Activated polo-like kinase Plx1 is required at multiple points during mitosis in Xenopus laevis.

Entry into mitosis depends upon activation of the dual-specificity phosphatase Cdc25C, which dephosphorylates and activates the cyclin B-Cdc2 complex. Previous work has shown that the Xenopus polo-like kinase Plx1 can phosphorylate and activate Cdc25C in vitro. In the work presented here, we demonstrate that Plx1 is activated in vivo during oocyte maturation with the same kinetics as Cdc25C. Microinjection of wild-type Plx1 into Xenopus oocytes accelerated the rate of activation of Cdc25C and cyclin B-Cdc2. Conversely, microinjection of either an antibody against Plx1 or kinase-dead Plx1 significantly inhibited the activation of Cdc25C and cyclin B-Cdc2. This effect could be reversed by injection of active Cdc25C, indicating that Plx1 is upstream of Cdc25C. However, injection of Cdc25C, which directly activates cyclin B-Cdc2, also caused activation of Plx1, suggesting that a positive feedback loop exists in the Plx1 activation pathway. Other experiments show that injection of Plx1 antibody into early embryos, which do not require Cdc25C for the activation of cyclin B-Cdc2, resulted in an arrest of cleavage that was associated with monopolar spindles. These results demonstrate that in Xenopus laevis, Plx1 plays important roles both in the activation of Cdc25C at the initiation of mitosis and in spindle assembly at late stages of mitosis.

Asymptomatic non-ulcerative genital tract infections in a rural Ugandan population.

OBJECTIVE: To document the prevalence of asymptomatic non-ulcerative genital tract infections (GTI) in a rural African cohort. METHODS: The study population consisted of all adults aged 15-59 residing in 56 rural communities of Rakai District, southwest Uganda, enrolled in the Rakai STD Control for AIDS Prevention Study. Participants were interviewed about the occurrence of vaginal or urethral discharge and frequent or painful urination in the previous 6 months. Respondents were asked to provide blood and a first catch urine sample. Serum was tested for HIV-1. Urine was tested with ligase chain reaction (LCR) for N gonorrhoeae and C trachomatis. Women provided two self administered vaginal swabs; one for T vaginalis culture and the other for a Gram stained slide for bacterial vaginosis (BV) diagnosis. RESULTS: A total of 12,827 men and women were enrolled. Among 5140 men providing specimens, 0.9% had gonorrhoea and 2.1% had chlamydia. Among 6356 women, 1.5% had gonorrhoea, 2.4% had chlamydia, 23.8% were infected with trichomonas and 50.9% had BV.53% of men and 66% of women with gonorrhoea did not report genital discharge or dysuria at anytime within the previous 6 months. 92% of men and 76% of women with chlamydia and over 80% of women with trichomonas or BV were asymptomatic. The sensitivities of dysuria or urethral discharge for detection of infection with either gonorrhoea or chlamydia among men were only 21.4% and 9.8% respectively; similarly, among women the sensitivity of dysuria was 21.0% while that of vaginal discharge was 11.6%. For trichomonas or BV the sensitivity of dysuria was 11.7% and that of vaginal discharge was 10.5%. CONCLUSION: The prevalence of non-ulcerative GTIs is very high in this rural African population and the majority are asymptomatic. Reliance on reported symptoms alone would have missed 80% of men and 72% of women with either gonorrhoea or chlamydia, and over 80% of women with trichomonas or BV. To achieve STD control in this and similar populations public health programmes must target asymptomatic infections.

Fos response of fetal sheep anterior circumventricular organs to osmotic challenge in late gestation.

We hypothesized that the anterior circumventricular organs (ACVO) and the supraoptic (SON) and hypothalamic paraventricular nuclei (PVN), among other structures that play a role in sensing extracellular body fluid volume and composition in postnatal animals (as demonstrated by Fos protein production by the immediate-early gene c-fos), would show similar activation in fetal sheep during an osmotic challenge. The brains of 10 fetal sheep [6 treated, 4 controls; 129-131 days of gestational age (dGA) = 0.87 gestation] were immunostained for Fos. Seventy-five minutes before tissue collection the dams were given intravenous 20% mannitol (1 ml . min-1 . kg-1 for 10 min). Subsequently, the ACVO, SON, and PVN were scored for the amount of neuronal Fos immunostaining. The subfornical organ (SFO; 24.5 +/- 9.0 vs. 1.7 +/- 1.2), the organum vasculosum of the lamina terminalis (OVLT; 26.8 +/- 5.6 vs. 7.0 +/- 2.0), the SON (39.8 +/- 3.0 vs. 0.15 +/- 0.1), and the PVN (59.8 +/- 7.9 vs. 0.7 +/- 0.7) had increases (P < 0.05) in the average number of Fos-positive cells per field compared with controls, whereas the median preoptic nucleus did not. Double immunostaining for Fos and arginine vasopressin (AVP) or oxytocin (OT) indicated that AVP- but not OT-immunopositive neurons in SON and PVN respond to osmotic challenge. These results demonstrate that the SFO, OVLT, SON, and PVN are activated by osmotic challenge in fetal sheep at 130 dGA.

