[Application of electronic larynx in speech rehabilitation after laryngectomy].

An electronic larynx has been developed at this center and used among laryngectomees for speaking rehabilitation. The phonation rate was 98% in 4,520 patients from all over the country. The frequency characteristics of the artificial larynx is close to that of human sound. Other advantages of the electronic larynx are: the best sound conducting point at the neck; natural phonation actions; and on/off switch synchronized with speaking attempts. The electronic larynx is easy to use and worth to be recommended as an alternative to surgical intervention in speech rehabilitation after laryngectomy.

[Preventive effect of gubao on hydrocortisone-induced osteoporosis in rats].

The antagonistic effect of Gubao (consisting of Herba Epimedii, Radix Astragali, etc.) on hydrocortisone-induced osteoporosis in rats was observed in quantitative bone histomorphometry study with stereological method. The results of experiment show that Gubao helps to reduce trbecular bone resorption and increase bone formation in ratsuith hydrocortisone-induced osteoporosis. It is suggested that Gubao is useful in preventing glucocorticoid-induced osteoporosis.

[Study on rearrangements of immunoglobulin genes in acute nonlymphocytic leukemia].

OBJECTIVE: To explore the mechanism of infidelity of immunoglobulin gene rearrangements in acute nonlymphocytic leukemia and its significance in genotyping of acute leukemia. METHODS: Immunoglobulin gene rearrangement was studied in 25 patients with acute nonlymphocytic leukemia in various stages by polymerase chain reaction. RESULTS: Immunoglobulin heavy chain gene rearrangements were detected in three of 25 patients, one of whom in complete remission was found to have tendency of relapse one month later. CONCLUSION: Lineage infidelity of immunoglobulin gene rearrangements does exist in acute nonlymphocytic leukemia, therefore, immunoglobulin rearrangements probably can not be regarded as an absolutely reliable marker for acute lymphocytic leukemia. Immunoglobulin rearrangements may have some significance in the detection of minimal residual disease of acute leukemia.

[Whole bladder wall laser irradiation to prevent bladder cancer recurrence with intravesical HpD and ascorbic acid].

OBJECTIVE: To evaluate the effect of whole bladder wall laser irradiation with intravesical HpD and ascorbic acid on bladder cancer recurrence. METHODS: Photodynamic therapy(PDT) was performed by intravesical instillation of hematoporphyrin derivative (HpD) PsD-007 containing ascorbic acid followed by whole bladder wall laser irradiation in 30 post-operative cases of bladder cancer. RESULTS: The tumour recurrence rate was 23.3% (7/30) on follow-up for 30 months after PDT. No photo-sensitizing effects was found on the skin of patients, nor was retraction of bladder observed in any case of this series. CONCLUSION: This method is effective in the prevention of bladder cancer recurrence. Protection from light is not necessary during treatment.

[Retroviral vector mediated gene transfer and expression of human GM-CSF in HL-60 leukemic cells].

OBJECTIVE: To study the condition and possibility for introducing foreign gene into myeloid leukemic cells and gene therapy for malignant tumors in hematology. METHODS: With Lipofectin technique, N2A/CMV/hGM-CSF was transfected into packaging cell line PA317 and recombinant retrovirus were obtained. Then the human myeloid leukemic cells HL-60 were infected with recombinant retroviral supernatant. Positive cells were obtained through the neomycin analogue G418 selection. RESULTS: Successful integration of GM-CSF gene into HL-60 cell genome was verified by PCR and Southern Blot, while no specific fragments were amplified by PCR test in HL-60 cells without being transferred or transferred with empty vector N2A. With measurement by GM-CSF dependent cell line TF-1, the level of GM-CSF released by the GM-CSF gene modi-fied HL-60 cell line was 60-200 ng.ml-1.10(-6).24 h-1, while the supernatant of HL-60 cells without being transferred or transferred with empty vector N2A. CONCLUSION: hGM-CSF gene mediated by retroviral vector can achieve efficient transfer and expression in HL-60 leukemic cells.

[Mechanism and treatment of cancer cachexia in tumor-bearing mice].

OBJECTIVE: To evaluate the role of TNF-alpha in the development of cancer cachexia, and the effects of TNF-alpha antibody and high agglutinated staphylococcin (BM828) in the treatment of experimental cancer cachexia. METHODS: T739 mice bearing LA795 tumor were used as murine tumor cachexia model and also the T739 receiving TNF-alpha. The level of serum TNF-alpha was assayed by TNF-alpha RIA in each group. Comparisons were made in the food intake and body weight of all the five groups including BM828 and TNF-alpha AB groups. RESULTS: Two weeks after being inoculated with LA795, the tumor-bearing mice developed significant anorexia and weight loss. The level of serum TNF-alpha in tumor-bearing mice was significantly higher than that in the non-tumor bearing mice (P < 0.01). TNF-alpha administration to mice resulted also in marked anorexia and weight loss as in cancer cachexia. CONCLUSION: The results suggested that the mechanism of cachexia be correlated with TNF-alpha. BM828 and TNF-alpha AB have the potentiality of attenuating the development of cachexia in murine models.

