mice with C. albicans infection is under way.

[Morphometric analysis of myocardial cells in experimental diabetic rats].

Ultrastructure of myocardial cells was studied with a morphometric technique in Wistar rats 6 and 12 months after induction of experimental diabetes with streptozotocin (70 mg/kg i.p.). Weight-matched and untreated rats served as controls. Morphometric analysis showed that volume densities of mitochondria, sarcoplasmic reticulum, lipid droplets and lysosomes were markedly increased and volume density of myofibril was significantly decreased in comparison with those of the control animals. Further measurements of mitochondria indicated that the number density and the transverse profile axis of mitochondria were obviously increased, and inner and cristae membrane surface areas of mitochondria were significantly decreased when compared with the controls. These changes became more serious when diabetic course was prolonged from 6 to 12 months. The results suggest that metabolic disturbances of the myocardial cells in diabetic state may be an important cause of the occurrence of diabetic cardiomyopathy.

[Studies of the herbal and botanical origins of seman Cuscutae].

Herbalogical study shows that Seman Cuscutae originated from Cuscuta chinensis is one of the most commonly used drugs in ancient times. Survey of botanical origins indicates that there are 9 species in Genus Cuscutae and 4 of them are commodities. The Cuscuta chinensis specified in current Chinese Pharmacopoeia (1985) is not the principal one.

[Effects of processing on volatile oil constituents in nutmeg and on the contents of myristicin].

This paper reports a GC analysis of the volatile oil contained in the nutmeg (Semen Myristicae) prepared by simmering wrapped in flour in hot purified talc, scalding in hot purified talc and stir-frying in smoking wheat bran. The experimental results showed that before and after processing the volatile oil compounds in nutmeg are the same and during processing no known compounds disappeared. The contents of the main volatile oil compounds in nutmeg changed, however, before and after processing, including the amounts of myristicin, one of the toxic compounds in nutmeg. It is suggested that the amounts of myristicin could be decreased notably under certain processing conditions.

[The correlation of central projection of the acupoints riyue, qimen with correlative viscera, in the rabbit].

Twenty two rabbits were selected for the experiment and were divided into three groups. CT-HRP or CB-HRP was injected respectively into the ampulla wall (group 1) or acupoint area Riyue (group 2) or Qimen (group 3). The results obtained are as follows: Group 1. The labelled cells appeared in the spinal ganglia from T3 to T11 on the left side and T2 to T10 on the right side, and a lot appeared in the nodose ganglion of the vagus nerve on both sides. The transganglionic labeled terminals were found in the region of the solitary nucleus on both sides. A lot of labelled cells appeared in the celiae ganglia on both sides, a few dorsal motor nucleus of the vagus nerve on both sides. Group 2. The labelled cells appeared in the spinal ganglia from T4 to T10 and C6 to C8 on the same side, the anterior horn of the spinal cord from T5 to T10 segments on the same side, and a lot appeared in the ganglia of the sympathetic trunk from T3 to T10 on the same side. Group 3. The labelled cells appeared in the spinal ganglia from T4 to T8 on the same side, the anterior horn of the spinal cord from T5 to T7 segments on the same side, and a lot appeared in the ganglia of the sympathetic trunk from T4 to T8 on the same side. The results of experiment shows: 1. There are 5-7 overlapping segments of sensory nerve projection between Riyue, Qimen acupoint area and ductus choledochus ampulla.(ABSTRACT TRUNCATED AT 250 WORDS)

[The effect of laser irradiation on arthritis in rats].

Based on the analgesic effect of laser therapy in clinic, the effect of laser irradiation at acupoints on the experimental arthritis in rats was further observed. The experiments were carried out on Wistar rats, which were divided into three groups: The normal group (n = 6): without any treatment. The laser group (n = 8): 24 hours after an injection of Freund's adjuvant, the typical symptoms of acute arthritis (red, swelling, pain and hyperalgesia) occurred. Then transcutaneous irradiation with a low power Helium-neon laser (3 mw, 6328 A) at ipsilateral Kun-Lun point was applied ten minutes every day for five days. The control group (n = 8): with the same treatment as the laser group except laser irradiation. All the indexes, such as foot volume, pain score and pain threshold (Heat-leg-withdrawal latency, HWL and electric-shock vocalization, VOC) were measured every day. Comparing with the symptoms in the control group, the intensity of pain as measured on pain score and the swelling of the ankles (foot volume) were significantly reduced by laser irradiation (P less than 0.01), but the pain threshold (HWL and VOC) did not show any improvement in laser group. However, immediately after laser irradiation, the average pain threshold (VOC) was significantly raised (P less than 0.01). It is concluded that low power laser irradiation at local points can produce the relief from arthralgia and reduction in the swelling of ankles, and it can also produce instant analgesic effect in test of pain threshold (VOC).

[Comparative study on the inhibitory elastase capacity of fructus Forsythiae, radix Salviae Miltiorrhizae, radix Angelicae Sinensis and Ligustrazini].