The acute suckling stimulus induces expression of neuropeptide Y (NPY) in cells in the dorsomedial hypothalamus and increases NPY expression in the arcuate nucleus.

Elevated neuropeptide Y (NPY) levels in the hypothalamus have been reported during lactation in the rat. The increase in NPY neuronal activity may be important in modulating a number of changes in hypothalamic neuronal function that are associated with lactation. The aims of the present study were to determine 1) if NPY neurons in the hypothalamus can be activated by the suckling stimulus; and 2) the time course of the activation in response to the suckling stimulus. In the first experiment, lactating rats were deprived of their 8-pup litters on day 9 post partum for 48 h. On day 11, the animals were divided into three groups and exposed to the suckling stimulus for varying periods of time up to 24 h. NPY neuronal activity was assessed by measuring changes in NPY messenger RNA (mRNA) levels, using in situ hybridization. NPY mRNA levels in the caudal portion of the hypothalamic arcuate nucleus (ARH) were approximately doubled by 24 h of suckling. NPY mRNA levels in the rostral portion of the ARH were not affected by suckling throughout the time examined. In addition to increased NPY mRNA in the ARH, resuckling for as little as 3 h induced NPY mRNA expression in cells located dorsal and lateral to the compact zone of the dorsomedial nucleus of the hypothalamus (DMH). NPY expression in these cells was not observed in the nonresuckled controls. These data demonstrate that the acute suckling stimulus activates two specific populations of NPY neurons in the hypothalamus: in the caudal portion of the ARH and in the DMH. The increased NPY neuronal activity may play an important role in modulating changes in hypothalamic regulation of hormone secretion and food intake.

The ontogeny of glucocorticoid negative feedback: influence of maternal deprivation.

Glucocorticoid feedback can be viewed as having two modes of operation: proactive and reactive. "Proactive" feedback maintains basal activity of the hypothalamic-pituitary-adrenal axis, whereas the termination of stress-induced hypothalamic-pituitary-adrenal activity is facilitated by "reactive" feedback. In the present study we studied the ontogeny of both feedback modes and tested the hypothesis that the development of feedback depends on mother-pup interaction. On postnatal day 9 or 12, pups were deprived (DEP) of the dam for 24 h; nondeprived pups of the same age served as controls. The pups were adrenalectomized (ADX) at the end of deprivation and given corticosterone (CORT) replacement by either injection or pellet implants using the following two designs: first at the time of adrenalectomy (ADX) to test the role of CORT in the maintenance of basal ACTH levels, and then 3 h after ADX, to investigate CORT suppression of elevated ACTH levels induced by prior ADX. Regarding proactive feedback, the results showed that injection of CORT at the time of ADX was only partially effective in preventing ACTH elevations, whereas CORT pellets maintained basal levels of ACTH in all ADX pups. The reactive mode of negative feedback in nondeprived pups was resistant to CORT injection, whereas the CORT pellet resulted in a return to basal levels within 60 min. Maternal deprivation did not affect proactive feedback, but caused a more sustained increase in ACTH levels and a failure to return to basal levels 3 h after ADX despite significantly higher levels of circulating CORT in these DEP pups. It is concluded that 1) proactive and reactive modes of negative feedback are operative, provided the pups are maintained on chronic replacement with CORT; 2) DEP impairs the reactive, rather than the proactive, mode of feedback inhibition in the neonate.

Fibroblast growth factor receptor 2 (FGFR2)-mediated reciprocal regulation loop between FGF8 and FGF10 is essential for limb induction.