[Effect of ascorbate on the permeation and photosensitizing activity of hematoporphyrin derivative (HPD) in tumor].

OBJECTIVE: To determine the depth of permeation and concentration of HPD in tumor tissue which had been immersed in HPD in the presence and absence of ascorbic acid, and to examine its photosensitizing effect. METHODS: Solid S-180 sarcoma of 1 cm3 in size was excised from tumor-bearing mice and immersed in HPD (PsD-007) 1 mg/ml normal saline (group I), or HPD (PsD-007) 1 mg/ml plus ascorbic acid 20 mg/ml normal saline (group II) for 1 hr. For comparison, HPD (PsD-007) 10 mg/kg was injected intravenously to mice bearing tumor of similar size and the tumor was excised 24 hr later(group III). RESULTS: When the tumors were cross-sectioned and UV (370 nm) irradiated, red fluorescence was mainly at the periphery of tumors that had been immersed in HPD whereas the fluorescence was weaker and more evenly distributed in tumors of mice that had received HPD i.v. Similar features were observed under fluorescence microscope in frozen sections of identically treated tumors. The concentration of PsD-007 (microgram/g) and malondialdehyde (nmol/L) in the tumor homogenates was higher in tumors that had been immersed in PsD-007 plus ascorbate than in tumors immersed in PsD-007 alone. Tumors of mice that had received PsD-007 i.v. had the lowest concentration of both PsD-007 and malondialdehyde. CONCLUSION: Ascorbate facilitates permeation of HPD into tumor and enhances the photo-dynamic effect of HPD.

[Outward character and inner quality of Bombyx Batryticatus, Carapax Trionycis, Squama Manitis and endothelium corneum Gigeriae Galli after frying].

According to the requirement for outward character in processing traditional Chinese medicines, Bombyx Batryticatus, Carapax Trionycis, Squama Manitis and Endothelium Corneum Gigeriae Galli, which are four kinds of often used traditional Chinese animal medicines, were fried, the contents of amino acids in each kind were determined, and investigate the inner quality of each kind of the processed product was investigated with reference to other standards.

[Bianticollagen membrane: preparation and analysis of properties].

This study was undertaken to assess the antiplaque and anticollagenase properties of bianticollagen membrane (BACM) for use in the guided tissue regeneration (GTR). First, preparing cross-linked collagen membrane (GLCM) from bone collagen by glutaraldehyde and ultraviolet irradiation. Then, the GLCM was coated with tetracycline (TC) delivery device, which is BACM. BACM's properties are as follows: under scan electron microscope (SEM), it is a three dimensional structure with small pores; the modulus of elasticity in low strain regions and swelling ratio are 20.4 g/mm2 and 0.141 respectively; immediate type and delayed type hypersensitivity are negative; BACM with TC 150 micrograms placed into pockets, the average intrasulcular TC concentration measured at the end of the 7-days is 46.76 +/- 5.69 micromol/L, which is 2 times higher than MIC of TC; the period against collagenase (100 u) digestion is over 30 days in vitro; 1 mg GLCM and BACM implanted in mice may maintain about 4 weeks and 7 weeks respectively, and BACM and GLCM placed onto patient's root surface in flap operation and taken at 7th day the bacteria on BACM are significantly less than that on GLCM under SEM. These results indicate that BACM has antiplaque and stronger antidegradation effects than GLCM.

[The high expression of human beta-nerve growth factor in E. coli].

The Hu beta-NGF gene encoding the beta subunit of mature human nerve growth factor was cloned into the pET11c vector under the control of T7 bacteriophage promoter. The SDS-PAGE electrophoresis results of nonreduced recombinant products showed a clear band corresponding to homodimeric (27 kD) form of the molecule. But the SDS-PAGE results of reduced expression protein showed a single band corresponding to monomeric form of Hu beta-NGF (13.5 kD). It represented approximately 14.5% of the total cellular protein. Both of them were immunopositive on Western-Blots with rabbit anti-m beta-NGF polyclonic antibodies. And the recombinant product was biologically active on cultured chicken dorsal root ganglion neurons. So it demonstrated the feasibility of synthesizing the biologically active forms of Hu beta-NGF in E. coli.

[Diuretic effect of liniaoling(KLP)].

Experimental results have shown that KLP taken orally helps promote the diuretic action of normal Wistar rats, decrease the urine volume of debilitated rats and increase the urine volume of BPH rats. It has also been shown that KLP(i.v.) can increase the urine volume of anesthetic rats and reduce the remnant urine. In addition, KLP can contract the smooth muscle of rats and rabbits.

[Amplification, clone and identification of the specific fragments of tumor metastasis-suppressor gene nm23-H1 and nm23-H2].

A series of DNA primers specific for the specific fragments of nm23-H1 and nm23-H2 were designed and synthesized. The specific fragments of nm23-H1(185 bp) and nm23-H2(145 bp) were amplified from human blood DNA by using polymerase chain reaction (PCR). The recovered PCR products were treated with Klenow fragment, inserted into pGEM-3zf(+) vector with blunt-end ligation, and then transformed into competent cell JM109. The positive colonies were directly identified by colour screening on indicator plates. The recombinant plasmids were digested by Alu I and identified by PCR. The results showed that the authors had obtained the specific fragments of nm23-H1 and nm23-H2 respectively, and these specific fragments could be used for study the expression of nm23-H1 and nm23-H2 seperately.

