[Experimental study on effect of Buyang Huanwu decoction on arterial blood platelet activating factor content pre- and post-arterial thrombosis of rats].

OBJECTIVE: To explore the effect of Buyang Huanwu Decoction (BHD) on arterial blood platelet factor (PAF) content in pre- and post-arterial thrombosis of rats. METHODS: Male Wistar rats were randomized into 3 groups, the BHD group treated with BHD 32 g/kg, or with BHD 16 g/kg daily for 14 days consecutively, the control group treated with dexamethasone 0.5-1 mg/kg and the blank group treated with distilled water 10 ml/kg. Arterial thrombosis was duplicated in the animals 2 hours after final medication, the blood content of PAF (by bioassay), dry weight (W) and occlusion time (OT) of thrombus, and weight of thrombus/body weight (TW/BW) ratio were observed. RESULTS: Consecutive BHD treatment could markedly lower the arterial blood content of PAF after thrombosis, increase the OT value of thrombus, reduce the dry weight of thrombus and the TW/BW ratio (P < 0.05), while no influence was found in rats treated with BHD once. CONCLUSIONS: BHD has certain antagonistic effect on pathologic PAF metabolism, and inhibitory effect on pathogenesis and development of arterial thrombosis.

[Clinical observation on effect of yangjing decoction in promoting follicular growth].

OBJECTIVE: To investigate the effect of Yangjing Decoction (YJD, a Chinese prescription of replenishing Kidney essence) in promoting follicular growth and its influence on blood supply of ovary and uterus. METHODS: Twenty-two cases with secondary amenorrhea and 4 cases of infertility with infrequent menstruation caused by follicle maldevelopment were treated with YJD. The blood supply of ovary and uterus in 7 cases was measured by colour Doppler before and after treatment. RESULTS: Of the 116 therapeutic cycles in 26 cases, there were 69 menstrual cycles (59.48%), 37 follicular maturation cycles (31.90%), 21 ovulation cycles (18.10%), and the follicular maturation rate was 84.62%, The disturbance in blood supply of ovary and uterus was improved remarkably along with follicular development by YJD in 7 cases. CONCLUSION: YJD could improve follicular development and blood supply of ovary and uterus.

[Preliminary study on therapeutic effect of oxymatrine in treating patients with chronic hepatitis C].

OBJECTIVE: To evaluate the efficacy of oxymatrine in treating chronic hepatitis C and its mechanism. METHODS: Forty-three patient were divided randomly into the treated group (20 cases) and the control group (23 cases). The treated group was given oxymatrine 600 mg per day intramuscularly, and the control group was given the general liver protective agents such as vitamins. The therapeutic course of both groups was 3 months. RESULTS: HCVRNA of 8 in 17 cases (47.1%) of the treated group converted to negative, while in 18 cases of the control group, the negative conversion only took place in 1 patient (5.6%), the negative conversion rate was significantly higher in the treated group than that in the control group (P < 0.05). The normalization rates of serum alanine transaminase (ALT) of the treated group after 1 month and 2 months treatment was higher than that of the control group, but after 3 months treatment, the normalization rates of the two groups were not different significantly. Plasma level of soluble interleukin-2 receptor and serum level of collagen type IV in the treated group were lowered significantly after treatment, but in the control group, there were no significant change, the difference between the two groups was significant (P < 0.01, P < 0.05). CONCLUSION: Oxymatrine is effective in inhibiting proliferation of HCV, antagonisting liver fibrosis and regulating immune reaction of the host, so it could be a safe, effective drug in treating chronic hepatitis C.

[Clinical study of jiuxin fumai injection in treatment of cardiogenic shock].

OBJECTIVE: To increase the cure and improvement rates and to decrease the mortality rate in the treatment of cardiogenic shock. METHODS: Thirty-one patients with cardiogenic shock were studied, and received Jiuxin Fumai injection (JXFM) in research group (n = 20) or dopamine (DA) in control group (n = 11). RESULTS: Eighteen effective cases (90.0%) and 2 ineffective cases (10.0%) in JXFM group, 9 effective cases (81.8%) and 2 ineffective cases (18.2%) in DA group. There was significant difference statistically, P > 0.05. CONCLUSIONS: JXFM and DA had the same effect in elevating blood pressure, enhancing pump function of the heart and ameliorating collapse syndrome. They are safe, effective, convenient for use and prompt in responding. However, JXFM showed raising blood pressure faster, higher as well as more stable, and the dual phase phenomenon in adjusting heart rate, which could slow down tachycardia as well as speed up bradycardia were observed.