FGFR2 is a membrane-spanning tyrosine kinase that serves as a high affinity receptor for several members of the fibroblast growth factor (FGF) family. To explore functions of FGF/FGFR2 signals in development, we have mutated FGFR2 by deleting the entire immunoglobin-like domain III of the receptor. We showed that murine FGFR2 is essential for chorioallantoic fusion and placenta trophoblast cell proliferation. Fgfr2(DeltaIgIII/DeltaIgIII) embryos displayed two distinct defects that resulted in failures in formation of a functional placenta. About one third of the mutants failed to form the chorioallantoic fusion junction and the remaining mutants did not have the labyrinthine portion of the placenta. Consequently, all mutants died at 10-11 days of gestation. Interestingly, Fgfr2(DeltaIgIII/DeltaIgIII) embryos do not form limb buds. Consistent with this defect, the expression of Fgf8, an apical ectodermal factor, is absent in the mutant presumptive limb ectoderm, and the expression of Fgf10, a mesenchymally expressed limb bud initiator, is down regulated in the underlying mesoderm. These findings provide direct genetic evidence that FGF/FGFR2 signals are absolutely required for vertebrate limb induction and that an FGFR2 signal is essential for the reciprocal regulation loop between FGF8 and FGF10 during limb induction.

Characteristics of delayed excretion of flavonoids in human urine after administration of Shosaiko-to, a herbal medicine.

There has been little explanation of herbal medicines by modern medical sciences, including pharmacokinetics, whereas physicians follow empirical indications written in classical literature. Recent reports of herb-induced adverse reactions compelled us to proceed the investigation of a herbal medicine Shosaiko-to (TJ-9) from a pharmacokinetic point of view. To five healthy volunteers, a single 5 g dose of TJ-9, consisting of 7 herbs, was administered. We conducted HPLC analysis of the timed-urine specimens to disclose the type and amount of compounds excreted. Excretion rate-time curves were analyzed individually. Four flavonoids, liquiritigenin, baicalein, wogonin and oroxylin A, were found both in the urine and TJ-9. The glycosides in TJ-9 were absorbed after microflora hydrolysis. Davidigenin, which was not found in TJ-9, was an intestinal metabolite of liquiritigenin. Also, two flavanones, S-dihydrowogonin and S-dihydrooroxylin A, were identified as the metabolites of wogonin and oroxylin A, respectively. Excretion rate-time curves of the flavonoids were divided into three types of structure-dependent absorption, i.e. (1) the fast absorption of herbal-origin aglycons, (2) the moderately-delayed absorption of aglycons derived from herbal glycosides, and (3) markedly-delayed absorption after the molecular transformation of herbal compounds. Individual excretion profiles seemed to depend on microflora activities. Two types of flavanones, S-dihydrowogonin and S-dihydrooroxylin A, were found in a half of the volunteers, suggesting there might be two kinds of volunteers, namely, rapid and poor metabolizers of flavonoids.

Stereochemistry and putative origins of flavanones found in post-administration urine of the traditional Chinese remedies shosaiko-to and daisaiko-to.

Optically active flavanones, dihydrowogonin and dihydrooroxylin A, were found in the urine of healthy volunteers who orally received the traditional Chinese remedies Shosaiko-to and Daisaiko-to on separate occasions. These remedies, which consisted of dried extracts of Scutellariae Radix and other herbs, contained the metabolic precursors of the flavanones, but not the flavanones themselves, in stoichiometrically sufficient amounts. Structures and stereochemistry of the flavanones were elucidated by UV, circular dichroism (CD), electron impact (EI)-MS and 1H-NMR analyses, showing that the biotransformations from the corresponding flavones, wogonin and oroxylin A, were stereoselective with a preference for the S-enantiomers. The putative origins of the flavanones were confirmed in terms of pharmacokinetics. Renal excretion-time data of the flavanones and the flavones suggested that the stereoselective transformations might have occurred in the intestinal tract as a result of microfloral metabolism before absorption.

Evaluation of a mucoadhesive buccal patch for delivery of peptides: in vitro screening of bioadhesion.