[mRNA expression of metastasis-suppressor gene nm23 in carcinoma of buccal mucosa I. Northern blotting study].

In this study the authors detected both nm23-H1 and nm23-H2 mRNA levels in 52 tissues samples of patients with carcinoma of buccal mucosa (CBM) by Northern blotting method. The expression levels of both nm23-H1 and nm23-H2 mRNA in CBM were higher than those in normal buccal mucosa, leukoplakia, adjacent nontumorous mucosa, and lymph nodes with or without metastasis. The nm23-H1 mRNA expression levels in CBM with lymph nodes metastases were lower than those in CBM without metastasis, and were lower in metastatic lympho nodes (P < 0.05). Of the 11 cases with metastases, 9 cases (81.8%) showed low expression of nm23-H1 mRNA, of 19 cases without metastasis, 15 cases (78.9%) were high expression. No significance of nm23-H2 mRNA expression levels were found between CBM with and CBM without metastasis (P > 0.05). The expression of nm23-H1 and nm23-H2 mRNA showed no significant correlation with each clinical-pathological parameter examed. The result suggested that the expression of nm23-H1 mRNA significantly correlated inversely with lymph node metastasis in CBM, while the expression of nm23-H2 mRNA not. nm23 mRNA expression levels can provide a valuable predictor of lymph node metastases in CBM.

[An outline medical history of Taiwan (I): the period of folklore medicine and witch doctor].

The paper makes a correlated analysis on the origin of health folklore between Chinese in mainland and Taiwan island. After quoting literatures written by authors living in the Qing dynasty in Taiwan, this paper analyses health condition among aboriginals of Taiwan during the witchcraft age. Along with the increasing immigration from China mainland to Taiwan island, health of folklore and gods from China mainland were introduced into Taiwan, hence the period of witch doctor in Taiwan, featuring the correlation of both. Though modern medicine in Taiwan is so advanced, yet there are still witch doctors elsewhere.

[Influence of leaf-picking on effective constituents of the Astragalus mongholicus roots].

After the application of the orthogonal design, experiments show that the proportion and time of leaf-picking have a great influence not only on the total content of 17 kinds of amino acids but also on that of Astragaloside. If the proportion is large and the time is delated, both contents of 17 kinds of amino acids and Astragaloside will be reduced accordingly compared to the control group. There is less influence on the contents if the time of leaf-picking is set on June 15 each year and the picking proportion is 1 out 3.

[Studies on the relationship between purified alpha class glutathione S-transferase (GST-alpha) and hepatocellular damage].

Using our ELISA kit for GST-alpha, we tested the serum GST-alpha levels of patients with hepatitis A, acute hepatitis B (HB), chronic active HB, chronic persistent HB and liver cirrhosis. Serum GST-alpha levels in all these groups of patients were significantly higher than those in the controls (P < 0.01). Serum GST-alpha levels was closely (P < 0.01) correlated with serum alanine transaminase (ALT) levels in various groups of patients except chronic persistant HB. The combined application of the two markers, GST-alpha and ALT, raised the sensitivity of detection for liver diseases. In detection of chronic persistent HB patients, the GST-alpha marker was more sensitive than ALT (P < 0.01). The follow-up data of GST-alpha and ALT markers in serum showed that the GST-alpha level could reflect the clinical progression of liver disease more exactly as observed in 79 liver cancer patients in then GST-alpha and ALT positive rate were 81% and 61% respectively (P < 0.01). In 30 persons with positive HBsAg, the positive rates of GST-alpha and ALT were 70% and 37%, respectively. These results indicated that, (1) the detection of GST-alpha combined with ALT was capable of increasing the sensitivity for recognizing hepatocellular damage; (2) the elevation of serum GST-alpha level mainly resulted from the increased expression of GST-alpha in liver cells during hepatocarcinogenesis, thus, the GST-alpha is thought to be a tumor marker for liver cancer; (3) GST-alpha measurement offers advantage over ALT for the detection of minor degree of hepatocellular damages. GST-alpha may act as an early, sensitive and specific enzyme marker.

[Study on enterovirus aetiology of CNS infection in Shantou].

Enterovirus isolation and serological assay were done in 236 cases of CNS infection in Shantou. 62 strains of virus (from 55 cases) were isolated from 257 samples of 236 cases (24.1%). 48 cases were positive among 57 cases (84.2%) by serological assay. 78 cases were positive for virus isolation and/or serological assay (33.1%). Virus infection included 33 cases of poliovirus (including 6 polio-I, 18 polio-II, 4 polio-III, 5 mixed infections), 21 cases of Coxsackie virus (including 6 CoxA24,7 CoxB1,2 CoxB4 and 6 CoxB5), 11 cases of ECHO virus (including 5 ECHO-7,2 ECHO-24 and each one of ECHO-11, 12, 15, 27), 10 cases of Enterovirus (including 3 EV-70, 7 EV-71), and 3 cases of mixed viral infection.