[Effect of turmeric volatile oil on the respiratory tract].

OBJECTIVE: Researching the effect of turmeric volatile oil on functions of the respiratory tract. METHOD: The action of turmeric volatile oil on sputum removing, cough and asthma relieving was observed by vapour inhaling. RESULT: The volatile oil is significantly active in removing sputum, relieving cough and preventing asthma. CONCLUSION: Turmeric volatile oil may be an efficacious drug for the treatment of respiratory diseases.

[Reversal effect of Ys-96, a bisbenzylisoquinoline, on adriamycin or vincristine resistance in human cancer cells in vitro].

OBJECTIVE: Reversal effect of Ys-96, a bisbenzylisoquinoline, on the resistance of human cancer cells to adriamycin or vincristine was studied in vitro. METHOD: In an in vitro culture system of human cancer cells MCF-7 and its adriamycin-resistant line MCF-7/Ad or KB and its vincristine-resistant line KBv200, the sensitivity(IC50) of the resistant cell lines to adriamycin or vincristine was evaluated with a MTT assay. RESULT: IC50 value of adriamycin or vincristine in combination with Ys-96 at a concentration of 1.00 mumol/L against MCF-7/Ad or KBv200 was found to be close to that of adriamycin alone or vincristine alone against the sensitive cell line MCF-7 or KB. CONCLUSION: The drug resistance of MCF-7/Ad or KBv200 could be essentially reversed by Ys-96 at a concentration of 1.00 mumol/L.

[The influence of side-to-side portacaval shunt plus enhancement of perfusion of hepatic artery on cirrhotic liver: an experimental study].

OBJECTIVE: To for improving the perfusion of liver, we designed an operation of portacaval shunt (PCS) plus enhancement of perfusion of hepatic artery (EPHA). METHOD: 48 wistar rats were divided into 4 groups randomly. group I (normal control), group II (liver cirrhotic control), group III (PCS on liver cirrhosis), and group IV (PCS + EPHA on liver cirrhosis). The EPHA was performed by the ligation of the left gastric and splenic artery. Liver function tests, liver biopsy, and nuclide hepatobiliary dynamic imaging (NHDI) were performed on the 2nd week and the 6th month after operation. RESULT: In NHDI, the peak time was short (P < 0.05) and the excretive rate was high (P < 0.01). The liver function and the proliferative degree of the fibrotic tissue of the liver were perfected significantly (P < 0.05) in the group IV compared with the group III after operation. CONCLUSION: PCS + EPHA can increase the perfusion of the hepatic artery, maintain the liver function and delay liver cirrhosis, and overcome the side effects of PCS. It is a new surgical technique in treating portal hypertension.

[The changes of pancreatic function after cardiopulmonary bypass].

OBJECTIVE: To evaluate the changes of endocrine and exocrine pancreatic function in patients who underwent heart valve replacement under cardiopulmonary bypass (CPB) in order to optimally conside clinical management perioperatively. METHOD: Sixteen patients with heart valve disease were selected. Serum glucose, lactate, insulin, C peptide, amylase and lipase were measured. RESULT: Hyperglycemia and hyperlactacemia were noted after CPB and peaked at 19:00 on the operative day. Their changes were correctly correlated. Serum glucose and lactate returned to the baseline level on the third postoperative day. The average level of serum insulin were normal, but higher on the third postoperative day. However, the level of serum C peptide was higher than the baseline. It doubled the baseline level at 19:00 on the operative day. The average level of serum amylase and lipase was normal, except the higher level of serum lipase on the fourth postoperative day. In our group, 12 patients had normal serum amylase level perioperatively, and 4 increased serum amylase level (25%), among them 2 patients accompanied with hyperlipasimia (16%). There were 8 patients with hyperlipasimia (50%). CONCLUSION: The endocrine and exocrine pancreatic functions were influenced by CPB and did produce pathophysiologic changes.

[Experimental study on regenerative capacity and form of corneal endothelial cells in the primate].

OBJECTIVE: To investigate the regenerative capacity and form of the corneal endothelial cells of the primate. METHODS: Chromosome, HE stainings and autoradiography were made for the cornea of human and monkey. RESULTS: After trauma, corneal endothelial cells of the adult monkey in vivo showed regenerative power. The regenerative form of corneal endothelial cells of the adult, infant and fetal primate was amitosis under both physiologic and pathologic conditions. CONCLUSION: The corneal endothelial cells of the primate have regenerative power, and the form is amitosis.