We have assessed the bioadhesive properties of several different mucoadhesive buccal patches. The patches consisted of custom coformulations of silicone polymers and Carbopol 974P. The contact angle of water was measured for each of the test formulations, using an ophthalmic shadow scope. The corresponding work of adhesion between the water and the patches (W1), and between the patches and freshly-excised rabbit buccal mucosa (W2) was then calculated, using a modification of Dupre's equation. The bioadhesive strength between the patches and excised rabbit buccal mucosa was also assessed. The results of the contact-angle measurements indicated that the contact angle decreased with an increase in the amount of Carbopol in the formulation. Additionally, the calculated values of both W1 and W2 increased with an increase in the amount of Carbopol in the buccal-patch formulations. A correlation (r not equal to 0.9808) was found between the measured contact angle and the calculated values for W2. The direct measurement of the force required to separate a buccal patch from excised rabbit buccal mucosa with the INSTRON demonstrated that the adhesive strength increased with an increase in the amount of Carbopol. This preliminary study has shown that the measurement of contact angles alone may provide a useful technique for estimating the work of adhesion, and may serve as a convenient and rapid screening procedure to identify potential mucoadhesive buccal-patch formulations.

Electrocardiogram pattern in hypercholesterolemic women: the influence of hormone replacement therapy.

OBJECTIVE: The aim of this study was to delineate electrocardiogram (ECG) patterns in postmenopausal women with hypercholesterolemia and to assess the possible influence of female sex hormones. STUDY DESIGN: A total of 72 postmenopausal women with moderately elevated total cholesterol levels constituted the case group, of which 48 came from a clinical trial and 24 from a cohort study. Some 236 women aged 50-59 years with normal levels of cholesterol were participants in the same cohort study in the local area. These 236 women had been subdivided into three groups: premenopausal, postmenopausal and postmenopausal with hormone replacement therapy (HRT). Of the 48 women in the clinical study group, 12 patients showed pathological ECG changes. Six of these patients were treated with HRT for 2 years (transdermal estradiol 50 micrograms/day and a daily dose of 5 mg medroxyprogesterone acetate, MPA) and the rest were non-users of HRT. RESULTS: In the women with hypercholesterolemia, 16 of 72 patients (22%) showed ischemic ECG changes, compared to nine of 88 (10%) with normal cholesterol levels (p = 0.04). We found no significant difference in the prevalence of ECG changes between postmenopausal women with and without HRT in the groups with normal levels of cholesterol. In the hormone treatment group, four of six patients showed an improvement in ECG pattern, in contrast to two of six non-users of HRT. CONCLUSIONS: This preliminary study revealed a higher prevalence of pathological ECG changes in postmenopausal women who had hypercholesterolemia than in normocholesterolemic women. These findings support the idea that hyperlipidemia contributes to the overall increase in cardiovascular disease, as this is also associated with ECG changes. Transdermal estradiol combined with MPA has a beneficial effect in reversing the process of atherosclerosis, as well as improving the ECG pattern. The prevalence of pathological ECG patterns was similar for HRT users and non-users. This outcome may be affected by several factors. Hence, further research is warranted.

[A study on dose-effect of suppression to gap junctional intercellular communication function by 50-Hz magnetic fields].

OBJECTIVE: To study the relationship between dose of exposure to 50-Hz magnetic field (MF) and its effects of suppression to gap junctional intercellular communication (GJIC) function. METHODS: Lucifer yellow, a kind of fluorescent dye, was led into single CHL cell by iontophoretic injection. Number of dye-coupled cells (DCC) five minutes after injection was used as an indicator for GJIC function. The effects of different intensities and irradiation of MF on GJIC in CHL cells were studied. RESULTS: Suppression to GJIC by 50-Hz MF depended on its flux intensity. Irradiation with lower flux intensity (0.05, 0.2 and 0.4 mT) of the MF for 24 hours could not inhibit GJIC, but that with higher intensity (0.8 and 1.6 mT) could. No "amplitude window" effect was appeared by irradiation with MF at intensities of zero to 1.6 mT. Exposure to MF at a fixed intensity of 0.8 mT for five minutes had no effects, but that for one hour was more than enough to inhibit GJIC, with the most apparent effects for 24-hour exposure. In addition, it was showed that suppression to GJIC was caused by direct action of 50-Hz MF rather then induced electrical field. CONCLUSION: MF of 50-Hz had suppression to GJIC with a dose-effect relationship.

[Antibody reaction to immunization with different recombinant hepatitis B vaccine in primary school children].