In this paper elastase capacity was measured with agar radial diffusion using elastin-agar plate. There was a linear correlation between elatase capacity and diameter of agar radial diffusion. Significant inhibitory effects of Fructus Forsythiae, Radix Salviae Miltiorrhizae, Radix Angelicae Sinensis and Ligustrazini Hydrochloridum on elastase were observed and Fructus Forsythiae proved the most effective.

[Human pulmonary ascariasis in Anhui Province: an epidemiological survey].

924 subjects, 542 males and 382 females working in grainstoring, or herb-storing, from different parts of Anhui Province were examined for the possible existence of pulmonary acariasis. 49 cases(5.3%), 31 males and 18 females, were found positive for mites in their sputum. 83.7% of the sufferers aged 16-45. 22 out of the 22 out of the 49 sufferers developed a marked eosinophilia ranging from 4% to 48% and a count of 320-5,050/mm3, whereas X-ray films revealed varied degrees of widening lung hilum shadow with increased and disordered lung markings. In some cases, the chest-film showed a lot of scattered nodular shadows 1-5 mm in diameter in the lung lobes. The symptoms presented were cough, expectoration, depression in the chest, restlessness, low fever, asthma, hemoptysis, etc. 10 species of mites were found in the sputum of the 49 sufferers, i.e. Acarus siro, Tyrophagus putreseltiae, Aleuroglyphus ovatus, Caloglyphus berlesei, C. myoophagus, Dermatophagoides farinae, D. pteronyssinus, Euroglyphus maynei, Tarsonemus granarius, Cheyletus eruditus.

TCM treatment of retinal vein occlusion in 216 cases.

Traditional Chinese Medicine was adopted in the treatment of 216 cases (219 eyes) of retinal vein occlusion. The period of treatment averaged 3 months; the total rate of efficacy was 80.4%. During an average follow-up of 3 years, 84.1% of the patients maintained a stable or slightly improved vision. The authors believed that treatment of the disease with TCM promoted the absorption of blood and formation of collateral circulation, and suppressed neovascularization, thus preventing repeated bleeding that would lead to serious complications.

Soft tissue osteochondroma. A report of three cases.

Three cases of benign soft tissue osteochondroma, a lesion of uncertain pathogenesis, are reported. Two cases were located in the subcutaneous tissues beneath the calcaneus. The other was located in the soft tissues near the left ankle joint. The diagnosis of soft tissue osteochondroma should be considered when a well-defined osseous mass is located in the soft tissues. The differential diagnosis includes myositis ossificans, tumoral calcinosis, synovial chondromatosis, and soft tissue osteosarcoma.

Increased sarcolemmal membrane permeability of cardiac muscle cells in rats fed grains cultivated in a Keshan disease endemic area: a study with horseradish peroxidase as a tracer.

The alteration of sarcolemmal membrane permeability of cardiac muscle cells in rats fed grain from a Keshan disease (KD) endemic area was studied with horseradish peroxidase (HRP) as a tracer. Weanling male rats were divided at random into three groups and fed the following three diets, respectively, for 3 months: a diet with grain from a KD endemic area (Group A), a diet with grain from a KD nonendemic area (Group B), and standard laboratory chow (Group C). At the end of the experiment, HRP was injected intravenously and localized in the ventricular myocardium by light microscopy. The experimental results showed that the percentage of cardiac muscle cells containing HRP reaction product in rats fed grain from the KD endemic area was significantly greater than that in rats fed grain from nonendemic area and standard laboratory chow. This suggests that the sarcolemmal membrane permeability of cardiac muscle cells in rats of Group A was markedly increased. The distribution of cardiac muscle cells with increased sarcolemmal membrane permeability was similar to that seen in the myocardium of KD patients. The present study suggests that the abnormal membrane permeability of cardiac muscle cells may play an important role in the pathogenesis of myocardial necrosis in KD.

Prospective, controlled, randomized trial of naloxone infusion in early hyperdynamic septic shock.

To determine whether naloxone infusion is efficacious in severe hyperdynamic septic shock, we conducted a prospective study of 22 patients randomly assigned to a naloxone or placebo group. Patients were treated 12 +/- 2 h (SEM) after the onset of shock, with a mean arterial pressure (MAP) of 63 +/- 3 mm Hg. All patients had clinical evidence of an infectious process and required dopamine 20 +/- 2 micrograms/kg.min. Five (46%) of 11 patients in the naloxone group and one (9%) of the other 11 patients in the placebo group responded clinically. The MAP among the five responders increased from 62 +/- 5 to 89 +/- 4 mm Hg within 20 min of naloxone treatment (p less than .01). This favorable hemodynamic response was sustained throughout the patients' clinical course. In contrast, the MAP did not change significantly in the nonresponders who received naloxone, nor did it change in the placebo group. More patients in the naloxone group than in the placebo group received steroids concurrently. Survival rate was 100% in those who responded to naloxone clinically. However, overall survival rate in each group was essentially the same. No adverse effects were observed, except for mild agitation in some of the patients receiving naloxone. We conclude that naloxone infusion is clinically efficacious in improving the hemodynamic profile of a subgroup of patients with severe early hyperdynamic septic shock, but does not appear to improve the overall survival rate.