[Determination and significance of catecholamines in aqueous humor, plasma and 24 hour urine of patients with acute angle-closure glaucoma].

OBJECTIVE: To study the functional state of ocular and systemic sympathetic nervous system and the effects of its related neurohumoral factor, catecholamines (CA), on intraocular pressure (IOP) in patients with acute angle-closure glaucoma (AACG). METHODS: The levels of CA in the aqueous humor, plasma and the total amount of CA in 24 hour urine of cases with AACG during attack stage were determined with fluorometry, and the results were compared to that of patients with senile cataract and normal adults. RESULTS: The CA levels within aqueous humor and plasma and in 24 hour urine of the patients with AACG during attack stage were elevated obviously as compared with that of the control subjects (P < 0.001), and positively correlated with the level of IOP. CONCLUSION: During attack stage of AACG, the ocular sympathetic nervous system is highly excited and a large amount of CA is released into the aqueous humor, in the meantime the systemic sympathetic nervous system is also excited and CA is released into the blood circulation, which may play a certain role in the regulation of IOP. Possibly, CA is an important supplemental factor within the attack stage of AACG.

[A preliminary observation on vitrectomy combined with autologous platelet concentrate for the treatment of idiopathic macular holes].

OBJECTIVE: To investigate the efficacy of vitrectomy combined with autologous platelet concentrate for the treatment of idiopathic macular holes. METHODS: Fifteen eyes of 14 patients with idiopathic macular hole were treated. The follow-up period ranged from 3 to 16 months. Postoperatively, visual acuity, macular threshold, Amsler grid, fluorescein angiography (FA) and complications were evaluated. RESULTS: Of the fifteen eyes, the closure of the holes was achieved in 13 eyes (86.7%), visual acuity improved for two lines or more in 12 eyes (80.0%). The postoperative visual acuities in patients with abnormal Amsler grid were improved. No significant differences were found in the macular threshold (P = 0.34). Postoperative FA showed that the blocked fluorescence at the site of the macular hole which was surrounded by a hyperfluorescence ring (window defect) in three eyes. The intraoperative complication was primarily the retinal hole, and the postoperative complication was chiefly the aggravation of cataract. CONCLUSIONS: These results indicate that vitrectomy combined with autologous platelet concentrate can seal the macular hole well and improve the visual acuity in most patients. A large multicenter randomized prospective study and a longer period of follow-up are needed to verify these results.

[Synthesis and structure-activity relationships of novel 14 beta-side chain taxol derivatives].

In order to develop new generation of taxol-like anticancer agents with fewer side effects, improved activity, superior pharmacological properties and broad antitumor spectrum, along with structure-activity relationship study, a series of new taxol derivatives are to be synthesized starting from sinenxan A--a biosynthetic taxane. Eight new 14 beta-side chain taxol derivatives with 4-OH and 4-Ac were synthesized in 5 and 6 steps from semisynthetic taxoid intermediate 7, respectively. These included taxol derivatives modified at C-2 position with benzoate, m-Cl benzoate, valerate and phenylacetate. All target compounds together with two other 14 beta-side chain taxol derivatives were tested in a microtubule assembly assay, and in an in vitro cytotoxicity assay (KB, A2780, HCT-8 cell line). All compounds had no effect in the microtubule assembly assay at 10 mumol.L-1. Most of them showed marginal activity in the in vitro cytotoxicity. The structure-activity relationship was different from taxol derivatives. 2-Aliphatic ester displayed similar activity to 2-aromatic ester, which indicated that the aromatic group at C-2 position was not important for cytotoxicity. Comparing among themselves, 4-OH derivatives possessed stronger activity than 4-Ac.

[In vitro sensitivity of oral gram-negative bacteria to the bactericidal activity of defensins].

Oral gram-negative bacteria, including Actinobacillus actinomycetemcomitans, Porphyromonas gingivalis, Capnocytophaga sputigena, Provotella intermedius, Provotella melaninogenicus, have been associated with destructive periodontal infection. An ultrasensitive antibacterial assay was applied to determine the sensitivity of a battery of oral gram-negative bacteria to the bactericidal effects of the isolated human neutrophil defensins (HNP) and two kinds of rabbit defensins NP1 and NP2. All species tested were killed by HNP, NP1 and NP2 except that porphyromonas gingivalis was resistant to HNP. However, there was strain-to-strain variation in sensitivity. It was concluded that the oral gram-negative bacteria were sensitive to the cidal mechanism involved in defensinmediated bacterial killing.