OBJECTIVE: To evaluate the safety and immunogenicity of local produced hepatitis B (HB) vaccine and to lay a basis for formulating an immunization strategy. METHODS: Two batches of local produced and two batches of imported yeast derived recombinant HB vaccine (YDV), and four batches of local produced Chinese hamster ovary cell derived recombinant HB vaccine (CHOV) were administered to 550 children in primary schools to study their immunogenicity one year after immunization. RESULTS: No severe adverse reaction was found in all vaccinee. Positive seroconversion rate was more than 90 percent one month after completion of three-dose immunization with all kinds of HB vaccine mentioned above, and then declined slowly. Titer of antibody against hepatitis B surface antigen (anti-HBs) in those vaccinated with two batches of local produced YDV, except four groups, was significantly lower than that in those with imported YDV and local produced CHOV seven months after first-dose immunization, but no difference between local produced YDV and imported Merck vaccine and CHOV was found one year after immunization. Mean geometric titer (GMT) of anti-HBs reached the peak seven months after immunization in those vaccinated with all batches of HB vaccine except for the YDV imported from Amgen Co. There was no significant difference in positive seroconversion and GMT of anti-HBs between different batches of local produced YDV and CHOV. CONCLUSION: Safety and immnogenicity of local produced recombinant HB vaccine is as good as those of imported one in immunization for primary school children.

Histological and electron-microscopic observations on the mucosa of pediculated duodenal wall graft transplanted to the stomach in Wistar rats.

OBJECTIVE: To study the mechanism of gastric metaplasia in duodenum through the transplantation of a flap of duodenal wall with vascular pedicle to the stomach. METHODS: The pediculated duodenal wall flaps of Wistar rats were transplanted to their stomachs. The rats were killed at the 3rd, 6th, 9th and 12th month after the operation respectively, and histological changes of the duodenal grafts were observed with optical and electron microscopy. RESULTS: Gastric metaplasia was found in the mucosa of duodenal grafts transplanted to the stomach at the 6th, 9th and 12th month. CONCLUSIONS: The formation of gastric metaplasia in the duodenal mucosa may be related to a change of the microenvironment around the tissues, and duodenal mucosa may differentiate into gastric epithelium by the decrease of pH value.

A clinical study on bing gan ling oral liquid for treatment of hepatitis C.

Based on the principles of cooling the blood to expel the pathogenic toxins and regulating the function of liver and activating the spleen in traditional Chinese medicine, 60 cases of chronic hepatitis C were treated with Bing Gan Ling (BGL), an oral liquid, which is composed mainly of Shuiniujiao (Cornu Bubali), Huzhang (Rhizoma Polygoni Cuspidati), Chishao (Radix Paeoniae Rubra), and Huangqi (Radix Astragali), etc. The total effective rate was 86.7%, which is considerably better than that of the control group (P < 0.01). The animal experiments and pathological studies suggest that the above-mentioned prescription has marked action of protecting the liver and lowering the activity of enzymes; and that it can ameliorate the acute liver injury induced by D-galactosamine.

Expression of human interleukin 12 (hIL-12) in insect cells.

Human interleukin-12(hIL-12) is a herterodimer cytokine, which consists of two disulfide-linked subunits, p40 and p35. This paper reports the expression of hIL-12 using the Baculovirus Expression System in insect cells. First, we constructed two expression vectors pVL1392-hp40 and pVL1393-hp35, and then they were used to co-transfect the insect cells (Sf9) separately with linearized polyhedrosis virus genomic DNA. Two kinds of recombinant viruses AcNPV-hp40 and AcNPV-hp35 were visually screened out. Biological activity of the recombinant hIL-12 (rhIL-12) was detected in the conditioned medium using proliferation assay of PHA-activated human lymphocytes and the expression level was about 1.5 approximately 2 microg/10(6) cells. The results of real-time Biomolecular Interaction Analysis (BIA) and Northern blot demonstrated that the subunits of rhIL-12, hp35, and hp40 were expressed successfully in the insect cells. The apparent molecular weights of rhIL-12 and hp40 homodimer were 76 kDa and 92 kDa under non-reducing conditions of Western blot, respectively. The recombinant hp40 can significantly inhibit the biological activity of hIL-12.

[Study on the psychological status of video display terminal operator].

Occupational psychological test designed by the Labour Hygiene Department of West China University of Medical Sciences was applied to survey the mental health of 516 video display terminal (VDT) workers and 396 control workers in Chengdu. The results showed that mental health level of VDT workers was higher than that of controls. In the somatic disorder, anxious and hostile are increased obviously. With the prolonging of operating hours per week, somatic disorders, depression and obsession are increased, especially when VDT operating time is more than 30 hours per week and operating on VDT is more than ten years. This indicates that the operating hours should not be more than 30 hours per week. It is found by multiple factors analysis of data that the main adverse effects on mental health are age and VDT operating time.