[Longitudinal surveillance of sensitivity of Plasmodium falciparum to pyronaridine in south Yunnan].

AIM: To understand the changes in the sensitivity of Plasmodium falciparum to pyronaridine in the south of Yunnan Province. METHODS: WHO standard in vivo and in vitro microtest was used. RESULTS: In 1984-1985 and 1995, 36 and 39 cases of acute falciparum malaria were observed, the average defervescence time being 32.7 +/- 16.0 h and 56.2 +/- 27.4 h, the mean asexual parasite clearance time, 64.2 +/- 22.9 h and 55.3 +/- 11.8 h, and the recrudescence rate of patients followed up for 28 days, 15.2%(5/33) and 37.5%(9/24), respectively. The resistance rates of resistant isolates by in vitro microtest were 0(0/24), 8.8%(3/34), 13.8%(4/29) and 36.4%(4/11) in 1988, 1990, 1992 and 1995, respectively, while the corresponding ID50 were 13.0, 20.0, 32.0 and 40.0 nmol/L, ID95 were 48.0, 136.0, 325.0 and 330.0 nmol/L, the mean concentrations for complete inhibition of schizont formation were 32.6, 99.5, 148.2 and 190.1 nmol/L, respectively. CONCLUSION: The sensitivity of Plasmodium falciparum to pyronaridine was found to have a tendency of continuous dropping in south Yunnan.

[An epidemiological survey of schistosome cercarial dermatitis among the residents living along the banks of the Huaihe River system].

AIM: To make an epidemiological survey of schistosome cercarial dermatitis among the residents living along the banks of the Huaihe River. METHODS: Making inquiries about signs and symptoms of schistosome cercarial dermatitis, and physical examinations of manifestation in the residents (fishermen and peasants). Cercariae shed from the Radix auricularia collected in the field were used to infect ducklings. The eggs and miracidia separated from the feces of the full grown ducks bred by fishermen were used to infect Radix auricularia. RESULTS: Schistosome dermatitis locally known as "duck itch" (meaning duck-fecesdermatitis) was discovered among the residents living along the Huaihe River bank. The main signs and symptoms of the dermatitis included diffuse erythema or red papular eruption with areorae, urticae, urticant itch or titillation around the affected areas (including thoracic part, abdomen and lower limbs etc). The cercariae with eye spot were found from Radix auricularia collected from Huaihe riverside and were identified to be the species of Trichobilharzia can both hatch miracidium, which laboratory-reared Radix auricularia were infected with miracidia hatched from the two types of eggs, rhomboid eggs and crescent eggs, found in the feces of full grown ducks bred by fishermen and shed the same cereariae with eye spot. Besides, adult worms of Trichobiharzia were obtained by dissecting both experimentally infected ducklings bred in the laboratory and naturally infected ducks bred in the Huaihe river-side area. The above-mentioned eggs, miracidia, cercariae and adult worms were all identified to belong to the life-phase of Trichobilharzia. CONCLUSION: The epidemic disease known as schistosoma cercarial dermatitis harmful to the health of the residents living along the banks of Huaihe River is caused by the cercariae of Trichobilharzia.

[Human herpesvirus-6 infection induces IL-10 production in monocytes].

In this report, interleukin 10(IL-10) and other cytokines were evaluated through reverse transcription-polymerase chain reaction (RT-PCR) and sandwich ELISA to explore possible immune pathogenesis of human herpesvirus 6 (HHV-6) infection. The results showed that IL-10 was expressed in HHV-6 infected monocytes. Kinetic studies found that transcription of IL-1 beta, IL-6 and TNF-alpha mRNA decreased with accumulation of IL-10 mRNA. When endogenous IL-10 induced by HHV-6 was blocked using anti-human IL-10 monoclonal antibody, the production and the expression of TNF-alpha, IL-1 beta and IL-6 markedly increased as compared with infection by HHV-6 alone, indicating that endogenous IL-10 inhibited cytokine production at the transcription level in HHV-6 infected monocytes. IL-10 has been shown to suppress Th1 response and down-regulate monocyte/macrophage function. The data from our studies suggest that disturbance of immune function and latent infection resulting from HHV-6 infection might partially contribute to endogenous IL-10 production.
